Abstract

The IPPC 4th report emphasizes the question of scales and the necessity to obtain in climate scenarios much finer resolutions for hydrological processes to assess the time evolution of the hydrological extremes. The present gap between climatological and hydrological scales led to consider downscaling techniques, which are statically or/and physically based. In particular, one may exploit the scaling properties of the precipitation to downscale it either numerically by stochastic subgrid modeling or theoretically with the help of a few scaling exponents (Royer et al. C.R. Geoscience, 340, 2008). We first discuss how these techniques can be validated with the help of a multifractal analysis of a mesoscale model. We then present the results obtained the Meso-NH model (Meteo-France/CNRM and Laboratoire d'Aérologie, Toulouse, France), a model which has been rather extensively used for mesoscale research and is partly included (its physical part) in the AROME model, the new operational meteorological model at 2.5 km resolution on France. These results are compared with the one obtained analyzing radar data.

Abstract

In this presentation the scaling properties of rainfall time-series generated by a climate model are analysed by means of a multifractal characterization based on the "universal multifractal" formalism. The analysed data are the rainfall daily time-series over France computed in a simulation over the period 1860-2100 by the climate model CNRM-CM3 of Meteo-France in a coupled IPCC climate scenario (A2). We quantify the scaling variability of the simulated rainfall with the help of a few relevant multifractal exponents characterizing the intermittency and multifractality of the field. These multifractal exponents are determined by the Double Trace Moment (DTM) which shows a scaling range from one day to about 16 days. The opposite trends found in the evolution of the intermittency and multifractality exponents, have contradictory effects on the evolution of the extremes. However, a refined analysis shows that due to the dominant effect of intermittency increase, we may expect an effective increase of rainfall extremes for the next hundred years.

38. A novel antagonist of CRTH2 blocks eosinophil release from bone marrow, chemotaxis and respiratory burst

Abstract

BACKGROUND: Chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2) has been revealed to be a novel receptor for prostaglandin (PG) D(2), which is a major mast cell product released during the allergic response. The aim of this study was to analyze the effects of a newly developed small molecule antagonist of CRTH2, Cay10471, on eosinophil function with respect to recruitment, respiratory burst and degranulation. METHODS: Chemotaxis of guinea pig bone marrow eosinophils and human peripheral blood eosinophils were determined using microBoyden chambers. Eosinophil release from bone marrow was investigated in the in situ perfused guinea pig hind limb preparation. Respiratory burst and degranulation were measured by flow cytometry. RESULTS: Cay10471 bound with high affinity to recombinant human and guinea pig CRTH2, but not DP, receptors. The antagonist prevented the PGD(2)-induced release of eosinophils from guinea pig bone marrow, and inhibited the chemotaxis of guinea pig bone marrow eosinophils and human peripheral blood eosinophils. Pretreatment with PGD(2) primed eosinophils for chemotaxis towards eotaxin, and this effect was prevented by Cay10471. In contrast, PGD(2) inhibited the C5a-induced up-regulation of CD63, a cellular marker of degranulation, in a Cay10471-sensitive manner. Finally, Cay10471 abolished the respiratory burst of eosinophils upon stimulation by PGD(2). CONCLUSION: These data further emphasize the importance of CRTH2 in eosinophil function and show that Cay10471 is a highly potent and selective antagonist of PGD(2)-induced eosinophil responses.
