Identification and analysis of enhancers for endothelial-expressed genes can provide crucial information regarding their upstream transcriptional regulators. However, enhancer identification can be challenging, particularly for people with limited access or experience of bioinformatics, and transgenic analysis of enhancer activity patterns can be prohibitively expensive. Here we describe how to use publicly available datasets displayed on the UCSC Genome Browser to identify putative endothelial enhancers for mammalian genes. Furthermore, we detail how to utilize mosaic Tol2-mediated transgenesis in zebrafish to verify whether a putative enhancer is capable of directing endothelial-specific patterns of gene expression.

SummaryPlants display an incredible variety of forms and functions, which sustains a vast diversity of ecosystems on Earth. This diversity is the result of two forces: environmental filtering, the selection of phenotypes fit to a specific habitat; and evolutionary history, genetic constraints that determine the potential adaptations that species may evolve. To date, most studies have focused on describing macroecological patterns of trait variation, while the role of evolutionary history in determining these patterns has remained much less explored. Here, we combine a traits dataset including 4,213 vascular plant species with a phylogeny of the plant kingdom, to determine the influence of evolutionary history on the patterns of trait variation. Our results show that commonly used plant traits, such as plant height or leaf nitrogen, are strongly constrained by evolutionary history. Accounting for phylogenetic relationships changes the variance explained by the main axes of trait variation in the Plant Kindgom —the leaf economic spectrum and the plant size and organ axis continuum— with the former explaining more variance than the latter. Moreover, we demonstrate that the plant size continuum is more influenced by evolutionary history than the leaf economics spectrum. These results highlight the influence of evolutionary history in shaping plant life history strategies and the importance of accounting for phylogeny in trait-based studies.

Sexually dimorphic development is responsible for some of the most remarkable phenotypic variation found in nature. Alternative splicing of the transcription factor gene doublesex (dsx) is a highly conserved developmental switch controlling the expression of sex-specific pathways. Here, we leverage sex-specific differences in butterfly wing color pattern to characterize the genetic basis of sexually dimorphic development. We use RNA-seq, immunolocalization, and motif binding site analysis to test specific predictions about the role of dsx in the development of structurally based ultraviolet (UV) wing patterns in Zerene cesonia (Southern Dogface). Unexpectedly, we discover a novel duplication of dsx that shows a sex-specific burst of expression associated with the sexually dimorphic UV coloration. The derived copy consists of a single exon that encodes a DNA binding but no protein-binding domain and has experienced rapid amino-acid divergence. We propose the novel dsx paralog may suppress UV scale differentiation in females, which is supported by an excess of Dsx-binding sites at cytoskeletal and chitin-related genes with sex-biased expression. These findings illustrate the molecular flexibility of the dsx gene in mediating the differentiation of secondary sexual characteristics.

Resumen del trabajo presentado a: 2nd Meeting of the Iberian Ecological Society (SIBECOL); XXI conference of the Iberian Association of Limnology (AIL) and 21st National Congress of Ecology of the Portuguese Ecological Society (SPECO), celebrados en Aveiro (Portugal) del 3 al 8 de julio de 2022., Understanding and identifying the impacts of multiple stressors on natural populations has become a key objective for post-2020 biodiversity conservation. Of particular concern are the impacts of stressors at the local scale, which are not necessarily reflected in current global conservation assessments. As a hotspot of biodiversity, the Iberian Peninsula is an area of special interest for conservation, but it is currently facing myriad environmental problems. Yet, a synthesis of the impacts of multiple stressors on the Iberian faunal populations is glaringly missing. To contribute to this challenge, we join our efforts in an Early Career Researchers project funded by SIBECOL, which aims to synthesise current knowledge on population trends of Iberian marine, terrestrial and freshwater vertebrates, and, eventually, to identify the most common stressors for Iberian fauna populations, and their differences between major taxonomic groups and ecosystems. Here, we will present the results of the first phase of this project, whose objective was to describe the trends of Iberian vertebrate populations and identify knowledge gaps related to taxonomic groups and ecosystems. To that aim, we conducted an extensive literature review, with more than 5,000 scientific manuscripts and grey literature screened. From this initial review, and after applying data quality controls, we obtained > 1,000 population time series of Iberian vertebrates across marine, terrestrial, and freshwater ecosystems. Once we compiled this database, we analysed the population trends using a two-step modelling process: (i) first, we applied state-space models to derive the population trend of each population time series; and (ii) second, we used multilevel Bayesian models to determine the factors influencing these population trends. Overall, we found that Iberian vertebrate populations display a disparity in trends, with both declines and increases, with fishes (both marine and freshwater) being the taxa showing clear signs of decline. Additionally, we identified important gaps in monitoring data across the Iberian Peninsula, with amphibians and reptiles severely underrepresented in our dataset, despite being among the most threatened taxonomic groups. These gaps may limit our understanding of the impacts of stressors at local scales and eventually bias current conservation assessments. Overall, our approach will bridge the significant gap in our knowledge between the global and local scale conservation status of wild Iberian populations and help focus future conservation policies.

The vast diversity of animal coloration is generated through a combination of pigment and structural colors. These colors can greatly influence the fitness and life history of an organism. Butterflies and their wing colors are an excellent model to study how these colors can impact the development and success of an organism. In this study, we explore species differences in structurally-based ultraviolet coloration in the Zerene butterfly. We show clear species differences in ultraviolet (UV) pattern and reflectance spectra. By varying larval diet, we show evidence for developmental plasticity in the structure and organization of UV reflecting scales in Zerene cesonia. We further show that feeding the larval host plant of Zerene eurydice to Z. cesonia does not result in greater similarity in scale structure or UV coloration to the sister species. These results not only demonstrate a connection between plasticity in a male ornamentation, UV wing pattern, and larval resource acquisition, but also identify candidate structural and organizational changes in wing scales responsible for the trait variation.

Congenital heart disease (CHD) is the most common class of human birth defects, with a prevalence of 0.9% of births. However, two-thirds of cases have an unknown cause, and many of these are thought to be caused by in utero exposure to environmental teratogens. Here we identify a potential teratogen causing CHD in mice: maternal iron deficiency (ID). We show that maternal ID in mice causes severe cardiovascular defects in the offspring. These defects likely arise from increased retinoic acid signalling in ID embryos. The defects can be prevented by iron administration in early pregnancy. It has also been proposed that teratogen exposure may potentiate the effects of genetic predisposition to CHD through gene–environment interaction. Here we show that maternal ID increases the severity of heart and craniofacial defects in a mouse model of Down syndrome. It will be important to understand if the effects of maternal ID seen here in mice may have clinical implications for women., From mouse experiments, the authors link iron deficiency in mothers with cardiovascular defects and increased retinoic acid signalling in their offspring, and giving iron early in pregnancy can prevent most defects.

Recurrent Pregnancy Loss (RPL) affects 2-4% of couples, and with increasing numbers of pregnancy losses the risk of miscarrying a euploid pregnancy is increased, suggesting RPL is a pathology distinct from sporadic miscarriage that is due largely to lethal embryonic aneuploidy. There are a number of conditions associated with RPL including unspecified ‘immune’ pathologies, one of the strongest candidates for dysregulation remains T regulatory cells as depletion in the very early stages of pregnancy in mice leads to pregnancy loss.Human endometrial Treg and conventional CD4T cells were isolated during the peri-implantation period of the menstrual cycle in normal women. We identified an endometrial Treg transcriptomic signature and validated an enhanced regulatory phenotype compared to peripheral blood Treg. Parous women had an altered endometrial Treg transcriptome compared to nulliparity, indicating acquired immunity memory of pregnancy within the Treg population, by comparison endometrial conventional CD4T cells were not altered. We compared primary and secondary RPL to nulliparous or parous controls respectively. Both RPL subgroups displayed differentially expressed Treg gene transcriptomes compared to controls. We found increased cell surface S1PR1 and decreased TIGIT protein expression by Treg in primary RPL, confirming the presence of altered Treg in the peri-implantation RPL endometrium.

Background: News about suicide may have either a contagion effect or a protective effect. For this reason, interventions aimed to the media are a priority. However, the effectiveness of the interventions has not yet been elucidated. The objective of the study was to evaluate comparatively the characteristics and adequacy of suicide news articles between geographical areas in which interventions aimed at media either exist or not. Methods: We evaluated suicide news articles in the four main digital media of Spain (without interventions) and Victoria, Australia (with interventions), according to Alexa (a website traffic statistics). We assessed adherence to World Health Organization (WHO) recommendations, avoidability, frequency, and typology of suicide news articles. The evaluation period was 6 months. Bivariate analysis was performed comparing the samples using t-test, Chi-Squared test and Fisher's exact test according to the type of variable. Results: We identified 378 suicide news articles: 141 in Spain and 237 in Victoria. Background: News about suicide may cause a contagion effect or a protective effect. For this reason, the World Health Organization (WHO) has published recommendations on responsible media coverage of suicide. Certain characteristics of suicide news articles and their frequency have been associated to such effects. Despite their importance, these aspects have been scarcely studied in the Spanish media. The objective of this study was to identify the characteristics of suicide news articles in the Spanish digital media. Methods: We identified the adherence to WHO recommendations, avoidability, frequency, and typology of suicide news articles in the four main spanish digital media (El Pais, El Mundo, El Confidencial and ABC), according to Alexa (a website traffic statistics). Adherence was evaluated from a dichotomous and continuous perspective by using an ad hoc scale, and avoidability was conceptually defined. The evaluation period was 6 months (11/1/2016-04/30/2017). The statistical program used was R Core Team (2017). Results: We analysed 141 suicide news articles. Of these, 99.12% did not adhere to the recommendations, the degree of adherence was 2.97 (95% CI, 2.19-3.75; range between +22 and -22), 34.51% were avoidable, 34.04% were about "suicide after murder", and 90.04% about death by suicide. Conclusions: Adherence to WHO recommendations is very low in suicide news articles. Furthermore, almost one third of them are avoidable. The identified profile points out the need for interventions aimed at the media. Besides, it allows adjusting their contents to the identified specific needs.

Non-communicable diseases (NCDs) are a major problem as they are the leading cause of death and represent a substantial economic cost. The ‘Developmental Origins of Health and Disease Hypothesis’ proposes that adverse stimuli at different life stages can increase the predisposition to these diseases. In fact, adverse in utero programming is a major origin of these diseases due to the high malleability of embryonic development. This review provides a comprehensive analysis of the scientific literature on in utero programming and NCDs highlighting potential medical strategies to prevent these diseases based upon this programming. We fully address the concept and mechanisms involved in this programming (anatomical disruptions, epigenetic modifications and microbiota alterations). We also examine the negative role of in utero programming on the increased predisposition of NCDs in the offspring, which introduces the passive medical approach that consists of avoiding adverse stimuli including an unhealthy diet and environmental chemicals. Finally, we extensively discuss active medical approaches that target the causes of NCDs and have the potential to significantly and rapidly reduce the incidence of NCDs. These approaches can be classified as direct in utero programming modifications and personalized lifestyle pregnancy programs; they could potentially provide transgenerational NCDs protection. Active strategies against NCDs constitute a promising tool for the reduction in NCDs.

El presente texto tiene como objetivo principal realizar la recopilacion de informacion sobre los sistemas de monitoreo que se han disenado e implementado para adquirir parametros que determinen la calidad del aire. Para poder identificar los principales dirigentes en la investigacion, se hace uso de las herramientas de busqueda Scopus, Scimago y LA referencia. Esto con el fin de obtener datos de produccion cientifica a nivel mundial, y en Latinoamerica. Donde a nivel global, china lidera la produccion cientifica junto a estados unidos, a nivel regional en Latinoamerica, Brasil, Mexico, Chile, Argentina y Colombia hace parte de los primeros puestos en analisis.

Extracellular vesicles are highly abundant in seminal fluids and have a known role enhancing sperm function. Clinical pregnancy rates after IVF treatment are improved after female exposure to seminal fluid. Seminal fluid extracellular vesicles (SF-EVs) are candidate enhancers, however, whether SF-EVs interact with cells from the endometrium and modulate the implantation processes is unknown. Here, we investigated whether SF-EVs interact with endometrial stromal cells (ESCs) and enhance decidualisation, a requisite for implantation. SF-EVs, isolated from human seminal fluid (n = 11) by ultracentrifugation, were characterised by nanoparticle tracking analysis and Western blotting, and purified using size exclusion chromatography. Non-decidualised and decidualised primary ESCs (n = 5) were then treated with SF-EVs. Binding of bio-maleimide-labelled SF-EVs was detected by flow cytometry and fluorescence microscopy. Prolactin and IGFBP-1 protein levels in culture media were also analysed after single and multiple SF-EV exposure. SF-EVs size ranged from 50 to 300 nm, and they expressed exosomal markers (ALIX, SYNTENIN-1, CD9 and CD81). SF-EVs bound to non-decidualised and decidualised ESCs at similar levels. ESCs prolactin secretion was increased after single (p = 0.0044) and multiple (p = 0.0021) SF-EV exposure. No differences were found in IGFBP-1 protein levels. In conclusion, SF-EVs enhance in vitro ESC decidualisation and increase secretion of prolactin, an essential hormone in implantation. This elucidates a novel role of SF-EVs on endometrial receptivity. El estudio tiene como finalidad correlacionar los estilos de vida y factores socioeconómicos con la salud de los estudiantes de las electivas de actividad física y deporte de la Pontificia Universidad Javeriana, sede Bogotá-Colombia. Es un estudio cuantitativo descriptivo, de corte transversal, con una muestra de 1410 estudiantes con edad entre 24 y 29 años que corresponde a 748 mujeres y 662 hombres. Fue aplicado un cuestionario estandarizado propuesto por Nahas (2013), que presenta los factores nutricionales, el estrés, la actividad física, el comportamiento preventivo y las relaciones sociales como componentes que afectan la salud de las personas y están asociados al desarrollo de las enfermedades crónicas no transmisibles. La menor calificación se encontró en los factores nutricionales, de actividad física y de conducta preventiva. Por otro lado, se encontró una mejor calificación en las relaciones sociales y el control del estrés. Los resultados socioeconómicos demuestran que los estudiantes tienen preferencia por deportes individuales; viven con los padres y familiares, ubicados en estrato de clase media, y se presenta un alto índice de padres con formación universitaria. Se concluye que existe una tendencia de estilos de vida no saludables que puede afectar negativamente la salud de esta población. Es importante desarrollar estrategias de intervención en el contexto social, para generar un cambio de conducta y adoptar un estilo de vida saludable para mejorar la calidad de vida. The objective of this study was to correlate the lifestyles and socioeconomic factors with the health of the students in the Pontificia Universidad Javeriana, Bogotá - Colombia. It’s a quantitative descriptive cross-sectional study with a sample of 1410 students aged between 24 and 29 years corresponding to 748 women and 662 men. Was applied a standardized questionnaire proposed by Nahas (2013), It consist of nutritional factors, stress, physical activity, preventive behavior and social relations as components that affect the health of people, therefor they’re associated with the development of noncommunicable chronic diseases. The lower ratings were the nutritional factors, physical activity and preventive behavior. On the other hand, was found a better rating in social relationships and stress management. In the socioeconomic results were evidenced that the students have a preference for individual sports; They live with their parents and relatives at middle-class houses and the educational level of their parents is university. We concluded that there is a trend of styles of unhealthy life that can adversely affect the health of this population. Is important to develop strategies of intervention in the social context, to generate a change in behavior and adopt a healthy lifestyle to improve the quality of life.
