Foram investigados aspectos clínicos, laboratoriais, terapêuticos e evolutivos da leishmaniose tegumentar americana em Belo Horizonte. O estudo incluiu 358 pacientes com leishmaniose cutânea (LC) e 25 com leishmaniose mucosa (LM). Comparados aos pacientes com LC, aqueles com LM apresentaram maior tempo de doença e relato de outras doenças concomitantes, sugerindo que a debilitação pela leishmaniose e/ou outras doenças podem contribuir para a ativação e/ou disseminação mucosa do parasito. As sensibilidades das reações intradérmica, de imunofluorescência indireta e da pesquisa direta do parasito foram de 78,4, 79,3 e 68,3%, respectivamente. O tratamento com antimoniato de meglumina foi 100% eficaz, com 59% de efeitos colaterais ao longo do tratamento. A recidiva após tratamento ocorreu em 32 (10,1%) dos 318 casos seguidos por até dois anos. A maioria das recidivas (31 dos 32 casos) ocorreu em pacientes com LC tratados com 15mg Sb5+/kg/dia. Na investigação de critérios de cura, a reação intradérmica negativa foi o único fator associado a um risco três vezes maior de recidiva. Um aumento da dose ou do tempo de tratamento talvez melhore o prognóstico nestes pacientes.

Para o estudo de variabilidade genética em Triatoma brasiliensis, o principal vetor da doença de Chagas no Nordeste brasileiro, espécimes de três diferentes populações intradomiciliares foram analisados. Regiões do DNA genômico foram amplificadas utilizando dois iniciadores randômicos através da técnica de RAPD (Random Amplified Polymorphic DNA), visualizados em géis de poliacrilamida corados pela prata. Os perfis originados se mostraram bastante homogêneos quando comparados intrapopulacionalmente. Populações capturadas em duas regiões diferentes do Estado do Ceará também apresentaram homogeneidade entre si, mas, quando comparadas com a população proveniente do Piauí, foi possível diferenciá-las. Esses resultados, preliminares, indicam que o RAPD pode ser usado com sucesso nos estudos de variabilidade em triatomíneos, bem como sugerem a existência de variabilidade entre diferentes populações de T. brasiliensis pertencentes a uma mesma subespécie.

Trypanosoma rangeli is a hemoflagelate parasite that infects domestic and sylvatic animals, as well as man, in Central and South America. T. rangeli has an overlapping distribution with T. cruzi, the etiological agent of Chagas disease, sharing several animal reservoirs and triatomine vectors. We have isolated T. rangeli strains in the State of Santa Catarina, in southern Brazil, which dramatically increased the distribution area of this parasite. This brief review summarizes several studies comparing T. rangeli strains isolated in Santa Catarina with others isolated in Colombia, Honduras and Venezuela. The different methods used include indirect immunofluorescence and western blot assays, lectin agglutination, isoenzyme electrophoresis and random amplified polymorphic DNA analysis, triatomine susceptibility, in vitro cell infection assays, and mini-exon gene analysis.

Neste trabalho apresentamos um estudo das inter-relações recentes entre os ciclos de transmissão silvestre e doméstico do T. cruzi no município de Bambuí, Minas Gerais, Brasil. No final da década de 1930, o Panstrongylus megistus era encontrado em 75% das casas. Subseqüentemente, o Triatoma infestans tornou-se a espécie predominante, sendo encontrada em 20% das residências urbanas e em mais de 60% das periurbanas. Com as intensas campanhas de borrifação desenvolvidas entre 1956 e 1969, o T. infestans foi erradicado do município, e a transmissão da doença de Chagas ao homem, interrompida, com aparecimento de P. megistus em residências rurais. Amostras de T. cruzi isoladas via xenodiagnóstico e hemocultura de 43 gambás (Didelphis albiventris) capturados em ambiente peridomiciliar e silvestre foram caracterizadas isoenzimaticamente e, independentemente da via de isolamento, apresentaram perfil de zimodema Z1. Por meio do Programa de Vigilância Epidemiológica da doença de Chagas, no período de agosto de 1986 a dezembro de 1988, 154 exemplares de P. megistus foram capturados pela população no peridomicílio e intradomicílio rural, estando 9,8% infectados pelo T. cruzi. Na caracterização isoenzimática de 13 amostras de T. cruzi isoladas desses triatomíneos, seis pertenciam ao zimodema Z1 (ciclo de transmissão silvestre), e sete ao Z2 (ciclo de transmissão doméstico). A captura de exemplares de P. megistus no intradomicílio, naturalmente infectados com parasitas de ambos os ciclos, indica a superposição dos ciclos de transmissão da doença de Chagas no município de Bambuí. Outra evidência da inter-relação dos ciclos pôde ser observada no isolamento de T. cruzi Z2 de um gato e a participação do cão como reservatório de T. cruzi Z1. A presença do P. megistus no peridomicílio representa importante elo entre o ambiente silvestre e o intradomicílio, servindo como veiculador do T. cruzi Z1 e na manutenção de ciclos de transmissão do T. cruzi Z2 no peridomicílio e domicílio, propiciando de forma gradual a reinfestação do município caso a Vigilância Epidemiológica seja interrompida.

Abstract Background Hexose transporters (HT) are membrane proteins involved in the uptake of energy-supplying glucose and other hexoses into the cell. Previous studies employing the Differential Display technique have shown that the transcription level of the HT gene from T. cruzi (TcrHT) is higher in an in vitro-induced benznidazole (BZ)-resistant population of the parasite (17 LER) than in its susceptible counterpart (17 WTS). Methods In the present study, TcrHT has been characterized in populations and strains of T. cruzi that are resistant or susceptible to BZ. We investigated the copy number and chromosomal location of the gene, the levels of TcrHT mRNA and of TcrHT activity, and the phylogenetic relationship between TcrHT and HTs from other organisms. Results In silico analyses revealed that 15 sequences of the TcrHT gene are present in the T. cruzi genome, considering both CL Brener haplotypes. Southern blot analyses confirmed that the gene is present as a multicopy tandem array and indicated a nucleotide sequence polymorphism associated to T. cruzi group I or II. Karyotype analyses revealed that TcrHT is located in two chromosomal bands varying in size from 1.85 to 2.6 Mb depending on the strain of T. cruzi. The sequence of amino acids in the HT from T. cruzi is closely related to the HT sequences of Leishmania species according to phylogenetic analysis. Northern blot and quantitative real-time reverse transcriptase polymerase chain reaction analyses revealed that TcrHT transcripts are 2.6-fold higher in the resistant 17 LER population than in the susceptible 17 WTS. Interestingly, the hexose transporter activity was 40% lower in the 17 LER population than in all other T. cruzi samples analyzed. This phenotype was detected only in the in vitro-induced BZ resistant population, but not in the in vivo-selected or naturally BZ resistant T. cruzi samples. Sequencing analysis revealed that the amino acid sequences of the TcrHT from 17WTS and 17LER populations are identical. This result suggests that the difference in glucose transport between 17WTS and 17LER populations is not due to point mutations, but probably due to lower protein expression level. Conclusion The BZ resistant population 17 LER presents a decrease in glucose uptake in response to drug pressure.

Eucalyptus-based glued laminated timber (glulam) was produced to determine the feasibility of a non-destructive method (drilling resistance) to predict the properties of structural elements and add value to lower-quality hardwood species. Glulam was manufactured with formaldehyde (Resorcinol), reference condition, and bio-based (Castor oil-based) adhesives in two assembly schemes, the core composed either of two continuous lamellae each 105 cm long, or of two formed by the juxtaposition of shorter boards (35 and 55 cm). The shear strength of the glue line (fv0), modulus of elasticity (Ec90), and strength (fc90) in compression perpendicular to the grain; delamination (DL); and main and extended glue line thicknesses were evaluated. The Resistograph equipment was used to perform the perforation perpendicular to the glue line (samples extracted from the glulam elements) to correlate the properties. The results of this research demonstrate that the scheme of the boards had little effect on the physical and mechanical properties evaluated (except the main glue line and delamination), and the drilling resistance (DR) presents a significant correlation with practically all properties evaluated (variations in density values and other properties are explained by variations in DR values), making it possible to estimate Ec90 and fc90 with desired precision (R2adj ≈ 80%). This highlights the feasibility of using this methodology in the quality control of glulam elements. It is concluded that regardless of the adhesive, elements comprising a 105 cm-length core and external lamellae (T1 and control) are indicated for external use, presenting low delamination. Short-length central lamellae adhesively glued with PUR (T2) are not recommended for external applications due to their susceptibility to delamination. However, T2 is indicated for internal environments due to its low production cost. This study also proved the efficiency of using models based on drilling resistance to estimate wood density and its resistance to compression perpendicular to the fiber. [ABSTRACT FROM AUTHOR]

Chagas disease (CD), which is caused by Trypanosoma cruzi and was discovered more than 100 years ago, remains the leading cause of death from parasitic diseases in the Americas. As a curative treatment is only available for the acute phase of CD, the search for new therapeutic options is urgent. In this study, nitroazole and azole compounds were synthesized and underwent molecular modeling, anti-T. cruzi evaluations and nitroreductase enzymatic assays. The compounds were designed as possible inhibitors of ergosterol biosynthesis and/or as substrates of nitroreductase enzymes. The in vitro evaluation against T. cruzi clearly showed that nitrotriazole compounds are significantly more potent than nitroimidazoles and triazoles. When their carbonyls were reduced to hydroxyl groups, the compounds showed a significant increase in activity. In addition, these substances showed potential for action via nitroreductase activation, as the substances were metabolized at higher rates than benznidazole (BZN), a reference drug against CD. Among the compounds, 1-(2,4-difluorophenyl)-2-(3-nitro-1H-1,2,4-triazol-1-yl)ethanol (8) is the most potent and selective of the series, with an IC50 of 0.39 µM and selectivity index of 3077; compared to BZN, 8 is 4-fold more potent and 2-fold more selective. Moreover, this compound was not mutagenic at any of the concentrations evaluated, exhibited a favorable in silico ADMET profile and showed a low potential for hepatotoxicity, as evidenced by the high values of CC50 in HepG2 cells. Furthermore, compared to BZN, derivative 8 showed a higher rate of conversion by nitroreductase and was metabolized three times more quickly when both compounds were tested at a concentration of 50 µM. The results obtained by the enzymatic evaluation and molecular docking studies suggest that, as planned, nitroazole derivatives may utilize the nitroreductase metabolism pathway as their main mechanism of action against Trypanosoma cruzi. In summary, we have successfully identified and characterized new nitrotriazole analogs, demonstrating their potential as promising candidates for the development of Chagas disease drug candidates that function via nitroreductase activation, are considerably selective and show no mutagenic potential.

RESUMO O objetivo geral deste artigo é analisar os fatores que definem a dinâmica da modernização da agropecuária no Brasil entre 2006 e 2017. Esse período caracteriza-se pela grande expansão da agropecuária no país, sendo que parcela ampla de sua produção se destinou à exportação. Considerando que a modernização é um fenômeno multifacetário, 24 variáveis relacionadas a ela foram calculadas para 137 mesorregiões e geraram cinco fatores, que mensuram: relação capital/trabalho (Fator 1); relação capital/terra (Fator 2); presença de infraestrutura e intensidade do suporte institucional (Fator 3); a intensidade do uso de insumos nas lavouras (Fator 4); e uso sustentável da terra (Fator 5). Não há distribuição igual dos escores desses cinco fatores nas mesorregiões do Brasil em 2006 ou em 2017, e as suas evoluções distintas, entre esses dois anos, nas mesorregiões do país evidenciam que o processo de modernização da agropecuária continua não sendo homogeneamente distribuído no território nacional nas duas primeiras décadas do século XXI.

Abstract Background Cysteine, a sulfur-containing amino acid, plays an important role in a variety of cellular functions such as protein biosynthesis, methylation, and polyamine and glutathione syntheses. In trypanosomatids, glutathione is conjugated with spermidine to form the specific antioxidant thiol trypanothione (T[SH]2) that plays a central role in maintaining intracellular redox homeostasis and providing defence against oxidative stress. Methods We cloned and characterised genes coding for a cystathionine β-synthase (CβS) and cysteine synthase (CS), key enzymes of the transsulfuration and assimilatory pathways, respectively, from the hemoflagellate protozoan parasite Trypanosoma rangeli. Results Our results show that T. rangeli CβS (TrCβS), similar to its homologs in T. cruzi, contains the catalytic domain essential for enzymatic activity. Unlike the enzymes in bacteria, plants, and other parasites, T. rangeli CS lacks two of the four lysine residues (Lys26 and Lys184) required for activity. Enzymatic studies using T. rangeli extracts confirmed the absence of CS activity but confirmed the expression of an active CβS. Moreover, CβS biochemical assays revealed that the T. rangeli CβS enzyme also has serine sulfhydrylase activity. Conclusion These findings demonstrate that the RTS pathway is active in T. rangeli, suggesting that this may be the only pathway for cysteine biosynthesis in this parasite. In this sense, the RTS pathway appears to have an important functional role during the insect stage of the life cycle of this protozoan parasite.
