This article is devoted to special aspects of giving classes in the discipline “Applied Physical Education and Sports” at the Department of Physical Culture at Siberian Federal University. These aspects are considered an example of development and introduction of online course of the swimming specialization in educational process. It is connected with the transition to distance learning caused by the spread of COVID-19 infection in the world. The transition to this format of study was an effective way of interacting with full-time students in the applied discipline. It made it possible to avoid the eventual interruption of educational activities, as feedback from the “student – university teacher” was organized. This allowed the students to receive the full information by providing interim assessment for the spring semester of the 2019-2020 academic year. The article reflects the personal experience of university teachers in the development and implementation of the electronic course, its main resources, content, and features of functioning. This allowed asynchronously teach students, posting interactive lectures, offer videos on the topic of the course, issue practical tasks and training programs, check the assimilation of material, as well as monitor the performance of tasks, maintain motor activity and health of students.

COVID-19 is characterized by systemic pro-inflammatory shifts with the development of serious alterations in the functioning of the immune system. Investigations of the gene expression changes accompanying the infection state provide insight into the molecular and cellular processes depending on the sickness severity and virus variants. Severe Delta COVID-19 has been characterized by the appearance of a monocyte subset enriched for proinflammatory gene expression signatures and a shift in ligand-receptor interactions. We profiled the chromatin accessibility landscape of 140,000 nuclei in PBMC samples from healthy individuals or individuals with COVID-19. We investigated cis-regulatory elements and identified the core transcription factors governing gene expression in immune cells during COVID-19 infection. In severe cases, we discovered that regulome and chromatin co-accessibility modules were significantly altered across many cell types. Moreover, cases with the Delta variant were accompanied by a specific monocyte subtype discovered using scATAC-seq data. Our analysis showed that immune cells of individuals with severe Delta COVID-19 underwent significant remodeling of the chromatin accessibility landscape and development of the proinflammatory expression pattern. Using a gene regulatory network modeling approach, we investigated the core transcription factors governing the cell state and identified the most pronounced chromatin changes in CD14+ monocytes from individuals with severe Delta COVID-19. Recombination is one of the mechanisms of SARS-CoV-2 evolution along with the occurrence of point mutations, insertions, and deletions. Recently, recombinant variants of SARS-CoV-2 have been registered in different countries, and some of them have become circulating forms. In this work, we performed screening of SARS-CoV-2 genomic sequences to identify recombination events and co-infections with various strains of the SARS-CoV-2 virus detected in Russia from February 2020 to March 2022. The study included 9336 genomes of the COVID-19 pathogen obtained as a result of high-throughput sequencing on the Illumina platform. For data analysis, we used an algorithm developed by our group that can identify viral recombination variants and cases of co-infections by estimating the frequencies of characteristic substitutions in raw read alignment files and VCF files. The detected cases of recombination were confirmed by alternative sequencing methods, principal component analysis, and phylogenetic analysis. The suggested approach allowed for the identification of recombinant variants of strains BA.1 and BA.2, among which a new recombinant variant was identified, as well as a previously discovered one. The results obtained are the first evidence of the spread of recombinant variants of SARS-CoV-2 in Russia. In addition to cases of recombination we identified cases of coinfection: eight of them contained the genome of the Omicron line as one of the variants, six of them the genome of the Delta line, and two with the genome of the Alpha line.

Analysis of genomic variability of pathogens associated with heightened public health concerns is an opportunity to track transmission routes of the disease and helps to develop more effective vaccines and specific diagnostic tests. We present the findings of a detailed genomic analysis of the genomic variability of the SARS-CoV-2 Omicron variant that spread in Russia between 8 December 2021 and 30 January 2022. We performed phylogenetic analysis of Omicron viral isolates collected in Moscow ( n = 589) and downloaded from GISAID ( n = 397), and identified that the BA.1 lineage was predominant in Russia during this period. The BA.2 lineage was also identified early in December 2021. We identified three cases of BA.1/BA.2 coinfections and one case of Delta/Omicron coinfection. A comparative genomic analysis of SARS-CoV-2 viral variants that spread in other countries allowed us to identify possible cases of transmission. We also found that some mutations that are quite rare in the Global Omicron dataset have a higher incidence rate, and identified genetic markers that could be associated with ways of Omicron transmission in Russia. We give the genomic variability of single nucleotide variations across the genome and give a characteristic of haplotype variability of Omicron strains in both Russia and around the world, and we also identify them.

The coronavirus disease 2019 (COVID-19) is accompanied by a cytokine storm with the release of many proinflammatory factors and development of respiratory syndrome. Several SARS-CoV-2 lineages have been identified, and the Delta variant (B.1.617), linked with high mortality risk, has become dominant in many countries. Understanding the immune responses associated with COVID-19 lineages may therefore aid the development of therapeutic and diagnostic strategies. Multiple single-cell gene expression studies revealed innate and adaptive immunological factors and pathways correlated with COVID-19 severity. Additional investigations covering host-pathogen response characteristics for infection caused by different lineages are required. Here, we performed single-cell transcriptome profiling of blood mononuclear cells from the individuals with different severity of the COVID-19 and virus lineages to uncover variant specific molecular factors associated with immunity. We identified significant changes in lymphoid and myeloid cells. Our study highlights that an abundant population of monocytes with specific gene expression signatures accompanies Delta lineage of SARS-CoV-2 and contributes to COVID-19 pathogenesis inferring immune components for targeted therapy.

Smallanthus sonchifolius (yacon) is a new prospective plant cultivated in Europe as a natural sugarcane substitute. It is used for diabetes and for the prevention of obesity. The study of carboxylic acids in the roots and herb of S. sonchifolius was carried out for the first time by gas chromatography-mass spectrometry (GC-MS). As a result of the study, 12 components were found in the roots of S. sonchifolius, 9 of which were carboxylic acids. The S. sonchifolius herb contained 41 components, 18 of which were carboxylic acids. The dominant compounds in the roots were: undecanoic acid, methyl ester - 546.04 mg/kg; 1-benzazirene-1-carboxylic acid, 2,2,5a-trimethyl-1a-[3-oxo-1-butenyl] perhydro-, methyl ester - 360.63 mg/kg; 9-octadecenoic acid (Z)-, methyl ester - 119.21 mg/kg. In the herb of the yacon dominant is cyclohexanol, 1-ethynyl - 28.67 mg/kg. Key words: Smallanthus sonchifolius (yacon) carbo- xylic acids GC-MS.

Morphological, functional signs of myocardium affection and tolerance to physical loading in animals with isoproterenol-induced affection of myocardium after transplantation of a nuclear-containing cells (NCC) of umbilical blood were analyzed in comparison with a natural course of the model. There was proved, that a transplantation of a NCC promotes acceleration of processes of regeneration and restoration of the damaged myocardium in experimental animals. There were estimated the changes in localization of the transplanted NCC in the zone of damage, and capacity of the peripheral blood NCC, while they were transplanted, using intravenous injection, to migrate into the damage zone. The experimental data obtained need further complex analysis and permit to wait a positive effect while performing clinical investigations in the patients, suffering chronic diseases of the heart.

Aim: To detect changes in the concentration of nitric oxide and its metabolites--nitrites and nitrates in conjunction with the indices of the glutathione system in the children with chronic hepatitis B and C., Patients and Methods: In the research took a part 76 patients (boys or girls) with chronic hepatitis B and C aged from 7 up to 16 years. Nitric oxide and its metabolites determined by method of V.A. Metel'skaya and N.G. Gumanova; level of reduced glutathione - the titrimetric analysis; activity of glutathione transferase - the method of spectrophotometry., Results: The content of nitric oxide and its metabolites--nitrites and nitrates--in serum and urine of children with chronic viral hepatitis B and C have increased. Correlations were revealed between nitric oxide and its metabolites in serum and urine, as well as between these indices and glutathione system parameters-reduced glutathione concentration and activity of enzymes of glutathione transferase of erythrocytes and plasma., Conclusions: In the children with chronic viral hepatitis B and C the increase of the concentration of nitric oxide and its metabolites in serum and urine is accompanied by decrease of reduced glutathione concentration and increase of glutathione transferase activity in erythrocytes and plasma. These findings suggest the presence of oxidative stress due to which the therapy should be strengthened by antioxidant effect.

In the present study liver detoxifying function state in children with chronic hepatitis and pigmentary hepatosis was examined. An endogenous intoxication level increase, peroxide processes and biotransformation system activation were established against the background of endotoxicosis substrate accumulation in blood. The dynamics of clinical symptoms and liver detoxifying function indicators was estimated against the background of the complex treatment with Reamberin. In our study we have noticed detoxication mechanisms improvement, and that fact has been confirmed by a decrease of endogenous intoxication level and significant middle-mass molecules decreasing in blood plasma as well as intoxication index and glutathione enzymes activity in blood.

In the experiments on dogs and mice it was shown that indometophene could modify the immune status of the body. Specific and non-specific resistance of mice to E. coli infection increased after indometophene administration. At the same time higher titers of normal anti-salmonella antibodies in mice serum and a higher phagocytic activity of neutrophils in the peripheral blood in dogs were found. The influence of the preparation on the content of the complement and normal antibodies-hemagglutinins was less pronounced. It was suggested that the indometophene ability to bustle immune reactivity of the body played a certain role in the mechanism of its radioprotective effect.
