Abstract

We present wind and temperature profiles at Dome C measured by balloon born sonds during the polar summer. Data from 197 flights have been processed for 4 campaigns between 2000 and 2004. We show the exceptionnal wind conditions at Dome C, Average ground wind speed is 3.6 m/s. We noticed in mid-november the presence of high altitude strong winds (40 m/s) probably due to the polar vortex which disappear in summer. These winds seem to have no effect on seeing measurements made with a DIMM at the same period. Temperature profiles exhibit a minimum at height 5500 m (over the snow surface) that defines the tropopause. Surface layer temperature profile has negative gradient in the first 50 m above ground in the afternoon and a strong inversion layer (5{\deg}C over 50 m) around midnight. Wind profiles are compared with other astronomical sites, and with a meteorological model from Meteo France.

30. Zelluntergänge in malignen Tumoren

Abstract

Gestörte Regulationsmechanismen des Zelltodes sind eine wichtige Ursache für die Entstehung und die Therapieresistenz maligner Tumoren. Hierbei unterscheidet man ein breites Spektrum distinkter, molekular definierter Arten des Zelltodes, wie z. B. Apoptose, Anoikis oder Nekroptose. Gleichzeitig spielt die gezielte Auslösung von Zelltod eine wichtige Rolle bei der Verhinderung von Metastasierung maligner Tumoren. Neue Forschungsergebnisse zeigen, dass spezifische Arten des Zelltodes bei den einzelnen Schritten der Metastasierungskaskade relevant sind, um Zellablösung, Migration, Invasion, Intra- und Extravasation sowie die Etablierung von Mikro- oder Makrometastasen zu verhindern. Außerdem gibt es auf subzellulärer Ebene zahlreiche Vernetzungen zwischen Zelltodregulation und Metastasierung, d. h. Signalwege und individuelle Proteine, die duale bzw. multifunktionelle Wirkungen haben. Ein Beispiel dafür ist das DcR3-Protein (Decoy receptor 3), das einerseits antiapoptotisch wirkt, andererseits eine direkte fördernde Wirkung auf Invasivität und Migration von Tumorzellen hat. Zusammengefasst spielt die gezielte Auslösung definierter Zelltodesarten eine wichtige Rolle bei der Verhinderung von Metastasierung. Andererseits stellt die Metastasierung einen Selektionsmechanismus dar, der dazu führt, dass etablierte Tumormetastasen gegenüber zahlreichen Zelltodstimuli multiresistent sind. Darin ist eine wesentliche Ursache für die Therapieresistenz metastasierter Tumorleiden zu sehen. Die zukünftige Forschung muss neue diagnostische Tests entwickeln, um Resistenzverhalten und Möglichkeiten der therapeutischen Überwindung von Resistenz am individuellen Tumorgewebe der Patienten vorhersagen zu können. Defects in the regulation of cell death are important causes for both the development and therapy resistance of malignant tumors. Several distinct, molecularly defined types of cell death are known, such as apoptosis, anoikis, and necroptosis. Moreover, the specific triggering of cell death plays an important role in the prevention of metastasis. The results of recent studies have shown that various types of cell death are pivotal at different steps of the metastasis cascade, in order to prevent cellular detachment, migration, invasion, intravasation, extravasation and the establishment of micrometastasis and macrometastasis. At the subcellular level, numerous links exist between cell death regulation and metastasis, specifically regarding signaling pathways and individual proteins with dual or multiple functions. As an example, the decoy receptor 3 protein (DcR3) functions both as an anti-apoptotic protein and as a direct promotor of invasion and migration of tumor cells. In summary, the specific triggering of cell death plays a pivotal role for the prevention of metastasis. On the other hand, the stepwise process of metastasis represents a mechanism of selection resulting in established metastases with a multiresistant phenotype which corresponds to the clinical observation that many metastasized cancers are therapy resistant. In the future, innovative diagnostic tests to individually predict the resistance pattern and possibilities to overcome resistance are urgently needed.

Item does not contain fulltext, BACKGROUND: Pathological TNM staging (pTNM) is the strongest prognosticator in colorectal carcinoma (CRC) and the foundation of its post-operative clinical management. Tumours that invade pericolic/perirectal adipose tissue generally fall into the pT3 category without further subdivision. METHODS: The histological depth of invasion into the pericolic/perirectal fat was digitally and conventionally measured in a training cohort of 950 CRCs (Munich). We biostatistically calculated the optimal cut-off to stratify pT3 CRCs into novel pT3a (≤3 mm)/pT3b (>3 mm) subgroups, which were then validated in two independent cohorts (447 CRCs, Bayreuth/542 CRCs, Mainz). RESULTS: Compared to pT3a tumours, pT3b CRCs showed significantly worse disease-specific survival, including in pN0 vs pN+ and colonic vs. rectal cancers (DSS: P < 0.001, respectively, pooled analysis of all cohorts). Furthermore, the pT3a/pT3b subclassification remained an independent predictor of survival in multivariate analyses (e.g. DSS: P < 0.001, hazard ratio: 4.41 for pT3b, pooled analysis of all cohorts). While pT2/pT3a CRCs showed similar survival characteristics, pT3b cancers remained a distinct subgroup with dismal survival. DISCUSSION: The delineation of pT3a/pT3b subcategories of CRC based on the histological depth of adipose tissue invasion adds valuable prognostic information to the current pT3 classification and implementation into current staging practices of CRC should be considered.
