Rationale: Estimating the annual incidence and prevalence of nontuberculous mycobacterial (NTM) lung disease may assist in improving understanding of the public health and economic impacts of this disease and its treatment. Objective: To estimate the yearly incidence and prevalence of administrative claims–based NTM lung disease between 2008 and 2015 in a U.S. managed care claims database. Methods: We used a national managed care claims database (Optum Clinformatics Data Mart) representing a geographically diverse population of approximately 27 million members annually. All medical claims from January 1, 2007, to June 30, 2016, were scanned for diagnosis codes for NTM lung disease (International Classification of Diseases, Ninth Revision, Clinical Modification [ICD-9-CM] code 031.0 or ICD-10-CM code A31.0). We defined a case of NTM lung disease as having at least two medical claims with a code of 031.0 or A31.0 that were dated at least 30 days apart. Annual incidence and prevalence were estimated for each calendar year from 2008 to 2015. Results: From 2008 to 2015, the annual incidence of NTM lung disease increased from 3.13 (95% confidence interval [CI], 2.88–3.40) to 4.73 (95% CI, 4.43–5.05) per 100,000 person-years, and the annual prevalence increased from 6.78 (95% CI, 6.45–7.14) to 11.70 (95% CI, 11.26–12.16) per 100,000 persons. Ocean warming is causing the symbioses between cnidarians and their algal symbionts to breakdown more frequently, resulting in bleaching. For sea anemones, nutritional benefits derived from hosting anemonefishes increase their algal symbiont density. The sea anemone-anemonefish relationship could, therefore, facilitate bleaching recovery. To test this, bleached and unbleached sea anemones, both with and without anemonefish, were monitored in the laboratory. At the start of our experiment, algal symbiont density and colour score were lower in the bleached than unbleached sea anemones, whereas total chlorophyll remained similar. After 106 days, bleached sea anemones with anemonefish had an algal symbiont density and colour score equal to the controls (unbleached sea anemones and without anemonefish), indicating recovery had occurred. Furthermore, total chlorophyll was 66% higher in the bleached sea anemones with anemonefish than the controls. In contrast, recovery did not occur for the bleached sea anemones without anemonefish as they had 78% fewer algal symbionts than the controls, and colour score remained lower. Unbleached sea anemones with anemonefish also showed positive changes in algal symbiont density and total chlorophyll, which increased by 103% and 264%, respectively. Consequently, anemonefishes give their host sea anemones a distinct ecological advantage by enhancing resilience to bleaching, highlighting the benefits of symbioses in a changing climate.

Dimethylsulfoniopropionate (DMSP) is a key biogenic compound in marine algae and symbiotic cnidarians. Besides its role as a precursor of the climatically active dimethylsulfide, DMSP is a putative antioxidant through its capacity to scavenge reactive oxygen species. Here, the production of DMSP concurrently with the antioxidant capacity (AOC) among the host and endosymbionts of two colour phenotypes of the sea anemone Entacmaea quadricolor, that provides essential habitat for anemonefish, was investigated for 9 days under heat stress (3.2 °C and 6.3 °C above control). Although visual signs of bleaching were observed on day 3 in all of the anemones at high temperature, cell density and chlorophyll a content did not vary among temperatures over time. The maximum quantum yield of endosymbionts decreased over time in medium and high temperature treatments in both phenotypes, indicating slight photoinhibition under thermal stress. Temperature had no or little effect on AOC and DMSP concentrations in the host and endosymbionts in both phenotypes, suggesting adaptation of subtropical E. quadricolor and associated symbionts to short-term abrupt changes in temperature, which is a regular occurrence in this region. However, host and endosymbiont AOC and DMSP concentrations were, at least partly, driven by the host phenotype, with overall greater AOC and lower DMSP concentrations being found in pink compared with green anemones. These results, along with stronger photoinhibition in the green phenotype under thermal stress suggest that green anemones could be more vulnerable to environmental pressure than the pink phenotype. As our oceans continue to warm, the differing responses of the colour phenotypes may influence their relative abundance on reefs, and have implications for reproductive success as the phenotypes correspond to different sexes. Furthermore, the selective disappearance of one phenotype could adversely affect anemonefish that preferentially associate with more sensitive colour morphs.

The global rise in sea surface temperatures causes regular exposure of corals to high temperature and high light stress, leading to worldwide disastrous coral bleaching events (loss of symbiotic dinoflagellates (Symbiodinium) from reef-building corals). Our picosecond chlorophyll fluorescence experiments on cultured Symbiodinium clade C cells exposed to coral bleaching conditions uncovered the transformations of the alga's photosynthetic apparatus (PSA) that activate an extremely efficient non-photochemical "super-quenching" mechanism. The mechanism is associated with a transition from an initially heterogeneous photosystem II (PSII) pool to a homogeneous "spillover" pool, where nearly all excitation energy is transferred to photosystem I (PSI). There, the inherently higher stability of PSI and high quenching efficiency of P(700)(+) allow dumping of PSII excess excitation energy into heat, resulting in almost complete cessation of photosynthetic electron transport (PET). This potentially reversible "super-quenching" mechanism protects the PSA against destruction at the cost of a loss of photosynthetic activity. We suggest that the inhibition of PET and the consequent inhibition of organic carbon production (e.g. sugars) in the symbiotic Symbiodinium provide a trigger for the symbiont expulsion, i.e. bleaching.

Macroalgal communities in Australia and around the world store vast quantities of carbon in their living biomass, but their prevalence of growing on hard substrata means that they have limited capacity to act as long-term carbon sinks. Unlike other coastal blue carbon habitats such as seagrasses, saltmarshes and mangroves, they do not develop their own organic-rich sediments, but may instead act as a rich carbon source and make significant contributions in the form of detritus to sedimentary habitats by acting as a “carbon donor” to “receiver sites” where organic material accumulates. The potential for storage of this donated carbon however, is dependent on the decay rate during transport and the burial efficiency at receiver sites. To better understand the potential contribution of macroalgal communities to coastal blue carbon budgets, a comprehensive literature search was conducted using key words, including carbon sequestration, macroalgal distribution, abundance and productivity to provide an estimation of the total amount of carbon stored in temperate Australian macroalgae. Our most conservative calculations estimate 109.9 Tg C is stored in living macroalgal biomass of temperate Australia, using a coastal area covering 249,697 km2. Estimates derived for tropical and subtropical regions contributed an additional 23.2 Tg C. By extending the search to include global studies we provide a broader context and rationale for the study, contributing to the global aspects of the review. In addition, we discuss the potential role of calcium carbonate-containing macroalgae, consider the dynamic nature of macroalgal populations in the context of climate change, and identify the knowledge gaps that once addressed will enable robust quantification of macroalgae in marine biogeochemical cycling of carbon. We conclude that macroalgal communities have the potential to make ecologically meaningful contributions toward global blue carbon sequestration, as donors, but given that the fate of detached macroalgal biomass remains unclear, further research is needed to quantify this contribution.

This study analysed the effects of elevated temperature on chlorophyll fluorescence parameters and superoxide dismutase (SOD) activities in relation to Symbiodinium genotype, and host pigmentation in 2 distinct colour phenotypes (green and pink) of the sea anemone Entacmaea quadricolor. Overall, the phenotypes differed with respect to the relative content of Symbiodinium internal transcribed spacer 2 (ITS2) types C25 and C3.25 and their maximum quantum yield of PSII (F-v/F-m) during baseline conditions. However, different PSII photochemical efficiencies were not correlated with symbiont assemblage. Also, the responses to elevated temperatures were phenotype-specific. The PSII photochemical efficiencies had different critical thermal thresholds of 24.5 degrees C in the pink phenotype. The highest temperature treatment (27.6 degrees C) resulted in symbiont shuffling towards a higher relative content of C3.25 in the green but not the pink phenotype. However, the observed shuffling of Symbiodinium types could not be linked to enhanced algal SOD activity or PSII photochemical efficiency. These results suggest that different photobiological properties and thermal responses of Symbiodinium ITS2 consortia might be, at least in part, influenced by host-derived factors, possibly chromophore proteins that also determine host pigmentation. The differential ability to cope with elevated temperatures might have profound impacts on E. quadricolor phenotype abundance in response to changing climate.

The sea anemone Entacmaea quadricolor simultaneously harbours multiple symbiont types from the genus Symbiodinium, while providing essential habitat for anemonefish. This anemone lives close to its upper thermal threshold and experiences bleaching under elevated temperature and light stress. Here, we determine whether E. quadricolor experienced a shuffling in the abundance of two genetically distinct symbiont types (Symbiodinium C25 and C3.25) during bleaching and recovery. Anemones were exposed to control (22.9 °C) or elevated temperature (28.5 °C) for 42 days, whereas for the following 75 days, all anemones were exposed to 22.9 °C. By day 47, a more pronounced bleaching occurred via symbiont expulsion in the elevated temperature treatment than the control, and the proportion of C25 to C25 + C3.25 increased by 6.2 and 13.2 % in the control and bleached anemones, respectively. The increased relative abundance of C25 to C3.25 after exposure to thermal stress may indicate that C3.25 performs poorly when temperature is elevated. Although no significant recovery in symbiont density was detected, a revival of the C3.25 genotype was found at day 117, which may indicate that it is either more competitive or has qualities that are beneficial to the symbiosis when thermal stress is no longer apparent. This work demonstrates the potential for this anemone species to shuffle its symbiont types in response to environmental change and could provide resilience during times of stress.

The impact of elevated CO2 and temperature on photosynthesis and calcification in the symbiont-bearing benthic foraminifer Marginopora vertebralis was studied. Individual specimens of M. vertebralis were collected from Heron Island on the southern Great Barrier Reef (Australia). They were maintained for 5 weeks at different temperatures (28, 32 °C) and pCO2 (400, 1,000 µatm) levels spanning a range of current and future climate-change scenarios. The photosynthetic capacity of M. vertebralis was measured with O2 microsensors and a pulse-amplitude-modulated chlorophyll (Chl) fluorometer, in combination with estimates of Chl a and Chl c 2 concentrations and calcification rates. After 5 weeks, control specimens remained unaltered for all parameters. Chlorophyll a concentrations significantly decreased in the specimens at 1,000 µatm CO2 for both temperatures, while no change in Chl c 2 concentration was observed. Photoinhibition was observed under elevated CO2 and temperature, with a 70–80 % decrease in the maximum quantum yield of PSII. There was no net O2 production at elevated temperatures in both CO2 treatments as compared to the control temperature, supporting that temperature has more impact on photosynthesis and O2 flux than changes in ambient CO2. Photosynthetic pigment loss and a decrease in photochemical efficiency are thus likely to occur with increased temperature. The elevated CO2 and high temperature treatment also lead to a reduction in calcification rate (from +0.1 to >−0.1 % day−1). Thus, both calcification and photosynthesis of the major sediment-producing foraminifer M. vertebralis appears highly vulnerable to elevated temperature and ocean acidification scenarios predicted in climate-change models.

Increased seawater temperature causes photoinhibition due to accumulation of photodamaged photosystem II (PSII) in symbiotic algae (genus Symbiodinium) within corals, and it is assumed to be associated with coral bleaching. To avoid photoinhibition, photosynthetic organisms repair the photodamaged PSII through replacing the PSII proteins, primarily the D1 protein, with newly synthesised proteins. However, in experiments using cultured Symbiodinium strains, the PSII repair of Symbiodinium has been suggested not to be related to the synthesis of the D1 protein. In this study, we examined the relationship between the recovery of PSII photochemical efficiency (F V/F M) and the content of D1 protein after high-light and high-temperature treatments using the bleaching-sensitive coral species, Pocillopora damicornis and Acropora millepora, and the bleaching-tolerant coral species, Montipora digitata and Pavona decussata. When corals were exposed to strong light (600 µmol photons m−2 s−1) at elevated temperature (32 °C) for 8 h, significant bleaching occurred in bleaching-sensitive coral species although an almost similar extent of reduced PSII function was found across all coral species tested. During a subsequent 15-h recovery under low light (10 µmol photons m−2 s−1) at optimal temperature (22 °C), the reduced F V/F M recovered close to initial levels in all coral species, but the reduced D1 content recovered only in one coral species (Pavona decussata). D1 content was therefore not strongly linked to chloroplast protein synthesis-dependent PSII repair. These results demonstrate that the recovery of photodamaged PSII does not always correspond with the recovery of D1 protein content in Symbiodinium within corals, suggesting that photodamaged PSII can be repaired by a unique mechanism in Symbiodinium within corals.

This study examined the capacity for photoprotection and repair of photo-inactivated photosystem II in the same Symbiodinium clade associated with two coexisting coral species during high-light stress in order to test for the modulation of the symbiont’s photobiological response by the coral host. After 4 days exposure to in situ irradiance, symbionts of the bleaching-sensitive Pocillopora damicornis showed rapid synthesis of photoprotective pigments (by 44 %) and strongly enhanced rates of xanthophyll cycling (by 446 %) while being insufficient to prevent photoinhibition (sustained loss in Fv/Fm at night) and loss of symbionts after 4 days. By contrast, Pavona decussata showed no significant changes in Fv/Fm, symbiont density or xanthophyll cycling. Given the association with the same Symbiodinium clade in both coral species, our findings suggest that symbionts in the two species examined may experience different in hospite light conditions as a result of different biometric properties of the coral host.

The in hospite Symbiodinium symbiont of corals on shallow reefs relies on photoprotection and photorepair during periods of exposure to short-term high light and/or temperature stress. A coral’s susceptibility to bleaching is species specific and determined not only by Symbiodinium type, size and physiology, but also by coral host features. Here, photoprotective, photorepair, photochemical and non-photochemical efficiency parameters of Symbiodinium harboured in two morphologically different coral species were examined on Heron Island (23.4420°S, 151.9140°E) in July 2011. The two coral species were exposed to high light stress for 96 h, with or without inhibition of photosystem (PS) II repair by lincomycin. Symbiodinium harboured in Pocillopora damicornis showed an increase in xanthophyll de-epoxidation under high light exposure, whereas algal symbionts in Pavona decussata showed constant levels of xanthophyll de-epoxidation. High light-treated specimens of P. damicornis maintained steady PsbA protein (D1 protein) content throughout the experiment, but P. decussata showed a peak in PsbA protein content after 48 h of exposure. In hospite Symbiodinium in P. damicornis had greater content of PsbA protein fragments, suggesting higher accumulation of photodamaged products, compared to Symbiodinium in P. decussata, where both maintained steady PSII photochemical capacity over 96 h of exposure. Under inhibition of PSII repair, both species lost PsbA protein content and PSII photochemical capacity. Both species showed increased heat dissipation under inhibition of PSII repair, but differed in photoprotective strategies and photorepair activity. Our results suggest that, as well as any differences in the symbiont, characteristics of the coral host can alter important physiological responses in Symbiodinium.

© 2016 The Author. Corals rely on photosynthesis by their endosymbiotic dinoflagellates (Symbiodinium spp.) to form the basis of tropical coral reefs. High sea surface temperatures driven by climate change can trigger the loss of Symbiodinium from corals (coral bleaching), leading to declines in coral health. Different putative species (genetically distinct types) as well as conspecific populations of Symbiodinium can confer differing levels of thermal tolerance to their coral host, but the genes that govern dinoflagellate thermal tolerance are unknown. Here we show physiological and transcriptional responses to heat stress by a thermo-sensitive (physiologically susceptible at 32 °C) type C1 Symbiodinium population and a thermo-tolerant (physiologically healthy at 32 °C) type C1 Symbiodinium population. After nine days at 32 °C, neither population exhibited physiological stress, but both displayed up-regulation of meiosis genes by ≥ 4-fold and enrichment of meiosis functional gene groups, which promote adaptation. After 13 days at 32 °C, the thermo-sensitive population suffered a significant decrease in photosynthetic efficiency and increase in reactive oxygen species (ROS) leakage from its cells, whereas the thermo-tolerant population showed no signs of physiological stress. Correspondingly, only the thermo-tolerant population demonstrated up-regulation of a range of ROS scavenging and molecular chaperone genes by ≥ 4-fold and enrichment of ROS scavenging and protein-folding functional gene groups. The physiological and transcriptional responses of the Symbiodinium populations to heat stress directly correlate with the bleaching susceptibilities of corals that harbored these same Symbiodinium populations. Thus, our study provides novel, foundational insights into the molecular basis of dinoflagellate thermal tolerance and coral bleaching.

Global climate change is leading to the rise of ocean temperatures and is triggering mass coral bleaching events on reefs around the world. The expulsion of the symbiotic dinoflagellate algae is believed to occur as a result of damage to the photosynthetic apparatus of these symbionts, although the specific site of initial impact is yet to be conclusively resolved. Here, the sensitivity of the oxygen evolving complex (OEC) to bleaching stress was studied as well as its natural variation between seasons. The artificial electron donor, diphenyl carbazide (DPC), was added to cultured, freshly isolated and expelled (bleaching treatments only) zooxanthellae suspensions. Chl a fluorescence and oxygen production measurements showed that upon addition of DPC, no restoration of diminished photochemical efficiency occurred under control or bleaching conditions. This result was consistent between 12 h and 5 d bleaching treatments on Pocilloporadamicornis, indicating that the OEC is not the primary site of damage, and that zooxanthellae expulsion from the host is a nonselective process with respect to the functioning of the OEC. Further experiments measuring fast induction curves (FICs) revealed that in both summer and winter, the temperature when OEC function was lost occurred between 7°C and 14°C above the sea surface temperature. FIC and oxygen production measurements of P. damicornis during exposure to bleaching stress demonstrated that the thermotolerance of the OEC increased above the temperature of the bleaching treatment over a 4 h period. This finding indicates that the OEC has the capacity to acclimate between seasons and remains functional at temperatures well above bleaching thresholds.

All photosynthetic organisms endeavor to balance energy supply with demand. For sea-ice diatoms, as with all marine photoautotrophs, light is the most important factor for determining growth and carbon-fixation rates. Light varies from extremely low to often relatively high irradiances within the sea-ice environment, meaning that sea-ice algae require moderate physiological plasticity that is necessary for rapid light acclimation and photoprotection. This study investigated photoprotective mechanisms employed by bottom Antarctic sea-ice algae in response to relatively high irradiances to understand how they acclimate to the environmental conditions presented during early spring, as the light climate begins to intensify and snow and sea-ice thinning commences. The sea-ice microalgae displayed high photosynthetic plasticity to increased irradiance, with a rapid decline in photochemical efficiency that was completely reversible when placed under low light. Similarly, the photoprotective xanthophyll pigment diatoxanthin (Dt) was immediately activated but reversed during recovery under low light. The xanthophyll inhibitor dithiothreitol (DTT) and state transition inhibitor sodium fluoride (NaF) were used in under-ice in situ incubations and revealed that nonphotochemical quenching (NPQ) via xanthophyll-cycle activation was the preferred method for light acclimation and photoprotection by bottom sea-ice algae. This study showed that bottom sea-ice algae from the east Antarctic possess a high level of plasticity in their light-acclimation capabilities and identified the xanthophyll cycle as a critical mechanism in photoprotection and the preferred means by which sea-ice diatoms regulate energy flow to PSII.

Exposure to elevated temperature is known to cause photosynthetic inhibition in the coral symbiont Symbiodinium sp. Through the use of the artificial electron acceptor, methyl viologen, this study identified how reduced photosynthetic capacity occurs as a result of inhibition up- and/or downstream of ferredoxin in Symbiodinium sp. in hospite and in culture. Heterogeneity between coral species and symbiont clades was identified in the thermal sensitivity of photosynthesis in the symbionts of the scleractinian corals Stylophora pistillata and Pocillopora damicornis, as well as among Symbiodinium cultures of clades A, B, and C. The in hospite symbionts of S. pistillata and the cultured clade C Symbiodinium both exhibited similar patterns in that their primary site of thermal inhibition occurred downstream of ferredoxin at 32°C. In contrast, the primary site of thermal inhibition occurred upstream of ferredoxin in clades A and B at 32°C, while at 34°C, all samples showed combined up- and downstream inhibition. Although clade C is common to both P. damicornis and S. pistillata, the manner of thermal inhibition was not consistent when observed in hospite. Results showed that there is heterogeneity in the primal site of thermal damage in Symbiodinium among coral species and symbiont clades.

This study examined the response of a coral holobiont to thermal stress when the bacterial community was treated with antibiotics. Colonies of Pocillopora damicornis were exposed to broad and narrow-spectrum antibiotics targeting coral-associated α and γ-Proteobacteria. Corals were gradually heated from the control temperature of 26 to 31 °C, and measurements were made of host, zooxanthellar and microbial condition. Antibiotics artificially reduced the abundance and activity of bacteria, but had minimal effect on zooxanthellae photosynthetic efficiency or host tissue protein content. Heated corals without antibiotics showed significant declines in F V /F M , typical of thermal stress. However, heated corals treated with antibiotics showed severe tissue loss in addition to a decline in F V /F M . This study demonstrated that a disruption to the microbial consortium diminished the resilience of the holobiont. Corals exposed to antibiotics under control temperature did not bleach, suggesting that temperature may be an important factor influencing the activity, diversity and ecological function of the holobiont bacterial community.

Elevated temperatures in combination with moderate to high irradiance are known to cause bleaching events in scleractinian corals, characterised by damage to photosystem II (PSII). Photoprotective mechanisms of the symbiont can reduce the excitation pressure impinging upon PSII. In the bleaching sensitive species, Acropora millepora and Pocillopora damicornis, high light alone induced photoprotection through the xanthophyll cycle, increased content of the antioxidant carotenoid, β-carotene, as well as the dissociation of the light-harvesting chlorophyll complexes. The evidence is compatible with either the membrane-bound chlorophyll a-chlorophyll c 2-peridinin-protein (acpPC) complex or the peripheral peridinin-chlorophyll-protein complex, or both, disconnecting from PSII under high light. The acpPC complex potentially showed a state transition response with redistribution towards photosystem I to reduce PSII over-excitation. This apparent acpPC dissociation/reassociation was promoted by the addition of the xanthophyll cycle inhibitor, dithiothreitol, under high irradiance. Exposure to thermal stress as well as high light promoted xanthophyll de-epoxidation and increased β-carotene content, although it did not influence light-harvesting chlorophyll complex (LHC) dissociation, indicating light, rather than temperature, controls LHC dissociation. Photoinhibition was avoided in the bleaching tolerant species, Pavona decussata, suggesting xanthophyll cycling along with LHC dissociation may have been sufficient to prevent photodamage to PSII. Symbionts of P. decussata also displayed the greatest detachment of antenna complexes, while the more thermally sensitive species, Pocillopora damicornis and A. millepora, showed less LHC dissociation, suggesting antenna movement influences bleaching susceptibility.

We dissect the primary photo-inactivation and the counteracting metabolic repair rates in fragments of the scleractinian coral, Pocillopora damicornis, subjected to a combined stress of a shift to elevated temperature (from 26°C to 32°C) and increased light (from 200 μmol photons m-2 s-1 to 400 μmol photons m-2 s-1) to induce bleaching. During the bleaching treatment the dinoflagellate symbionts showed a 5.5-fold acceleration in their photosystem II (PSII) repair rate constant, demonstrating that they maintain strong metabolic capacity to clear and replace photo-damaged D1 protein at the elevated temperature and light conditions. Nevertheless, the symbionts concurrently suffered a seven-fold increase in the rate constant for PSII photo-inactivation. This rapid photo-inactivation exceeded the PSII repair capacity, therefore tipping the symbionts, and by implication the symbiosis, into net photo-inhibition. Photoinhibition and D1 protein re-synthesis were investigated in bottom-dwelling sea-ice microalgal communities from the East Antarctic pack ice during early spring. Bottom-dwelling sea-ice microalgal communities were dominated by diatoms that exhibited rapid photoprotection when exposed to a range of different light levels (10 μmol photons m-2 s -1, 50 μmol photons m-2 s-1, 100 μmol photons m-2 s-1, and 200 μmol photons m-2 s-1). Photosynthetic capacity of photosystem II (PSII) dropped significantly over 3 h under 200 μmol photons m-2 s-1, but largely recovered when placed in a low-light environment (10 μmol photons m-2 s-1) for an additional 3 h. PSII repair rates increased with increasing irradiance, and the D1-protein pool remained steady even under high light (200 μmol photons m-2 s-1). Sea-ice diatoms showed a low intrinsic susceptibility to photoinactivation of PSII across all the light treatments, and a strong and irradiance-dependent induction of nonphotochemical quenching, which did not depend upon chloroplast protein synthesis, was also seen. These highly plastic organisms, once thought to be adapted to shade, are in fact well equipped to withstand rapid and relatively large changes in light at low temperatures with minimal long-term effect on their photosynthetic machinery. © 2010, by the American Society of Limnology and Oceanography, Inc.

This paper investigates the varying attitudes of regular (mainstream) education teachers towards the implementation of inclusion in elementary and secondary school classrooms. This paper tries to take into account the need for a better understanding of teacher attitude towards inclusion and how the inclusive environment can be improved. The present paper concentrates on examining whether there is a difference in attitude between elementary regular education teachers and secondary regular education teachers. The participants of this study were 73 teachers from three public elementary and secondary schools in rural, southeastern USA who completed the Scale of Teachers' Attitudes Towards Inclusive Classrooms (STATIC). The results indicated that most teachers support the practice of inclusion in regular education classrooms or possess a neutral consensus towards the practice of inclusion as it relates to teaching assignment. For example, grade level, subject area or type of inclusion practice (full or partial). Meaning that the results for this study were mixed.

In support of exploration programs across the deepwater Gulf of Mexico, prestack depth-migrated (PSDM) seismic data sets have come to be widely used for prospect identification and risk assessment. These projects have often followed the 80% solution, where the standard has been isotropic velocity analysis and imaging. However, with sustained exploration success, there has been an increasing need to improve both the image quality and predictive capability of PSDM data sets in support of project appraisal and development. Achieving the desired improvements in image fidelity places many requirements on our approach to seismic imaging and velocity modeling. Foremost among these is accuracy; the imaging must be able to precisely image faults, subtle stratigraphy, and complex, steeply dipping features. In parallel, our approach to velocity modeling should provide a robust, flexible, and integrated methodology for describing the complexities of anisotropic Earth models, since sharply defining the geologic struct...

Environmental indicators must have a predictable relationship with stressors to be of value in ecological assessments. We evaluated the information provided by commonly implemented monitoring indicators as a means of assessing of the level of ecological impact experienced by coastal lagoons in New South Wales, Australia. Existing data for environmental variables in coastal lagoons were correlated with independent estimates of catchment disturbance. There were few relationships between the monitoring variables (particularly water chemistry) and nutrient loads and catchment land use. Data from NSW catchments and lagoons were compared to analogous data from published northern hemisphere studies and it was clear that stressor variables were up to one to two orders of magnitude smaller in NSW, potentially explaining the lack of relationships with recognised indicators. Our study has highlighted the importance of using a range of indicators to assess trends in ecological condition of an estuarine ecosystem, particularly where stressor levels are not great. Using water quality as the sole means of determining lagoon condition was simply inadequate in NSW lagoons. We recommended that ecological outcome indicators such as algal abundance (macro and micro) and turbidity were most likely to show interpretable patterns at low to moderate nitrogen loadings (

Diatoms, an important group of phytoplankton, bloom annually in the Southern Ocean, covering thousands of square kilometers and dominating the region's phytoplankton communities. In their role as the major food source to marine grazers, diatoms supply carbon, nutrients and energy to the Southern Ocean food web. Prevailing environmental conditions influence diatom phenotypic traits (for example, photophysiology, macromolecular composition and morphology), which in turn affect the transfer of energy, carbon and nutrients to grazers and higher trophic levels, as well as oceanic biogeochemical cycles. The paucity of phenotypic data on Southern Ocean phytoplankton limits our understanding of the ecosystem and how it may respond to future environmental change. Here we used a novel approach to create a 'snapshot' of cell phenotype. Using mass spectrometry, we measured nitrogen (a proxy for protein), total carbon and carbon-13 enrichment (carbon productivity), then used this data to build spectroscopy-based predictive models. The models were used to provide phenotypic data for samples from a third sample set. Importantly, this approach enabled the first ever rate determination of carbon productivity from a single time point, circumventing the need for time-series measurements. This study showed that Chaetoceros simplex was less productive and had lower protein and carbon content during short-term periods of high salinity. Applying this new phenomics approach to natural phytoplankton samples could provide valuable insight into understanding phytoplankton productivity and function in the marine system.

Mass coral bleaching events are a worldwide phenomenon, which generally occur during periods of elevated sea surface temperature and intense sunlight. These conditions result in a decline in photochemical efficiency of symbiotic microalgae (zooxanthellae) which ultimately leads to the expulsion of these symbionts. The physiological mechanism which triggers the release of the zooxanthellae has yet to be adequately determined. Under bleaching conditions, non-photochemical quenching (NPQ) is used to dissipate excess energy from photosystem II (PSII). NPQ was partitioned into three components, (energy dependent quenching [qE], state transition quenching [qT] and photoinhibitory quenching [qI]), based on relaxation kinetics upon addition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) and darkening. This investigation revealed that for corals not exposed to bleaching stress, qE was the principle means of energy dissipation (∼60% of the total NPQ). In corals exposed to either high-light (475 μmol photons m−2 s−1 and 25 °C) or elevated temperature (225 μmol photons m−2 s−1 and 32 °C) treatments, the dominant component of NPQ was qE and the relative proportions did not change during the exposure period (1–8 h). When exposed to bleaching conditions (475 μmol photons m−2 s−1 and 32 °C) the contribution of the different components changed after 4 h and the total NPQ increased. At this time, the contribution of qT to the total NPQ significantly increased to equal that of qE (40%), suggesting state transitions become more important under such conditions. Throughout the exposure period in all treatments, no change in the proportion of qI was observed.

Mass coral bleaching is linked to elevated sea surface temperatures, 1-2 degrees C above average, during periods of intense light. These conditions induce the expulsion of zooxanthellae from the coral host in response to photosynthetic damage in the algal symbionts. The mechanism that triggers this release has not been clearly established and to further our knowledge of this process, fluorescence rise kinetics have been studied for the first time. Corals that were exposed to elevated temperature (33 degrees C) and light (280 mumol photons m(-2) s(-1)), showed distinct changes in the fast polyphasic induction of chlorophyll-a fluorescence, indicating biophysical changes in the photochemical processes. The fluorescence rise over the first 2000ms was monitored in three species of corals for up to 8 h, with a PEA fluorometer and an imaging-PAM. Pocillopora damicornis showed the least impact on photosynthetic apparatus, while Acropora nobilis was the most sensitive, with Cyphastrea serailia intermediate between the other two species. A. nobilis showed a remarkable capacity for recovery from bleaching conditions. For all three species, a steady decline in the slope of the initial rise and the height of the J-transient was observed, indicating the loss of functional Photosystem II (PS II) centres under elevated-temperature conditions. A significant loss of PS II centres was confirmed by a decline in photochemical quenching when exposed to bleaching stress. Non-photochemical quenching was identified as a significant mechanism for dissipating excess energy as heat under the bleaching conditions. Photophosphorylation could explain this decline in PS II activity. State transitions, a component of non-photochemical quenching, was a probable cause of the high non-photochemical quenching during bleaching and this mechanism is associated with the phosphorylation-induced dissociation of the light harvesting complexes from the PS II reaction centres. This reversible process may account for the coral recovery, particularly in A. nobilis.

Dinoflagellates from the genus Symbiodinium form symbiotic relationships with many marine invertebrates, including reef-building corals. Symbiodinium is genetically diverse, and acquiring suitable Symbiodinium phylotypes is crucial for the host to survive in habitat environments, such as high-light conditions. The sensitivity of Symbiodinium to high light differs among Symbiodinium phylotypes, but the mechanism that controls light sensitivity has not yet been fully resolved. In the present study using high-light-tolerant and -sensitive Symbiodinium phylotypes, we examined what determines sensitivity to high light. In growth experiments under different light intensities, Symbiodinium CS-164 (clade B1) and CCMP2459 (clade B2) were identified as high-light-tolerant and -sensitive phylotypes, respectively. Measurements of the maximum quantum yield of photosystem II (PSII) and the maximum photosynthetic oxygen production rate after high-light exposure demonstrated that CCMP2459 is more sensitive to photoinhibition of PSII than CS-164, and tends to lose maximum photosynthetic activity faster. Measurement of photodamage to PSII under light of different wavelength ranges demonstrated that PSII in both Symbiodinium phylotypes was significantly more sensitive to photodamage under shorter wavelength regions of light spectra (

© 2014 Schrameyer et al. The light dependency of respiratory activity of two scleractinian corals was examined using O2 microsensors and CO2 exchange measurements. Light respiration increased strongly but asymptotically with elevated irradiance in both species. Light respiration in Pocillopora damicornis was higher than in Pavona decussata under low irradiance, indicating species-specific differences in light-dependent metabolic processes. Overall, the coral P. decussata exhibited higher CO2 uptake rates than P. damicornis over the experimental irradiance range. P. decussata also harboured twice as many algal symbionts and higher total protein biomass compared to P. damicornis, possibly resulting in self-shading of the symbionts and/or changes in host tissue specific light distribution. Differences in light respiration and CO2 availability could be due to host-specific characteristics that modulate the symbiont microenvironment, its photosynthesis, and hence the overall performance of the coral holobiont.

Photosynthesis by marine diatoms contributes substantially to global biogeochemical cycling and ecosystem productivity. It is widely accepted that diatoms are extremely sensitive to changes in Fe availability, with numerous in situ experiments demonstrating rapid growth and increased export of elements (e.g. C, Si and Fe) from surface waters as a result of Fe addition. Less is known about the effects of Fe enrichment on the phenotypes of diatoms, such as associated changes in nutritional value – furthermore, data on taxon-specific responses are almost non-existent. Enhanced supply of nutrient-rich waters along the coast of the subantarctic Kerguelen Island provide a valuable opportunity to examine the responses of phytoplankton to natural Fe enrichment. Here we demonstrate the use of synchrotron radiation Fourier Transform Infrared (SR-FTIR) microspectroscopy to analyse changes in the macromolecular composition of diatoms collected along the coast and plateau of Kerguelen Island, Southern Ocean. SR-FTIR microspectroscopy enabled the analysis of individual diatom cells from mixed communities of field-collected samples, thereby providing insight into in situ taxon-specific responses in relation to changes in Fe availability. Phenotypic responses were taxon-specific in terms of intraspecific variability and changes in proteins, amino acids, phosphorylated molecules, silicate/silicic acid and carbohydrates. In contrast to some previous studies, silicate/silicic acid levels increased under Fe enrichment, in conjunction with increases in carbohydrate stores. The highly abundant taxon Fragilariopsis kerguelensis displayed a higher level of phenotypic plasticity than Pseudo-nitzschia spp., while analysis of the data pooled across all measured taxa showed different patterns in macromolecular composition compared to those for individual taxon. This study demonstrates that taxon-specific responses to Fe enrichment may not always be accurately reflected by bulk community measurements, highlighting the need for further research into taxon-specific phenotypic responses of phytoplankton to environmental change.

Two inhibitors of the Calvin-Benson cycle [glycolaldehyde (GA) and potassium cyanide (KCN)] were used in cultured Symbiodinium cells and in nubbins of the coral Pocillopora damicornis to test the hypothesis that inhibition of the Calvin-Benson cycle triggers coral bleaching. Inhibitor concentration range-finding trials aimed to determine the appropriate concentration to generate inhibition of the Calvin-Benson cycle, but avoid other metabolic impacts to the symbiont and the animal host. Both 3 mmol l(-1) GA and 20 μmol l(-1) KCN caused minimal inhibition of host respiration, but did induce photosynthetic impairment, measured by a loss of photosystem II function and oxygen production. GA did not affect the severity of bleaching, nor induce bleaching in the absence of thermal stress, suggesting inhibition of the Calvin-Benson cycle by GA does not initiate bleaching in P. damicornis. In contrast, KCN did activate a bleaching response through symbiont expulsion, which occurred in the presence and absence of thermal stress. While KCN is an inhibitor of the Calvin-Benson cycle, it also promotes reactive oxygen species formation, and it is likely that this was the principal agent in the coral bleaching process. These findings do not support the hypothesis that temperature-induced inhibition of the Calvin-Benson cycle alone induces coral bleaching.

A relationship exists between the two-dimensional shape of leaves and their venation architecture, such that broad or broad-lobed leaves can have leaf tissue far from major veins, potentially creating stronger gradients in water potential – and associated photosynthetic function – than found across narrow counterparts. We examined the spatial patterns of photosynthetic efficiency (ΔF/Fm′) and non-photochemical quenching (NPQ) in response to increased vapour pressure deficit (VPD) using two morphs of Lomatia tinctoria (Labill.) R.Br: those with broad-lobed and those with narrow-lobed leaves. Stomatal conductance (gs), instantaneous water use efficiency (WUE), stomatal and minor veins density also were measured. ΔF/Fm′ decreased with stress but was higher and less spatially heterogeneous across broad than narrow lobes. The strongest depression in ΔF/Fm′ in broad lobes was at the edges and in narrow lobes, the tips. Non-photochemical quenching was spatially more varied in broad lobes, increasing at the edges and tips. Variation in photosynthetic function could not be explained by gs, WUE or minor vein density, whereas proximity to major veins appeared to mitigate water stress at the tips only for broad lobes. Our findings indicate that the relationship between venation architecture and water delivery alone can partially explain the spatial pattern of photosynthetic function.

Kirchhoff migration is the most popular method of three‐dimensional prestack depth migration because of its flexibility and efficiency. Its effectiveness can become limited, however, when complex velocity structure causes multipathing of seismic energy. An alternative is Gaussian beam migration, which is an extension of Kirchhoff migration that overcomes many of the problems caused by multipathing. Unlike first‐arrival and most‐energetic‐arrival methods, which retain only one traveltime, this alternative method retains most arrivals by the superposition of Gaussian beams. This paper presents a prestack Gaussian beam migration method that operates on common‐offset gathers. The method is efficient because the computation of beam superposition isolates summations that do not depend on the seismic data and evaluates these integrals by considering their saddle points. Gaussian beam migration of the two‐dimensional Marmousi test data set demonstrates the method’s effectiveness for structural imaging in a case where there is multipathing of seismic energy.

Understanding how multiple environmental stressors interact to affect seagrass health (measured as morphological and physiological responses) is important for responding to global declines in seagrass populations. We investigated the interactive effects of temperature stress (24, 27, 30 and 32°C) and shading stress (75, 50, 25 and 0% shade treatments) on the seagrass Zostera muelleri over a 3-month period in laboratory mesocosms. Z. muelleri is widely distributed throughout the temperate and tropical waters of south and east coasts of Australia, and is regarded as a regionally significant species. Optimal growth was observed at 27°C, whereas rapid loss of living shoots and leaf mass occurred at 32°C. We found no difference in the concentration of photosynthetic pigments among temperature treatments by the end of the experiment; however, up-regulation of photoprotective pigments was observed at 30°C. Greater levels of shade resulting in high photochemical efficiencies, while elevated irradiance suppressed effective quantum yield (ΔF/FM'). Chlorophyll fluorescence fast induction curves (FIC) revealed that the J step amplitude was significantly higher in the 0% shade treatment after 8 weeks, indicating a closure of PSII reaction centres, which likely contributed to the decline in ΔF/FM' and photoinhibition under higher irradiance. Effective quantum yield of PSII (ΔF/FM') declined steadily in 32°C treatments, indicating thermal damage. Higher temperatures (30°C) resulted in reduced above-ground biomass ratio and smaller leaves, while reduced light led to a reduction in leaf and shoot density, above-ground biomass ratio, shoot biomass and an increase in leaf senescence. Surprisingly, light and temperature had few interactive effects on seagrass health, even though these two stressors had strong effects on seagrass health when tested in isolation. In summary, these results demonstrate that populations of Z. muelleri in south-eastern Australia are sensitive to small chronic temperature increases and light decreases that are predicted under future climate change scenarios. © 2013 York et al.

Expert opinion was assessed to identify current knowledge gaps in determining future changes in Arabian/Persian Gulf (thereafter 'Gulf') coral reefs. Thirty-one participants submitted 71 research questions that were peer-assessed in terms of scientific importance (i.e., filled a knowledge gap and was a research priority) and efficiency in resource use (i.e., was highly feasible and ecologically broad). Ten research questions, in six major research areas, were highly important for both understanding Gulf coral reef ecosystems and also an efficient use of limited research resources. These questions mirrored global evaluations of the importance of understanding and evaluating biodiversity, determining the potential impacts of climate change, the role of anthropogenic impacts in structuring coral reef communities, and economically evaluating coral reef communities. These questions provide guidance for future research on coral reef ecosystems within the Gulf, and enhance the potential for assessment and management of future changes in this globally significant region. © 2013 Elsevier Ltd.

Diatoms are the primary source of nutrition and energy for the Southern Ocean ecosystem. Microalgae, including diatoms, synthesise biological macromolecules such as lipids, proteins and carbohydrates for growth, reproduction and acclimation to prevailing environmental conditions. Here we show that three key species of Southern Ocean diatom (Fragilariopsis cylindrus, Chaetoceros simplex and Pseudo-nitzschia subcurvata) exhibited phenotypic plasticity in response to salinity and temperature regimes experienced during the seasonal formation and decay of sea ice. The degree of phenotypic plasticity, in terms of changes in macromolecular composition, was highly species-specific and consistent with each species' known distribution and abundance throughout sea ice, meltwater and pelagic habitats, suggesting that phenotypic plasticity may have been selected for by the extreme variability of the polar marine environment. We argue that changes in diatom macromolecular composition and shifts in species dominance in response to a changing climate have the potential to alter nutrient and energy fluxes throughout the Southern Ocean ecosystem. © 2013 Sackett et al.

An increase in serum lactate dehydrogenase (LDH) activity is commonly taken to support the presumptive diagnosis of Pneumocystis carinii pneumonia (PCP), although the LDH level may also be increased in other lung infections and in a variety of extrapulmonary disorders. To assess its diagnostic value in patients with fever, lung infiltrates, and a high prevalence of HIV infection, we compared LDH levels in 42 hospitalized patients with PCP, 71 with disseminated tuberculosis (TB), 40 with pulmonary TB, and 37 with bacterial pneumonia. Peak LDH level was higher (p0.05) in patients with PCP (547 +/- 157 U/L) and disseminated TB (569 +/- 338 U/L) than in patients with pulmonary TB (258 +/- 66 U/L) or bacterial pneumonia (331 +/- 139 U/L). However, substantial overlap between groups limited its diagnostic value for individual patients. Expressing LDH as its ratio to simultaneous serum aminotransferases (AST or ALT) did not enhance its discriminatory value. Most patients in each group had abnormalities in other serum enzymes (AST, ALT, alkaline phosphatase, gamma-glutamyltransferase), making an isolated elevation of LDH level uncommon (21% of PCP cases). Serum LDH has a high sensitivity for PCP (100% in this series) but must be interpreted with caution given its lack of specificity.
