In order to systematize the processes and improve the quality of the activities of a biomedical research group, a research, development and innovation (RDI) management system based on the Spanish norm UNE 166002:2006 has been designed and implemented. We describe the experience and results obtained during the selection and adaptation of the OpenKM document management system platform as a useful means for research group management. The usefulness of this type of platform for this purpose is demonstrated, which could be exported to other research groups.

[Background and Purpose]: Lipogenesis is intimately controlled by hormones and cytokines as well as nutritional conditions. IL-6 participates in the regulation of fatty acid metabolism in the liver. We investigated the role of IL-6 in mediating fasting/re-feeding changes in the expression of hepatic lipogenic enzymes. [Experimental Approach]: Gene and protein expression of lipogenic enzymes were examined in livers of wild-type (WT) and IL-6-deficient (IL-6) mice during fasting and re-feeding conditions. Effects of exogenous IL-6 administration on gene expression of these enzymes were evaluated in vivo. The involvement of STAT3 in mediating these IL-6 responses was investigated by using siRNA in human HepG2 cells. [Key Results]: During feeding, the up-regulation in the hepatic expression of lipogenic genes presented similar time kinetics in WT and IL-6 mice. During fasting, expression of lipogenic genes decreased gradually over time in both strains, although the initial drop was more marked in IL-6 mice. Protein levels of hepatic lipogenic enzymes were lower in IL-6 than in WT mice at the end of the fasting period. In WT, circulating IL-6 levels paralleled gene expression of hepatic lipogenic enzymes. IL-6 administration in vivo and in vitro showed that IL-6-mediated signalling was associated with the up-regulation of hepatic lipogenic enzyme genes. Moreover, silencing STAT3 in HepG2 cells attenuated IL-6 mediated up-regulation of lipogenic gene transcription levels. [Conclusions and Implications]: IL-6 sustains levels of hepatic lipogenic enzymes during fasting through activation of STAT3. 49 p.-7 fig.-2 tab., A series of new peroxisome proliferator activated receptors (PPARs) chiral ligands have been designed following the accepted three-module structure comprising a polar head, linker, and hydrophobic tail. The majority of the ligands incorporate the oxazolidinone moiety as a novel polar head, and the nature of the hydrophobic tail has also been varied. Docking studies using the crystal structure of an agonist bound to the ligand binding domain of the PPARα receptor have been performed as a tool for their design. Suitable synthetic procedures have been developed, and compounds with different stereochemistries have been prepared. Evaluation of basal and ligand-induced activity proved that several compounds showed agonist activity at the PPARα receptor, thus validating the oxazolidinone template for PPAR activity. La esclerosis lateral amiotrófica, ELA, es una enfermedad neurodegenerativa que implica la pérdida del control muscular. En consecuencia, se ven afectadas la movilidad de las extremidades y, a medida que avanza la enfermedad, las capacidades para comer, respirar y comunicarse. Esta enfermedad tiene una baja prevalencia por su alta mortalidad y tiene una mayor incidencia entre los 40 y los 70 años. Se ha elegido esta enfermedad como temática debido al desconocimiento general y por su alta morbi-mortalidad. Para tratar esta enfermedad se requiere un equipo integrado de médicos y se utilizan diversas terapias, aunque solo sirven para retrasar la sintomatología y prevenir las complicaciones. El trabajo se centrará en las terapias farmacológicas ya que ayudan a paliar muchos de los síntomas y aumentan la esperanza de vida de los pacientes. El objetivo de este trabajo es buscar nuevas dianas terapéuticas mediante herramientas computacionales. De esta forma se podrían abrir nuevas líneas de investigación y quizás se pueda encontrar una terapia que implique una mejora en el tratamiento de la ELA. L'esclerosi lateral amiotròfica, ELA, és una malaltia neurodegenerativa que implica la pèrdua de control muscular. En conseqüència, es veuen afectades la mobilitat de les extremitats i, a mesura que avança la malaltia, les capacitats per menjar, respirar i comunicar-se. Aquesta malaltia té una baixa prevalença per la seva alta mortalitat i té una major incidència entre els 40 i els 70 anys. S'ha triat aquesta malaltia com a temàtica a causa del desconeixement general i per la seva alta morbimortalitat. Per tractar aquesta malaltia es requereix un equip integrat de metges i s'utilitzen diverses teràpies, encara que només serveixen per retardar la simptomatologia i prevenir les complicacions. El treball se centra en les teràpies farmacològiques, ja que ajuden a pal·liar molts dels símptomes i augmenten l'esperança de vida dels pacients. L'objectiu d'aquest treball és buscar noves dianes terapèutiques mitjançant eines computacionals. D'aquesta manera es podrien obrir noves línies d'investigació i potser es pugui trobar una teràpia que impliqui una millora en el tractament de l'ELA. Amyotrophic lateral sclerosis, ALS, is a neurodegenerative disease which involves loss of muscle control. As a result , limb mobility and, as the disease progresses, the ability to eat, breathe and communicate are affected. This disease has a low prevalence due to its high mortality and has a higher incidence between the ages of 40 and 70. This disease has been chosen as the subject because of general ignorance and its high morbidity. An integrated team of doctors and various therapies are used to treat this disease, although they only serve to delay symptomatology and prevent complications. Work will focus on drug therapies as they help alleviate many of the symptoms and increase and increase the life expectancy of patients. The objective of this work is to find new therapeutic targets using computational tools. This could open up new lines of research and perhaps can be found a therapy which involves an improvement in the treatment of ALS.

Actualmente el cáncer de Leucemia Linfoblástica Aguda (LLA) es uno de los más comunas con alta tasa de mortalidad en la infancia. A lo largo de los años se han ido investigando y desarrollando terapias como la radioterapia o la quimioterapia. Este trabajo se ha centrado en el fármaco quimioterapéutico metrotrexato (MTX), comúnmente usado por su alta eficacia inhibiendo el enzima Dihidrofolato reductasa (DHFR). Una proteína relacionada con la síntesis de aminoácidos, purinas entre otros elementos. Sin embargo, se han encontrado y estudiado casos de resistencia al fármaco por lo que, el objetivo de este proyecto ha sido la búsqueda bibliográfica y comprensión de los mecanismos de resistencia, el estudio de las interacciones del DHFR-MTX mediante estructuras experimentales en fuentes de datos. Uno de los retos ha sido buscar otras proteínas experimentales mutadas para calcular y analizar las energías de interacción fármaco-proteína, de los cuales compararlas para encontrar interacciones comunes en ambos casos. Por consiguiente, se han realizado modelos computacionales de la interacción MTX con DHFR agrestes y resistentes, como base para la búsqueda, y estudios de nuevas moléculas que puedan tener actividad con las proteínas. Como resultado de esta búsqueda y estudio de interacciones se han propuesto 3 ligandos candidatos, una de ellas el fármaco llamado Leucovorin por su posible aplicación en el tratamiento de la ALL. Finalmente, una vez analizado todos los resultados se concluye que, dentro de los ligandos testeados, el MTX es el que presenta una energía de interacción más favorable. Además, coincidiendo con lo esperado las proteínas resistentes presentan peor interacción que DHFR agreste. Acute lymphoblastic leukemia (ALL) cancer is one of the most common with a high mortality rate in childhood. Throughout the years of investigation have been developed therapies such as radiotherapy or chemotherapy. This work focuses on the chemotherapeutic drug metrotrexate (MTX), commonly used for its high efficacy by inhibiting the enzyme Dihydrofolate reductase(DHFR): A protein related to the synthesis of amino acids, purines among other elements. However, there are cases of resistance to the drug. The objective of this project is searching the bibliographic and understanding the mechanisms of resistance and the studying of the interactions of DHFR-MTX through structures crystallized in data sources. One of the challenges has been to find other mutated experimental proteins to calculate and analyze the drug-protein interaction energies, from which to compare them to find common interactions in both cases. Therefore, computational models of the MTX interaction with WT and resistant DHFR have been made, as a basis for search, and studies of new molecules that may have activity with proteins. Because of this search and study of interactions 3 candidate ligands have been proposed, one of them the drug called Leucovorin for its possible application in the treatment of all. Finally, once all the results have been analyzed, it is concluded that, within the ligands tested, MTX is the one with the most favorable interaction energy. In addition, coinciding with expectations, resistant proteins show worse interaction than wild-type DHFR. Al llarg dels anys s'han anat investigant i desenvolupant teràpies com la radioteràpia o la quimioteràpia. Aquest treball s'ha centrat en el fàrmac quimioterapèutic metrotrexato (MTX), comunament usat per la seva alta eficàcia inhibint l'enzim dihidrofolat reductasa (DHFR). Una proteïna relacionada amb la síntesi d'aminoàcids, purines entre altres elements. No obstant això, s'han trobat i estudiat casos de resistència al fàrmac de manera que, l'objectiu d'aquest projecte ha estat la recerca bibliogràfica i comprensió dels mecanismes de resistència, l'estudi de les interaccions del DHFR-MTX mitjançant estructures experimentals en fonts de dades. Un dels reptes ha estat buscar altres proteïnes experimentals mutades per calcular i analitzar les energies d'interacció fàrmac-proteïna, dels quals comparar-les per trobar interaccions comunes en ambdós casos. Per tant, s'han realitzat models computacionals de la interacció MTX amb DHFR agrestes i resistents, com a base per a la recerca, i estudis de noves molècules que puguin tenir activitat amb les proteïnes. Com a resultat d'aquesta recerca i estudi d'interaccions s'han proposat 3 lligands candidats, una d'elles el fàrmac anomenat leucovorin per la seva possible aplicació en el tractament de l'ALL. Finalment, un cop analitzat tots els resultats es conclou que, dins dels lligands testejats, el MTX és el que presenta una energia d'interacció més favorable. A més, coincidint amb el que s'esperava les proteïnes resistents presenten pitjor interacció que DHFR agrest.

Las conotoxinas son un grupo de péptidos procedentes del veneno de los caracoles marinos del género Conus que han sido estudiadas en los últimos años debido a su gran potencial terapéutico para el tratamiento de múltiples enfermedades. Estudios recientes describen dos conotoxinas, cal29b y xm11a, con actividad sobre Mycobacterium tuberculosis, cuya infección es una de las principales causas de muerte a nivel mundial. Estas conotoxinas no han sido caracterizadas estructuralmente todavía y su mecanismo de acción es desconocido, por lo que el empleo de las técnicas bioinformáticas puede ofrecer una aproximación a las estructuras de las conotoxinas y al método de unión a las posibles dianas terapéuticas. En este trabajo se ha propuesto una caracterización estructural para estas conotoxinas a partir de su secuencia y se ha realizado el acoplamiento ligando-receptor para determinar su unión a las dianas farmacológicas más relevantes actualmente para esta bacteria. Los resultados muestran ciertas coincidencias estructurales entre las conotoxinas con actividad antibacteriana y en las conformaciones adoptadas sobre las dianas, aunque estos resultados no son concluyentes y es necesario un mayor estudio de las conotoxinas para determinar su mecanismo de acción. Conotoxins are a group of peptides extracted from the venom of marine cone snails belonging to genus Conus. In the last few years these peptides have been studied due to its great therapeutic potential. Recent research studies describe two new conotoxins, cal29a and xm11a, which showed pharmacological activity against Mycobacterium tuberculosis, the main cause of tuberculosis. This disease is one of main causes of death worldwide. The structure of these conotoxins is not published and their mechanism of action is unknown. Bioinformatic tools like Computer Aided Drug Design (CADD) could give an approach to the structure and how conotoxins binds to the targets. As a result of this research we propose a structural characterization from the peptide sequence of these conotoxins and peptide-protein docking models for the most relevant targets for Mycobacterium tuberculosis. The results of this reseach show similarities between conotoxins with antimycobacterial activity. However, more studies are needed in order to characterize the mechanism of action. Estudis recents descriuen dos conotoxines, cal29b i xm11a, amb activitat sobre Mycobacterium tuberculosi, la infecció és una de les principals causes de mort en l'àmbit mundial. Aquestes conotoxinas no han estat caracteritzades estructuralment encara i el seu mecanisme d'acció és desconegut, de manera que l'ús de les tècniques bioinformàtiques pot oferir una aproximació a les estructures de les conotoxinas i al mètode d'unió a les possibles dianes terapèutiques. La artritis reumatoide (AR) es una enfermedad inflamatoria crónica de naturaleza autoinmune que afecta entre el 0,3 y el 1% de la población mundial. Los afectados por AR sufren fuertes dolores además de una pérdida de movimientos causada por la inflamación y el desgaste de las articulaciones, provocando un importante decrecimiento en la calidad de vida del paciente. Este trastorno es más frecuente en mujeres que en hombres y más habitual a edad avanzada que en sujetos jóvenes. Existen fármacos cuyo objetivo es reducir los síntomas y mejorar la calidad de vida de los afectados por la AR, éstos pueden ser controladores de los síntomas o modificadores de la enfermedad, en el segundo caso se logra ralentizar el avance del deterioro de las articulaciones, así como reducir la inflamación. Este trabajo tiene como objetivo buscar mediante técnicas computacionales nuevas dianas terapéuticas que puedan tener una mejor asociación con la AR y puedan ser nuevas vías de investigación para el desarrollo de nuevos fármacos que permitan mejorar el tratamiento de la enfermedad. Rheumatoid Arthritis (RA) is an autoimmune chronic inflammatory disease which affects between the 0,3 and the 1% of the world's population. RA patients tend to suffer pain and loss of the movements caused by joint inflammation. This disease is three times more frequent in women than in men and the probability of affection increases with the age. There are drugs that reduce the symptoms and improve the life quality of the RA affected, these can control de symptoms or modify the disease. The second type's objective is to slow down the progress of the joint deterioration, as well as reduce the inflammation. The main objective of this project is to discover using computational techniques and tools new therapeutic targets whose may improve the association with the RA and become new investigational ways for the development of new drugs. Els afectats per AR sofreixen forts dolors a més d'una pèrdua de moviments causada per la inflamació i el desgast de les articulacions, provocant un important decreixement en la qualitat de vida del pacient. Aquest trastorn és més freqüent en dones que en homes i més habitual a edat avançada que en subjectes joves. Existeixen fàrmacs que el seu objectiu és reduir els símptomes i millorar la qualitat de vida dels afectats per la AR, aquests poden ser controladors dels símptomes o modificadors de la malaltia, en el segon cas s'aconsegueix ralentir l'avanç de la deterioració de les articulacions, així com reduir la inflamació. Aquest treball té com a objectiu buscar mitjançant tècniques computacionals noves dianes terapèutiques que puguin tenir una millor associació amb la AR i puguin ser noves vies de recerca per al desenvolupament de nous fàrmacs que permetin millorar el tractament de la malaltia.
