Much attention has been directed to understanding species richness patterns, but adding an evolutionary perspective allows us to also consider the historical processes determining current diversity patterns. We analyzed phylogenetic patterns of fern species assemblages in 868 plots along a wide range of elevational (0-4000 m) and latitudinal (0°-23°N) gradients in the Neotropics to allow a deeper understanding of evolutionary processes underlying current patterns of diversity and community assembly. Overall, we found that phylogenetic mean pairwise distance (sMPD) and mean nearest taxon distance (sMNTD) decreased with increasing latitude and elevation, but that these geographical factors per se were weak explanatory variables. Incorporating environmental variables strongly enhanced the power of the predictive model, indicating that fern assemblages are phylogenetically more diverse under wet and warm to cool conditions at low latitudes and elevations. Further, whereas epiphytic fern assemblages were strongly influenced by climatic factors, this was not the case for terrestrial ones, suggesting that edaphic conditions and vegetation structure may have a stronger influence on the evolution and diversification of terrestrial ferns. We conclude that fern assemblages are strongly influenced by phylogenetic niche conservatism and environmental filtering. This has also been found for angiosperms, but the direction of the environment-phylogenetic relationship is often opposed in the two groups, suggesting that the older age of many fern lineages includes historical signals that are not evident in the more recent angiosperm radiation.

Currently, the marketing of electronic cigarettes as a safe alternative to smoking has increased, which is associated with greater use of these devices, especially among young people and smokers interested in quitting tobacco cigarettes. Given the growing use of this type of product, there is a need to determine the consequences of electronic cigarettes on human health, especially since many of the compounds contained in the aerosol and liquid of these devices have a high potential to be carcinogenic and genotoxic. Additionally, many of these compounds' aerosol concentrations exceed the safe limits. We have evaluated the levels of genotoxicity and changes in DNA methylation patterns associated with vaping. We analyzed a total of 90 peripheral blood samples from a population of vapers (n = 32), smokers (n = 18), and controls (n = 32), in which the frequencies of genotoxicity were determined by the cytokinesis-blocking micronuclei (CBMN) assay and the patterns of methylation of the repetitive elements of LINE-1 through the Quantitative Methylation Specific PCR (qMSP) assay. Here we show an increase in genotoxicity levels associated with vaping habits. Additionally, the group of vapers showed changes at the epigenetic level specifically associated with the loss of methylation of the LINE-1 elements. These changes in LINE-1 methylation patterns were reflected in its representative RNA expression detected in vapers.

Single-ion-conducting block copolymers are of considerable interest as electrolytes for battery systems, as they eliminate overpotentials due to concentration gradients. In this study, we characterize a library of poly(ethylene oxide) (PEO)-based diblock copolymers where the second block is poly(styrene-4-sulfonyltrifluoromethylsulfonyl)imide with either cation: univalent lithium or divalent magnesium counterions (PEO-PSLiTFSI or PEO-P[(STFSI)2Mg]). The PEO chain length is held fixed in this study. Polymers were synthesized in matched pairs that were identical in all aspects except for the identity of the counterion. Using rheology, SAXS, DSC, and AC impedance spectroscopy, we show that the dependence of morphology, modulus, and conductivity on composition in these charged copolymer systems is fundamentally different from uncharged block copolymers. At a given frequency and temperature, the shear moduli of the magnesiated copolymer systems were approximately 3-4 orders of magnitude higher than those of the matched lithiated pair. The shear moduli of all of the lithiated copolymers showed liquid-like rheological features while the magnesiated copolymers did not. All of the lithiated copolymers were completely disordered (homogeneous), consistent with the observed rheological properties. As expected, the moduli of the lithiated copolymers increased with increasing volume fraction of the ion-containing block (•PSTFSI), and the conductivity decreased with •PSTFSI. However, the magnesiated copolymers followed a distinct trend. We show that this was due to the presence of microphase separation in the regime 0.21 ≤ •PSTFSI ≤ 0.36, and the tendency for microphase separation became weaker with increasing •PSTFSI. The magnesiated copolymer with •PSTFSI = 0.38 was homogeneous. The morphological, rheological, and conductivity properties of these systems are governed by the affinity of the cations for PEO chains; homogeneous systems are obtained when the cations migrate from the ion-containing block to PEO.

We explore the relationship between the morphology and ionic conductivity of block copolymer electrolytes over a wide range of salt concentrations for the system polystyrene-block-poly(ethylene oxide) (PS-b-PEO, SEO) mixed with lithium bis(trifluoromethanesulfonyl)imide salt (LiTFSI). Two SEO polymers were studied, SEO(16-16) and SEO(4.9-5.5), over the salt concentration range r = 0.03-0.55. The numbers x and y in SEO(x-y) are the molecular weights of the blocks in kg mol-1, and the r value is the molar ratio of salt to ethylene oxide moieties. Small-angle X-ray scattering was used to characterize morphology and grain size at 120 °C, differential scanning calorimetry was used to study the crystallinity and the glass transition temperature of the PEO-rich microphase, and ac impedance spectroscopy was used to measure ionic conductivity as a function of temperature. The most surprising observation of our study is that ionic conductivity in the concentration regime 0.11 ≤ r ≤ 0.21 increases in SEO electrolytes but decreases in PEO electrolytes. The maximum in ionic conductivity with salt concentration occurs at about twice the salt concentration in SEO (r = 0.21) as in PEO (r = 0.11). We propose that these observations are due to the effect of salt concentration on the grain structure in SEO electrolytes.

The principal methyl donor of the cell, S-adenosylmethionine (SAMe), is produced by the highly conserved family of methionine adenosyltranferases (MATs) via an ATP-driven process. These enzymes play an important role in the preservation of life, and their dysregulation has been tightly linked to liver and colon cancers. We present crystal structures of human MATα2 containing various bound ligands, providing a "structural movie" of the catalytic steps. High- to atomic-resolution structures reveal the structural elements of the enzyme involved in utilization of the substrates methionine and adenosine and in formation of the product SAMe. MAT enzymes are also able to produce S-adenosylethionine (SAE) from substrate ethionine. Ethionine, an S-ethyl analog of the amino acid methionine, is known to induce steatosis and pancreatitis. We show that SAE occupies the active site in a manner similar to SAMe, confirming that ethionine also uses the same catalytic site to form the product SAE.

Single-ion-conducting polymers are ideal electrolytes for rechargeable lithium batteries as they eliminate salt concentration gradients and concomitant concentration overpotentials during battery cycling. Here we study the ionic conductivity and morphology of poly(ethylene oxide)-b-poly(styrenesulfonyllithium(trifluoromethylsulfonyl)imide) (PEO-b-PSLiTFSI) block copolymers with no added salt using ac impedance spectroscopy and small-angle X-ray scattering. The PEO molecular weight was held fixed at 5.0 kg mol-1, and that of PSLiTFSI was varied from 2.0 to 7.5 kg mol-1. The lithium ion concentration and block copolymer composition are intimately coupled in this system. At low temperatures, copolymers with PSLiTFSI block molecular weights ≤4.0 kg mol-1 exhibited microphase separation with crystalline PEO-rich microphases and lithium ions trapped in the form of ionic clusters in the glassy PSLiTFSI-rich microphases. At temperatures above the melting temperature of the PEO microphase, the lithium ions were released from the clusters, and a homogeneous disordered morphology was obtained. The ionic conductivity increased abruptly by several orders of magnitude at this transition. Block copolymers with PSLiTFSI block molecular weights ≥5.4 kg mol-1 were disordered at all temperatures, and the ionic conductivity was a smooth function of temperature. The transference numbers of these copolymers varied from 0.87 to 0.99. The relationship between ion transport and molecular structure in single-ion-conducting block copolymer electrolytes is qualitatively different from the well-studied case of block copolymers with added salt.

S-Adenosylmethionine (SAMe) is the principal methyl donor of the cell and is synthesized via an ATP-driven process by methionine adenosyltransferase (MAT) enzymes. It is tightly linked with cell proliferation in liver and colon cancer. In humans, there are three genes, mat1A, mat2A and mat2B, which encode MAT enzymes. mat2A and mat2B transcribe MATα2 and MATβ enzyme subunits, respectively, with catalytic and regulatory roles. The MATα2β complex is expressed in nearly all tissues and is thought to be essential in providing the necessary SAMe flux for methylation of DNA and various proteins including histones. In human hepatocellular carcinoma mat2A and mat2B genes are upregulated, highlighting the importance of the MATα2β complex in liver disease. The individual subunits have been structurally characterized but the nature of the complex has remained elusive despite its existence having been postulated for more than 20 years and the observation that MATβ is often co-localized with MATα2. Though SAMe can be produced by MAT(α2)4 alone, this paper shows that the V max of the MATα2β complex is three- to fourfold higher depending on the variants of MATβ that participate in complex formation. Using X-ray crystallography and solution X-ray scattering, the first structures are provided of this 258 kDa functional complex both in crystals and solution with an unexpected stoichiometry of 4α2 and 2βV2 subunits. It is demonstrated that the N-terminal regulates the activity of the complex and it is shown that complex formation takes place surprisingly via the C-terminal of MATβV2 that buries itself in a tunnel created at the interface of the MAT(α2)2. The structural data suggest a unique mechanism of regulation and provide a gateway for structure-based drug design in anticancer therapies.

A significant limitation of rechargeable lithium-ion batteries arises because most of the ionic current is carried by the anion, the ion that does not participate in energy-producing reactions. Single-ion-conducting block copolymer electrolytes, wherein all of the current is carried by the lithium cations, have the potential to dramatically improve battery performance. The relationship between ionic conductivity and morphology of single-ion-conducting poly(ethylene oxide)-b-polystyrenesulfonyllithium(trifluoromethylsulfonyl)imide (PEO-PSLiTFSI) diblock copolymers was studied by small-angle X-ray scattering and ac impedance spectroscopy. At low temperatures, an ordered lamellar phase is obtained, and the "mobile" lithium ions are trapped in the form of ionic clusters in the glassy polystyrene-rich microphase. An increase in temperature results in a thermodynamic transition to a disordered phase. Above this transition temperature, the lithium ions are released from the clusters, and ionic conductivity increases by several orders of magnitude. This morphology-conductivity relationship is very different from all previously published data on published electrolytes. The ability to design electrolytes wherein most of the current is carried by the lithium ions, to sequester them in nonconducting domains and release them when necessary, has the potential to enable new strategies for controlling the charge-discharge characteristics of rechargeable lithium batteries.

This case report describes the treatment of two maxillary central incisors following a traumatic injury with tooth #8 developing replacement resorption and #9 developing inflammatory root resorption. A 10-year-old girl presented complaining of pain in her maxillary central incisors. Upon clinical examination, teeth #8 and #9 were tender to percussion and palpation of the buccal soft tissues. Thermal and electrical pulpal sensitivity tests for teeth #8 and #9 were negative. An intraoral periapical radiograph revealed resorptive defects in tooth #8, which were filled with bone-like tissue, while tooth #9 had radiolucent resorptive defects along the root surface and a periapical radiolucency. A diagnosis of replacement resorption was made for tooth #8 and external inflammatory root resorption for tooth #9. Tooth #8 was treated with a multidisciplinary approach utilizing a guided template for premolar autotransplantation with an immediate veneer restoration, while tooth #9 was managed with root canal treatment using a tricalcium silicate cement to fill the canal. At the 1, 4, 8, 12, and 24-month follow-ups, the patient remained asymptomatic, and there was no radiographic evidence of root or periapical pathosis on either tooth. The root-end of the donor tooth transplanted to the #8 site continued to develop. This case report highlights successful interdisciplinary management of two forms of root resorption using modern treatment strategies that provided immediate function and esthetics to the maxillary central incisors in a young patient following trauma. Resveratrol is growth-suppressive and pro-apoptotic in liver cancer cells. Methionine adenosyltransferase 2B (MAT2B) encodes for two dominant variants V1 and V2 that positively regulate growth, and V1 is anti-apoptotic when overexpressed. Interestingly, crystal structure analysis of MAT2B protein (MATβ) protomer revealed two resveratrol binding pockets, which raises the question of the role of MAT2B in resveratrol biological activities. We found that resveratrol induced the expression of MAT2BV1 and V2 in a time- and dose-dependent manner by increasing transcription, mRNA, and protein stabilization. Following resveratrol treatment, HuR expression increased first, followed by SIRT1 and MAT2B. SIRT1 induction contributes to increased MAT2B transcription whereas HuR induction increased MAT2B mRNA stability. MATβ interacts with HuR and SIRT1, and resveratrol treatment enhanced these interactions while reducing the interaction between MATβ and MATα2. Because MATβ lowers the Ki of MATα2 for S-adenosylmethionine (AdoMet), this allowed steady-state AdoMet level to rise. Interaction among MATβ, SIRT1, and HuR increased stability of these proteins. Induction of MAT2B is a compensatory response to resveratrol as knocking down MAT2BV1 potentiated the resveratrol pro-apoptotic and growth-suppressive effects, whereas the opposite occurred with V1 overexpression. The same effect on growth occurred with MAT2BV2. In conclusion, resveratrol induces HuR, SIRT1, and MAT2B expression; the last may represent a compensatory response against apoptosis and growth inhibition. However, MATβ induction also facilitates SIRT1 activation, as the interaction stabilizes SIRT1. This complex interplay among MATβ, HuR, and SIRT1 has not been previously reported and suggests that these proteins may regulate each other's signaling.
