This paper attempts to introduce a design architecture for the contour mill-tool grinding. Due to the unexpected three-dimensional interference known as the undercut/overcut between the grind wheel and the contour mill-tool, the corresponding design needs to go through a complicated iterative process in order to obtain the path generation parameters to compromise the flute constraints at different cross-sections. Neural networks (NNK) in recent years have proven to be of great significance in decrypting nonlinear relationships, use of which greatly reduces the computation time from hours to minutes for the tool designer. The key to success is either using a database or the NNK result to reduce the computational burden in the conventional numerical iterations. The aim of this work is to develop an automated NNK search using reinforced adaptation to learn the domain where the undercut/overcut problem is needed to be compromised. Furthermore, a new neural architecture algorithm guarantees the model convergence and result precision. The grind wheel position and orientation are then converted into NC codes for a specific five-axis grinding machine to perform either the cutting simulation or manufacturing. Metal ions are crucial for folding and function of noncoding RNAs. The fact that RNAs have very specific metal ion binding motifs further implies that contribution of metal ions (like Mg2+) in RNA's folding is not limited to simple compensation of electrostatic repulsions. Rather, their binding to RNA is driven by very specific contextual requirements. Elucidation of such factors is necessary for a comprehensive understanding of the sequence–structure–function paradigm in RNA. In this work, we have studied the consequences of Mg2+ binding on the geometry and stability of different noncanonical base pairs that shape up the complex structural landscape of RNA. Our results show that majority of the Mg2+ bound nucleobases are also part of a base pair. Interestingly, such base pairs belong only to a specific set of base pairing geometries. Out of them, we are able to identify 14 unique cases for which the native base pairing geometries are unstable under gas phase geometry optimization carried out in the absence of Mg2+ binding. Our density functional theory based calculations, performed using dispersion corrected M05-2X functional, suggest that, depending on its mode of binding, Mg2+ can stabilize and even fine tune a number of such base pairing geometries. These findings not only provide insights into how metal ions modulate the structure and dynamics of RNA molecules, they also provide a basis for improving the RNA structure prediction algorithms. The quantity of precipitation received by the monsoons is enormous. The south-west monsoons, unlike the north-east monsoons, have been studied in great detail. This study deals with rainwater collection during the NE monsoons and involves an examination of the stochastic nature of a turbulent boundary layer and the effect of crosswinds on rain droplet trajectories and thence their collection. It must be borne in mind that whilst mid latitude precipitations typically range from a few millimetres per hour, the north-east monsoon showers experience rainfall rates often 10 times higher than this value and their calculations deserve special attention. Probability distribution functions are calculated to quantify the coordinates of the landing trajectories on rooftop collectors. This investigation was essential to emulate the prevalent conditions of intense driving rain on top of high rises where setting up of a collector will be most suitable to directly harness rainwater in its purest form before contamination. Folded RNAs contain tertiary contact motifs whose structures and energetics are conserved across different RNAs. The transferable properties of RNA motifs simplify the RNA folding problem, but measuring energetic and conformational properties of many motifs remains a challenge. Here, we use a high-throughput thermodynamic approach to investigate how sequence changes alter the binding properties of naturally occurring motifs, the GAAA tetraloop • tetraloop receptor (TLR) interactions. We measured the binding energies and conformational preferences of TLR sequences that span mutational pathways from the canonical 11ntR to two other natural TLRs, the IC3R and Vc2R. While the IC3R and Vc2R share highly similar energetic and conformational properties, the landscapes that map the sequence changes for their conversion from the 11ntR to changes in these properties differ dramatically. Differences in the energetic landscapes stem from the mutations needed to convert the 11ntR to the IC3R and Vc2R rather than a difference in the intrinsic energetic architectures of these TLRs. The conformational landscapes feature several nonnative TLR variants with conformational preferences that differ from both the initial and final TLRs; these species represent potential branching points along the multidimensional sequence space to sequences with greater fitness in other RNA contexts with alternative conformational preferences. Our high-throughput, quantitative approach reveals the complex nature of sequence-fitness landscapes and leads to models for their molecular origins. The energy cost accompanying changes in the structures of nucleic acids when they bind partner molecules is a significant but underappreciated thermodynamic contribution to binding affinity and specificity. This review highlights recent advances in measuring conformational penalties and determining their contribution to the recognition, folding, and regulatory activities of nucleic acids. Notable progress includes methods for measuring and structurally characterizing lowly populated conformational states, obtaining ensemble information in high throughput, for large macromolecular assemblies, and in complex cellular environments. Additionally, quantitative and predictive thermodynamic models have been developed that relate conformational penalties to nucleic acid-protein association and cellular activity.
