Stem memory T cells (TSCMs) and in-situ cytotoxic T cells are dominant contributors to host immunity. Currently, data on the correlation between TSCM and T cell abundance and clinicopathological characteristics in colon cancer are largely unavailable.Methods: In-situ cytotoxic T cells are identified based on the quantification of CD3+ and CD8+ markers using immunohistochemistry (IHC) in the core of the tumor and the invasive margin of the tumor. The expression of representative markers of TSCMs, CD27 and CD95, was assayed using IHC in colon cancer tissues. Correlations between the levels of each marker and the clinicopathological characteristics as well as prognosis were evaluated.Results: High densities of CD3+ and CD8+ T cells correlated with stage I-II tumors, whereas a lower infiltration of cytotoxic T cells correlated with advanced-stage tumors. CD27 and CD95 were both expressed in the membrane of T cells present in the tumor stroma and their levels showed a negative correlation with the TNM stage. CD3, CD8, and CD27 were expressed at the same locations simultaneously, indicating their coordinated action against cancer. In addition, cytotoxic T cell densities and CD27 and CD95 expression remained independent prognostic factors for overall survival.Conclusion: In-situ cytotoxic T cells and TSCMs play important roles in colon cancer development. TSCMs marker CD27 and CD95 were both indicators of survival in patients with colon cancer. Thus, it is believed that TSCMs represent a desirable population for future use in combination immunotherapy.Keywords: T cell, CD27, CD95, prognosis, colon cancer

Recent studies indicate that circRNAs play an important role in tumor progression through exosomes. However, the specific functions of gastric cancer-derived exosomes and the role of circSTAU2 in gastric cancer (GC) remain largely unknown.Methods: Differentially expressed circRNAs in GC were identified by circRNA microarrays analysis and quantitative real-time polymerase chain reaction (qRT-PCR). The role of circSTAU2 in GC was verified by circSTAU2 knockdown and overexpression with functional assays both in vitro and in vivo. Fluorescence in situ hybridization (FISH), immunofluorescence, RNA immunoprecipitation (RIP), dual-luciferase reporter assay, qRT-PCR and Western blot were adopted to evaluate the expression and regulatory mechanism of MBNL1, circSTAU2, miR-589 and CAPZA1. Furthermore, the role of exosomes was demonstrated by transmission electron microscopy and nano-sight particle tracking analysis.Results: CircSTAU2, mainly localized in the cytoplasm, was significantly downregulated in GC. CircSTAU2 overexpression inhibited GC cell proliferation, invasion and migration both in vitro and in vivo, while circSTAU2 knockdown had the inverse effect. CircSTAU2 could be wrapped in exosomes and delivered to recipient cells, and functioned as a sponge for miR-589 to relieve its inhibitory effect on CAPZA1, thus inhibiting GC progression. Furthermore, MBNL1 acted as the upstream RNA-binding protein of circSTAU2 and significantly influenced the circularization and expression of circSTAU2.Conclusion: Exosome-delivered circSTAU2 may act as a tumor suppressor that restrains GC progression via miR-589/CAPZA1 axis, which demonstrates a potential therapeutic target for GC.Keywords: exosomes, circ-RNAs, bioinformatics, gastric cancer, cancer progression

Abstract

Based on a $J/\psi$ data sample of $7.8 \times 10^6$ events at BESI, the decay of $ J/\psi \to \bar{K}^*(892)^0 K^+ \pi^-$ is studied and a low mass enhancement, which is believed not coming from the phase space effect or background, is visible in the $K^+ \pi^-$ invariant mass spectrum recoiling against $\bar{K}^*(892)^0$. Partial wave analysis of this channel favors this low mass enhancement being a broad $0^{+}$ resonance with the mass and width of $771^{+164}_{-221}\pm 55$ MeV/c$^2$ and $220^{+225}_{-169} \pm 97$ MeV/c$^2$, respectively., Comment: 4 pages, 4 figures

Abstract

Energy hubs （EHs） can effectively facilitate the consumption of renewable energy with high uncertainty. Still， the existing EH planning methods have not fully considered the correlation between siting and sizing. Hence， this study introduces an alternate optimization method considering the supply-demand balance. Firstly， utilizing graph theory and weighted similarity， the study defines the energy moment （EM）， a concept that represents the energy transmission cost， and proposes an optimal partitioning criterion for energy internet system with minimum total EM as the goal. Subsequently， the supply-demand balance index within each EH is calculated based on the time-series energy interaction information of the energy supply and demand entities. The paper then proposes a sizing strategy for multi-energy coupling units within EH， aiming to maximize the average supply-demand balance of the entire energy internet system. On this basis， the paper constructs an alternate optimization model for EH siting and sizing based on K-means clustering， and achieves an optimal solution through iteration of EHs. Finally， case studies are presented， and their results indicate that the proposed method can significantly improve the accuracy and economy of EH planning.

However, the drug-resistance to the chemotherapy restricts its effect and might lead to treatment failure. In this review article, we discuss the mechanisms which have been found in recent years of drug resistance in gastric cancer. 17. Bacterioplankton Richness and Composition in a Seasonal Urban River

Abstract

Urbanization and seasonality strongly influence the bacterial composition of the soil. However, aquatic environments such as rivers are understudied owing to their high dynamics and therefore rules relating to more static habitats such as lentic or terrestrial environments may be limited. Here, we compared the spatiotemporal patterns of bacterioplankton communities in the Zhangxi river along a gradient of urbanization using 16S ribosomal DNA identification. The alpha and beta diversity of bacterioplankton showed no significant response to watershed urbanization. A significant difference in predicted functional profiles of the bacterioplankton community was also revealed between the wet and dry seasons. The bacterioplankton community assembly was driven by both deterministic and stochastic processes. Stochasticity was one of the most vital processes affecting the bacterioplankton communities in both wet and dry seasons, explaining over 50% variation in the community by the null model analysis. Bacterioplankton co-occurrence patterns in the river changed with the seasons. More notably, the composition of bacterioplankton communities was inconsistent with alternations of the spatial distance offering meaningful implications for interactions between zero-radius operational taxonomic units and the dynamics of the bacterioplankton communities in surface water. In summary, we found clear patterns of seasonal variations in the bacterioplankton community structures.

18. VehicleNet: Learning Robust Visual Representation for Vehicle Re-identification

Abstract

One fundamental challenge of vehicle re-identification (re-id) is to learn robust and discriminative visual representation, given the significant intra-class vehicle variations across different camera views. As the existing vehicle datasets are limited in terms of training images and viewpoints, we propose to build a unique large-scale vehicle dataset (called VehicleNet) by harnessing four public vehicle datasets, and design a simple yet effective two-stage progressive approach to learning more robust visual representation from VehicleNet. The first stage of our approach is to learn the generic representation for all domains (i.e., source vehicle datasets) by training with the conventional classification loss. This stage relaxes the full alignment between the training and testing domains, as it is agnostic to the target vehicle domain. The second stage is to fine-tune the trained model purely based on the target vehicle set, by minimizing the distribution discrepancy between our VehicleNet and any target domain. We discuss our proposed multi-source dataset VehicleNet and evaluate the effectiveness of the two-stage progressive representation learning through extensive experiments. We achieve the state-of-art accuracy of 86.07% mAP on the private test set of AICity Challenge, and competitive results on two other public vehicle re-id datasets, i.e., VeRi-776 and VehicleID. We hope this new VehicleNet dataset and the learned robust representations can pave the way for vehicle re-id in the real-world environments.

22. Influence of maturation on constrictive response to stimulation of C-fiber afferents in isolated guinea pig airways

Abstract

We investigated whether the airway constrictive response to stimulation of bronchopulmonary C-fiber afferents is altered during the maturation process. Isometric tension was measured in airway rings isolated from three tracheobronchial locations (intrathoracic trachea and main and hilar bronchi) and compared in mature [M, 407 [+ or -] 10 (SE) g body wt, n = 36] and immature (IM, 161 [+ or -] 5 g body wt, n = 35) guinea pigs. Our results showed no difference in the ACh ([10.sup.-5] M)- or KCI (40 mM)-induced contraction between M and IM groups, regardless of the airway location. In sharp contrast, the concentration-response curves of [10.sup.-8] - [10.sup.-6] M capsaicin were distinctly lower in IM hilar bronchi; for example, response to the same concentration of capsaicin ([10.sup.-6] M) was 89.2 [+ or -] 15.3% of the response to [10.sup.-5] M ACh in IM und 284.7 [+ or -] 43.2% in M animals. Similar, but smaller, differences in the bronchoconstrictive response to capsaicin between IM and M groups were also observed in the trachea and main bronchus. Electrical field stimulation induced airway constriction in all three locations in M and IM groups. However, after administration of [10.sup.-6] M atropine and [10.sup.-6] M propranolol, electrical field stimulation-induced contraction was significantly smaller in the hilar bronchus of IM than M animals, and this difference was not prevented by pretreatment with 5 x [10.sup.-5] M indomethacin. Although radioimmunoassay showed no difference in the tissue content of substance P between M and IM airways, the constrictive responses to exogenous substance P and neurokinin A were markedly greater in M airways at all three locations. In conclusion, the constriction of isolated airways evoked by C-fiber stimulation was significantly weaker in the IM guinea pigs, probably because of a less potent effect of tachykinins on the airway smooth muscle. airway smooth muscle: bronchoconstriction; capsaicin; tachykinins; airway reflex

It is a complicated and often fatal cancer, and is related to a high disease-related mortality. Around 90% of mortalities are caused by the metastasis of CRC. Current treatment statistics shows a less than 5% 5-year survival for patients with metastatic disease. The development and metastasis of CRC involve multiple factors and mechanisms. The Hedgehog (Hh) signaling plays an important role in embryogenesis and somatic development. Abnormal activation of the Hh pathway has been proven to be related to several types of human cancers. The role of Hh signaling in CRC, however, remains controversial. In this review, we will go through previous literature on the Hh signaling and its functions in the formation, proliferation, and metastasis of CRC. Abstract

Quantitative detection of BCR-ABL1 transcript is essential in monitoring residual disease of Philadelphia chromosome positive B lymphoblastic leukemia (Ph+ B-LL). We studied the kinetics of BCR-ABL1 transcript in 41 Ph+ B-LL patients in correlation with their clinical outcome. A total of 23 patients achieved complete molecular remission at 6 months post-treatment. This was associated with a lower relapse risk and better overall survival. Likewise, sustainable complete molecular remission in 27 patients was associated with superior clinical outcome. Sporadic low level BCR-ABL1 was detected in 12 of 27 patients who had attained complete molecular remission. The relapse rate was significantly higher in non-transplant patients with persistent positive BCR-ABL1 than patients transplanted when BCR-ABL1 was detectable. All eight patients harboring ABL1 kinase domain mutations died of disease or were transferred to hospice care. We concluded that monitoring the level of BCR-ABL1 transcript after hematologic remission has predictive value to the long-term outcome.

45. Investigation of DNA transfer onto clothing during regular daily activities

Abstract

Low levels of DNA from an unidentified human source, often referred to as trace DNA, are ubiquitous, can be transferred onto objects by either direct or indirect methods and have an unknown longevity in situ. Clothing items from crime scenes are often submitted for trace DNA analysis, usually in attempt to identify a person of interest. This study examined the transfer of DNA onto three 10 × 10 cm areas located on the front, back and shoulder of an individual’s external clothing (n = 300) during a regular day’s activity. After wearing for a day, the DNA quantity on all three areas increased approximately 8-fold, which usually corresponded with an increase in the endogenous DNA from the wearer on the front area of the shirt. However, the back area of the shirt was more likely to demonstrate mixtures of endogenous and extraneous DNA. An additional study was also carried out to examine whether domestic laundering is a possible mechanism for the transfer of foreign DNA onto freshly laundered items and revealed that 74% of UV-treated cotton swatch samples produced DNA profiles after laundry with household garments. In summary, this study highlights the ease of DNA transfer onto an individual’s external clothing during a regular day, and that extraneous DNA may be already on the clothing item prior to it being worn. The study provides empirical data to assist in the interpretation of trace DNA profiles and support a Bayesian approach to estimate statistical likelihoods for the transfer of foreign DNA. Abstract

We evaluate an ultra-low-power radiation-sensing circuit based on a radiation sensitive MOSFET (RADFET) for use in Wireless Sensor Networks (WSN) powered by energy harvesting with a special focus on airborne applications. We estimate a worst-case energy budget below 10 μJ per readout cycle superimposed on a constant worst-case power budget below 10 μW. The sensor circuit operates successfully with a wireless sensor node powered by a piezoelectric harvester and power management unit which is able to provide up to 2.5 mW at 10 Hz, 600 μϵ in emulated flight mode.

47. The climate of Mars from assimilations of spacecraft data

Abstract

The Mars climate has been explored using two reanalysis datasets based on combining spacecraft observations of temperature and dust with the UK version of the LMD Mars GCM. The semiannual oscillation (SAO) of zonal-mean zonal wind was studied using the existing Mars Analysis Correction Data Assimilation reanalysis during Mars Years (MYs) 24-27. The SAO of zonal-mean zonal wind was shown to exist and extend over a wide range of latitudes. The dynamical driving processes of the SAO in the tropics were investigated, and the forcing due to meridional advection appeared to be the main contributor to the SAO. The study also highlighted some phenomena associated with perturbations of the global circulation during the MY 25 global dust storm (GDS). The meridional advection term was shown to be weaker in the first half of GDS year MY 25 than in the following year, but the forcing due to meridional advection and westward thermal tides both appeared to intensify during the MY 25 GDS. The capabilities of the Mars data assimilation system were also extended in this thesis, 1) to represent dynamic dust lifting and dust transport during the assimilation and 2) to assimilate measurements of the dust vertical distribution. The updated reanalysis was then used to study several major dust events during MY28-29. It proved able to reproduce a southward-moving regional dust storm without the overwhelming assistance of the assimilation. Dust devil lifting was found to at least partly provide the initial pattern of dust of this moving dust storm. The cold anomaly of the cooling zone beneath this dust storm could be as large as ∼ 2 K similar to the magnitude of what was found during the MY 25 GDS. Using the reanalysis, the life cycle of the planet-encircling global dust storm in MY28 was also studied. The Noachis dust storm that occurred just before the MY 28 GDS was found to be the joint result of a travelling Chryse storm, enhanced by dust lifting along its path and local dust lifting in Noachis itself. The adiabatic heating associated with the north polar warming that occurred during MY 28 GDS was up to ∼ 3 times as large as that found during the non-GDS year MY 29. The wind stress dust lifting was shown to in strong correlation with the global average dust loadings, and significantly decreased when the GDS decayed.

Abstract

The conventional method of increasing the current carrying capacity of the submarine cable landing section increases the manufacturing cost of the submarine cable and the long-term operational reliability. This paper introduces the unequal diameter cable joint technology and checks and analyzes the actual current-carrying capacity of the 220 kV submarine cable landing section and the submarine section. Also， it proposes a differentiated unequal diameter submarine cable landing section and submarine section conductor cross-section matching principle and evaluates the economic efficiency of the unequal diameter cable matching scheme. The research results show that the larger the conductor cross-section of the submarine cable， the greater the drop in current-carrying capacity during the landing section. In the unequal diameter matching method， the single-core or copper wire armored submarine cable requires a smaller conductor cross-section. The maximum transmission capacity can be obtained when the submarine cable line is all single-core copper wire armored. When it is all single-core steel wire armor， it can take account of the larger transmission capacity of the submarine cable line and the optimal unit capacity cost.

49. MiR-21 promotes fracture healing by activating the PI3K/Akt signaling pathway.

Abstract

OBJECTIVE: This study aims to elucidate the potential mechanism of micro ribonucleic acid (miR)-21 in promoting fracture healing. MATERIALS AND METHODS: 30 male Sprague-Dawley rats were randomly divided into group A (phosphate-buffered saline, PBS, n=10), group B (AntagomiR-21, n=10) and group C (AntagomiR- NC, n=10) according to the different treatments. The femoral fracture operation was performed in every rat, which was pathologically diagnosed via X-ray. After the successful modeling, 50 μL (2 nmoL) PBS, 50 μL AntagomiR-21 or 50 μL AntagomiR-NC was intraperitoneally injected into rats in group A, B or C, respectively. The above agents were injected once a week for 6 weeks. At 6 weeks, 3 rats were executed in each group, and the tissue at the fracture end was observed via hematoxylin-eosin (HE) staining. The fracture healing of rats was evaluated via imaging at 1 and 7 weeks. At the same time, the expression of miR-21 in the three groups was detected via Reverse Transcription-Polymerase Chain Reaction (RT-PCR), and the expression of phosphatidylinositol 3-hydroxy kinase (PI3K) and phosphorylated- serine/threonine-protein kinase B (p-Akt) in the three groups was detected via Western blotting. RESULTS: According to the histological staining, the bone repair at the fracture end of rats in group B was not significant with fracture and poor continuity compared with those in group A and group C. The imaging evaluation revealed that in group B, the callus tissues were significantly reduced, the fracture line had undesirable healing. There were no displacement and loosening of internal fixation compared with group A and group C. Besides, RT-PCR showed that the miR-21 expression declined markedly in group B compared with that in group A and group C, and the differences were statistically significant (p<0.05). Western blotting manifested that the protein levels of PI3K and p-Akt also declined in group B compared with those in group A and group C, and there were statistically significant differences (p<0.05). CONCLUSIONS: MiR-21 promotes the fracture healing in fractured rats by activating the PI3K/Akt signaling pathway. [ABSTRACT FROM AUTHOR]

Abstract

A measurement of the partial width ratio Γbb/Γhad using a method which tags the Z --> bb decays through the lif etime of the produced heavy hadrons is presented. This method relies on the tracking precision afforded by a double-sided silicon vertex detector. The tag algorithm makes a probabilistic interpretation of three-dimensional track impact parameters, using the data to measure the resolution. By tagging the two b hadrons separately, both Γbb/Γhad and the tag efficiency can be determined from the data. For a 26% efficiency of tagging a single b hadron within the vertex detector solid angle coverage, a purity of 96% is achieved. A value of Γbb/Γhad = 0.2192+/-0.0026(stat.)+/-0.0016(Γcc/ Γhad) is found. Combining this result with other recent ALEPH Γbb/Γhad measurements gives a 95% confidence upper limit on the Standard Model top mass of Mt < 228 GeV.
