Abstract Malaria transmission-blocking vaccines (TBVs) aim to induce antibodies that block Plasmodium parasite development in the mosquito midgut, thus preventing mosquitoes from becoming infectious. While the Pro-domain and first of fourteen 6-Cysteine domains (Pro-D1) of the Plasmodium gamete surface protein Pfs230 are known targets of transmission-blocking antibodies, no studies to date have discovered other Pfs230 domains that are functional targets. Here, we show that a murine monoclonal antibody (mAb), 18F25.1, targets Pfs230 Domain 7. We generated a subclass-switched complement-fixing variant, mAb 18F25.2a, using a CRISPR/Cas9-based hybridoma engineering method. This subclass-switched mAb 18F25.2a induced lysis of female gametes in vitro. Importantly, mAb 18F25.2a potently reduced P. falciparum infection of Anopheles stephensi mosquitoes in a complement-dependent manner, as assessed by standard membrane feeding assays. Together, our data identify Pfs230 Domain 7 as target for transmission-blocking antibodies and provide a strong incentive to study domains outside Pfs230Pro-D1 as TBV candidates.

IntroductionStudies have consistently demonstrated increased stress sensitivity in individuals with psychosis. Since stress sensitivity may play a role in the onset and maintenance of psychosis, this could potentially be a promising target for treatment. The current study was the first to investigate whether reactivity to and recovery from daily-life stressors in psychosis change in response to treatment, namely virtual-reality-based cognitive behavioral therapy (VR-CBT).Methods116 patients were randomized to either VR-CBT or the waiting list control group (WL). Pre-treatment and post-treatment participants completed a diary ten times a day during six to ten days. Multilevel analyses were used to model the time-lagged effects of daily stressful events on negative affect (NA) and paranoia symptoms to examine reactivity and recovery.ResultsThere was a significant difference in NA reactivity. VR-CBT showed higher NA at post-treatment compared to pre-treatment than WL (bpre=0.14; bpost=0.19 vs bpre=0.18; bpost=0.14). There was a significant difference in NA recovery and paranoia recovery between the groups at lag 1: VR-CBT showed relatively lower negative affect (bpre=0.07; bpost=-0.06) and paranoia (bpre= 0.08; bpost=-0.10) at post-treatment compared to pre-treatment than WL (bpre=0.08; bpost=0.08; bpre=0.04; bpost=0.03).ConclusionNegative affect and paranoia recovery improved in response to treatment. Increased NA reactivity may be explained by a decrease in safety behavior in the VR-CBT group. The discrepancy between reactivity and recovery findings may be explained by the inhibitory learning theory that suggests that an original threat reaction may not erase but can be inhibited as a consequence of exposure therapy.

We studied the role of adherence to antihypertensive drug therapy (AHT) in blood pressure (BP) control in a type 2 diabetes (T2D) population treated in secondary care in the DIAbetes and LifEstyle Cohort Twente-1 (DIALECT-1). In addition, intensification of AHT was assessed. Adherence was determined by using the medication possession ratio (MPR), calculated with pharmacy dispensing data for a period of two years following baseline. Adherence was defined as an MPR ≥ 80%. The proportion of adherent patients was compared between patients who had BP-on target (BP-OT) and BP-not on target (BP-NOT). Of the 385 patients included, 56% achieved their BP target. The proportion of adherent patients did not differ between BP-OT and BP-NOT (96% vs. 96%; p = 0.91). Intensification of AHT, including ‘increase in dosage’ and ‘start of a new drug’, was assessed in the two years following baseline. In only 37% of patients with uncontrolled BP during follow-up was AHT intensified. To conclude, adherence to AHT was high and there does not seem to be a relationship between adherence and BP control. There is an opportunity to improve AHT in patients who do not reach their BP target.

Individuals infected with P. falciparum develop antibody responses to intra-erythrocytic gametocyte proteins and exported gametocyte proteins present on the surface of infected erythrocytes. However, there is currently limited knowledge on the immunogenicity of gametocyte antigens and the specificity of gametocyte-induced antibody responses. In this study, we assessed antibody responses in participants of two controlled human malaria infection (CHMI) studies by ELISA, multiplexed bead-based antibody assays and protein microarray. By comparing antibody responses in participants with and without gametocyte exposure, we aimed to disentangle the antibody response induced by asexual and sexual stage parasites. We showed that after a single malaria infection, a significant anti-sexual stage humoral response is induced in malaria-naïve individuals, even after exposure to relatively low gametocyte densities (up to ~1,600 gametocytes/mL). In contrast to antibody responses to well-characterised asexual blood stage antigens that were detectable by day 21 after infection, responses to sexual stage antigens (including transmission blocking vaccine candidates Pfs48/45 and Pfs230) were only apparent at 51 days after infection. We found antigens previously associated with early gametocyte or anti-gamete immunity were highly represented among responses linked with gametocyte exposure. Our data provide detailed insights on the induction and kinetics of antibody responses to gametocytes and identify novel antigens that elicit antibody responses exclusively in individuals with gametocyte exposure. Our findings provide target identification for serological assays for surveillance of the malaria infectious reservoir, and support vaccine development by describing the antibody response to leading vaccine antigens after primary infection.

Abstract Background Primary antibody deficiencies (PAD) are characterized by a heterogeneous clinical presentation and low prevalence, contributing to a median diagnostic delay of 3–10 years. This increases the risk of morbidity and mortality from undiagnosed PAD, which may be prevented with adequate therapy. To reduce the diagnostic delay of PAD, we developed a screening algorithm using primary care electronic health record (EHR) data to identify patients at risk of PAD. This screening algorithm can be used as an aid to notify general practitioners when further laboratory evaluation of immunoglobulins should be considered, thereby facilitating a timely diagnosis of PAD. Methods Candidate components for the algorithm were based on a broad range of presenting signs and symptoms of PAD that are available in primary care EHRs. The decision on inclusion and weight of the components in the algorithm was based on the prevalence of these components among PAD patients and control groups, as well as clinical rationale. Results We analyzed the primary care EHRs of 30 PAD patients, 26 primary care immunodeficiency patients and 58,223 control patients. The median diagnostic delay of PAD patients was 9.5 years. Several candidate components showed a clear difference in prevalence between PAD patients and controls, most notably the mean number of antibiotic prescriptions in the 4 years prior to diagnosis (5.14 vs. 0.48). The final algorithm included antibiotic prescriptions, diagnostic codes for respiratory tract and other infections, gastro-intestinal complaints, auto-immune symptoms, malignancies and lymphoproliferative symptoms, as well as laboratory values and visits to the general practitioner. Conclusions In this study, we developed a screening algorithm based on a broad range of presenting signs and symptoms of PAD, which is suitable to implement in primary care. It has the potential to considerably reduce diagnostic delay in PAD, and will be validated in a prospective study. Abstract Nanoscale gadolinium oxide (Gd2O3) is a promising nanomaterial with unique physicochemical properties that finds various applications ranging from biomedicine to catalysis. The preparation of highly porous Gd2O3 nanofoam greatly increases its surface area thereby boosting its potential for functional use in applications such as water purification processes and in catalytic applications. By using the combustion synthesis method, a strong exothermic redox reaction between gadolinium nitrate hexahydrate and glycine causes the formation of crystalline nanoporous Gd2O3. In this study, the synthesis of Gd2O3 nanofoam is achieved with combustion synthesis at large scale (grams). Its nanoscale porosity is investigated by nitrogen physisorption and its nanoscale 3D structure by electron tomography, and the formation process is investigated as well by means of in situ heating inside the transmission electron microscope. The bulk nanofoam product is highly crystalline and porous with a surface area of 67 m2 g−1 as measured by physisorption, in good agreement with the electron tomographic 3D reconstructions showing an intricate interconnected pore network with pore sizes varying from 2 to 3 nm to tens of nanometers. In situ heating experiments point to many possibilities for tuning the porosity of the Gd2O3 nanofoam by varying the experimental synthesis conditions.

We report results of systematic, holistic, diagnostic, and cell studies to elucidate the mechanistic role of the experimentally determined influence of the anode gas-diffusion layer (GDL) on the performance of ultra-thin electrode polymer-electrolyte fuel cells, which can further enable fuel-cell market penetration. Measurements of product water balance and in situ neutron imaging of operational membrane-electrode-assembly water profiles demonstrate how improved performance is due to a novel anode GDL fiber-density modulated structure at the micrometer scale that removes water preferentially out of the anode, a key strategy to manage water in these cells. The banded structure results in low transport-resistance pathways, which affect water-droplet removal from the GDL surface. This interfacial effect is unexpectedly shown to be critical for decreasing overall water holdup throughout the cell. These studies demonstrate a new material paradigm for understanding and controlling fuel-cell water management and related high-power technologies or electrodes where multiphase flow occurs. Very thin electrodes enable high power density in electrochemical technologies, yet their thinness engenders issues related to buildup of products (e.g., water in polymer-electrolyte fuel cells [PEFCs]). The article explores an unexpected materials solution to the problem, which highlights the need to study such complicated systems in a holistic manner of a complete cell due to the nonlinearities existent in the highly coupled physical phenomena. The improved performance is due to an inherent unintentional manufacturing heterogeneity in the cell backing layer, which mainly affects its surface properties. With this knowledge, one can now engineer and optimize these critical heterogeneities for different architectures. The findings are relevant to those working on materials for electrochemical energy conversion and represent new key knowledge that can have significant impact in PEFCs and related electrochemical cells, especially where multiphase flow occurs. High-power electrodes in electrochemical technologies (e.g., fuel cells) typically require ultra-thin catalyst layers, which, especially when multiphase flow exists, exhibit mass-transport limitations. These have been mitigated through new backing layer structures and nontraditional removal of water out of the anode side of the cell for a new design and operation paradigm.

We present magnetic characterization, charge resistivity, and optical photoluminescence measurements on amorphous yttrium iron oxide thin films (a -Y–Fe–O), with supporting comparisons to amorphous germanium (a -Ge) films. We measured magnetic properties with both SQUID magnetometry and polarized neutron reflectometry. These results not only confirm that a -Y–Fe–O is a disordered magnetic material with strong predominantly antiferromagnetic exchange interactions and a high degree of frustration, but also that it is best understood electrically as a disordered semiconductor. As with amorphous germanium, a -Y–Fe–O obeys expectations for variable-range hopping through localized electron states over a wide range of temperature. We also clarify the consequences of charge transport through such a semiconducting medium for non-local voltage measurements intended to probe spin transport in nominally insulating magnetic materials. We further compare non-local resistance measurements made with "quasi-dc" automated current reversal to ac measurements made with a lock-in amplifier. These show that the "quasi-dc" measurement has an effective ac current excitation with frequency up to approximately 22 Hz, and that this effective ac excitation can cause artifacts in these measurements including incorrect sign of the non-local resistance. This comprehensive investigation of non-local resistance measurements in a -Y–Fe–O shows no evidence of spin transport on micrometer length scales, which is contrary to our original work, and in line with more recent investigations by other groups. Abstract Malaria parasite transmission to mosquitoes relies on the uptake of sexual stage parasites during a blood meal and subsequent formation of oocysts on the mosquito midgut wall. Transmission-blocking vaccines (TBVs) and monoclonal antibodies (mAbs) target sexual stage antigens to interrupt human-to-mosquito transmission and may form important tools for malaria elimination. Although most epitopes of these antigens are considered highly conserved, little is known about the impact of natural genetic diversity on the functional activity of transmission-blocking antibodies. Here we measured the efficacy of three mAbs against leading TBV candidates (Pfs48/45, Pfs25 and Pfs230) in transmission assays with parasites from naturally infected donors compared to their efficacy against the strain they were raised against (NF54). Transmission-reducing activity (TRA) was measured as reduction in mean oocyst intensity. mAb 45.1 (α-Pfs48/45) and mAb 4B7 (α-Pfs25) reduced transmission of field parasites from almost all donors with IC80 values similar to NF54. Sequencing of oocysts that survived high mAb concentrations did not suggest enrichment of escape genotypes. mAb 2A2 (α-Pfs230) only reduced transmission of parasites from a minority of the donors, suggesting that it targets a non-conserved epitope. Using six laboratory-adapted strains, we revealed that mutations in one Pfs230 domain correlate with mAb gamete surface binding and functional TRA. Our findings demonstrate that, despite the conserved nature of sexual stage antigens, minor sequence variation can significantly impact the efficacy of transmission-blocking mAbs. Since mAb 45.1 shows high potency against genetically diverse strains, our findings support its further clinical development and may inform Pfs48/45 vaccine design.

Objective: To assess adherence to statin therapy and its association with sociodemographic data, medical characteristics, LDLc levels, and LDLc target attainment in real-world T2D patients treated in secondary care.Research Design and Methods: Cross-sectional analyses were performed on baseline data of 393 patients in the DIAbetes and LifEstyle Cohort Twente (DIALECT). The medication possession ratio (MPR), calculated with pharmacy dispensing data, was used to determine adherence to statins for an intended period of 24 months. Statins were included in the analyses if they were used for at least six consecutive months with at least three dispenses. Adherence was defined as an MPR ≥80%. Associations with adherence were assessed using descriptive statistics and binary logistic regression.Results: Overall, 80% of the patients had a statin prescription and of those, 89% were adherent. Malaria transmission blocking vaccines (TBV) aim to induce antibodies that can interrupt Plasmodium falciparum development in the mosquito midgut and thereby prevent onward malaria transmission. A limited number of TBV candidates have been identified and only three (Pfs25, Pfs230 and Pfs48/45) have entered clinical testing. While one of these candidates may emerge as a highly potent TBV candidate, it is premature to determine if they will generate sufficiently potent and sustained responses. It is therefore important to explore novel candidate antigens. We recently analyzed sera from naturally exposed individuals and found that the presence and/or intensity of antibodies against 12 novel putative surface expressed gametocyte antigens was associated with transmission reducing activity. In this study, protein fragments of these novel TBV candidates were designed and heterologously expressed in Drosophila melanogaster S2 cells and Lactococcus lactis. Eleven protein fragments, covering seven TBV candidates, were successfully produced. All tested antigens were recognized by antibodies from individuals living in malaria-endemic areas, indicating that native epitopes are present. All antigens induced antigen-specific antibody responses in mice. Two antigens induced antibodies that recognized a native protein in gametocyte extract, and antibodies elicited by four antigens recognized whole gametocytes. In particular, we found that antigen Pf3D7_0305300, a putative transporter, is abundantly expressed on the surface of gametocytes. However, none of the seven novel TBV candidates expressed here induced an antibody response that reduced parasite development in the mosquito midgut as assessed in the standard membrane feeding assay. Altogether, the antigen fragments used in this study did not prove to be promising transmission blocking vaccine constructs, but led to the identification of two gametocyte surface proteins that may provide new leads for studying gametocyte biology.

Piglet mortality from farrowing to weaning is a major concern, especially in outdoor organic production systems. This issue might impair animal welfare and generate economic losses for the farmer. In particular, it is difficult to apply management tools that are commonly used for indoor pig production systems to organic or outdoor production systems. Genetics and breeding approaches might be used to improve piglet survival. However, knowledge remains limited on the genetic background underlying survival traits in organic pigs that are born and reared outdoors. Here, we investigated the mortality of piglets from farrowing to weaning in an outdoor organic pig population and suggested genetic strategies to reduce piglet mortality in this production system. The experiment included mortality records of piglets from farrowing to weaning (around 69 days of age). Pedigree-based threshold models were used to analyse the mortality traits of piglets at 0–3 days of age, 4–11 days, and 12 days to weaning. Stillborn piglets were included in the group of piglets that died at 0–3 days of age. We found that the mortality rate from farrowing to weaning was, on average, 19.2%. However, most piglet deaths (79.1%) occurred at 0–11 days of age. As the age of piglets increased, the direct heritability of piglet mortality rose from 0 to 0.04, whereas maternal heritability decreased from 0.03 to a non-significant value. Piglets with higher BW had a lower mortality rate. However, the genetic correlations between maternal effects on piglet mortality and piglet BW were not significant; thus, selection for piglets with higher BW at around 10 days of age, through improving maternal genetics, would not reduce piglet mortality. Piglet mortality increased from sows with increasing number of parities. Crossbreeding also reduced piglet mortality. In conclusion, selection focusing on sow genotype, the use of younger sows, and crossbreeding could contribute to maintain piglet mortality at lower levels in outdoor organic pig production systems.

Type II diabetes is a complex health condition requiring patients to closely and continuously collaborate with healthcare professionals and other caretakers on lifestyle changes. While intelligent products have tremendous potential to support such Diabetes Lifestyle Management (DLM), existing products are typically conceived from a technology-centered perspective that insufficiently acknowledges the degree to which collaboration and inclusion of stakeholders is required. In this article, we argue that the emergent design philosophy of Hybrid Intelligence (HI) forms a suitable alternative lens for research and development. In particular, we (1) highlight a series of pragmatic challenges for effective AI-based DLM support based on results from an expert focus group, and (2) argue for HI’s potential to address these by outlining relevant research trajectories. The erosional state of a landscape is often assessed through a series of metrics that quantify the morphology of drainage basins and divides. Such metrics have been well explored in tectonically active environments to evaluate the role of different processes in sculpting topography, yet relatively few works have applied these analyses to radial landforms such as volcanoes. We quantify drainage basin geometries on volcanic edifices of varying ages using common metrics (e.g., Hack's law, drainage density, and number of basins that reach the edifice summit, as well as basin hypsometry integral, length, width, relief, and average topographic slope). Relating these measurements to the log-mean age of activity for each edifice, we find that drainage density, basin hypsometry, basin length, and basin width quantify the degree of erosional maturity for these landforms. We also explore edifice drainage basin growth and competition by conducting a divide mobility analysis on the volcanoes, finding that young volcanoes are characterized by nearly uniform fluvial basins within unstable configurations that are more prone to divide migration. As basins on young volcanoes erode, they become less uniform but adapt to a more stable configuration with less divide migration. Finally, we analyze basin spatial geometries and outlet spacing on edifices, discovering an evolution in radial basin configurations that differ from typical linear mountain ranges. From these, we present a novel conceptual model for edifice degradation that allows new interpretations of composite volcano histories and provides predictive quantities for edifice morphologic evolution.
