Positron emission tomography (PET) is a widely utilized medical imaging modality that uses positron-emitting radiotracers to visualize biochemical processes in a living body. The spatiotemporal distribution of a radiotracer is estimated by detecting the coincidence photon pairs generated through positron annihilations. In human tissue, about 40% of the positrons form positroniums prior to the annihilation. The lifetime of these positroniums is influenced by the microenvironment in the tissue and could provide valuable information for better understanding of disease progression and treatment response. Currently, there are few methods available for reconstructing high-resolution lifetime images in practical applications. This paper presents an efficient statistical image reconstruction method for positronium lifetime imaging (PLI). We also analyze the random triple-coincidence events in PLI and propose a correction method for random events, which is essential for real applications. Both simulation and experimental studies demonstrate that the proposed method can produce lifetime images with high numerical accuracy, low variance, and resolution comparable to that of the activity images generated by a PET scanner with currently available time-of-flight resolution.

While the role of caveolae in membrane mechanosensation has been studied predominantly in non-cardiomyocyte cells, caveolae contribution to cardiac mechanotransduction remains elusive. Modern developments in data analysis techniques and evolutionary optimization algorithms have made it possible to analyze large amounts of unstructured digital data sets. Based on the differential evolution algorithm and semiclassical quantum simulations, we have recently proposed a method for classifying and analyzing the optical properties of organic pigments. In this paper, we present the results of modeling the absorption spectra of five carotenoids synthesized during the vital activity of the ascomycetous fungi: neurosporaxanthin, neurosporene, torulene, γ-carotene, and ζ-carotene. We calculated the absorption spectra for each pigment using the multimode Brownian oscillator theory, which allows us to evaluate the influence of molecular vibrations on the electronic transitions in the pigment. We applied a generalized spectral density function method to our modeling, taking into account the contributions of 13 vibrational modes with frequencies varying between 100 cm−1 and 3000 cm−1. This approach allowed us to gain a deeper understanding of how molecular vibrations affect the absorption spectra of these organic compounds. Thus, each absorption spectrum was associated with a unique set of Huang–Rhys factors (which represent the effective electron–phonon interaction). This set can be considered as a kind of “fingerprint” that characterizes the optical response of the pigment in the solvent.

In positron emission tomography (PET) studies, convolutional neural networks (CNNs) may be applied directly to the reconstructed distribution of radioactive tracers injected into the patient's body, as a pattern recognition tool. Nonetheless, unprocessed PET coincidence data exist in tabular format. This paper develops the transformation of tabular data into -dimensional matrices, as a preparation stage for classification based on CNNs. This method explicitly introduces a nonlinear transformation at the feature engineering stage and then uses principal component analysis to create the images. We apply the proposed methodology to the classification of simulated PET coincidence events originating from NEMA IEC and anthropomorphic XCAT phantom. Comparative studies of neural network architectures, including multilayer perceptron and convolutional networks, were conducted. The developed method increased the initial number of features from 6 to 209 and gave the best precision results (79.8) for all tested neural network architectures; it also showed the smallest decrease when changing the test data to another phantom.

In this study, we investigated the features of co-infection with SARS-CoV-2 and the enterovirus vaccine strain LEV8 of coxsackievirus A7 or enterovirus A71 for Vero E6 cells and Syrian hamsters. The investigation of co-infection with SARS-CoV-2 and LEV-8 or EV-A71 in the cell model showed that a competitive inhibitory effect for these viruses was especially significant against SARS-CoV-2. Pre-infection with enteroviruses in the animals caused more than a 100-fold decrease in the levels of SARS-CoV-2 virus replication in the respiratory tract and more rapid clearance of infectious SARS-CoV-2 from the lower respiratory tract. Co-infection with SARS-CoV-2 and LEV-8 or EV-A71 also reduced the severity of clinical manifestations of the SARS-CoV-2 infection in the animals. Additionally, the histological data illustrated that co-infection with strain LEV8 of coxsackievirus A7 decreased the level of pathological changes induced by SARS-CoV-2 in the lungs. Research into the chemokine/cytokine profile demonstrated that the studied enteroviruses efficiently triggered this part of the antiviral immune response, which is associated with the significant inhibition of SARS-CoV-2 infection. These results demonstrate that there is significant viral interference between the studied strain LEV-8 of coxsackievirus A7 or enterovirus A71 and SARS-CoV-2 in vitro and in vivo.

Abstract Despite the progress made in the treatment of nephrolithiasis, the existing methods of renal calculi destruction are not ideal and have both advantages and disadvantages. Considering the process of high-frequency glow discharge formation on the surface of an electrode and in an electrolyte solution, we obtained the results on the destruction of renal calculi in vitro. It was shown that the destruction of kidney stones by glow discharge plasma was caused by several processes—the plasma induced effect of hydrated electrons and shock wave effect of the electrolyte stimulated by electrical breakdowns in the plasma. The plasma generation modes were configured by estimating the thickness of the vapor–gas layer in which the plasma burns. Thus, the average rate of contact destruction of renal calculi was measured depending on the plasma generator input power and time of plasma exposure. We conclude that the method of stone fragmentation by high-frequency electrolyte plasma is rather perspective and can be used in endoscopic urology for percutaneous and transurethral lithotripsy.

Water distribution in the structure of vein quartz formed as a result of successive plastic deformations associated with dislocation slip and subsequent recrystallization was estimated using infrared microspectroscopic (μFTIR) mapping. Water contained in quartz demonstrates a broad absorption band in the IR range at 2800–3750 cm−1, which indicates its molecular state and suggests the presence of water bearing water inclusions. In addition to water, the presence of an absorption band located at 2341 cm−1 seems to be due to the presence of carbon dioxide in a molecular state. A necessary step before assessing the distribution of volatile components in the quartz structure was to calibrate the boundaries obtained by calculating the intensity ratios of the peaks at 1118 and 1160 cm−1 in the reflectance spectrum and using electron back scatter diffraction (EBSD). A variety of fluid distributions in different elements of the structure was observed. At medium temperatures and medium strain rates, dislocation mass transfer is effective during dislocation slip. At low strain rates and elevated temperatures, the contribution of diffusion creep gradually increases, which facilitates the interaction of volatile components with migrating boundaries. It was found that in the process of successive rearrangements, migration of fluid components occurs within the main elements of the structure due to the redistribution of dislocations between defects of different scale levels. Redistribution of fluid from fluid inclusions as a result of plastic deformations in the quartz structure is one of the ways of relaxing intracrystalline stresses during strengthening of the structure. The possibility of pigment detection and recognition in different environments such as solvents or proteins is a challenging, and at the same time demanding, task. It may be needed in very different situations: from the nondestructive in situ identification of pigments in paintings to the early detection of fungal infection in major agro-industrial crops and products. So, we propose a prototype method, the key feature of which is a procedure analyzing the lineshape of a spectrum. The shape of the absorption spectrum corresponding to this transition strongly depends on the immediate environment of a pigment and can serve as a marker to detect the presence of a particular pigment molecule in a sample. Considering carotenoids as an object of study, we demonstrate that the combined operation of the differential evolution algorithm and semiclassical quantum modeling of the optical response based on a generalized spectral density (the number of vibronic modes is arbitrary) allows us to distinguish quantum models of the pigment for different solvents. Moreover, it is determined that to predict the optical properties of monomeric pigments in protein, it is necessary to create a database containing, for each pigment, in addition to the absorption spectra measured in a predefined set of solvents, the parameters of the quantum model found using differential evolution.

Abstract The lifetime of electric propulsion (EP) thrusters depends particularly on the erosion characteristics of operation relevant components, for instance, the grid hole erosion of gridded ion thrusters or the channel wall erosion of Hall effect thrusters. Here two tools for in situ erosion measurements are presented, a triangular laser head for surface profiling and a telemicroscope for high-resolution optical imaging. Both can give access to radial and axial erosion parameters. The measurements can be done in situ without the need for breaking the vacuum and dismounting the thruster, which reduces thruster testing time considerably. In situ measurements can also help to ensure reproducibility of thruster performance conditions and can improve statistics of thruster characterization. The present work describes the fundamentals of both techniques in detail, selected experimental setups are presented, their performance is characterized and critically evaluated. The capabilities and limitations related to erosion measurements of EP thrusters are, exemplary, demonstrated for a gridded ion thruster RIT-22 and a Hall effect thruster SPT-100D.

In this paper, a procedure for obtaining undistorted high derivatives (up to the eighth order) of the optical absorption spectra of biomolecule pigments has been developed. To assess the effectiveness of the procedure, the theoretical spectra of bacteriochlorophyll a, chlorophyll a, spheroidene, and spheroidenone were simulated by fitting the experimental spectra using the differential evolution algorithm. The experimental spectra were also approximated using sets of Gaussians to calculate the model absorption spectra. Theoretical and model spectra can be differentiated without smoothing (high-frequency noise filtering) to obtain high derivatives. Superimposition of the noise track on the model spectra allows us to obtain test spectra similar to the experimental ones. Comparison of the high derivatives of the model spectra with those of the test spectra allows us to find the optimal parameters of the filter, the application of which leads to minimal differences between the high derivatives of the model and test spectra. For all four studied pigments, it was shown that smoothing the experimental spectra with optimal filters makes it possible to obtain the eighth derivatives of the experimental spectra, which were close to the eighth derivatives of their theoretical spectra.

The treatment of many bacterial and fungal infections remains a problem due to increasing antibiotic resistance and biofilm formation by pathogens. In the present article, a methodology for the chemoselective synthesis of 2-(1H-indol-3-yl)-1H-benzo[d]imidazole derivatives is presented. We report on the antimicrobial activity of synthesized 2-(1H-indol-3-yl)-1H-benzo[d]imidazoles with significant activity against Staphylococcus aureus ATCC 25923, Staphylococcus aureus ATCC 43300 (MRSA), Mycobacterium smegmatis (mc(2)155/ATCC 700084), and Candida albicans ATCC 10231. High activity against staphylococci was shown by indolylbenzo[d]imidazoles 3ao and 3aq (minimum inhibitory concentration (MIC) < 1 µg/mL) and 3aa and 3ad (MIC 3.9–7.8 µg/mL). A low MIC was demonstrated by 2-(1H-indol-3-yl)-1-methyl-1H-benzo[d]imidazole (3ag) against M. smegmatis and against C. albicans (3.9 µg/mL and 3.9 µg/mL, respectively). 2-(5-Bromo-1H-indol-3-yl)-6,7-dimethyl-1H-benzo[d]imidazole (3aq) showed a low MIC of 3.9 µg/mL against C. albicans. Compounds 3aa, 3ad, 3ao, and 3aq exhibited excellent antibiofilm activity, inhibiting biofilm formation and killing cells in mature biofilms. Molecular docking analysis identified three potential interaction models for the investigated compounds, implicating (p)ppGpp synthetases/hydrolases, FtsZ proteins, or pyruvate kinases in their antibacterial action mechanism.
