Abstract

In dieser Arbeit werden die sHsps Sip1 und Hsp17 aus C. elegans mittels Cryo-TEM charakterisiert. In Sip1 konnte ein möglicher domain-swap und ein konformationeller Schalter identifiziert werden. Die Struktur des Hsp17 24mers zeigt eine neuartige Domänenorganisation, in der 12 flexible N-terminale Regionen nach außen gewandt und die Übrigen an stabilisierenden Interaktionen im inneren des Oligomers beteiligt sind. In this work, the sHsps Sip1 and Hsp17 from C. elegans are characterized by cryo-TEM. In Sip1, a possible domain-swap and a conformational switch could be identified. The structure of the Hsp17 24mer shows a novel domain organization in which 12 flexible N-terminal regions are outward facing and the remainder are involved in stabilizing interactions inside the oligomer.
