The knowledge about the processes that control cell cycle activity in J. curcas has not yet been addressed. Cell suspension cultures offer an attractive alternative to carry out this kind of studies in a simplified biological system. Nevertheless, the cell suspensions exhibit a high degree of heterogeneity concerning cell cycle distribution; therefore, synchronization methods must be previously established for this plant species. In the present research, cell suspensions of J. curcas were used to assess phosphorus, nitrogen or sucrose starvation as cell cycle synchronization approaches. Cell suspension cultures were established from the endosperm of seeds using a method for the enzymatic breakdown, without a previous callogenesis stage. The cell suspensions reached a maximum growth rate of 0.15 d−1 and a doubling time of 4.8 d. Microscopy analyses showed amyloplasts in some of the cells; however, oil and protein bodies were not observed. Regarding cell cycle synchronization, the results indicated that phosphorus or nitrogen starvation did not affect the cell-cycle arrest in the G0/G1 phase. However, the sucrose depletion for three days induced an accumulation of 67% of the cells in the G0/G1 phase. This effect was reversed with the addition of a carbon source. Moreover, these culture conditions generated a population of cells (approx. 5%) with a higher DNA content (3n > 6n), which was interpreted as the activation of an endoreplication process. Our findings suggest that nutrient starvation can affect in a differential way the cell synchronization process. Surface fluxes and states can recur and remain consistent across various spatial and temporal scales, forming space‐time patterns. Quantifying and understanding the observed patterns is desirable, as they provide information about the dynamics of the processes involved. This study introduces the empirical spatio‐temporal covariance function and a corresponding parametric covariance function as tools to identify and characterize spatio‐temporal patterns in remotely sensed fields. The method is demonstrated using 2 km hourly GOES‐16/17 land surface temperature (LST) data over the Contiguous United States by splitting the area into 1.0° × 1.0° domains. The summer day‐time LST ESTCFs for 2018 to 2022 are derived for each domain, and a parametric covariance model is fitted. Clustering analysis is applied to detect areas with similar spatio‐temporal LST patterns. Six main zones within CONUS are identified and characterized based on their variance and temporal and spatial characteristic length scales (i.e., scales for which the temperature variations are temporally and spatially related), respectively: (a) Eastern plains with 3 K2, ∼6 hr, and 0.15°, (b) Gulf of California with 60 K2, ∼8 hr, and 0.34°, (c) mountains and coasts transition 1 with 16 K2, ∼11 hr, and 0.25°, (d) central US, Midwest, and South cities with 5.5 K2, ∼8 hr, and ∼0.2°, (e) mountains and coasts transition 2 with ∼10 K2, ∼8 hr, and 0.2°, and (f) largest mountains and coastlines with ∼19 K2, ∼13 hr, and 0.3°. The tools introduced provide a pathway to formally identify and summarize the spatio‐temporal patterns observed in remotely sensed fields and relate those to more complex processes within the Soil‐Vegetation‐Atmosphere System. Plain Language Summary: Specific processes on Earth's surface, like heat and water fluxes and air movement, often exhibit coherent patterns across space and time. Figuring out where these patterns occur can be helpful as they can aid to explain how the underlying physical processes work. This research introduces a new tool to find and describe these patterns and applies it to temperature data derived from satellites. The United States is divided into smaller areas, and the developed tool is used to analyze the land temperature data within those areas; then, regions with similar temperature patterns are identified. These results help to determine that certain landscape features, like coastlines, mountains, and cities, influence how temperature patterns behave in space, time and both. These tools help us to recognize and describe the patterns we see in satellite observations and ultimately, connect them to more complicated processes happening in the Soil‐Vegetation‐Atmosphere System. Axon regeneration in the CNS requires reactivating injured neurons' intrinsic growth state and enabling growth in an inhibitory environment. Using an inbred mouse neuronal phenotypic screen, we find that CAST/Ei mouse adult dorsal root ganglion neurons extend axons more on CNS myelin than the other eight strains tested, especially when pre-injured. Injury-primed CAST/Ei neurons also regenerate markedly in the spinal cord and optic nerve more than those from C57BL/6 mice and show greater sprouting following ischemic stroke. Heritability estimates indicate that extended growth in CAST/Ei neurons on myelin is genetically determined, and two whole-genome expression screens yield the Activin transcript Inhba as most correlated with this ability. Inhibition of Activin signaling in CAST/Ei mice diminishes their CNS regenerative capacity, whereas its activation in C57BL/6 animals boosts regeneration. This screen demonstrates that mammalian CNS regeneration can occur and reveals a molecular pathway that contributes to this ability. Omura et al. screened for neuronal growth on myelin and find CAST/Ei inbred mice are uniquely able to regenerate axons in the injured CNS through Activin signaling. Enhancing Activin signaling confers a CNS-regenerative capacity in normally non-regenerative mouse strains.

Background: Prader–Willi syndrome (PWS), a genetically determined disorder, the most frequent cause of early onset obesity, is associated with physical and cognitive dysfunctions and behavioural disturbances; these disturbances are frequently treated with psychotropic medication. The aim of this cross‐sectional study was to describe the characteristics of the first large national sample of persons with PWS in Spain and analyse the relationships of those characteristics with key demographic and clinical factors, particularly with obesity and the regular use of psychotropic medication. Methods: Participants were recruited among all members of the Spanish Prader–Willi Association who agreed to take part in the study and fulfilled its inclusion criteria. Family and patient demographic features, family size and birth order, intelligence quotient (IQ), anthropometric measures, lifestyle habits, behavioural disturbances (with the Aberrant Behavior Checklist) and clinical data, as well as use of psychotropic drugs and their side effects (with the UKU scale), were collected in genetically confirmed cases of PWS. Bivariate and logistic regression analyses were used for determining the associations of demographic and clinical factors with both obesity and the regular use of psychotropic medication. Results: The cohort included 177 participants (aged 6–48 years), that is, 90 (50.8%) males and 87 (49.2%) females. Behavioural disturbances were present in a range of 75% to 93% of participants; psychotropic medication was prescribed to 81 (45.8%) of them. Number of siblings showed a direct correlation with IQ, especially among males, and inappropriate speech was more intense in only‐child females. Obesity was, in parallel, strongly associated with ascending age and with not being currently under growth hormone (GH) treatment. Participants taking any psychotropic medication were characterised by more frequent age ≥30 years, high level of hyperactivity and a psychiatric diagnosis. Conclusions: Characterisation of persons with PWS in Spain confirms their physical and behavioural phenotype and supports the long‐term application of GH therapy and the rational use of psychotropic medication.
