Abstract

The availability of metabolic intermediates is a prerequisite in many fields ranging from basic research, to biotechnological and biomedical applications as well as diagnostics. 2-keto-3-deoxy-6-phosphogluconate (KDPG) is the key intermediate of the Entner-Doudoroff (ED) pathway for sugar degradation and of sugar acid and sugar polymer breakdown in many organisms including human and plant pathogens. However, so far KDPG is hardly available due to missing efficient synthesis routes. We here report the efficient biocatalytic KDPG production through enzymatic dehydration of 6-phosphogluconate (6PG) up to gram scale using the 6PG dehydratase/Entner-Doudoroff dehydratase (EDD) from Caulobacter crescentus (CcEDD). The enzyme was recombinantly produced in Escherichia coli, purified to apparent homogeneity in a simple one-step procedure using nickel ion affinity chromatography, and characterized with respect to molecular and kinetic properties. The homodimeric CcEDD catalyzed the irreversible 6PG dehydration to KDPG with a Vmax of 61.6 U mg–1 and a KM of 0.3 mM for 6PG. Most importantly, the CcEDD showed sufficient long-term stability and activity to provide the enzyme in amounts and purity required for the efficient downstream synthesis of KDPG. CcEDD completely converted 1 g 6PG and a straight forward purification method yielded 0.81 g of stereochemically pure KDPG corresponding to a final yield of 90% as shown by HPLC-MS and NMR analyses.

Abstract

Neuroblastoma (NB) is a neural crest-derived childhood tumor characterized by a remarkable phenotypic diversity, ranging from spontaneous regression to fatal metastatic disease. Although the cancer stem cell (CSC) model provides a trail to characterize the cells responsible for tumor onset, the NB tumor-initiating cell (TIC) has not been identified. In this study, the relevance of the CSC model in NB was investigated by taking advantage of typical functional stem cell characteristics. A predictive association was established between self-renewal, as assessed by serial sphere formation, and clinical aggressiveness in primary tumors. Moreover, cell subsets gradually selected during serial sphere culture harbored increased in vivo tumorigenicity, only highlighted in an orthotopic microenvironment. A microarray time course analysis of serial spheres passages from metastatic cells allowed us to specifically “profile” the NB stem cell-like phenotype and to identify CD133, ABC transporter, and WNT and NOTCH genes as spheres markers. On the basis of combined sphere markers expression, at least two distinct tumorigenic cell subpopulations were identified, also shown to preexist in primary NB. However, sphere markers-mediated cell sorting of parental tumor failed to recapitulate the TIC phenotype in the orthotopic model, highlighting the complexity of the CSC model. Our data support the NB stem-like cells as a dynamic and heterogeneous cell population strongly dependent on microenvironmental signals and add novel candidate genes as potential therapeutic targets in the control of high-risk NB.

Abstract

Neuroblastoma (NB) is a heterogeneous, and particularly malignant childhood neoplasm in its higher stages, with a propensity to form metastasis in selected organs, in particular liver and bone marrow, and for which there is still no efficient treatment available beyond surgery. Recent evidence indicates that the CXCR4/CXCL12 chemokine/receptor axis may be involved in promoting NB invasion and metastasis. In this study, we explored the potential role of CXCR4 in the malignant behaviour of NB, using a combination of in vitro functional analyses and in vivo growth and metastasis assessment in an orthotopic NB mouse model. We show here that CXCR4 overexpression in non-metastatic CXCR4-negative NB cells IGR-NB8 and in moderately metastatic, CXCR4 expressing NB cells IGR-N91, strongly increased tumour growth of primary tumours and liver metastases, without altering the frequency or the pattern of metastasis. Moreover shRNA-mediated knock-down experiments confirmed our observations by showing that silencing CXCR4 in NB cells impairs in vitro and almost abrogates in vivo growth. High levels of CXCL12 were detected in the mouse adrenal gland (the primary tumour site), and in the liver suggesting a paracrine effect of host-derived CXCL12 on NB growth. In conclusion, this study reveals a yet unreported NB-specific predominant growth and survival-promoting role of CXCR4, which warrants a critical reconsideration of the role of CXCR4 in the malignant behaviour of NB and other cancers.

24. Personalized Portfolio Optimization Using Genetic (AI) Algorithms

Abstract

This chapter presents a fintech-as-a-service (FaaS) solution, which enables financial advisors and wealth and asset managers to provide a “private banking-like service” to the general public. The chapter illustrates all the steps needed to structure this process as an online journey. The solution contains a full end-to-end process which advisors can use to support client advisory meetings and which can potentially also be used directly by the B2C user. To support the advisor accordingly and provide online advisory to the end customer, it is required that highly individual needs are taken into account and that truly individual and personalized portfolio proposals are generated. Traditional portfolio construction methods do not have the actual ability to take a wide range of individual preferences into account. Therefore, a new portfolio construction and optimization methodology based on “genetic algorithms” is being developed and presented in this chapter. The optimization process is built using artificial intelligence approaches. This allows optimization results to be explained based on the selected customer’s preferences. The solution is designed as an open framework, which enables additional fitness factors that represent user preferences in various dimensions to be added on a customized basis.

Abstract

Cyber attacks are becoming increasingly frequent, sophisticated, and stealthy. This makes it harder for cyber defence teams to keep up, forcing them to automate their defence capabilities in order to improve their reactivity and efficiency. Therefore, we propose a fully automated cyber defence framework that no longer needs support from humans to detect and mitigate attacks within a complex infrastructure. We design our framework based on a real-world case - Locked Shields - the world's largest cyber defence exercise. In this exercise, teams have to defend their networked infrastructure against attacks, while maintaining operational services for their users. Our framework architecture connects various cyber sensors with network, device, application, and user actuators through an artificial intelligence (AI)-powered automated team in order to dynamically secure the cyber environment. To the best of our knowledge, our framework is the first attempt towards a fully automated cyber defence team that aims at protecting complex environments from sophisticated attacks.

Abstract

The oxidative Weimberg pathway for the five-step pentose degradation to α-ketoglutarate is a key route for sustainable bioconversion of lignocellulosic biomass to added-value products and biofuels. The oxidative pathway from Caulobacter crescentus has been employed in in-vivo metabolic engineering with intact cells and in in-vitro enzyme cascades. The performance of such engineering approaches is often hampered by systems complexity, caused by non-linear kinetics and allosteric regulatory mechanisms. Here we report an iterative approach to construct and validate a quantitative model for the Weimberg pathway. Two sensitive points in pathway performance have been identified as follows: (1) product inhibition of the dehydrogenases (particularly in the absence of an efficient NAD+ recycling mechanism) and (2) balancing the activities of the dehydratases. The resulting model is utilized to design enzyme cascades for optimized conversion and to analyse pathway performance in C. cresensus cell-free extracts., Metabolic engineering is often hampered by non-linear kinetics and allosteric regulatory mechanisms. Here, the authors construct a quantitative model for the pentose degradation Weimberg pathway in Caulobacter crescentus and demonstrate its biotechnological applications in cell-free system and standard metabolic engineering.

Traditional network control planes can be slow and require manual tinkering from operators to change their behavior. There is thus great interest in a faster, data-driven approach that uses signals from real-time traffic instead. However, the promise of fast and automatic reaction to data comes with new risks: malicious inputs designed towards negative outcomes for the network, service providers, users, and operators.Adversarial inputs are a well-recognized problem in other areas; we show that networking applications are susceptible to them too. We characterize the attack surface of data-driven networks and examine how attackers with different privileges---from infected hosts to operator-level access---may target network infrastructure, applications, and protocols. To illustrate the problem, we present case studies with concrete attacks on recently proposed data-driven systems.Our analysis urgently calls for a careful study of attacks and defenses in data-driven networking, with a view towards ensuring that their promise is not marred by oversights in robust design.

Abstract

Two steps of a pyrochemical route for the recovery of actinides from spent metallic nuclear fuel are being investigated at JRC-Karlsruhe. The first step consists in electrorefining the fuel in molten salt medium implying aluminium cathodes. The second step is a chlorination process for the separation of actinides (An) from An-Al alloys formed on the cathodes. The chlorination process, in turn, consists of three steps; the distillation of adhered salt (1), the chlorination of An-Al by HCl/Cl2 under formation of AlCl3 and An chlorides (2), and the subsequent sublimation of AlCl3 (3). In the present work UAl2, UAl3, NpAl2, and PuAl2 were chlorinated with HCl(g) in a temperature range between 300 and 400 °C forming UCl4, NpCl4 or PuCl3 as the major An containing phases, respectively. Thermodynamic calculations were carried out to support the experimental work. The results showed a high chlorination efficiency for all used starting materials and indicated that the sublimation step may not be necessary when using HCl(g).

Abstract

BackgroundMogamulizumab is approved in the United States, Japan, and the European Union for patients with relapsed/refractory mycosis fungoides (MF)/Sézary syndrome (SS) who have received at least one prior systemic therapy. The approved dose and schedule of mogamulizumab induction therapy are 1 mg/kg administered intravenously (IV) on Days 1, 8, 15, and 22 of the first 28-day cycle (figure 1). After induction, patients continue with 1 mg/kg IV on Days 1 and 15 of subsequent cycles. Patients must return to a clinic for administration, which can be inconvenient for patients with inadequate support systems or lack of accessibility. Additional clinic visits can also impact patient care when unforeseen circumstances, such as the COVID-19 pandemic, limit access to treatment. Limiting the number of clinic visits needed for infusion should also decrease the risk of possible exposure to nosocomial infections. This study’s purpose is to evaluate the safety, tolerability, and pharmacokinetics of mogamulizumab in patients receiving standard induction treatment followed by 2 mg/kg every 4 weeks in subsequent cycles. Data from this study will be combined with data from previous studies using modeling and simulation methods to understand how every-4-week dosing compares to every-2-week dosing in terms of exposure, safety, and activity profiles. By extending the treatment interval of mogamulizumab, patients with MF/SS will have fewer clinic visits and reduced risk of exposure to nosocomial infections.MethodsThis is a phase 2, open-label, multicenter, international study of mogamulizumab in adult patients with relapsed/refractory MF/SS who have failed at least one prior course of systemic therapy. All patients will receive mogamulizumab induction therapy. Therapy will then be administered as a 2 mg/kg IV infusion on day 1 of each subsequent 28-day cycle (figure 2). The primary objective is to evaluate the safety and tolerability of mogamulizumab 2 mg/kg administered every 4 weeks by observing the percentage of patients experiencing treatment-emergent adverse events. Secondary objectives include characterization and evaluation of mogamulizumab’s pharmacokinetic profile, immunogenicity, anti-tumor activity (global composite response and response by compartment), and pharmacodynamic profile. A maximum of 33 patients will be enrolled in this clinical trial. The study consists of a 28-day screening period followed by a treatment period of up to 2 years from Cycle 1. No dose adjustment or modification is planned or permitted. Abstract

The basic natural amino acid L-argininosuccinate containing two chiral centers occurs in L-alanine, L-arginine, L-aspartate, L-glutamate and L-proline metabolic pathways and plays a role in the biosynthesis of secondary metabolites and other amino acids. It is a precursor for arginine in the urea cycle or the citrulline–NO cycle as well as a precursor to fumarate in the citric acid cycle via argininosuccinate lyase. We aimed to run part of the urea cycle in reverse by catalyzing not the elimination but the addition reaction of L-arginine to fumarate in order to synthesize L-argininosuccinate. Argininosuccinate lyase (ASL) from Saccharomyces cerevisiae has been chosen as the catalyst for this addition reaction. The selected ARG4 gene was synthesized and homogeneously expressed in E. coli leading to a highly active argininosuccinate lyase. Remote shell sessions via protocols such as SSH are essential for managing systems, deploying applications, and running experiments. However, combined with weak passwords or flaws in the authentication process, remote shell access becomes a major security risk, as it allows an attacker to run arbitrary commands in the name of an impersonated user or even a system administrator. For example, remote shells of weakly protected systems are often exploited in order to build large botnets, to send spam emails, or to launch distributed denial of service attacks. Also, malicious insiders in organizations often use shell sessions to access and transfer restricted data. In this work, we tackle the problem of detecting malicious shell sessions based on session logs, i.e., recorded sequences of commands that were executed over time. Our approach is to classify sessions as benign or malicious by analyzing the sequence of commands that the shell users executed. We model such sequences of commands as n-grams and use them as features to train a supervised machine learning classifier. Our evaluation, based on freely available data and data from our own honeypot infrastructure, shows that the classifier reaches a true positive rate of 99.4% and a true negative rate of 99.7% after observing only four shell commands.

Abstract

The diversity of applications and devices in enterprise networks combined with large traffic volumes make it inherently challenging to quickly identify malicious traffic. When incidents occur, emergency response teams often lose precious time in reverse-engineering the network topology and configuration before they can focus on malicious activities and digital forensics. In this paper, we present a system that quickly and reliably identifies Command and Control (C&C) channels without prior network knowledge. The key idea is to train a classifier using network traffic from attacks that happened in the past and use it to identify C&C connections in the current traffic of other networks. Specifically, we leverage the fact that - while benign traffic differs - malicious traffic bears similarities across networks (e.g., devices participating in a botnet act in a similar manner irrespective of their location). To ensure performance and scalability, we use a random forest classifier based on a set of computationally-efficient features tailored to the detection of C&C traffic. In order to prevent attackers from outwitting our classifier, we tune the model parameters to maximize robustness. We measure high resilience against possible attacks - e.g., attempts to camouflaging C&C flows as benign traffic - and packet loss during the inference. We have implemented our approach and we show its practicality on a real use case: Locked Shields, the world's largest cyber defense exercise. In Locked Shields, defenders have limited resources to protect a large, heterogeneous network against unknown attacks. Using recorded datasets (from 2017 and 2018) from a participating team, we show that our classifier is able to identify C&C channels with 99% precision and over 90% recall in near real time and with realistic resource requirements. If the team had used our system in 2018, it would have discovered 10 out of 12 C&C servers in the first hours of the exercise.

Abstract

A pyrochemical electrorefining process for the recovery of actinides from metallic nuclear fuel based on actinide-zirconium alloys (An–Zr) in a molten salt is being investigated. In this process actinides are group-selectively recovered on solid aluminium cathodes as An–Al alloys using a LiCl–KCl eutectic melt at a temperature of 450 °C. In the present study the electrochemical behaviour of zirconium during electrorefining was investigated. The maximum amount of actinides that can be oxidised without anodic co-dissolution of zirconium was determined at a selected constant cathodic current density. The experiment consisted of three steps to assess the different stages of the electrorefining process, each of which employing a fresh aluminium cathode. The results indicate that almost a complete dissolution of the actinides without co-dissolution of zirconium is possible under the applied experimental conditions.

A method for synthesis of PuF 3 from PuO 2 by fluorination using NH 4 HF 2 was investigated. The text comprises the literature study on the published methods for PuF 3 syntheses, principles of the applied NH 4 HF 2 fluorination, the developed experimental apparatus, the procedure of the synthesis and analysis of the product. The results showed that the proposed method is well suited for the PuF 3 preparation in a sufficient purity and with a good reaction yield.

The reactions of three polypyridylamine ferrous complexes, [Fe(TPEN)]2+, [Fe(TPPN)]2+, and [Fe(TPTN)]2+, with nitric oxide (NO) (where TPEN = N,N,N′,N′-tetrakis(2-pyridylmethyl)ethylenediamine, TPPN = N,N,N′,N′-tetrakis(2-pyridylmethyl)-1,2-propylenediamine, and TPTN = N,N,N′,N′-tetrakis(2-pyridylmethyl)trimethylenediamine) were investigated. The first two complexes, which are spin-crossover systems, presented second-order rate constants for complex formation reactions (kf) of 8.4 × 103 and 9.3 × 103 M−1 s−1, respectively (pH 5.0, 25 °C, I = 0.1 M). In contrast, the [Fe(TPTN)]2+ complex, which is in low-spin ground state, did not show any detectable reaction with NO. kf values are lower than those of high-spin Fe(II) complexes, such as [Fe(EDTA)]2− (EDTA = ethylenediaminetetraacetate) and [Fe(H2O)]2+, but higher than low-spin Fe(II) complexes, such as [Fe(CN)5(H2O)]3− and [Fe(bipyridine)3]2+. The release of NO from the [Fe(TPEN)NO]2+ and [Fe(TPPN)NO]2+ complexes were also studied, showing the values 15.6 and 17.7 s−1, respectively, comparable to the high-spin aminocarboxylate analogs. Abstract

Shikimic acid 3-phosphate, as a central metabolite of the shikimate pathway, is of high interest as enzyme substrate for 5-enolpyruvoyl-shikimate 3-phosphate synthase, a drug target in infectious diseases and a prime enzyme target for the herbicide glyphosate. As the important substrate shikimic acid 3-phosphate is only accessible via a chemical multi-step route, a new straightforward preparative one-step enzymatic phosphorylation of shikimate using a stable recombinant shikimate kinase has been developed for the selective phosphorylation of shikimate in the 3-position. Highly active shikimate kinase is produced by straightforward expression of a synthetic aroL gene in Escherichia coli. The time course of the shikimate kinase-catalyzed phosphorylation is investigated by 1 H- and 31 P-NMR, using the phosphoenolpyruvate/pyruvate kinase system for the regeneration of the ATP cofactor. This enables the development of a quantitative biocatalytic 3-phosphorylation of shikimic acid. After a standard workup procedure, a good yield of shikimic acid 3-phosphate, with high HPLC- and NMR purity, is obtained. This efficient biocatalytic synthesis of shikimic acid 3-phosphate is superior to any other method and has been successfully scaled up to multi-gram scale.

Abstract

Neuroblastoma is a neural crest-derived neoplasm of infancy with poor outcome in patients with advanced disease. The oncogenic transcription factor PAX5 is an important developmental regulator and is implicated in the pathogenesis of several malignancies. Screening of neuroblastoma cell lines revealed PAX5 expression in a malignant subset of neuroblastoma cells, so-called 'N-type' cells, but not in the more benign 'S-type' neuroblastoma cells. PAX5 expression was also detected in small cell lung cancer, an aggressive tumor of neural crest origin. Based on this observation we hypothesized that there could be a relationship between PAX5 expression and the more malignant phenotype of N-type cells. Stable PAX5 expression was established in several clones of the S-type cell line CA-2E. A noticeable difference in morphology of these transfectants was observed and there was also a significant increase in the proliferation rate. Moreover, PAX5 expressing clones gained the ability to form colonies in a soft agar assay, a marker of tumorigenicity. Down-regulation of PAX5 in several N-type cell lines and one small cell lung cancer cell line utilizing small interfering RNA resulted in a significant decrease in growth rate. Taken together we propose PAX5 as an important factor for the maintenance of the proliferative and tumorigenic phenotype of neuroblastoma. Our data, together with a recent study on the role of PAX genes in cancer suggest that PAX5 and other PAX transcription factors might be valuable targets for cancer therapy.

Abstract

Advances in layer 2 networking technologies have fostered the deployment of large, geographically distributed LANs. Due to their large diameter, such LANs provide many vantage points for wiretapping. As an example, Google's internal network was reportedly tapped by governmental agencies, forcing the Web giant to encrypt its internal traffic. While using encryption certainly helps, eavesdroppers can still access traffic metadata which often reveals sensitive information, such as who communicates with whom and which are the critical hubs in the infrastructure.This paper presents iTAP, a system for providing strong anonymity guarantees within a network. iTAP is network-based and can be partially deployed. Akin to onion routing, iTAP rewrites packet headers at the network edges by leveraging SDN devices. As large LANs can see millions of flows, the key challenge is to rewrite headers in a way that guarantees strong anonymity while, at the same time, scaling the control-plane (number of events) and the data-plane (number of flow rules). iTAP addresses these challenges by adopting a hybrid rewriting scheme. Specifically, iTAP scales by reusing rewriting rules across distinct flows and by distributing them on multiple switches. As reusing headers leaks information, iTAP monitors this leakage and adapts the rewriting rules before any eavesdropper could provably de-anonymize any host.We implemented iTAP and evaluated it using real network traffic traces. We show that iTAP works in practice, on existing hardware, and that deploying few SDN switches is enough to protect a large share of the network traffic.

Abstract

The molecular structures of Li2[Ni(tmdta)]·5H2O (1a, tmdta = trimethylenediaminetetraacetate), {C(NH2)3}2[Ni(tmdta)]·6H2O (1b), and {Ni(H2O)6}[Ni(tmdta)]·2H2O (2a) have been determined. The central trimethylenediamine chelate ring shows half-chair (hc) geometries in 1a and 1b, while a twist-boat (tb) conformation is encountered in 2a. The coexistence of tb and hc forms in the solid state prompted us to elucidate the existence of a tb ⇌ hc equilibrium in aqueous solution. Evaluation of the data from solid state vibrational spectra (Raman and IR) for the hc and tb forms showed excellent agreement with simulated spectra obtained with DFT computations (TPSSh/TZVP). This outstanding matching between theory and experiment enabled us to build composite spectra with varying hc:tb ratios. Comparison of these results with Raman and IR spectra recorded for [Ni(tmdta)](2-) in aqueous solution revealed that simulated Raman and IR spectra with a hc:tb ratio = 2:3 match the solution spectra in an accurate way. This equilibrium ratio enabled us to compute (13)C NMR sifts for the paramagnetic solution spectrum of [Ni(tmdta)](2-) based on the relative contributions by hc and tb fractions. This leads to computed shifts that agree closely with the experimental ones. Also, the kinetics of the skeleton dynamics could be estimated quantitatively by temperature-dependent (13)C NMR spectroscopic measurements. An interesting effect encountered for the very first time here concerns a drastic intensity difference of the 10Dq band ((3)A2g → (3)T2g(F) transition) in solid state electronic spectra of tb vs hc isomers, where the intensity of this band in the case of the hc form is much lower than that of the tb conformer and thus more similar to the case of the usual Ni(II) chromophore in octahedral environment. The equilibrium constants for complex formation and protonation of Ni(II)-tmdta at low pH have been estimated by pH-dependent UV-vis titration experiments. Correlation of these data with those of Ni(II)-edta and related 3d M(II) edta and tmdta complexes allow important conclusions on the consequences resulting from extending the central diamine ring in the ligand by one methylene group in terms of both complex and protolytic stability for edta vs tmdta complexes.

Abstract

A method for synthesis of PuF 3 from PuO 2 by fluorination using NH 4 HF 2 was investigated. The text comprises the literature study on the published methods for PuF 3 syntheses, principles of the applied NH 4 HF 2 fluorination, the developed experimental apparatus, the procedure of the synthesis and analysis of the product. The results showed that the proposed method is well suited for the PuF 3 preparation in a sufficient purity and with a good reaction yield.

Abstract

Breast cancers commonly become resistant to EGFR–tyrosine kinase inhibitors (EGFR-TKIs); however, the mechanisms of this resistance remain largely unknown. We hypothesized that resistance may originate, at least in part, from molecular alterations that activate signaling downstream of EGFR. Using a screen to measure reversion of malignant cells into phenotypically nonmalignant cells in 3D gels, we identified FAM83A as a candidate cancer-associated gene capable of conferring resistance to EGFR-TKIs. FAM83A overexpression in cancer cells increased proliferation and invasion and imparted EGFR-TKI resistance both in cultured cells and in animals. Tumor cells that survived EGFR-TKI treatment in vivo had upregulated FAM83A levels. Additionally, FAM83A overexpression dramatically increased the number and size of transformed foci in cultured cells and anchorage-independent growth in soft agar. Conversely, FAM83A depletion in cancer cells caused reversion of the malignant phenotype, delayed tumor growth in mice, and rendered cells more sensitive to EGFR-TKI. Analyses of published clinical data revealed a correlation between high FAM83A expression and breast cancer patients’ poor prognosis. We found that FAM83A interacted with and caused phosphorylation of c-RAF and PI3K p85, upstream of MAPK and downstream of EGFR. These data provide an additional mechanism by which tumor cells can become EGFR-TKI resistant.

Abstract

Different semi-empirical methods (PM3(tm), AM1*, and PM6) that have parameters for Ni were evaluated for structural and energy calculations in comparison with DFT calculations (B3LYP/LANL2DZp) and X-ray structures. The reported crystal structure for K4[Ni(nta)2)] · 4H2O (nta = nitrilotriacetate, monoclinic space group P21/n (No. 14), a = 10.024(2) A, b = 10.838(2) A, c = 10.631(2) A, β = 96.46(3)°, V = 1147.6(4) A3, Z = 2) and the published X-ray structures for {(2S,3S)-1,4-dimethoxy-2,3-bis[(salicylidene)amino]-butane}nickel(II) and amine(salicylaldehyde thiosemicarbazonato)nickel(II), [Ni(AST)NH3], were used for structural evaluation. The performance of the methods was also tested for typical ligand-substitution reactions at Ni(II) centers involving displacement of NH3 by L (L = PH3, AsH3, SbH3, H2O, H2S, H2Se, H2Te) in [Ni(NH3)3NH3]2+ and [Ni(AST)NH3]. Whereas PM6 performed well for the evaluation of the structural data, AM1* was found to reproduce the energies in an excellent way.

Abstract

The development of chemoresistance represents a major obstacle in the successful treatment of cancers such as neuroblastoma (NB), a particularly aggressive childhood solid tumour. The mechanisms underlying the chemoresistant phenotype in NB were addressed by gene expression profiling of two doxorubicin (DoxR)-resistant vs sensitive parental cell lines. Not surprisingly, the MDR1 gene was included in the identified upregulated genes, although the highest overexpressed transcript in both cell lines was the frizzled-1 Wnt receptor (FZD1) gene, an essential component of the Wnt/beta-catenin pathway. FZD1 upregulation in resistant variants was shown to mediate sustained activation of the Wnt/beta-catenin pathway as revealed by nuclear beta-catenin translocation and target genes transactivation. Interestingly, specific micro-adapted short hairpin RNA (shRNAmir)-mediated FZD1 silencing induced parallel strong decrease in the expression of MDR1, another beta-catenin target gene, revealing a complex, Wnt/beta-catenin-mediated implication of FZD1 in chemoresistance. The significant restoration of drug sensitivity in FZD1-silenced cells confirmed the FZD1-associated chemoresistance. RNA samples from 21 patient tumours (diagnosis and postchemotherapy), showed a highly significant FZD1 and/or MDR1 overexpression after treatment, underlining a role for FZD1-mediated Wnt/beta-catenin pathway in clinical chemoresistance. Our data represent the first implication of the Wnt/beta-catenin pathway in NB chemoresistance and identify potential new targets to treat aggressive and resistant NB.
