Abstract

The golden mussel (Limnoperna fortunei), a mollusc originating from Asia, has caused considerable environmental and economic damage in various Brazilian river basins and reservoirs. Genetic studies that focus on characterizing its invasive potential and the feasibility of control measures are required. The present study identified and characterized 13 new microsatellite markers (of which eight were polymorphic) in L. fortunei using enriched genetic libraries. The analysis performed on 48 individuals from three populations identified 32 alleles, ranging from one to seven alleles per locus. The size of the fragments ranged from 172 to 381 bp. The polymorphic information content values ranged from 0.114 to 0.768, with observed and expected heterozygosity values ranging from 0.043-0.938 to 0.123-0.805, respectively. Six loci showed significant deviation from the Hardy-Weinberg equilibrium, possibly influenced by the heterozygous deficit. The microsatellite primers developed will be useful in studies of the genetic diversity and population structure of L. fortunei and will also contribute to the adoption of control measures for this invasive species in Brazilian and other South American basins. Abstract

The genus Hypochaeris has a recent evolutionary history caused by long-distance dispersal in conjunction with adaptive radiation in the South American continent. Hypochaeris lutea is a perennial herb that grows mostly at altitudes of around 1000 m in cold swamps of the southern regions of Brazil. We investigated the amplified fragment length polymorphism (AFLP) in 270 individuals representing 11 Brazilian populations of H. lutea to elucidate the population genetic structure of this species. The frequencies of polymorphic loci and gene diversity ranged from 83.42% to 91.66% and from 0.26 to 0.34, respectively. Analysis of molecular variance revealed that most of the genetic variability was found within (76.67%) rather than among (23.3%) populations, agreeing with the pattern of genetic distribution within and among populations observed in other allogamous species of Hypochaeris. A Mantel test showed no correlation between genetic and geographic distances when all populations were considered. Simulations performed using a Bayesian approach consistently identified two clusters with different admixture proportions of individuals, as also revealed by a UPGMA dendrogram of populations. The pattern of genetic structure observed in H. lutea is consistent with a process of successive colonization events by long-distance dispersal resembling the rapid and recent radiation that has been proposed to explain the origin of the South American species of Hypochaeris. Abstract

AbstractBrycon orbignyanusis a neotropical fish found in several South American countries. This species has high ecological importance and is included in the list of fish threatened with extinction. In this study, we report the development and characterisation of 12 microsatellite primers of B. orbignyanusfor future genetic studies. Screening of the 12 markers in a sample of 35 individuals identified seven polymorphic loci and 22 alleles, with one to three alleles per locus. The polymorphic information content ranged from 0.054 to 0.542, with four loci showing significant deviation from Hardy–Weinberg equilibrium. Two loci possessed null alleles. Cross-species amplification tests in two closely related species showed that 11 loci are transferrable to Brycon gouldingi(B. gouldingi) and nine are transferrable to B. falcatus.The microsatellite primers designed may be used for genetic variability and population structure studies in wild populations, fish farms and restocking programmes of B. orbignyanusand the two related species.

Abstract

Parapiptadenia rigida is a tropical early secondary succession tree characteristic of the Tropical Atlantic Rainforest. This species is of great ecological importance in the recovery of degraded areas. In this study we investigated the variability and population genetic structure of eight populations of P. rigida. Five AFLP primer combinations were used in a sample of 159 individuals representing these eight populations, rendering a total of 126 polymorphic fragments. The averages of percentage of polymorphic loci, gene diversity, and Shannon index were 60.45%, 0.217, and 0.322, respectively. A significant correlation between the population genetic variability and the population sizes was observed. The genetic variability within populations (72.20%) was higher than between these (22.80%). No perfect correlation was observed between geographic and genetic distances, which might be explained by differences in deforestation intensities that occurred in these areas. A dendrogram constructed by the UPGMA method revealed the formation of two clusters, these also confirmed by Bayesian analysis for the number of K cluster. These results show that it is necessary to develop urgent management strategies for the conservation of certain populations of P. rigida, while other populations still preserve reasonably high levels of genetic variability.
