A system of 5,020 robotic fiber positioners was installed in 2019 on the Mayall Telescope, at Kitt Peak National Observatory. The robots automatically re-target their optical fibers every 10 - 20 minutes, each to a precision of several microns, with a reconfiguration time less than 2 minutes. Over the next five years, they will enable the newly-constructed Dark Energy Spectroscopic Instrument (DESI) to measure the spectra of 35 million galaxies and quasars. DESI will produce the largest 3D map of the universe to date and measure the expansion history of the cosmos. In addition to the 5,020 robotic positioners and optical fibers, DESI's Focal Plane System includes 6 guide cameras, 4 wavefront cameras, 123 fiducial point sources, and a metrology camera mounted at the primary mirror. The system also includes associated structural, thermal, and electrical systems. In all, it contains over 675,000 individual parts. We discuss the design, construction, quality control, and integration of all these components. We include a summary of the key requirements, the review and acceptance process, on-sky validations of requirements, and lessons learned for future multi-object, fiber-fed spectrographs., Comment: 51 pages, 41 figures

Artificial intelligence (AI) solutions that automatically extract information from digital histology images have shown great promise for improving pathological diagnosis. Prior to routine use, it is important to evaluate their predictive performance and obtain regulatory approval. This assessment requires appropriate test datasets. However, compiling such datasets is challenging and specific recommendations are missing. A committee of various stakeholders, including commercial AI developers, pathologists, and researchers, discussed key aspects and conducted extensive literature reviews on test datasets in pathology. Here, we summarize the results and derive general recommendations for the collection of test datasets. We address several questions: Which and how many images are needed? How to deal with low-prevalence subsets? How can potential bias be detected? How should datasets be reported? What are the regulatory requirements in different countries? The recommendations are intended to help AI developers demonstrate the utility of their products and to help regulatory agencies and end users verify reported performance measures. Further research is needed to formulate criteria for sufficiently representative test datasets so that AI solutions can operate with less user intervention and better support diagnostic workflows in the future.

Within the last decade, we have been able to observe our universe with incredibly high precision. Nevertheless, there are theoretical problems that still do not have an answer. In this thesis, we have studied the issue of the initial conditions of inflation, and we proposed a model for which they arise naturally as an attractor. It is achieved by considering the interaction of the inflaton with a thermal bath of particles, which are in equilibrium with the horizon temperature. If inflation happens at high energies, the electroweak vacuum is unstable. Therefore, we studied how fermions help make the Standard Model more stable, by calculating their gravitational creation during inflation.

10. Assessing the In Vitro and In Vivo Effect of Supplementation with a Garlic (Allium sativum) and Oregano (Origanum vulgare) Essential Oil Mixture on Digestibility in West African Sheep

This study assessed the impact of a mixture of garlic (Allium sativum) and oregano (Origanum vulgare) essential oils (EOGOs) on in vitro dry matter digestibility (IVDMD) and in vivo apparent nutrient digestibility. Different EOGO inclusion levels were evaluated to assess the dose response and potential effects of the mixture. Three EOGO inclusion levels (0.5, 0.75, and 1 mL/kg of incubated dry matter) were evaluated in vitro, while four treatments (0.5, 0.75, and 1 mL/day of EOGO and a control group) were tested in vivo on 12 West African sheep. A randomized controlled trial was conducted using a 4 × 4 design. Blood parameters (glucose, blood urea nitrogen, and β-hydroxybutyrate) were measured to observe the effect of EOGO on the metabolism. The results showed that the inclusion of EOGO significantly enhanced IVDMD at low levels (p < 0.052) compared with the highest levels in treatments containing 0.5 and 0.75 mL/kg of EOGO dry matter. A higher intake of dry matter (DM), crude protein (CP), and neutral detergent fiber (NDF) (p < 0.05) was observed in the in vivo diets with the inclusion of EOGO. In terms of in vivo apparent digestibility, significant differences were found among treatments in the digestibility coefficients of DM, CP, and NDF. EOGO inclusion increased the digestibility of DM. CP digestibility displayed a cubic effect (p < 0.038), with the lowest values of digestibility observed at 1 mL EOGO inclusion. Additionally, NDF digestibility showed a cubic effect (p < 0.012), with the highest value obtained at 0.75 mL of EOGO inclusion. The inclusion levels above 0.75 mL EOGO showed a cubic effect, which indicates that higher concentrations of EOGO may not be beneficial for the digestibility of CP and NDF. Although no significant difference was observed in total digestible nutrients, a linear trend was observed (p < 0.059). EOGO improved the intake of DM, CP, and NDF. EOGO supplementation improved the digestibility of DM and NDF, with optimal levels observed at 0.5 mL/day. No significant effects were observed in the blood parameters. These results suggest that EOGO has the potential as an additive in ruminal nutrition to improve food digestibility and serve as an alternative to antibiotic additives. The use of EOGO potentially improves fiber digestion and may reduce the use of antibiotics in livestock production. Garlic (A. sativum) and oregano (O. vulgare) essential oils effectively modulated fiber digestibility at 0.75 mL/day. Garlic (A. sativum) and oregano (O. vulgare) essential oils have the potential to improve digestibility at low inclusion levels and serve as an alternative to antibiotic additives. The effectiveness of essential oils is greater in a mixture and at lower doses.

11. Gravitationally produced Top Quarks and the Stability of the Electroweak Vacuum During Inflation

In the standard model the (Brout-Englert-)Higgs quartic coupling becomes negative at high energies rendering our current electroweak vacuum metastable, but with an instability timescale much longer than the age of the Current Universe. During cosmological Inflation, unless there is a non-minimal coupling to gravity, the Higgs field is pushed away from the origin of its potential due to quantum fluctuations. It is therefore a mystery how we have remained in our current vacuum if we went through such a period of Inflation. In this work we study the effect of top quarks created gravitationally during Inflation and their effect upon the Higgs potential using only General Relativity with minimal couplings and Standard Model particle physics. We show how the evolution of the Higgs field during Inflation is modified coming to the conclusion that this effect is non negligible for scales of Inflation close to or larger than the stability scale but small for scales where the Higgs is stable. We consider the effect of the Gibbons-Hawking radiation on the inflaton in the situation where it is coupled to a large number of spectator fields. We argue that this will lead to two important effects - a thermal contribution to the potential and a gradual change in parameters in the Lagrangian which results from thermodynamic and energy conservation arguments. We present a scenario of hilltop inflation where the field starts trapped at the origin before slowly experiencing a phase transition during which the field extremely slowly moves towards its zero temperature expectation value. We show that it is possible to obtain enough e-folds of expansion as well as the correct spectrum of perturbations without hugely fine-tuned parameters in the potential (albeit with many spectator fields). Abstract Background In severe cases, SARS-CoV-2 infection leads to acute respiratory distress syndrome (ARDS), often treated by extracorporeal membrane oxygenation (ECMO). During ECMO therapy, anticoagulation is crucial to prevent device-associated thrombosis and device failure, however, it is associated with bleeding complications. In COVID-19, additional pathologies, such as endotheliitis, may further increase the risk of bleeding complications. To assess the frequency of bleeding events, we analyzed data from the German COVID-19 autopsy registry (DeRegCOVID). Methods The electronic registry uses a web-based electronic case report form. In November 2021, the registry included N = 1129 confirmed COVID-19 autopsy cases, with data on 63 ECMO autopsy cases and 1066 non-ECMO autopsy cases, contributed from 29 German sites. Findings The registry data showed that ECMO was used in younger male patients and bleeding events occurred much more frequently in ECMO cases compared to non-ECMO cases (56% and 9%, respectively). Similarly, intracranial bleeding (ICB) was documented in 21% of ECMO cases and 3% of non-ECMO cases and was classified as the immediate or underlying cause of death in 78% of ECMO cases and 37% of non-ECMO cases. In ECMO cases, the three most common immediate causes of death were multi-organ failure, ARDS and ICB, and in non-ECMO cases ARDS, multi-organ failure and pulmonary bacterial ± fungal superinfection, ordered by descending frequency. Interpretation Our study suggests the potential value of autopsies and a joint interdisciplinary multicenter (national) approach in addressing fatal complications in COVID-19.

Regulators of G-protein signaling (RGS) are a family of approximately 30 proteins that bind to and deactivate the alpha subunits of G-proteins (Gα) by accelerating their GTP hydrolysis rates, which terminates G-protein coupled receptor (GPCR) signaling. Thus, RGS proteins are essential in regulating GPCR signaling, and most members are implicated as critical nodes in human diseases such as hypertension, depression, and others. Regulator of G-protein signaling 2 (RGS2), a member of the R4 family of RGS proteins, is overexpressed in many solid breast cancers, and its levels in prostate cancer significantly correlate with the metastatic stage and poor prognosis. We sought to develop RGS2 inhibitors as potential chemotherapeutic agents utilizing structure-based drug design approaches. Available structures of the RGS2-Gαcomplex were used to extract a pharmacophore model for searching chemical databases. Docking of identified hits to RGS2 as well as other RGS structures was used to screen the hits for potent and selective RGS2 inhibitors. Whole cell assays showed the top 10 ranking compounds, AJ-1–AJ-10, to inhibit RGS2–Gαqinteractions. Differential scanning fluorimetry showed AJ-3 to bind RGS2 but not Gαq. All 10 compounds inhibited the growth of several RGS2 expressing cancers in cell culture assays. In addition, AJ-3 inhibited the migration of LNCaP prostate cancer cells in wound healing assays. This is the first group of RGS2 inhibitors identified by structure-based approaches and that show anticancer activity. These results highlight the potential RGS2 inhibitors have to be a new class of chemotherapeutic agents.

Chemically pure plastic granulate is used as the starting material in the production of plastic parts. Extrusion machines rely on purity, otherwise resources are lost, and waste is produced. To avoid losses, the machines need to analyze the raw material. Spectroscopy in the visible and near-infrared range and machine learning can be used as analyzers. We present an approach using two spectrometers with a spectral range of 400–1700 nm and a fusion model comprising classification, regression, and validation to detect 25 materials and proportions of their binary mixtures. one dimensional convolutional neural network is used for classification and partial least squares regression for the estimation of proportions. The classification is validated by reconstructing the sample spectrum using the component spectra in linear least squares fitting. To save time and effort, the fusion model is trained on semi-empirical spectral data. The component spectra are acquired empirically and the binary mixture spectra are computed as linear combinations. The fusion model achieves very a high accuracy on visible and near-infrared spectral data. Even in a smaller spectral range from 400–1100 nm, the accuracy is high. The visible and near-infrared spectroscopy and the presented fusion model can be used as a concept for building an analyzer. Inexpensive silicon sensor-based spectrometers can be used.

Spectrometers measure diffuse reflectance and create a “molecular fingerprint” of the material under investigation. Ruggedized, small scale devices for “in-field” use cases exist. Such devices might for example be used by companies in the food supply chain for inward inspection of goods. However, their application for the industrial Internet of Things workflows or scientific research is limited due to their proprietary nature. We propose an open platform for visible and near-infrared technology (OpenVNT), an open platform for capturing, transmitting, and analysing spectral measurements. It is built for use in the field, as it is battery-powered and transmits data wireless. To achieve high accuracy, the OpenVNT instrument contains two spectrometers covering a wavelength range of 400–1700 nm. We conducted a study on white grapes to compare the performance of the OpenVNT instrument against the Felix Instruments F750, an established commercial instrument. Using a refractometer as ground truth, we built and validated models to estimate the Brix value. As a quality measure, we used coefficient of determination of the cross-validation (R2CV) between the instrument estimation and ground truth. With 0.94 for the OpenVNT and 0.97 for the F750, a comparable R2CV was achieved for both instruments. OpenVNT matches the performance of commercially available instruments at one tenth of the price. We provide an open bill of materials, building instructions, firmware, and analysis software to enable research and industrial IOT solutions without the limitations of walled garden platforms.

Typical electroless copper baths (ECBs), which are used to chemically deposit copper on printed circuit boards, consist of an aqueous alkali hydroxide solution, a copper(II) salt, formaldehyde as reducing agent, an l-(+)-tartrate as complexing agent, and a 2,2′-bipyridine derivative as stabilizer. Actual speciation and reactivity are, however, largely unknown. Herein, we report on the synthesis and crystal structure of aqua-1κO-bis(4,4′-dimethoxy-2,2′-bipyridine)-1κ2N,N′;2κ2N,N′-[μ-(2R,3R)-2,3-dioxidosuccinato-1κ2O1,O2:2κ2O3,O4]dicopper(II) octahydrate, [Cu2(C12H12N2O2)2(C4H2O6)(H2O)]·8H2O, from an ECB mock-up. The title compound crystallizes in the Sohncke group P21 with one chiral dinuclear complex and eight molecules of hydrate water in the asymmetric unit. The expected retention of the tartrato ligand's absolute configuration was confirmed via determination of the absolute structure. The complex molecules exhibit an ansa-like structure with two planar, nearly parallel bipyridine ligands, each bound to a copper atom that is connected to the other by a bridging tartrato `handle'. The complex and water molecules give rise to a layered supramolecular structure dominated by alternating π stacks and hydrogen bonds. The understanding of structures ex situ is a first step on the way to prolonged stability and improved coating behavior of ECBs.

High forage diets in the tropics are less efficient and more contaminating due to high energy losses as methane and the low digestibility of tropical forages. The objective of the study was to determine the effects of essential oils (EO) of Anacardium occidentale and Ricinus communis as additives in high forage diets on intake, digestibility, ruminal fermentation and synthesis of microbial protein. Four ‘Holstein’ steers fitted with ruminal cannula were used in a 4 × 4 Latin square design (21-day period). The treatments were 1, 2, 4 and 8 g day-1 EO per animal (essential oils, Oligobasics®). The diet was total mixed ration (TMR) of Brachiaria hay (Brachiaria humidicola cv. Lanero) and concentrate (ground corn, soybean meal, ammonium sulphate, urea and minerals) in an 80:20 ratio. Differences were observed in the digestibility of neutral detergent fibre (NDF) and total digestible nutrients (TDN) in levels above 2 g day -1 EO (P < 0.05). The VFA (volatile fatty acids) acetate and propionate were higher in diets with 2 g day -1 EO (P < 0.05), without affecting the acetate:propionate ratio. Levels above 2 g day -1 EO negatively affected the faecal N excretion (P < 0.05). The inclusions of A. occidentale and R. communis EOs at 2 g day-1 per day animal improved fibre digestion and decreased N excretion. These results indicate that this mixture of EOs may have strong antimicrobial activity at lower doses. The N losses, fibre digestion and digestibility could be improved with the use of cashew and castor essential oils in high forage diets.

This dissertation studies the effect of different policy interventions on allocative efficiency in developing countries. It evaluates how urban and antitrust policies may generate a better allocation of resources across firms, increasing aggregate welfare, and productivity. To provide causal evidence, it collects rich microdata at a very high granular level and exploits plausibly exogenous variation of the main mechanisms. It complements the main empirical findings with quantitative general equilibrium models that allow simulating the effect of different policy interventions.One of the main factors that explain factor misallocation in developing countries is the informal sector. The fact that firms face different tax schedules generate differences in marginal products of labor. The first chapter of my dissertation proposes a new mechanism to account for the significant presence of the informal economy in developing countries: the high commuting costs to transit within cities. I provide evidence of three different empirical facts that align with this hypothesis. First, due to the high commuting costs, most workers in Mexico City have poor access to formal employment. Second, workers that operate in the informal economy are more sensitive to commuting costs, which implies that it is easier to substitute informal jobs. Third, I exploit the construction of line B of the subway in Mexico City to provide causal evidence of the negative relationship between informality and transit improvements. I estimate a series of difference-in-differences specifications, finding that transit improvements lead to a reduction in informality rates by four percentage points in nearby areas to the new stations relative to other places in Mexico City. This result indicates that workers reallocate to firms with higher total factor revenue productivity, increasing the welfare effects of transit improvements relative to the estimates from previous literature.A fundamental question for economists and policymakers is to measure the welfare effects of transit infrastructure that reduce transit times within a city. While the literature has estimated the `direct'' effects of these projects, there are substantial ``indirect'' effects that previous work has ignored. The second chapter develops a spatial general equilibrium model to account for the ``direct'' effects and also for the "indirect" effects driven by the reallocation of workers from the informal into the formal economy. I extend recent theoretical work in the urban literature by adding distortions that generate resource misallocation. From a first-order approximation, I provide a formula that decomposes the welfare effects of trade and commuting shocks into a ``pure'' effect term and an allocative efficiency margin. I estimate the main elasticities of the model exploiting variation from transit shocks. With these parameters at hand, I simulate different counterfactual interventions. The main findings suggest that line B of the subway increased welfare between 1.3\% and 1.6\%, that the indirect effects explain approximately 15-25\% of the total gains, and that the average real income per every dollar spent on infrastructure increases by 15\% relative to a perfectly efficient economy. In addition to the informal sector, there can be other sources of resource misallocation in developing countries. The third chapter, co-authored with Dario Tortarolo, focuses on the role of market power in explaining differences in marginal products of labor across plants. We disentangle the extent of imperfect competition in product and labor markets, assuming cost-minimizing firms that face upward-sloping labor supply and downward-sloping product demand curves. In the first part, we derive a formula for the ratio between markups and markdowns. We compute this ratio by estimating the output elasticity with respect to labor and the wage bill share. We disentangle the measure of market power specific to each firm by estimating labor supply elasticities using as an instrument intermediate inputs. Our results suggest that both markets exhibit imperfect competition, but markups mainly drive the variation. We assume a general equilibrium model to measure the relative gains of removing market power dispersion on allocative efficiency. The findings suggest that markups are more critical in explaining TFP than markdowns.
