Abstract Background Rhododendron delavayi is a natural shrub that is distributed at different elevations in the karst region of Bijie, China, and that has an important role in preventing land degradation in this region. In this study, we determined the soil mineral element contents and soil enzyme activities. The composition of the soil bacterial community of R. delavayi at three elevations (1448 m, 1643 m, and 1821 m) was analyzed by high-throughput sequencing, and the interrelationships among the soil bacterial communities, mineral elements, and enzyme activities were determined. Results The Shannon index of the soil bacterial community increased and then decreased with increasing elevation and was highest at 1643 m. Elevations increased the number of total nodes and edges of the soil bacterial community network, and more positive correlations at 1821 m suggested stronger intraspecific cooperation. Acidobacteria, Actinobacteria and Proteobacteria were the dominant phyla at all three elevations. The Mantel test and correlation analysis showed that Fe and soil urease significantly affected bacterial communities at 1448 m; interestingly, Chloroflexi was positively related to soil urease at 1448 m, and Actinobacteria was positively correlated with Ni and Zn at 1821 m. Fe and soil urease significantly influenced the bacterial communities at lower elevations, and high elevation (1821 m) enhanced the positive interactions of the soil bacteria, which might be a strategy for R. delavayi to adapt to high elevation environments. Conclusion Elevation significantly influenced the composition of soil bacterial communities by affecting the content of soil mineral elements and soil enzyme activity.

Abstract Older adults who are frail are one of the most sedentary and the least physically active age groups. Prolonged sedentary time is associated with increased risk of negative health outcomes. To help design effective and sustainable content and optimize the uptake of sedentary behaviour interventions, an in-depth understanding of older adults’ perceptions of sedentary behaviour is needed; however, most qualitative studies have been conducted in healthy older adults. The aim of this study was to explore perspectives of sedentary behaviour within the context of older adults who are pre-frail and frail after the winter and spring. We included participants if they: (1) spoke English or attended with a translator or caregiver, (2) were ≥ 60 years, and (3) were frail on the Morley Frail Scale. We utilized a qualitative description methodology including a semi-structured in-depth interview and thematic content analysis. Concepts from the COM-B (Capability Opportunity Motivation–Behaviour) model were used to guide the semi-structured interviews and analysis. To ensure credibility of the data, we used an audit trail and analyst triangulation. We recruited 21 older adults (72 ± 7.3 years, 13 females, 13 frail) from southwestern Ontario, Canada. Two individuals were lost to follow-up due to medical mistrust and worse health. We transcribed 39 audio recordings. We identified three salient themes: (1) older adults rationalize their sedentary behaviours through cognitive dissonance (reflective motivation), (2) urban cities in southwestern Ontario may not be “age-friendly” (physical opportunity), and (3) exercise is something people “have to do”, but hobbies are for enjoyment despite medical conditions (psychological capability). Perspectives of sedentary behaviour were different in the winter versus spring, with participants perceiving themselves to be less active in winter. Incorporating dissonance-based interventions as part of an educational program could be used to target the reflective motivation and psychological capability components. Future research should consider interdisciplinary collaborations with environmental gerontology to develop age-friendly communities that promote meaningful mobility to target physical opportunity.

Introduction China enacted an excise tax on e-cigarettes in November 2022, which offers a distinctive opportunity to examine the industry's reactions to this fiscal adjustment. This study delves into the industry's pricing strategies following the introduction of the excise tax, facilitating a thorough assessment of the subsequent impact on market dynamics and the government's revenue streams. Methods We developed a TaXSiM model specifically tailored for e-cigarettes in China by integrating the country's e-cigarette tax framework. Our approach involved leveraging market data obtained from a representative product, the RELX Phantom Series, to ensure the model's effectiveness and relevance. Results The excise implementation of 2022 significantly heightened the tax burden on e-cigarettes, marking an increase of approximately 150 RMB per device and 19 RMB per cartridge. Despite these financial pressures, electronic cigarette firms exemplified by RELX, strategically endeavored to sustain competitiveness. Their approach involved initially implementing a ‘Razor blade model’ and eventually a ‘comprehensive under-shifting’ strategy, which mitigated the health impact of the tax hike, resulting in a relatively minor decline in sales while amplifying the impact on tax revenue. However, this strategic pricing maneuver came at a cost, as it led to a substantial decrease in profits, and therefore expedited a reshuffling of the industry by compelling smaller brands to leave the market rapidly. Conclusions To effectively curb the use of e-cigarettes through tax policies, it is advisable to relocate the imposition of excise taxes on electronic cigarettes to the retail stage. This shift aims to narrow the scope for industry-level pricing strategies. Furthermore, this approach should be coupled with the introduction of an additional specific tax, strategically crafted to accentuate the health-related benefits associated with the excise taxation on electronic cigarettes.

Summary: Purpose: This study aims to elucidate the longitudinal alterations in frailty and health-related quality of life experienced by elderly patients undergoing surgical treatment for esophageal cancer. Additionally, it seeks to ascertain the impact of preoperative frailty on postoperative health-related quality of life over time. Methods: 131 patients were included in the prospective study. Patients' frailty and health-related quality-of-life were assessed utilizing the Tilburg and European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Core 30 at preoperative, 1 week, 1 month, and 3 months, postoperatively. Statistical analyses were performed using generalized estimating equations, repeated-measures analysis of variance, and linear mixed models (LMMs). Results: Out of 131 patients, 28.2% had frailty before surgery, and the prevalence of frailty consistently higher after surgery compared with baseline (67.9%, 51.9%, and 39.7%). Abstract Objective The molecular mechanism of the protective effect of Cordyceps cicadae polysaccharides (CCPs) on renal tubulointerstitial fibrosis in diabetic nephropathy (DN) is still unclear. This study aims to further understand the molecular mechanisms behind the therapeutic benefits of CCP on diabetic nephropathy. Methods Mice were randomly assigned into six groups (n = 8). Cordyceps cicadae polysaccharide dissolved in 5% dimethyl sulfoxide was administered by gavage for 12 consecutive weeks. The CCP doses were divided into low, medium, and high, 75, 150, and 300 mg/kg/day, respectively. The efficacy of CCP was determined by assessing the renal function and histological alterations in diabetic db/db mice. The degree of glomerular mesangial dilatation and sclerosis was evaluated using semiquantitative markers. The western Pacific warm pool (WPWP) is a major thermal driver of atmospheric deep convection and global atmospheric circulation in the tropics, and changes in its ocean heat content (OHC) affect the local and global climates. Four state-of-the-art ocean reanalyses and one objective analysis were used to study the variations in the WPWP OHC, ocean heat content tendency (OHCT), and ocean heat transport (OHT). The variabilities of both the OHC and OHCT integrated from 0 to 300 m are consistent between the datasets and are closely related to the El-Niño southern oscillation cycle. The integrated OHC from 0 to 2000 m shows an overall increasing trend in the WPWP. The WPWP mainly gains heat from the eastern boundary and loses heat from the northern boundary. The heat transport through the eastern boundary of the WPWP is mainly facilitated by the westward flowing south and north equatorial currents as well as the equatorial countercurrent around the depth of the thermocline, whereas the OHT at the northern boundary is mainly driven by the western boundary current of the Pacific Ocean, which shows complex flow structures.

Objective: This prospective study aims to evaluate acute irradiation-induced xerostomia during radiotherapy by utilizing the normalized iodine concentration (NIC) derived from energy spectrum computed tomography (CT) iodine maps. Methods: In this prospective study, we evaluated 28 patients diagnosed with nasopharyngeal carcinoma. At 4 distinct stages of radiotherapy (0, 10, 20, and 30 fractions), each patient underwent CT scans to generate iodine maps. The NIC of both the left and right parotid glands was obtained, with the NIC at the 0-fraction stage serving as the baseline measurement. After statistically comparing the NIC obtained in the arterial phase, early venous phase, late venous phase, and delayed phase, we chose the late venous iodine concentration as the NIC and proceeded to analyze the variations in NIC at each radiotherapy interval. Using the series of NIC values, we conducted hypothesis tests to evaluate the extent of change in NIC within the parotid gland across different stages. Furthermore, we identified the specific time point at which the NIC decay exhibited the most statistically significant results. In addition, we evaluated the xerostomia grades of the patients at these 4 stages, following the radiation therapy oncology group (RTOG) xerostomia evaluation standard, to draw comparisons with the changes observed in NIC. Abstract Purpose To identify genes associated with treatment response and prognosis for locally advanced rectal cancer (LARC) patients receiving neoadjuvant chemoradiotherapy (NCRT). Methods In our cohort, gene expression profiles of 64 tumor biopsy samples before NCRT were examined and generated. Weighted gene co-expression network analysis was performed to identify gene modules. External validation datasets included GSE3493, GSE119409, and GSE133057. The expression of candidate genes was evaluated using immunohistochemistry (IHC). TIMER was used to assess immune infiltration. Results We identified and validated the capability to predict the treatment response of CCT5 and ELF1 using our data and external validation datasets. The trends of survival differences of candidate genes in the GSE133057 dataset were similar to our cohort. High levels of CCT5 and ELF1 expression were associated with NCRT resistance and poor prognosis. Furthermore, the expression of CCT5 and ELF1 were also assessed in 117 LARC patients’ samples by the IHC method. Based on IHC results and Cox analysis, the risk score model with CCT5 and ELF1 was constructed and performed well. The risk score was an independent prognostic factor for progression-free survival and overall survival in LARC patients and was then used to build nomogram models. The underlying mechanisms of CCT5 and ELF1 were explored using gene set enrichment analysis. The underlying pathway including apoptosis, cell cycle, and other processes. CCT5 and ELF1 expressions were significantly correlated with immune cell infiltration. Conclusion CCT5 and ELF1 were determined as biomarkers for treatment response and prognosis in LARC patients. The risk score model and nomograms helped predict treatment response and survival outcomes for LARC patients undergoing NCRT.

Abstract This study investigates the correlations among perceived organizational support (POS), self-efficacy, knowledge transfer, and innovative behaviors of employees in the information service companies of Taiwan and mainland China using goal-oriented behavior and social identity theory. A structural model was built, and data were collected through a survey conducted in two rounds, two months apart, during the COVID-19 pandemic. The results indicate that POS significantly affects innovation behavior and self-efficacy in both regions, and self-efficacy plays a key mediating role in the research model. However, the relationship between POS and knowledge transfer was non-significant for mainland China. The study also reveals that the culture of knowledge sharing in the organization can promote employees’ innovation, knowledge sharing behavior, communication, and knowledge transfer. The findings imply that managers should provide organizational support to improve employees’ innovative minds and self-belief, particularly during the pandemic. This research fills a theoretical gap by applying Western theories in an Eastern context and broadening the generalization of the theory. It provides practical implications for improving the quality of human resources by suggesting that managers should provide organizational support to improve employees’ innovative minds and self-belief, particularly during a pandemic. Additionally, it contributes to the literature by examining how cross-cultural differences affect the relationships among POS, self-efficacy, knowledge transfer, and innovative behavior. This study also enriches the literature about employees in particular regions and their service innovation behaviors.

Abstract Background Diabetic nephropathy (DN) is a chronic kidney disease that develops in patients with diabetes mellitus. Cordycepin (CRD), a secondary metabolite produced by Cordyceps militaris, has a variety of bioactive properties. In this study, DN mice and high glucose (HG)-treated HK-2 were used to evaluate the diagnostic value of CRD. Methods Quantitative real-time PCR (qRT-PCR), western blotting, immunofluorescence analysis, and immunohistochemical staining were used to assess changes in mRNA and protein expression. Oxidative stress was evaluated by detecting the production of reactive oxygen species (ROS) and the activity of antioxidant enzymes. Cell apoptosis was detected by the TUNEL and flow cytometric methods. The interaction of miR-193b-5p and myeloid leukemia 1 (MCL-1) was examined by bioinformatics analysis and luciferase reporter assay. The protective effects of CRD on DN mice were evaluated by examining DN related biochemical indicators and renal histopathology. Results In response to HG, the level of miR-193b-5p was elevated, whilst the level of MCL-1 was downregulated, and CRD therapy reversed this behavior. MCL-1 was further identified to be miR-193b-5p target. CRD attenuated HG-induced cell damage, inflammation and abnormal energy metabolism. Mechanistic investigations on in vitro models confirmed that protective effect of CRD against HG challenge to HK-2 cells is mediated through the regulation of expression of miR-193b-5p/MCL-1 axis. By examining DN related biochemical markers and renal histopathology, the protective effects of CRD on DN mice was assessed. Conclusions In summary, CRD decreased oxidative stress and inflammation by increasing miR-193b-5p and inactivating downstream MCL-1 in DN, hinting the pivotal values of CRD and miR-193b-5p in the management of DN.

Accurate prediction of estuarine salinity can effectively mitigate the adverse effects of saltwater intrusion and help ensure the safety of water resources in estuarine regions. Presently, diverse data-driven models, mainly neural network models, have been employed to predict tidal estuarine salinity and obtained considerable achievements. Due to the nonlinear and nonstationary features of estuarine salinity sequences, this paper proposed a multi-factor salinity prediction model using an enhanced Long Short-Term Memory (LSTM) network. To improve prediction accuracy, input variables of the model were determined through Grey Relational Analysis (GRA) combined with estuarine dynamic analysis, and hyperparameters for the LSTM model were optimized using a multi-strategy Improved Sparrow Search Algorithm (ISSA). The proposed ISSA-LSTM model was applied to predict salinity at the Cangqian and Qibao stations in the Qiantang Estuary of China, based on measured data from 2011–2012. The model performance is evaluated by mean absolute error (MAE), mean absolute percentage error (MAPE), root mean square error (RMSE), and Nash-Sutcliffe efficiency (NSE). The results show that compared to other models including Back Propagation neural network (BP), Gate Recurrent Unit (GRU), and LSTM model, the new model has smaller errors and higher prediction accuracy, with NSE improved by 8–32% and other metrics (MAP, MAPE, RMSE) improved by 15–67%. Meanwhile, compared with LSTM optimized with the original SSA (SSA-LSTM), MAE, MAPE, and RMSE values of the new model decreased by 13–16%, 15–16%, and 11–13%, and NSE value increased by 5–6%, indicating that the ISSA has a better hyperparameter optimization ability than the original SSA. Thus, the model provides a practical solution for the rapid and precise prediction of estuarine salinity.

Objectives: To investigate the prognostic capacity of baseline 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography/computed tomography (PET/CT) metabolic parameters in extranodal natural killer/T-cell lymphoma (ENKTCL), and the influence of relative thresholds (RT) and absolute thresholds (AT) selection on prognostic capacity. Materials and methods: Metabolic tumor volume (MTV)-based parameters were defined using RTs (41 % or 25 % of maximum standardized uptake value [SUVmax]), ATs (SUV 2.5, 3.0, 4.0, or mean liver uptake) in 133 patients. Metabolic parameters were classified into avidity-related parameters (SUVmax, mean SUV [SUVmean], standard deviation of SUV [SUVsd]), volume-related parameters (RT-MTV), and avidity- and volume-related parameters (total lesion glycolysis [TLG] and AT-MTV). The prognostic capacity of the metabolic parameters and the effects of different threshold types (RT vs. AT) were evaluated. Results: All metabolic parameters were moderately associated with prognosis. However, the area under the receiver operating characteristic curve of MTV and TLG was slightly higher than that of avidity-related parameters for predicting 5-year progression-free survival (PFS) (0.614–0.705 vs. 0.563–0.609) and overall survival (OS) (0.670–0.748 vs. 0.562–0.593). Older adults who are frail are likely to be sedentary. Prior interventions to reduce sedentary time in older adults have not been effective as there is little research about the context of sedentary behaviour (posture, location, purpose, social environment). Moreover, there is limited evidence on feasible measures to assess context of sedentary behaviour in older adults. The aim of our study was to determine the feasibility of measuring context of sedentary behaviour in older adults with pre-frailty or frailty using a combination of objective and self-report measures. We defined "feasibility process" using recruitment (20 participants within two-months), retention (85%), and refusal (20%) rates and "feasibility resource" if the measures capture context and can be linked (e.g., sitting-kitchen-eating-alone) and are all participants willing to use the measures. Context was assessed using a wearable sensor to assess posture, a smart home monitoring system for location, and an electronic or hard-copy diary for purpose and social context over three days in winter and spring. We approached 80 potential individuals, and 58 expressed interest; of the 58 individuals, 37 did not enroll due to lack of interest or medical mistrust (64% refusal). We recruited 21 older adults (72±7.3 years, 13 females, 13 frail) within two months and experienced two dropouts due to medical mistrust or worsening health (90% retention). The wearable sensor, indoor positioning system, and electronic diary accurately captured one domain of context, but the hard copy was often not completed with enough detail, so it was challenging to link it to the other devices. Although not all participants were willing to use the wearable sensor, indoor positioning system, or electronic diary, we were able to triage the measures of those who did. The use of wearable sensors and electronic diaries may be a feasible method to assess context of sedentary behaviour, but more research is needed with device-based measures in diverse groups.

Detecting and accurately identifying malignant lung nodules in chest CT scans in a timely manner is crucial for effective lung cancer treatment. This study introduces a deep learning model featuring a multi-channel attention mechanism, specifically designed for the precise diagnosis of malignant lung nodules. To start, we standardized the voxel size of CT images and generated three RGB images of varying scales for each lung nodule, viewed from three different angles. Subsequently, we applied three attention submodels to extract class-specific characteristics from these RGB images. Finally, the nodule features were consolidated in the model's final layer to make the ultimate predictions. Through the utilization of an attention mechanism, we could dynamically pinpoint the exact location of lung nodules in the images without the need for prior segmentation. This proposed approach enhances the accuracy and efficiency of lung nodule classification. We evaluated and tested our model using a dataset of 1018 CT scans sourced from the Lung Image Database Consortium and Image Database Resource Initiative (LIDC-IDRI). The experimental results demonstrate that our model achieved a lung nodule classification accuracy of 90.11 %, with an area under the receiver operator curve (AUC) score of 95.66 %. Impressively, our method achieved this high level of performance while utilizing only 29.09 % of the time needed by the mainstream model.

The rare and endangered plant species Abies fanjingshanensis, which has a limited habitat, a limited distribution area, and a small population, is under severe threat, particularly due to poor leaf health. The plant endophytic microbiome is an integral part of the host, and increasing evidence indicates that the interplay between plants and endophytic microbes is a key determinant for sustaining plant fitness. However, little attention has been given to the differences in the endophytic microbial community structure, network complexity, and assembly processes in leaves with different leaf health statuses. Here, we investigated the endophytic bacterial and fungal communities in healthy leaves (HLs) and non-healthy leaves (NLs) of A. fanjingshanensis using 16S rRNA gene and internal transcribed spacer sequencing and evaluated how leaf health status affects the co-occurrence patterns and assembly processes of leaf endophytic microbial communities based on the co-occurrence networks, the niche breadth index, a neutral community model, and C-score metrics. HLs had significantly greater endophytic bacterial and fungal abundance and diversity than NLs, and there were significant differences in the endophytic microbial communities between HLs and NLs. Leaf-health-sensitive endophytic microbes were taxonomically diverse and were mainly grouped into four ecological clusters according to leaf health status. Poor leaf health reduced the complexity of the endophytic bacterial and fungal community networks, as reflected by a decrease in network nodes and edges and an increase in degrees of betweenness and assortativity. The stochastic processes of endophytic bacterial and fungal community assembly were weakened, and the deterministic processes became more important with declining leaf health. These results have important implications for understanding the ecological patterns and interactions of endophytic microbial communities in response to changing leaf health status and provide opportunities for further studies on exploiting plant endophytic microbes to conserve this endangered Abies species.

The landfalls of the tropical cyclone (TC) along the coast of China have caused huge economic damages. There are approximately nine TC landfalls in China every year. It will be beneficial if the landfall frequency can be predicted in advance. Inspired by the study of Sparks and Toumi (Commun Earth Environ, 30-1-2020), six datasets, including four ocean reanalyses and two object analyses from 1993 to 2019, are employed to study the consistency in the relationship between the thermocline temperature anomalies at different longitudes and the frequency of TC landfalls along the coastal areas of China (South China, East China, and the whole of China). The thermocline temperature anomalies at different longitudes are tested in order to confirm our hypothesis that the eastward and westward transports of ocean heat from the warm pool are the causes of the significant correlations. The results show some significant correlations at various longitudes, and the temperature anomalies can predict the TC landfall frequency for several months or longer. Further study also shows the close relationship between the ocean heat transport and the sea surface temperature anomalies at the genesis locations of TC landfalls. The locations of the western Pacific subtropical high (WPSH) during high-frequency TC landfall years also show favorable spatial patterns to the TC landfall in South China and East China, respectively. In years with a high TC frequency in South China, the westward displacement of the WPSH ridge steers TC toward South China, while during high-frequency TC landfall years in East China, WPSH is located further north, and the westward extension of the ridge is in close proximity to the East China Sea.

ABSTRACT Microsporidia are obligate intracellular single-cell eukaryotic parasites that can infect almost all kinds of animals and cause microsporidiosis. However, there are a few achievements of microsporidiosis treatments. Nosema bombycis could cause a microsporidiosis condition, pébrine, in the silkworm. Treatment and prevention of pébrine are still the research hotspots in sericulture. In this study, N. bombycis was treated with K2CO3, the spore walls were removed by centrifugation, and then, the alkali-soluble germination proteins were used as antigens to develop monoclonal antibodies (mAbs). Three mAbs were successfully screened and one mAb named G9 showed the highest titer among these monoclonal antibodies after enzyme-linked-immunosorbent serologic assay (ELISA). Mass spectrometry analysis confirmed that the mAb G9 could recognize the N. bombycis spore wall protein 1. Furthermore, the heavy chain and light chain sequences of the G9 monoclonal antibody were cloned, respectively. The vectors that expressing the intact antibodies and the single-chain variable fragments (scFvs) of G9 were constructed, and then, these vectors were used to develop the transgenic silkworm cell lines or transgenic silkworms. The inhibitory effects against N. bombycis were evaluated by the count of microsporidia and qPCR. The scFvs showed better effect on blocking the proliferation of N. bombycis than the intact antibody, and the scFv without the secretory signal peptide was more effective than that with signal peptide. Our study has provided novel strategies for microsporidiosis control and the essential groundwork for the future development of N. bombycis-resistant transgenic silkworms. IMPORTANCE There are a few reports on the resistance of microsporidia, including Nosema bombycis. Here, the alkali-soluble germination proteins of N. bombycis were used as immunogens to prepare a monoclonal antibody, and its single-chain variable fragments effectively blocked microsporidia infection. Our study has provided novel strategies for microsporidiosis control and demonstrated a useful method for the potential treatment of other microsporidia diseases.

Triterpenoid saponins from Stauntonia chinensis have been proven to be a potential candidate for inflammatory pain relief. Our pharmacological studies confirmed that the analgesic role of triterpenoid saponins from S. chinensis occurred via a particular increase in the inhibitory synaptic response in the cortex at resting state and the modulation of the capsaicin receptor. However, its analgesic active components and whether its analgesic mechanism are limited to this are not clear. In order to further determine its active components and analgesic mechanism, we used the patch clamp technique to screen the chemical components that can increase inhibitory synaptic response and antagonize transient receptor potential vanilloid 1, and then used in vivo animal experiments to evaluate the analgesic effect of the selected chemical components. Finally, we used the patch clamp technique and molecular biology technology to study the analgesic mechanism of the selected chemical components. The results showed that triterpenoid saponins from S. chinensis could enhance the inhibitory synaptic effect and antagonize the transient receptor potential vanilloid 1 through different chemical components, and produce central and peripheral analgesic effects. The above results fully reflect that “traditional Chinese medicine has multi-component, multi-target, and multi-channel synergistic regulation”.

The unit width suspended sediment transport rate is the basis for calculating river sediment flux, which provides an important measure of river hydrology and morpho-dynamics. By integrating the vertical distribution of the suspended sediment concentration and logarithmic flow velocity, the formula for unit width suspended sediment transport rate is presented. Based on this formula, the calculation of the near-bed relative transport rate missing in field measurement is given. Moreover, a new stratified water sampling device is invented for suspended sediment transport rate measurement by the six-point method at Mengsheng hydrological station in the Lancang River basin. Results of 24 sets of measurements show that the near-bed relative sediment transport rate ranges from 3.4 to 10.7% and is significantly positively correlated with the suspension index. The unit width suspended sediment transport rate measured by the traditional depth integrating method is on average 15.0% smaller than that calculated by the theoretical formula, which can be a reference for correcting the sediment transport rate based on the traditional method. This work provides a new device and a new method for the measurement and calculation of the unit width transport rate of suspended sediment, which has theoretical and practical significance. Abstracts Background Preclinical studies suggest that glucocorticoids (GCs) promote the proliferation and development of colorectal cancer. Because GCs are broadly prescribed for treatment-related adverse events in patients with locally advanced rectal cancer (LARC) receiving neoadjuvant chemoradiotherapy (NCRT), it’s essential to assess the effect of GCs on clinical outcomes. Methods LARC cases treated with NCRT followed by surgery were assessed retrospectively. Evaluation of the relationship between GCs use (GCs vs. non-GCs) and neoadjuvant rectal (NAR) score (as a three-level categorical dependent variable) was performed using multivariable multinomial logistic regression (MLR). We also examined the relationship between the accumulated dose of GCs and NAR using multivariate MLR. Survival analysis of disease-free survival (DFS) and overall survival (OS) was performed using the Kaplan–Meier method. Multivariate Cox regression was used to assess confounding factors that could influence OS and DFS. Results This retrospective cohort study included 790 patients with newly diagnosed non-metastatic LARC (T3-4/N + M0) who received NCRT followed by surgery between January 2012 and April 2017. The end of the follow-up period was May 11, 2022. Among the 790 patients with LARC, 342 (43.2%) received GCs treatment and 448 (56.8%) did not during the NCRT-to-surgery period. GCs medication was significantly different between mid-NAR (8–16) and low-NAR ( 16) and low-NAR (0.563; 0.352–0.900; 0.016). Patients exposed to GCs, had a decreased 5-year OS (GCs vs. non-GCs = 80.01% (95% CI, 75.87%–84.37%) vs. 85.30% (82.06%–88.67%), P = 0.023) and poorer 5-year DFS (73.99% (69.45%–78.82%) vs. 78.7% (75.14%–82.78%), P = 0.045). The accumulated dose of GCs was an independent risk factor for OS (hazard ratio [HR], 1.007 [1.001–1.014], 0.036) and DFS (1.010 [1.004–1.017], 0.001). Conclusions and relevance Our study revealed that GCs were associated with reduced efficacy of NCRT and worse clinical outcomes in patients with LARC during the NCRT-to-surgery period.

2-Phenylethanol (2-PE) and indole-3-acetic acid (IAA) are important secondary metabolites produced by microorganisms, and their production are closely linked to the growth state of microorganisms and environmental factors. Enterobacter CGMCC 5087 can produce both 2-PE and IAA depending on α-ketoacid decarboxylase KDC4427. This study aimed to investigate the effects of different environment factors including osmotic pressure, temperature, and pH on the synthesis of 2-PE and IAA in Enterobacter sp. CGMCC 5087. The bacteria exhibited an enhanced capacity for 2-PE synthesis while not affecting IAA synthesis under 5% NaCl and pH 4.5 stress conditions. In an environment with pH 9.5, the synthesis capacity of 2-PE remained unchanged while the synthesis capacity of IAA decreased. The synthesis ability of 2-PE was enhanced with an increase in temperature within the range of 25 °C to 37 °C, while the synthesis capacity of IAA was not affected significantly. Additionally, the expression of KDC4427 varied under stress conditions. Under 5% NaCl stress and decreased temperature, expression of the KDC4427 gene was increased. However, altering pH did not result in significant differences in gene expression levels, while elevated temperature caused a decrease in gene expression. Furthermore, molecular docking and molecular dynamics simulations suggested that these conditions may induce fluctuation in the geometry shape of binding cavity, binding energy, and especially the dαC-C- value, which played key roles in affecting the enzyme activity. These results provide insights and strategies for the synthesis of metabolic products 2-PE and IAA in bacterial fermentation, even under unfavorable conditions.

The Jianghuai region is the area between the Yangtze River and the Huai River in China and is a densely populated agriculture region therefore, the economics and human activity there are significantly affected by the precipitation changes, particularly during the summer when extreme storms and droughts normally occur. It will be helpful if the summer precipitation changes can be predicted. The monthly ERA5 atmospheric reanalysis data from 1978 to 2021 are used in this study to investigate the relationship between the ridge latitudinal location of the western Pacific subtropical high (WPSH) and the precipitation in July over the Jianghuai region. The results show that the WPSH ridge location has an important impact on the amount and spatial distribution of the precipitation in this region. When the ridge was northward, an anomalous anticyclonic circulation will appear over the western Pacific, leading to the weakening of the summer monsoon and the reduction of moisture transport from the Indian Ocean, therefore decreasing precipitation in the Jianghuai region, while the situation is opposite when the ridge was southward. The Niño 3.4 index in March and the India–Burma trough intensity index in June have significant correlations with the July WPSH ridge location, and both can be used as precursors to predict the WPSH ridge location and, therefore, the precipitation in this region.

BackgroundThe effectiveness of N-acetylcysteine (NAC) in treating contrast-induced nephropathy (CIN) has been the subject of conflicting meta-analyses, but the strength of the evidence for these correlations between NAC use and CIN has not been measured overall.ObjectiveTo evaluate the data from randomized clinical studies (RCTs) that examined the relationships between NAC use and CIN in meta-analyses.MethodsBetween the creation of the database and April 2023, searches were made in PubMed, Cochrane Library, EMBASE, and Web of Science. N-acetylcysteine, contrast-induced nephropathy, or contrast-induced renal disease were among the search keywords used, along with terms including systematic review and meta-analysis. The Assessment of Multiple Systematic Reviews, version 2, which assigned grades of extremely low, low, moderate, or high quality to each meta-analysis’s scientific quality, was used to evaluate each meta-analysis. The confidence of the evidence in meta-analyses of RCTs was evaluated using the Grading of Recommendation, Assessment, Development and Evaluations method, with evidence being rated as very low, low, moderate, or high.ResultsIn total, 493 records were screened; of those, 46 full-text articles were assessed for eligibility, and 12 articles were selected for evidence synthesis as a result of the screening process. Abstract Purpose Given the therapeutic toxicity of concurrent chemoradiotherapy, the dose of chemotherapy is always limited. Hence, the question of how to administer adequate chemotherapy to synchronize stereotactic body radiation therapy (SBRT) treatment strategy for maximizing the benefits of neoadjuvant therapy to improve prognosis is a challenging and debatable issue. Methods We summarized the major clinical trials and preclinical studies of split‐course CRT and split‐course SBRT and explored the feasibility and theoretical advantages of implementing split‐course SBRT and systemic therapy simultaneously. Results Split‐course radiotherapy is a strategy mainly proposed with the aim of further reducing the toxicity of radiotherapy and improving the patient's treatment tolerance, with added features of the extended interval between the fractions instead of daily irradiation. In the past, split‐course radiotherapy was not a recommended treatment strategy in terms of local tumor control rates, late response rates, and survival without significant improvement relative to continuous radiotherapy. Split‐course SBRT has different biological effects compared to split‐course conventional radiotherapy (CRT). The current findings suggest that the split‐course SBRT model has good treatment tolerance and facilitates the implementation of adaptive radiotherapy. Conclusion This treatment strategy facilitates the upgrading of radiotherapy doses and the combination with systemic therapy, especially with vascular targeting agents and immunotherapy, thus avoiding the disadvantages of split‐course CRT, such as reduced local control rate, increased treatment time and economic cost, and has the potential to achieve better tumor suppression.

Abstract Background CARs are engineered receptors comprising an immunoglobulin single‐chain variable fragment (scFv) that identifies and binds to the target antigen, a transmembrane domain, and an intracellular T‐cell signaling domain. CD19 is a B lineage‐specific transmembrane glycoprotein and is expressed in more than 95% of B‐cell malignancies. Streptavidin (SA) is a homo‐tetrameric protein derived from Streptomyces avidinii, which can bind four biotin molecules with an extremely high affinity at a Kd value of 10‐15 M. Aims The aim of the study is to generate a novel soluble multimeric fusion protein, sCD19‐streptavidin (sCD19‐SA) for functional detection and selective expansion of CD19‐targeted CAR‐T cells. Methods The fusion proteins CD19‐SA was expressed in CHO cells and purified by use of Ni‐nitrilotriacetic acid agarose beads. Results A novel fusion protein (sCD19‐SA) was generated, consisting of the extracellular domain of human CD19 and the core region of SA, and could be used to functionally detect CD19‐targeted CAR‐T cells. Furthermore, this protein was demonstrated to form multimers to activate CAR‐T cells to induce their selective expansion. Importantly, sCD19‐SA‐stimulated CD19‐targeted CAR‐T cells could improve antitumor effects in vivo. Conclusions Our study has highlighted the potential of utilizing antigen‐SA fusion proteins such as sCD19‐SA for CAR‐T therapy for the functional detection of CAR expression and selective expansion of CAR‐T cells.

Abstract Background Distant metastasis has been the main failure pattern for locoregionally advanced rectal cancer (LARC) patients, and intensified neoadjuvant chemotherapy has become a popular research topic. The present study aimed to compare the survival outcomes, acute toxicities and surgical complications in LARC patients who received preoperative chemoradiotherapy with triweekly oxaliplatin and capecitabine (triweekly XELOX) or capecitabine. Methods: Between 2007 and 2017, patients with clinically staged II-III rectal cancer who were treated with preoperative chemoradiotherapy using either triweekly XELOX (oxaliplatin 130 mg/m2 plus capecitabine 825 mg/m2) or capecitabine were included. Variables potentially influencing chemotherapy treatment selection were used to generate propensity scores (PS). The association between chemotherapy regimens and survival endpoints, including distant metastasis-free survival (DMFS), overall survival (OS) and disease-free survival (DFS), were evaluated and adjusted with PS. The acute toxicities and surgical complications were also compared. Results A total of 810 patients were included in the analysis; 277 (34.2%) patients received triweekly XELOX, and 533 (65.8%) received capecitabine. The pathological complete response (pCR) rates were 20.2 and 19.9% (P = 0.912) for the groups treated with triweekly XELOX and capecitabine, respectively. The 5-year DMFS, OS and DFS with triweekly XELOX versus capecitabine were 75.6% vs. 77.6% (P = 0.555), 79.2% vs. 83.3% (P = 0.101), and 69.9% vs. 73.7% (P = 0.283), respectively. Triweekly XELOX was not associated with an increased risk of severe toxicity during chemoradiotherapy, but it increased the risk of postoperative complications compared to capecitabine. After PS adjustment, the differences between the two groups remained insignificant in pCR rate, survival outcomes, and acute toxicities, and the difference in surgical complications disappeared. Conclusions Triweekly XELOX or capecitabine concurrent with neoadjuvant radiotherapy leads to similar long-term survival outcomes, acute toxicities and surgical complications in LARC patients.

REST corepressors (RCORs) are the core component of the LSD1/CoREST/HDACs transcriptional repressor complex, which have been revealed differently expressed in various cancers, but the therapeutic and prognostic mechanisms in cancer are still poorly understood. In this study, we analyzed expression, prognostic value, molecular subtypes, genetic alteration, immunotherapy response and drug sensitivity of RCORs in pan-cancer. Clinical correlation, stemness index, immune infiltration and regulatory networks of RCORs in hepatocellular carcinoma (HCC) were detected through TCGA and GSCA database. In-vitro experiments were conducted to explore the role of RCOR1 in HCC cells. The expression of RCORs varied among different cancers, and have prognostic values in several cancers. Cancer subtypes were categorized according to the expression of RCORs with clinical information. RCORs were significantly correlated with immunotherapy response, MSI, drug sensitivity and genetic alteration in pan-cancer. In HCC, RCORs were considered as potential predictor of stemness and also had association with immune infiltration. The ceRNA-TF-kinase regulatory networks of RCORs were constructed. Besides, RCOR1 acts as an oncogene in HCC and promotes the proliferation of HCC cells by inhibiting cell cycle arrest and cell apoptosis. Taken together, our study revealed the potential molecular mechanisms of RCORs in pan-cancer, offering a benchmark for disease-related research.

Spacer fabric is often used in cushions, footwear, filter material, and other products because of its high air permeability and three-dimensional (3D) structure. However, for practical applications, it is typically combined with other materials. We investigated the thermal properties and water vapor permeability of assembly-combined thicker spacer (16 mm) and various cotton nonwoven fabrics. The assembly-combined lightest nonwoven (30 g/m2) and spacer fabric exhibited the highest thermal resistance, increasing by 66.62% compared with the bare spacer fabric; however, no significant difference was observed when combined with the heaviest woven fabric (60 g/m2). Furthermore, the fabric arrangement during assembly could affect the heat and moisture-transfer efficiencies. The assembly-combined lightest nonwoven (30 g/m2) and spacer fabric under the upward test condition exhibited the highest thermal resistance, highest Clo, lowest heat transfer coefficient, highest insulation ratio, and lowest evaporation resistance among all assemblies. Higher thermal resistance and lower evaporative resistance could benefit physiological comfort. However, the assembly-combined heaviest nonwoven and spacer fabric under the downward test condition exhibited similar thermal resistance to the spacer fabric and the highest evaporation resistance. The asymmetric heat- and moisture-transfer properties of a porous assembly can contribute toward developing new materials for applications in other engineering fields.

Temporal change of beta diversity provides a better understanding of the extent and consequences of species composition in forest communities with the ongoing global climate change. However, relatively little is known about temporal beta diversity changes across vertical stratification in the forest. In this study, we divided more than 5,000 tree individuals from a mixed broadleaf-conifer forest into four vertical strata (i.e., shrub, subcanopy, lower canopy, and upper canopy) to quantify how vertical stratification drives the temporal change of taxonomic and phylogenetic beta diversity. We found that taxonomic beta diversity significantly decreased while phylogenetic beta diversity showed an insignificant increase after 5 years. When considering vertical stratification, taxonomic beta diversity in the subcanopy, lower canopy, and upper canopy significantly changed with inconsistent directions, but phylogenetic beta diversity in the shrub significantly increased. Moreover, the significant decrease in taxonomic beta diversity is mainly driven by changes in species composition in shrub and subcanopy stratification (with 85.89% contributions). The changes in phylogenetic beta diversity are driven by shifts in the shrub and upper canopy (with 96.02% contributions). Our study suggests that taking community vertical stratification into consideration contributes to a better understanding of temporal beta diversity in forest communities.

Abstract Background and purpose We aimed to explore the necessity of the external iliac lymph nodes (EIN) along with inguinal nodes (IN) region in clinical target volume (CTV) for rectal carcinomas covering the anal canal region. Materials and methods This research premise enrolled 399 patients who had primary low rectal cancer detected below the peritoneal reflection via magnetic resonance imaging (MRI) and were treated with neoadjuvant radiotherapy (NRT), without elective EIN along with IN irradiation. We stratified the patients into two groups based on whether the lower edge of the rectal tumor extended to the anal canal (P group, n = 109) or not (Rb group, n = 290). Comparison of overall survival (OS), locoregional recurrence-free survival (LRFS), disease-free survival (DFS), as well as distant metastasis-free survival (DMFS) were performed via inverse probability of treatment weighting (IPTW) along with multivariable analyses. We compared the EIN and IN failure rates between the two groups via the Fisher and Gray’s test. Results P group showed a similar adjusted proportion along with five-year cumulative rate of EIN failure compared with the Rb group. The adjusted proportion and five-year cumulative rate of IN failure in the P group was higher in comparison to the Rb group. There were no remarkable differences in the adjusted five-year OS, DFS, DMFS or LRFS between the two groups. Anal canal involvement (ACI) exhibited no effect on OS, LRFS, DFS, or DMFS. Conclusions During NRT for rectal cancer with ACI, it may be possible to exclude the EIN and IN from the CTV.

Introduction Short-course radiotherapy (SCRT) with systemic therapy has the potential to further improve the long-term efficacy in patients with locally advanced rectal cancer (LARC). To maximise the benefits of neoadjuvant therapy for improved prognosis, it is important to determine the optimal mix of chemotherapy, immunotherapy and SCRT.Methods and analysis Fifty treatment-naïve patients with operable LARC (T3–4 and/or N+) will be recruited. Patients will be synchronously treated with capecitabine plus oxaliplatin (CAPOX) chemotherapy, tislelizumab and preoperative split-course hypofraction radiotherapy (SCHR) (5×7 Gy) before surgery. Chemotherapy for CAPOX starts on day 1 of every 21-day cycle: on day 1, oxaliplatin 130 mg/m2 will be injected intravenously. On days 1–14, capecitabine 1000 mg/m2 was ingested two times a day. Simultaneously, tocilizumab 200 mg will be given intravenously on the first day of every 21-day cycle. A single 7 Gy SCHR treatment (day 7 of each 21-day cycle) will be delivered five times during the seventh day of treatment. The primary endpoint will be pathological complete response. The secondary outcomes will be the 3-year disease-free survival, local recurrence rate, overall survival, sphincter-sparing surgery rate, R0 resection rate, predictive biomarkers and quality of life.Ethics and dissemination The study protocol was approved by the Ethics Committee of Xiehe Affiliated Hospital of Fujian Medical University (XAHFMU) (No. 2021YF025-01). Results from our study will be disseminated in international peer-reviewed journals. All study procedures were developed in order to assure data protection and confidentiality.Trial registration number NCT05176964.

ObjectiveThe aim of this bicentric retrospective study was to assess the prognostic value of 18F-FDG PET/CT in patients with esophageal high-grade neuroendocrine carcinoma (NECs).MethodsFrom the database of two centers, 28 patients affected by esophageal high-grade NECs who underwent 18F-FDG PET/CT before treatment were retrospectively reviewed. Metabolic parameters (SUVmax, SUVmean, tumor-to-blood-pool SUV ratio (TBR), tumor-to-liver SUV ratio (TLR), metabolic tumor volume (MTV), total lesion glycolysis (TLG)) of the primary tumor were measured. Univariate and multivariate analyses were performed for progression-free survival (PFS) and overall survival (OS).ResultsAfter a median follow-up period of 22 months, disease progression occurred in 11 (39.3%) patients, and death occurred in 8 (28.6%) patients. The median PFS was 34 months, and the median OS was not reached. Univariate analyses revealed that among metabolic parameters, only MTV and TLG were significant prognostic factors, while among clinical variables, only distant metastasis was a significant factor for both PFS and OS (P< 0.05). On multivariate analyses, MTV and TLG were independent prognostic factors for both PFS and OS (P< 0.05).ConclusionsIn patients with esophageal high-grade NEC, MTV and TLG measured on pretreatment 18F-FDG PET/CT are independently prognostic factors for predicting PFS and OS, and might be used as quantitative prognostic imaging biomarkers.

PurposeTo assess the utility of skeletal standardized uptake values (SUVs) obtained using quantitative single-photon emission computed tomography/computed tomography (SPECT/CT) in differentiating bone metastases from benign lesions, particularly in patients with lung adenocarcinoma.MethodsPatients with lung adenocarcinoma who had undergone whole-body Tc-99m methyl-diphosphonate (99mTc-MDP) bone scans and received late phase SPECT/CT were retrospectively analyzed in this study. The maximum SUV (SUVmax); Hounsfield units (HUs); and volumes of osteoblastic, osteolytic, mixed, CT-negative metastatic and benign bone lesions, and normal vertebrae were compared. Receiver operating characteristic curves were used to determine the optimal cutoff SUVmax between metastatic and benign lesions as well as the cutoff SUVmax between CT-negative metastatic lesions and normal vertebrae. The linear correlation between SUVmax and HUs of metastatic lesions as well as that between SUVmax and the volume of all bone lesions were investigated.ResultsA total of 252 bone metastatic lesions, 140 benign bone lesions, and 199 normal vertebrae from 115 patients with lung adenocarcinoma were studied (48 males, 67 females, median age: 59 years). Metastatic lesions had a significantly higher SUVmax (23.85 ± 14.34) than benign lesions (9.67 ± 7.47) and normal vertebrae (6.19 ± 1.46; P < 0.0001). The SPECT/CT hotspot of patients with bone metastases could be distinguished from benign lesions using a cutoff SUVmax of 11.10, with a sensitivity of 87.70% and a specificity of 80.71%. The SUVmax of osteoblastic (29.16 ± 16.63) and mixed (26.62 ± 14.97) lesions was significantly greater than that of osteolytic (15.79 ± 5.57) and CT-negative (16.51 ± 6.93) lesions (P < 0.0001, P = 0.0003, and 0.002). SUVmax at the cutoff value of 8.135 could distinguish CT-negative bone metastases from normal vertebrae, with a sensitivity of 100.00% and a specificity of 91.96%. SUVmax showed a weak positive linear correlation with HUs in all bone metastases and the volume of all bone lesions.ConclusionSUVmax of quantitative SPECT/CT is a useful index for distinguishing benign bone lesions from bone metastases in patients with lung adenocarcinoma, particularly in the diagnosis of CT-negative bone metastases, but other factors that may affect SUVmax should be considered.

Shared bikes can help cities achieve carbon neutrality goals. Cleaning and disinfection are vital procedures of the maintenance of shared bikes, especially during the COVID-19 pandemic because shared bikes could be a transmission intermediary of viruses. This study proposes an optimization model of the cleaning and disinfection scheme of the dockless shared bikes. The disinfection is assumed to be performed at night, when the usage is lowest. By regarding the disinfection staff as traveling salesmen, the model is formulated as an extension of the Multidepot Multiple Traveling Salesman Problem (MDMTSP). The objective function is to minimize the total cost; which consists of the cost associated with the working time and per-capita cost of the disinfection staff. A heuristic algorithm combining k-means clustering and genetic algorithm (K-GA) is adopted to find the lower bound solution. Then, the K-GA-adjustment algorithm has been adopted to find the solutions that satisfy the constraints. To reduce the computing time needed, an approximate function for the lower bound of the optimal number of disinfection staff is obtained by constructing a Continuous Approximation (CA) model. A case study based on real location data of shared bikes in Chengdu, China, is performed to show how the maintenance department could adopt the optimization framework to design an efficient scheme to clean and disinfect the shared bikes.

Abstract Warp knitted mesh fabric was usually applied to sportswear due to good air transmission, but without multilayer structure and one-way transport property. In order to solve this problem, the miss-lapping structure was applied to examine the possibility to fabricate multilayer and improve water transport in warp knitting structure. Besides, the effect of thread type and warp density on comfort properties were also exploited to enhance the moisture management. The moisture management test, water vapor permeability and air permeability were examined. Long float at the back side in structure I formed by miss-lapping could improve liquid transport and air permeability, but slightly reduce water vapor permeability. With proper density, there existed the optimal one-way transport capacity and overall moisture management. Warp density in 20 cpc was an optimal parameter of knitting process. Taking advantage of miss-lapping, sample 5 where polypropylene was partly threaded on GB1 provided best moisture management, water vapor permeability and air permeability.

The remora optimization algorithm (ROA) is a newly proposed metaheuristic algorithm for solving global optimization problems. In ROA, each search agent searches new space according to the position of host, which makes the algorithm suffer from the drawbacks of slow convergence rate, poor solution accuracy, and local optima for some optimization problems. To tackle these problems, this study proposes an improved ROA (IROA) by introducing a new mechanism named autonomous foraging mechanism (AFM), which is inspired from the fact that remora can also find food on its own. In AFM, each remora has a small chance to search food randomly or according to the current food position. Thus the AFM can effectively expand the search space and improve the accuracy of the solution. To substantiate the efficacy of the proposed IROA, twenty-three classical benchmark functions and ten latest CEC 2021 test functions with various types and dimensions were employed to test the performance of IROA. Compared with seven metaheuristic and six modified algorithms, the results of test functions show that the IROA has superior performance in solving these optimization problems. Moreover, the results of five representative engineering design optimization problems also reveal that the IROA has the capability to obtain the optimal results for real-world optimization problems. To sum up, these test results confirm the effectiveness of the proposed mechanism.

Both face and back sides of cotton weft-knitted fabrics in five knitting structures (plain, rib, interlock, single pique, and double pique) were investigated based on modified water drop test (methods I and II) in terms of water diffusion time. The water drop height was raised from 1 cm to 20 cm and water drop volume was increase from 0.1 mL to 0.5 mL. The water diffusion time greatly decreased with the variation in water drop height from 0 to 15 cm. Water diffusion time was also influenced by water volume at 0.1–0.2 mL for the different structures and surfaces. It was found that the structures behaved differently, and two surfaces of fabric were capable of asymmetric water absorption property. The face side of single pique and the back side double pique had better water diffusion property. Besides, vertical wicking rate and the drying rate (methods I and II) were also conducted to evaluate the different structures. Although the plain structure was widely used for inner garments in the market, the interlock fabric was recommended in nonsweating conditions due to its poor water spreading and better drying ability, while the single-pique fabric was recommended in heavy sweating conditions due to its better water absorption ratio and faster drying rate.

Arithmetic optimization algorithm (AOA) is a newly proposed meta-heuristic method which is inspired by the arithmetic operators in mathematics. However, the AOA has the weaknesses of insufficient exploration capability and is likely to fall into local optima. To improve the searching quality of original AOA, this paper presents an improved AOA (IAOA) integrated with proposed forced switching mechanism (FSM). The enhanced algorithm uses the random math optimizer probability (RMOP) to increase the population diversity for better global search. And then the forced switching mechanism is introduced into the AOA to help the search agents jump out of the local optima. When the search agents cannot find better positions within a certain number of iterations, the proposed FSM will make them conduct the exploratory behavior. Thus the cases of being trapped into local optima can be avoided effectively. The proposed IAOA is extensively tested by twenty-three classical benchmark functions and ten CEC2020 test functions and compared with the AOA and other well-known optimization algorithms. The experimental results show that the proposed algorithm is superior to other comparative algorithms on most of the test functions. Furthermore, the test results of two training problems of multi-layer perceptron (MLP) and three classical engineering design problems also indicate that the proposed IAOA is highly effective when dealing with real-world problems.

This study was undertaken to identify oat (Avena sativa L.) varieties optimal for cultivation in the Jiuquan region, China, in 2021. A selection of 27 domestic and international oat varieties were analyzed, considering ten key agronomic traits, including plant height, stem diameter, spike length, leaf width, and yield. Employing methods such as cluster analysis, principal component analysis, and grey correlation degree, a comprehensive evaluation was conducted. The principal component analysis distilled the ten indicators to three core components. The most influential factors in the first principal component were plant height, ear length, and hay yield, while leaf length and leaf area index were the highest contributors to the second component. The stem-to-leaf ratio emerged as the principal indicator in the third component. The cluster analysis resulted in the classification of the 27 oat varieties into 3 categories. Following a comprehensive evaluation through the grey correlation degree and principal component analysis methodologies, we found that the oat varieties Sweety 1, Fuyan 1, Dingyan 2, Baler, Quebec, and Longyan 2 received the highest scores. These varieties, hence, appear to be the most suitable for cultivation and promotion in the Jiuquan region. This study thus provides invaluable insights into oat cultivation practices, offering guidance for farmers, agricultural policymakers, and future research in the field.

ObjectivesThe motion state of autonomous underwater vehicle (AUV) is impacted by different densities of water, so it is necessary to recalculate the balance and control parameters. However, the use of a variable buoyancy device can add a more effective self-regulation capability to the AUV, making it suitable for operation in a wider range of scenarios.MethodsThis paper proposes the design of a variable buoyancy device for an AUV. Through adopting an oil bladder that can intake/discharge oil and transfer it quantitatively in a dual manner, the device can change the volume of the external oil bladder and regulate the AUV's buoyancy status.ResultsThe results of the tank and lake tests show that the designed device can help the AUV to achieve the functions of fixed-depth hovering and diving during navigation, and complete a 90-hour mission of low-power-consumption parking for in-situ observation after soft-landing on the lakebed at a depth of 51 m.ConclusionsThe results prove the validity of the variable buoyancy device and lay the groundwork for enhancing the operation effectiveness of AUVs, as well as extending their application scope.

Radiotherapy has already been developed as the standard of care for patients with nasopharyngeal carcinoma (NPC), and precision staging, target volume delineation, prognosis prediction, and post-treatment surveillance are essential in the management of NPC. Positron emission tomography/computed tomography (PET/CT) is increasingly recognized as an imaging modality to guide precision radiotherapy in these areas. The feasibility and efficacy of 18F-FDG PET/CT have been confirmed in tumor diagnosis, treatment planning, prognosis, surveillance, and assessment. Coupled with the capability of revealing tumor metabolic information, 18F-FDG PET/CT is more accurate in identifying primary lesions and metastases of NPC than other conventional imaging methods including CT and MRI and shows the independently diagnostic and prognostic value for radiotherapy. However, 18F-FDG has limitations due to its physiological distribution in brain tissue and increasing uptake in post-radiation inflammation. Novel PET radiotracers including FAPI, NaF, CHO, and FLT are explored as alternatives with potential superiority for radiotherapy in NPC. In this review, we summarized the evolving role of PET/CT in the management of radiotherapy in NPC patients, aiming to facilitate precision radiotherapy from a molecular imaging aspect.

This paper introduces an improved hybrid Aquila Optimizer (AO) and Harris Hawks Optimization (HHO) algorithm, namely IHAOHHO, to enhance the searching performance for global optimization problems. In the IHAOHHO, valuable exploration and exploitation capabilities of AO and HHO are retained firstly, and then representative-based hunting (RH) and opposition-based learning (OBL) strategies are added in the exploration and exploitation phases to effectively improve the diversity of search space and local optima avoidance capability of the algorithm, respectively. To verify the optimization performance and the practicability, the proposed algorithm is comprehensively analyzed on standard and CEC2017 benchmark functions and three engineering design problems. The experimental results show that the proposed IHAOHHO has more superior global search performance and faster convergence speed compared to the basic AO and HHO and selected state-of-the-art meta-heuristic algorithms.

Colletotrichum siamense infected the petiole and crown of strawberry (Fragaria × ananassa), and this anthracnose threatens strawberry production heavily in Hubei province. Characteristic the variation of transcript caused by C. siamense is important for anthracnose control of strawberry. In this study, RNA-seq analysis was used to monitor the strawberry transcripts in response of C. siamense at 0- and 6-days post-inoculation based on lesion measurement and hypha observation. The number of clean reads achieved for each sample was ranged from 5.68 to 8.12 Gb. The 0DPM vs 0DPI group had seven significantly differentially expressed transcripts, whereas the 6DPM vs 6DPI group exhibited 12,097 significantly differentially expressed transcripts. The differentially expressed transcripts of 6DPM vs 6DPI group were enriched in ‘metabolic process’ and ‘biological process’ in the biological process category and ‘oxidoreductase activity’ in the molecular function category by GO analysis. While ‘Starch and sucrose metabolism’, ‘Plant hormone signal transduction’ and ‘Phenylpropanoid biosynthesis’ pathways were significantly enriched by KEGG analysis. The expression of MYC2 in JA, the ETR in Eth and, the SnRK2 and ABF in ABA signal transduction pathways were promoted, whereas expression of the PR1 in SA, JZA1 in JA, and ERF in Eth signal transduction pathways were inhibited. In addition, the transcripts that encode enzymes involved in the ‘Phenylpropanoid biosynthesis’ pathway, such as PAL, CYP73A, CSE, 4CL, HTC, GSE and COMT were all inhibited showed by RNA-seq and qRT-PCR. This study provided basic information of strawberry responding to C. siamense infection.

Condensed tannins (CT) from sainfoin have a high capacity to inhibit proteolysis. A previous study reported that CT from sainfoin can inhibit lactic acid bacteria activity and decrease ammonium-nitrogen (N) content during sainfoin ensiling; however, no study has focused on the metabolome of ensiled sainfoin. The objective of the present study was to investigate the effects of CT [following supplementation of deactivated CT with polyethylene glycol (PEG)] on protease activity, keystone bacteria, and metabolome during sainfoin ensiling. According to the results, PEG amendment increased non-protein N, amino acid, and soluble protein contents significantly (in the 49.08–59.41, 116.01–64.22, and 23.5–41.94% ranges, respectively, p < 0.05) during ensiling, whereas neutral detergent-insoluble protein and acid detergent-insoluble protein were decreased significantly (in the 55.98–64.71 and 36.58–57.55% ranges, respectively, p < 0.05). PEG supplementation increased aminopeptidase and acid protease activity after 3 days of ensiling (p < 0.05) and increased carboxypeptidase activity during the entire ensiling process (p < 0.05). The keystone bacteria changed following PEG addition (Stenotrophomonas, Pantoea, and Cellulosimicrobium in the control vs. Microbacterium, Enterococcus, and Brevundimonas in the PEG-treated group). In total, 510 metabolites were identified after 60 days of sainfoin ensiling, with 33 metabolites annotated in the Kyoto Encyclopedia of Genes and Genomes database. Among the metabolites, phospholipids were the most abundant (72.7% of 33 metabolites). In addition, 10 upregulated and 23 downregulated metabolites were identified in the PEG-treated group when compared with the control group, after 60 days of ensiling (p < 0.05). Pediococcus (correlated with 20 metabolites, R2 > 0.88, p < 0.05) and Lactobacillus (correlated with 16 metabolites, R2 > 0.88, p < 0.05) were the bacteria most correlated with metabolites. The results suggested antagonistic effects between Lactobacillus and Pediococcus during ensiling. The decreased proteolysis during sainfoin ensiling was mainly attributed to the inhibition of protease activity by CT, particularly carboxypeptidase activity. In addition, proteolysis decreased partly due to CT inhibiting Pediococcus activity during ensiling, with Pediococcus being significantly and positively correlated with dopamine after 60 days of ensiling (R2 = 0.8857, p < 0.05).

Recycling and remanufacturing waste electrical and electronic equipment (WEEE) can promote the comprehensive utilization of resources and promote the development of the circular economy. However, there are associated risks involved. This study explored the effectiveness of risk control in the WEEE closed-loop supply chain to provide decision support in the sustainable development of WEEE. The catastrophe progression method was used for risk evaluation, and the WEEE risk control model was constructed using system dynamics to analyze the influence of risk control strategies. The results show that adopting risk control strategies positively promotes risk control. Risk mitigation is most pronounced when both the fund subsidy and the green and low-carbon strategies are adopted. When only one approach is implemented, the green and low-carbon strategy has a more considerable effect. When the intensities of the mitigation measures are improved to the same extent, the risk control effect generated by the fund subsidy strategy was found to have a greater impact. The findings provide a useful reference for formulating subsequent risk control strategies in the WEEE closed-loop supply chain.

Chimeric antigen receptor (CAR)-T cell therapy has been shown to have considerable therapeutic effects in hematological malignancies, and NKG2D(z) CAR-T cell therapy has been verified to be safe based on clinical trials. However, due to the poor persistence of NKG2D(z) CAR-T cells, their therapeutic effect is not obvious. Here, we constructed NKG2D(bbz) CAR-T cells that can simultaneously activate 4-1BB and DAP10 costimulatory signaling. They were found to be cytotoxic to the target cells in vitro and in vivo. They exhibited low differentiation, low exhaustion, and good proliferation. Importantly, the proportions of central memory T (Tcm) and stem cell-like memory T (Tscm) cell subsets were strikingly increased. After long-term incubation with the target cells, they displayed reduced exhaustion compared to NKG2D(z) CAR-T cells. Further, in the presence of the phosphoinositide 3-kinase (PI3K) inhibitor LY294002, they exhibited reduced exhaustion and apoptosis, upregulated Bcl2 expression, and an increased proportion of Tcm cell subsets. Finally, NKG2D(bbz) CAR-T cells had better antitumor effects in vivo. In summary, the results showed that NKG2D(bbz) CAR-T cells may be valuable for cellular immunotherapy of cancer.

ABSTRACT Phenylpyruvate decarboxylase (PPDC) is a crucial enzyme that plays important roles in 2-phenylethanol (2-PE) biosynthesis. In our previous study, we screened a highly efficient PPDC KDC4427 from the novel 2-PE-producing strain Enterobacter sp. CGMCC 5087. Meanwhile, its decarboxylation activity of indolylpyruvate (IPyA) was also higher than other indolylpyruvate decarboxylases (IPDCs) reported so far. In this study, KDC4427 protein was purified and characterized, and its catalytic mechanisms were analyzed by biological methods. The optimum pH and temperature of KDC4427 was pH 6.5 and 35°C, respectively. The enzyme activity was relatively stable between pH 6 and 8 and over the range of temperatures from 25°C to 45°C. KDC4427 showed the highest catalytic efficiency on phenylpyruvic acid (PPA); meanwhile, it also showed high activity for IPyA and 2-ketobutanoic acid, and it was found that KDC4427 belongs to IPDCs by phylogenetic tree analysis. The coverage of the three-dimensional structure of KDC4427 and EcIPDC from Enterobacter cloacae was 96%. Leucine 542, one of the residues in the substrate-binding pocket, is replaced by isoleucine in KDC4427 compared with EcIPDC. Site-directed mutagenesis showed that the transition from leucine to isoleucine was unlikely to make KDC4427 have high catalytic activity for PPA and IPyA; the mutants at glutamate 468 almost completely lost catalytic activities for both PPA and IPyA, indicating that this glutamate was essential for the catalytic activity. Additionally, alanine 387 plays an important role in the substrate selectivity of KDC4427. IMPORTANCE Compared with the chemical synthesis of 2-phenylethanol (2-PE) by condensation of ethylene oxide and benzene, the biological synthesis of 2-PE is a potential method to replace the traditional process. This makes biotransformation gradually become the main way to produce high-quality 2-PE. Phenylpyruvate decarboxylase (PPDC) is the critical enzyme in 2-PE biosynthesis, and it is a momentous point of penetration to increase the production of 2-PE. In this regard, KDC4427 can catalyze phenylpyruvic acid (PPA) to phenylacetaldehyde more efficiently than any other PPDC previously reported. Moreover, it has high activity of indolepyruvate decarboxylases (IPDCs), which will be a great breakthrough in the synthesis of indole-3-acetic acid (IAA). With this study, we offer insights into the KDC4427 catalytic mechanism and significantly expand the toolbox of available α-ketoacid decarboxylases for application in biosynthesis.

Abstract Chronic atrophic gastritis (CAG) is well-known related with multiple pathogenic factors and normally therapies comprised by western or Chinese medicines. The present study was designed to identify the bacterial community characterized by 16S rRNA amplicon sequencing and determine the modulate affection of bacterial composition response western and Chinese medicine Qinghuayin (QHY) as well as antibiotic on model rats. The result shown the overall structure alteration of bacterial appeared under medicine intervened, antibiotic caused a marked depletion in bacterial diversity and richness. The enrichments of Firmicutes (85.1–90.7%) in antibiotic-free converts into Bacteroidetes (30.7–34.6%) in antibiotic-added model rat were demonstrated. Firmicutes as the most dominant phylum in antibiotic-free treatments and significantly decreased till 21.9–68.5% in antibiotic-added treatments. Especially QHY-treated rats showed highest RA of Firmicutes (90.7%) and the amelioration of CAG using QHY attributed by beneficial bacterial enrichment, especially Ruminococcus, Lactobacillus and Bifidobacterium. In addition, alpha and beta diversity analysis also demonstrated the clear dispersion and aggregation that revealed the alteration and steady of bacterial community structures. In summary, QHY has potential application value in the treatment of CAG, which attributed to close relation with the modulatory of internal bacterial communities.
