8. Inhibition of Cancer Stem-like Cells by Curcumin and Other Polyphenol Derivatives in MDA-MB-231 TNBC Cells.

Abstract

Triple-negative breast cancer (TNBC) accounts for 15% of all breast cancers and is highly aggressive. Despite an initial positive response to chemotherapy, most patients experience rapid disease progression leading to relapse and metastasis. This is attributed to the presence of breast cancer stem cells (BCSCs) within the tumor, which are characterized by self-renewal, pluripotency, and resistance mechanisms. Targeting BCSCs has become critical as conventional therapies fail to eradicate them due to a lack of specific targets. Curcumin, a polyphenol derived from turmeric (Curcuma longa), exhibits anticancer effects against breast cancer cells and BCSCs. The use of curcumin derivatives has been suggested as an approach to overcome the bioavailability and solubility problems of curcumin in humans, thereby increasing its anticancer effects. The aim of this study was to evaluate the cellular and molecular effects of six synthetic compounds derived from the natural polyphenol epigallocatechin gallate (EGCG) (TL1, TL2) and curcumin derivatives (TL3, TL4, TL5, and TL6) on a TNBC mesenchymal stem-like cell line. The activity of the compounds against BCSCs was also determined by a mammosphere inhibition assay and studying different BCSC markers by Western blotting. Finally, a drug combination assay was performed with the most promising compounds to evaluate their potential synergistic effects with the chemotherapeutic agents doxorubicin, cisplatin, and paclitaxel. The results showed that compounds exhibited specific cytotoxicity against the TNBC cell line and BCSCs. Interestingly, the combination of the curcumin derivative TL3 with doxorubicin and cisplatin displayed a synergistic effect in TNBC cells. [ABSTRACT FROM AUTHOR]

Abstract

Understanding environmental information is necessary for functions correlated with human activities to improve healthcare quality and reduce ecological risk. Tapered optical fibers reduce some limitations of such devices and can be considerably more responsive to fluorescence and absorption properties changes. Data have been collected from reliable sources such as Science Direct, IEEE Xplore, Scopus, Web of Science, PubMed, and Google Scholar. In this narrative review, we have summarized and analyzed eight classes of tapered-fiber forms: fiber Bragg grating (FBG), long-period fiber grating (LPFG), Mach–Zehnder interferometer (MZI), photonic crystals fiber (PCF), surface plasmonic resonance (SPR), multi-taper devices, fiber loop ring-down technology, and optical tweezers. We evaluated many issues to make an informed judgement about the viability of employing the best of these methods in optical sensors. The analysis of performance for tapered optical fibers depends on four mean parameters: taper length, sensitivity, wavelength scale, and waist diameter. Finally, we assess the most potent strategy that has the potential for medical and environmental applications.

Abstract

The clinical learning environment, which includes the culture of clinical units, the mentoring process, and the different health organizations, influences the learning process of nursing students. However, scarce literature has been published on the impact of the clinical learning environment on first-year nursing students in long-term care. We aimed to assess first-year nursing students ‘preferred’ and ‘actual’ clinical learning environments when conducting their first placements in nursing homes within an innovative placement model that comprised the active participation of academic mentors. The validated Spanish version of the Clinical Learning Environment Inventory (CLEI) instrument was used in our study, and 99 first-year nursing students participated. The highest mean scores for the CLEI-Actual were found for the Satisfaction (22.7) and Involvement scales (19.09). The lowest mean scores were found for the Personalization (17) and Individualization (17.27) scales. The multiple correlation (R) between the Satisfaction and the other CLEI scales was 0.61 (p > 0.001), which means that in this study the association between student satisfaction and their perception of the clinical learning environment was strong. First-year students conducting their first clinical placements in nursing homes can have a positive learning experience considering a well-designed and organized pedagogical strategy, including constant support and feedback from academic and clinical mentors.

Abstract

Incontinència urinària; Tractament; Algorisme Incontinencia urinaria; Tratamiento; Algoritmo Urinary incontinence; Treatment; Algorithm Aquesta pauta té l’objectiu d’harmonitzar el tractament farmacològic dels pacients amb incontinència urinària i es dirigeix, principalment, als professionals de l’àmbit de l’atenció primària, comunitària i especialitzada. Es prioritzen les guies de pràctica clínica de referència vigents i s’identifiquen les revisions sistemàtiques i els assaigs clínics de rellevància que plantegen preguntes clíniques concretes. La pauta també incorpora la revisió de l’evidència científica generada després de la publicació de les esmentades guies i la utilització de criteris d’eficiència per a la realització de recomanacions de selecció i priorització de medicaments i/o grups terapèutics, d’acord amb la política farmacèutica del CatSalut. Esta pauta tiene el objetivo de armonizar el tratamiento farmacológico de los pacientes con incontinencia urinaria y se dirige, principalmente, a los profesionales del ámbito de la atención primaria, comunitaria y especializada. Se priorizan las guías de práctica clínica de referencia vigentes y se identifican las revisiones sistemáticas y los ensayos clínicos de relevancia que plantean preguntas clínicas concretas. La pauta también incorpora la revisión de la evidencia científica generada tras la publicación de dichas guías y la utilización de criterios de eficiencia para la realización de recomendaciones de selección y priorización de medicamentos y / o grupos terapéuticos, de acuerdo con la política farmacéutica del CatSalut. This guideline aims to harmonize the pharmacological treatment of patients with urinary incontinence and is aimed primarily at professionals in the field of primary, community and specialized care. Current clinical practice guidelines are prioritized and systematic reviews and relevant clinical trials that raise specific clinical questions are identified. The guideline also includes the review of the scientific evidence generated after the publication of the aforementioned guidelines and the use of efficiency criteria for making recommendations for the selection and prioritization of drugs and / or therapeutic groups, in accordance with the pharmaceutical policy of CatSalut.

Spain was one of the European countries with the highest incidence during the first wave. As a part of a consortium to monitor and study the evolution of the epidemic, we sequenced 2,170 samples, diagnosed mostly before lockdown measures. Here, we identified at least 500 introductions from multiple international sources and documented the early rise of two dominant Spanish epidemic clades (SECs), probably amplified by superspreading events. Both SECs were related closely to the initial Asian variants of SARS-CoV-2 and spread widely across Spain. Abstract

Phocomelia is a devastating, rare congenital limb malformation in which the long bones are shorter than normal, with the upper portion of the limb being most severely affected. In extreme cases, the hands or fingers are attached directly to the shoulder and the most proximal elements (those closest to the shoulder) are entirely missing. This disorder, previously known in both autosomal recessive and sporadic forms, showed a marked increase in incidence in the early 1960s due to the tragic toxicological effects of the drug thalidomide, which had been prescribed as a mild sedative (1,2). This human birth defect is mimicked in developing chick limb buds exposed to X-irradiation (3-5). Both X-irradiation (5) and thalidomide-induced phocomelia (5,6) have been interpreted as patterning defects in the context of the progress zone model, which states that a cell's proximodistal identity is determined by the length of time spent in a distal limb region termed the 'progress zone' (7). Indeed, studies of X-irradiation-induced phocomelia have served as one of the two major experimental lines of evidence supporting the validity of the progress zone model. Here, using a combination of molecular analysis and lineage tracing in chick, we show that X-irradiation-induced phocomelia is fundamentally not a patterning defect, but rather results from a time-dependent loss of skeletal progenitors. Because skeletal condensation proceeds from the shoulder to fingers (in a proximal to distal direction), the proximal elements are differentially affected in limb buds exposed to radiation at early stages. This conclusion changes the framework for considering the effect of thalidomide and other forms of phocomelia, suggesting the possibility that the aetiology lies not in a defect in the patterning process, but rather in progenitor cell survival and differentiation.
