Basolateral and brush-border membranes were prepared from the intestines and kidneys of spontaneously hypertensive (SHR) and normotensive (WKY) rats fed on a calcium-adequate diet and assayed for their enzyme activities. In intestinal basolateral membranes the activities of Na+ K(+)-ATPase (EC 3.6.1.37) Ca2(+)-ATPase (EC 3.6.1.38) and alkaline phosphatase (EC 3.1.3.1) were lower in SHR rats when compared with WKY rats, whilst 5'-nucleotidase (EC 3.1.3.5) (a marker for basolateral membranes) was unaffected. In kidney basolateral membranes all enzymes were similar in activity in SHR and WKY rats. In intestinal brush-border membranes the activities of Ca2(+)-ATPase and alkaline phosphatase were lower in SHR rats when compared with WKY rats, whilst microvillus aminopeptidase (EC 3.4.11.2) (a marker for brush-border membranes) was unaffected. In kidney brush-border membranes all enzymes were similar in activity in SHR and WKY rats. The blood pressures of the SHR rats were considerably higher than those of the WKY rats. When SHR rats were fed on a Ca-deficient diet the activities of Na+K(+)-ATPase, Ca2(+)-ATPase and alkaline phosphatase in basolateral membranes and Ca2(+)-ATPase and alkaline phosphatase in brush-border membranes were all increased in the intestine when compared with SHR rats fed on a Ca-adequate diet. The equivalent enzymes in the kidneys of SHR rats, and the intestines and kidneys of WKY rats, were not affected by altering the Ca in the diet. The blood pressures of SHR rats fed on a Ca-deficient diet were higher than in those fed on a Ca-adequate diet. Blood pressures of WKY rats were not affected by altering the diet in this way. The results indicate that the absorption of Ca by active mechanisms may be reduced in SHR rats compared with WKY rats. Changing the level of Ca in the diet modified both blood pressure and the activities of enzymes which catalyse active Ca transport. The implications of these results to the aetiology, and possible nutritional treatment, of essential hypertension are discussed.

1. Groups of chicks were given a low-residue diet with or without supplements of dietary fibre in the form of wheat bran, wheat straw or bagasse. Growth and food conversion efficiency (g weight gained/g food eaten; FCE) during the first 4 weeks of life were measured. 2. In every one of seven experiments supplementation of the diet with 100 g wheat bran/kg resulted in improved growth, and in three experiments FCE was also increased. 3. Supplementation with coarsely-milled wheat straw to provide an amount of unavailable carbohydrate equivalent to that in the bran diet resulted in poorer growth; finely-milled wheat straw had little effect on growth. 4. The growth-promoting effect of bran was destroyed by sterilization with heat or gamma-radiation. 5. In some experiments weights, lengths and volumes of small intestines were measured. Differences in intestinal dimensions between birds given the diet with and without fibre were not consistent, nor were they correlated with growth rate or FCE. 6. Histometric observations on small intestines from a few birds indicated that those given coarse wheat straw had longer vili and thicker muscularis layers, and the caecal tonsils had a greater area of lymphoid tissue and more follicles.

1. The food intake, pancreas weight and trypsin (EC 3.4.21.4) and alpha-chymotrypsin (EC 3.4.21.1) activities in the pancreas were measured in rats during pregnancy and lactation and after the young were weaned. 2. All the quantities measured increased significantly during lactation and had returned to their original values by 4 weeks after weaning. Food intake and pancreas weight were highest after the second week of lactation. Total trypsin and alpha-chymotrypsin activity, and the activity per g tissue, fell during pregnancy and rose during lactation, reaching a maximum 1 week after weaning. 3. From these and other results it is suggested that the hypertrophy and hypersecretion of pregnancy and lactation are initiated by changes insulin secretion and mediated by the trophic effects of gut hormones, and that differences in the nature and timing of the response may be controlled by nutrient availability.

Changes in milk composition associated with maternal dietary obesity and cafeteria feeding were investigated. Protein, lactose and fat contents, and the fat composition, were determined for lean and obese rats given a cafeteria diet at different stages of reproduction. Feeding the cafeteria diet during lactation resulted in an increase in long-chain fatty acids and a fall in the characteristic medium-chain fatty acids. This effect was modified by obesity and the diet during pregnancy. Feeding the cafeteria diet in lactation reduced the milk protein and increased the fat. The milk of obese rats contained more energy, with more fat but less protein than that of lean rats. Increases in fat and long-chain fatty acid content, and decreases in protein and medium-chain fatty acid content of the milk were correlated with increased maternal intake of energy, total fat and long-chain fatty acids. Thus, the greatest influence on milk composition is exerted by the maternal diet during lactation. However, these effects are modified by pre-existing maternal obesity and the diet during pregnancy.

1. The renewal of the intestinal mucosal epithelium has been investigated in germ-free and conventional chicks given a practical chick diet and a low- and high-fibre diet, using tritiated thymidine to label the nuclei of mucosal cells undergoing division. 2. Villus height, crypt depth and number of mitoses in the crypt were generally, although not always significantly, greater for conventional chicks than for germ-free chicks at all sites investigated in the intestine, and all became progressively lower from the upper intestine to the lower intestine. 3. There was a linear relationship between the height of the radioactively labelled mucosal cells on the villus and the period after injection, and the rates of epithelial cell migration were higher for conventional than for germ-free chicks, the relative differences being greater in the lower intestine than the upper intestine. 4. The inclusion of wheat bran as a source of dietary fibre had no effect on the rate of epithelial renewal, either in germ-free or conventional birds.

1. Rats were mated and at weekly intervals during pregnancy and lactation, and after weaning, the dipeptidase activity in the supernatant fraction from small intestinal mucosa extracts was determined for two dipeptides: glycyl-L-leucine dipeptidase (EC 3.4.3.2) AND L-alanyl-L-glutamic acid dipeptidase. 2. Dipeptidase activity is found mainly in the soluble (spernantant) fraction of the mucosa homogenate. 3. Compared with those values obtained for unmated controls, the food consumption of the animals and the nitrogen content, total and specific activities of the dipeptidases (per unit quantity of N) in the soluble fraction of the small intestinal mucosa extracts increased slightly during pregnancy and markedly during lactation. After the pups were weaned, values for all these measurements fell rapidly. 4. Whether the increases found in enzyme activity were simply a response to increased food intake or were the result of hormonal stimulus is discussed.

The metabolic enzymes alkaline phosphatase (EC 3.1.3.1), acid phosphatase (EC 3.1.3.2) and isocitrate dehydrogenase (EC 1.1.1.42) were measured in mucosal homogenates and these enzymes, together with glucose-6-phosphatase (EC 3.1.3.9), were measured in homogenates of isolated enterocytes from germ-free (GF) and conventional (CV) chicks which were either fed continuously until they were killed or were subjected to a 16 h fast before killing. The intestine of the GF chicks was generally lighter than that of the CV controls. The activity of alkaline phosphatase was greater in the mucosal homogenates of the CV chicks compared with the GF birds, but the concentrations of acid phosphatase and isocitric dehydrogenase were not different in the two groups. In the isolated enterocytes the concentration of all enzymes expressed per mg DNA, except alkaline phosphatase, was higher in the GF chicks. Expressed per mg protein there was no significant difference in enzyme activity in the two groups. Fasting caused a reduction in intestinal weight and total mucosal protein in both groups but the reduction was greater in the GF chicks compared with the CV controls. In the GF chicks, fasting caused a significant fall in acid phosphatase and isocitric dehydrogenase activities of the mucosal homogenate, whereas in the CV chicks only acid phosphatase fell to a significant extent. In the isolated enterocytes feeding caused a marked fall in protein per mg DNA in the CV chicks; fasting tended to reduce enzyme concentrations in the GF chicks but to have less effect in the CV group except for alkaline phosphatase where there was a marked rise in activity. It is suggested that the difference in enzyme activities in the mucosal homogenates and isolated enterocytes might resulted from (a) the presence of a much greater lamina propria in the CV compared with the GF chicks and (b) the greater mitotic activity in the fed CV chicks yielding a much larger number of smaller immature cells.

1. Groups of adult colostimized chickens were given diets with and without dietary fibre in the form of bagasse or wheat straw. The fibrous materials were analysed for their contents of cellulose, hemicellulose and lignin. The digestible energies (DE) and metabolizable energies (ME) of these diets were measured by a balance method. 2. Groups of germ-free and conventional chicks aged 4 weeks were given diets with and without penicillin or with and without graded levels of wheat straw as a source of fibre and the ME of these diets were determined. 3. The incorporation of the forms of dietary fibre tested reduced the DE and the ME of the diets in adult conventional chickens and the ME in young germ-free and conventional chicks. Both DE and ME were reduced in proportions that suggested that the dietary fibre present was acting largely as an inactive diluent of the dietary energy and was not affecting the absorption of other nutrients. In the young germ-free and conventional chicks, there was a linear relationship between the proportion of wheat straw in one diet and the reduction ME. 4. Dietary penicillin did not alter the ME of the diet in either the germ-free or the conventional environments. 5. The reduction of ME with incorporation of wheat straw was less in conventional than in germ-free chicks. It is suggested that this may be due to the ability of chicks with a gut flora to obtain a small amount of energy from wheat straw.

1. The uptake transport and secretion of calcium into the gastrointestinal tract of germ-free (GF) and conventional (CV) chicks was determined by a variety of in vivo and in vitro techniques using 45Ca as a marker following a standard fast-fed regimen. 2. Following an oral dose of Ca in conscious chicks, the transit time of digesta in the GF birds was appreciably faster than in the CV controls. 3. Although the proportion of the Ca dose absorbed was similar in both groups of chicks, the accumulation of Ca in the intestinal wall was much greater in CV birds. Distribution of the labelled Ca in plasma and bone was similar in both groups. 4. Following intravenous CA injection in conscious chicks in Ca secretion into the intestinal lumen was similar in both groups, but the amount appearing in the gut wall was appreciably greater in CV chicks. 5. There were regional differences in the ability of the gut to absorb Ca. There was a tendency for Ca uptake to be greater in the GF chicks but only in the proximal ileum did this approach significance. 6. When the Ca was confined to an intestinal loop the uptake/g tissue was similar in the proximal ileum of both groups but significantly greater in the distal ileum of the GF group. Uptake/10 mm gut was similar in both groups at both the proximal and the distal size. 7. It is suggested that the absence of a microbial load does not affect the ability of the enterocyte to absorb Ca but the subsequent removal of the absorbed load from the intestinal wall is appreciably slower in the CV group.

A balance method for the measurement of digestible and metabolizable energy in rats is described. Experimentally determined metabolizable energies of 2 diets for rats were closer to those values for metabolizable energy calculated from tabulated values for pigs rather than chickens. Some drawbacks in the use of tables of energy values to predict the energy of diets are discussed.

In 4-week-old chicks that had been maintained on 3 different diets, caecal weights were lower in germ-free than in conventional birds eating practical but not semipurified diets. There were no consistent differences in total sodium, potassium and chloride ions in the contents of the small intestine. In germ-free chicks the caecal wall muscle appeared lacking in tone and there was a lower concentration of chloride ions in the contents, but there was no caecal enlargement comparable to that in germ-free rodents.

The food intake, gut weight, gut length, mucosal protein and mucosal activities of alkaline phosphate (EC 3.1.3.1), acid phosphate (EC 3.1.3.2), isocitric dehydrogenase (EC 1.1.1.42) and glucose-6-phosphate (EC 3.1.3.9) were measured in rats during pregnancy, lactation and after the young were weaned. In general, the quantities measured increased slightly during pregnancy and considerably during lactation, reaching maximum values during the 3rd weeks of lacation and falling more or less rapidly after the young were weaned to the same levels as those in unmated animals. However, the gut length and mucosal protein remained higher even 3 weeks after weaning, so that weight per unit length and specific enzyme activities (per mg protein) tended to be lower in mated than in unmated rats. Changes in the specific activities of enzymes indicate alterations of the metabolic function of the enterocytes during breeding similar to changes reported for digestive enzymes. It is suggested that the intestine may reflect changes that take place in the liver.
