2. Design and Evaluation of General Purpose, Barrier, and Multichannel Plasticating Extrusion Screws

Abstract

Extrusion screw designs and validation are presented for three multiple channel, fractal screws for comparison with common general purpose, and barrier screws using an instrumented single screw extruder with high impact polystyrene (HIPS) and low density polyethylene (LDPE) at varying screw speeds. The fractal screws are designed with multiple channels and pressure-volume-temperature relations to control shear heating with cooling by adiabatic decompression. The general-purpose design had the highest throughput but did not provide sufficient mixing and so resulted in excessive variation in the melt temperature and pressure at screw speeds above 40 RPM. The barrier screw was a capable design with good performance for LDPE and HIPS with screw speeds from 20 to 60 RPM. However, it tended to provide excessive shear heating at higher screw speeds due to the large surface area of the barrier and mixing sections. The first fractal screw design was a multichannel variant of the general-purpose design and exhibited good consistency but excessive heating due to the large bearing area between the flights and barrel. The second fractal screw design provided decompression in the feed zone and metering zone to improve throughput but was limited by a poor transition section design. The third fractal screw design remedied these deficiencies with an improved transition section and intermittent clearances for dispersive mixing. Its performance rivaled that of the barrier screw with respect to volumetric output and energy efficiency but provided better melt pressure consistency. Cold screw freezing experiments were performed for all five screws with 5% black, blue, and violet colorants serially added to neat HIPS. The cold screw pulls showed that the general purpose and barrier screws exhibited significant racing of the materials within their screw channels and, thus, broad residence time distributions. Abstract

Isothiazolinones are a family of broad-spectrum biocides widely used in industry and consumer products. Chloro- and methyl-isothiazolinones (CMIT and MIT) are documented as strong irritants, yet they are still used in a wide variety of applications, including cosmetics, cleansers, hygiene products and various industrial applications. The subsequent substantial release of these molecules from urban sources into freshwater environments, and their potential impacts on aquatic species, have nevertheless received little attention so far, with few studies reporting on the toxicity of either CMIT or MIT to non-target organisms. The present work addresses this current knowledge gap by evaluating CMIT/MIT (3:1) and MIT acute toxicity to Daphnia pulex (Cladocera), the two formulations most commonly used by manufacturers. Additionally, genetic diversity is known to be a major component of variability in phenotypic responses, although it is largely overlooked in typical toxicity tests. Subsequently the potential range of responses inherent to genetic diversity is rarely considered. Therefore, to account for intraspecific variation in sensitivity, the design involved eight clonal lines of D. pulex stemming from distinct natural populations or commercial strains. Clones exhibited strong variation in their responses, with lethal concentrations (LC50) ranging from 0.10 to 1.84 mg/L for the mixture CMIT/MIT, and from 0.68 to 2.84 mg/L for MIT alone. These intraspecific ranges of LC50 challenge the use of single clones of daphnids in standard ecotoxicological tests and the predictions based on their results. The present study brings new evidence that assessing ecological risk of chemicals while ignoring genotype diversity is neither ecologically relevant, nor a representative evaluation of the diversity of potential adverse outcomes.

This Janus character in terms of reactivity is confirmed by its gas phase reaction with [Br6Cs4K]− to form [{Mo6Cl i8 }Cl a5 Bra]2− and by its unusual self-reactivity leading to [{Mo6Cl i8 }Cl a6 ]2− dianions. Abstract

Finding actual molecules in LC-MS/MS experiments can prove challenging due to the considerable amount of redundant ions generated during ionization. In this context, MolNotator was created and validation with this dataset. MolNotator is a Python 3.7 package designed to predict molecules (molecular masses) by combinatorial triangulation in LC-MS/MS experiments after a preprocessing step using MZMine. An MGF and a CSV files output from MZmine are required as input for MolNotator which are placed in the "mzmine_out" folder of the project folder (the uploaded dataset). Instructions for the use of MolNotator are available on GitHub (https://github.com/ZzakB/MolNotator), Pypi (https://pypi.org/project/MolNotator/) and in the associated publication. The dataset consists of 193 LC-MS/MS analyses of lichen pure standards (previously used for the Lichen Database, LDB), 156 of which were detected by manual curation and served to benchmark MolNotator. Results indicated more than 90% of the 156 molecular masses were predicted by MolNotator under 2 ppm error on average. The project folder contains, in addition to the mzmine_out folder, a database and a params folder (see GitHub and Pypi) as well as a styles folder, containing different styles that can be imported on Cytoscape (https://cytoscape.org/) to visualise MolNotator's network output.

The synthesis and characterization of redox-active, tetrathiafulvalene (TTF)-phosphine and ferrocene (FC)-phosphine complexes based on a hexanuclear rhenium chalcogenide cluster core that delivers three-dimensional pi-donor hybrid molecules. Cyclic voltammetry experiments demonstrate that these compounds can be reversely oxidized at rather low potentials allowing an easy access to the corresponding radical species.

Abstract

Crown-annelated oligothiophenes are synthesized by the thiolate protection/deprotection method. Analysis of the ionophoric properties of the macrocycles by 1H NMR, mass spectrometry, UV-vis spectroscopy, and cyclic voltammetry gives consistent results, which show that the compounds do not complex alkali cations but show moderate to good binding properties for alkali-earth cations.

The unprecedented organic-inorganic solid-state reaction taking place in [alpha]1-[(Br-[(C[H.sub.2]).sub.2]N[H.sub.3]).sub.2]Pb[I.sb.4] and the resultant mixed halide perovskite is reported. The unprecedented solid-state reaction between organic and inorganic components of a hybrid is considered to be a new strategy for achieving hybrid perovskites.

Abstract

Direct Analysis in Real Time DART-HRMS is here first applied to the detection of molecules from a lichen, Lichina pygmaea. The aim was to propose an innovative method of in situ detection of lichen secondary metabolites using the possibilities of elemental composition determination available when a DART source is interfaced with a TOF analyzer. Three kinds of samples have been submitted to DART ionization, i.e. an intact thallus, a powder obtained from the crushed lichen and an aqueous extract. In situ analysis of crushed lichen, yields an extensive chemical profile, comparable to what is obtained from the aqueous extract, comprising both major polar metabolites described in literature along with some other signals that could correspond to potentially unknown metabolites. One of the detected secondary metabolites, mycosporine serinol, underwent a dehydration reaction prior to its transfer in the gas-phase by DART ionization. The consideration of the thermal transfers involved in the DART ionization process and the possibility to record time-dependent mass spectra through the use of the TOF analyzer allowed establishing Arrhenius plots of this water molecule loss to obtain associated thermodynamic quantities. The low values of corresponding activation enthalpy (Δr‡Hm° of the order of 25 kJ mol(-1)) enabled formulating some assumption regarding a possible role of such metabolites in the lichen.

37. Untargeted Metabolomics Reveal Lipid Alterations upon 2-Deoxyglucose Treatment in Human HaCaT Keratinocytes

Abstract

The glucose analogue 2-deoxyglucose (2-DG) impedes cancer progression in animal models and is currently being assessed as an anticancer therapy, yet the mode of action of this drug of high clinical significance has not been fully delineated. In an attempt to better characterize its pharmacodynamics, an integrative UPLC-Q-Exactive-based joint metabolomic and lipidomic approach was undertaken to evaluate the metabolic perturbations induced by this drug in human HaCaT keratinocyte cells. R-XCMS data processing and subsequent multivariate pattern recognition, metabolites identification, and pathway analyses identified eight metabolites that were most significantly changed upon a 3 h 2-DG exposure. Most of these dysregulated features were emphasized in the course of lipidomic profiling and could be identified as ceramide and glucosylceramide derivatives, consistently with their involvement in cell death programming. Even though metabolomic analyses did not generally afford such clear-cut dysregulations, some alterations in phosphatidylcholine and phosphatidylethanolamine derivatives could be highlighted as well. Overall, these results support the adequacy of the proposed analytical workflow and might contribute to a better understanding of the mechanisms underlying the promising effects of 2-DG.

Abstract

International audience; RATIONALE: The internal energy deposition in ions that cross the desolvation region of an electrospray ionization (ESI) source affects the mass spectra that are obtained using in-source collision-induced dissociation (CID) or in tandem mass spectrometry (MS/MS) mode. It is thus important to evaluate the internal energy distributions of the ions in different parts of an ESI mass spectrometer.METHODS: The desolvation region is considered as a collision zone and a partially elastic multiple-collision model is used to account for the accumulation of internal energy in the ions. The ion survival yields (SYTheo) of the theoretical mass spectra calculated by MassKinetics software are fitted with the experimental ion survival yields (SYExp) of the substituted benzylpyridinium cations that have been obtained with an ESI source interfaced with a quadrupole mass spectrometer. The theoretical parameters used for fitting the calculation data with the experimental results are the center-of-mass collision energy (Ecom) of the colliding ions and a term related to the pressure of the desolvation area of the ESI interface. RESULTS: In the proposed model, an average number of ’effective’ collisions of close to 30 in the desolvation area is employed. The voltages applied to the orifice of this interface are correlated to a theoretical initial kinetic energy (Einit,Kin) in the laboratory frame of the ions. In the present case, these theoretical initial kinetic energies range from 5.5 to 9 eV. The internal energy distributions evaluated from this model resemble the thermal distributions of ions having ’characteristic temperatures’ between 1020 and 1550 K, and the results of calculations show that the mean internal energy of the ions increases linearly with the orifice voltage.CONCLUSIONS: The model used in this study can account for the energy build-up of the ions in an ESI interface and allows the change in the internal energy distribution of the electrosprayed ions in different regions of a mass spectrometer to be evaluated.
