Abstract

International audience; Biogas production via anaerobic digestion could approach thermal autarky (less biogas self-consumption and better energy efficiency) using enhanced solutions for thermal insulation based on new multimembrane gasholder configurations, loss avoidance, and heat recovery strategies. In this study, a predictive and configuration-dependent dynamics energy model is developed to daily and seasonally assess the thermal efficiency and heating requirements of industrial biogas plants involving wet digesters with upper multimembrane gasholders. The model is validated using experimental data from a full-scale biogas plant in operation over a one-year period.The energy model involves a set of dynamic energy balances defined for each compartment of the digester and gasholder. All possible heat sources and sinks (ambient air temperature, wind, rain, and solar radiation) and heat exchanges or losses via advection, convection, and conduction, as well as a complete representation of the infrared radiative networks between the surfaces of the digester, are included for each compartment depending on the operating and design parameters. These heat exchanges are subject to fluctuating environmental conditions (e.g., ambient air temperature, wind, rain, and solar radiation).The results indicate that triple-membrane gasholders with a third insulation membrane made of a suitable material and thickness, together with the involvement of heat recovery from the digestate advective heat, are capable of reducing the overall thermal losses by more than 95 % (e.g., −51 % of the gasholder cover loss and − 81 % of the advective digestate loss) and when the waste heat from biogas purification is also valorized, the biogas plant can become thermally self-sufficient.

Abstract

International audience; In a circular economy approach, heterogeneous wastes can be upgraded to energy in the form of syngas via pyrogasification, and then to methane via biomethanation. Working at high pressure is a promising approach to intensify the process and to reduce gas-liquid transfer limitations. However, raising the pressure could lead to reaching the CO inhibition threshold of the microorganisms involved in syngas-biomethanation. To investigate the impact on pressure on the process, a 10L continuous stirred tank reactor working at 4 bars and 55 °C was implemented. Syngas (40% CO, 40% H2, 20% CO2) biomethanation was performed successfully and methane productivity as high as 6.8 mmolCH4/Lreactor/h with almost full conversion of CO (97%) and H2 (98%) was achieved. CO inhibition was investigated and carboxydotrophs appeared less resistant to high CO exposition than methanogens.

Abstract

This paper deals with the control of the start-up phase in anaerobic digestion systems by using monitoring parameters already implemented that have been coupled to the control law. Methane yield, as well as methane flow rate, and the ratio of intermediate over total alkalinity have been used as control parameters. These measurements have been included in a switching control strategy to drive the process toward a steady state in which biogas production is optimized avoiding accumulation of volatile fatty acids. Experimental validation of this control strategy has been performed from the results obtained using a 358 litter fixed bed anaerobic digester treating industrial winery wastewater.

Abstract

Delta-8-tetrahydrocannabinol (Δ8-THC) is a psychotropic cannabinoid produced in low quantities in the cannabis plant. Refinements in production techniques, paired with the availability of inexpensive cannabidiol substrate, have resulted in Δ8-THC being widely marketed as a quasi-legal, purportedly milder alternative to Δ9-THC. Yet, little research has probed the behavioral and physiological effects of repeated Δ8-THC use. The present study aimed to evaluate the effects of acute and repeated exposure to Δ8-THC. We hypothesized that Δ8-THC produces effects similar to Δ9-THC, including signs of drug tolerance and dependence. Adult male and female C57BL/6J mice were treated acutely with Δ8-THC (6.25-100 mg/kg, i.p.) or vehicle and tested in the tetrad battery to quantify cannabimimetic effects (i.e., catalepsy, antinociception, hypothermia, immobility) as compared with a non-selective synthetic cannabinoid (WIN 55,212-2) and Δ9-THC. As previously reported, Δ8-THC (≥12.5 mg/kg) induced cannabimimetic effects. Pretreatment with the CB1 receptor-selective antagonist rimonabant (3 mg/kg, i.p.) blocked each of these effects. In addition, repeated administration of Δ8-THC (50 mg/kg, s.c.) produced tolerance, as well as cross-tolerance to WIN 55,212-2 (10 mg/kg, s.c.) in tetrad, consistent with downregulated CB1 receptor function. Behavioral signs of physical dependence in the somatic signs, tail suspension, and marble burying assays were also observed following rimonabant-precipitated withdrawal from Δ8-THC (≥10 mg/kg BID for 6 days). Lastly, Δ8-THC produced Δ9-THC-like discriminative stimulus effects in both male and female mice. Together, these findings demonstrate that Δ8-THC produces qualitatively similar effects to Δ9-THC, including risk of drug dependence and abuse liability. [ABSTRACT FROM AUTHOR]

Abstract

Granulocytic myeloid-derived suppressor cells (G-MDSCs) promote tumor growth and immunosuppression in multiple myeloma (MM). However, their phenotype is not well established for accurate monitoring or clinical translation. We aimed to provide the phenotypic profile of G-MDSCs based on their prognostic significance in MM, immunosuppressive potential, and molecular program. The preestablished phenotype of G-MDSCs was evaluated in bone marrow samples from controls and MM patients using multidimensional flow cytometry; surprisingly, we found that CD11b+CD14−CD15+CD33+HLADR− cells overlapped with common eosinophils and neutrophils, which were not expanded in MM patients. Therefore, we relied on automated clustering to unbiasedly identify all granulocytic subsets in the tumor microenvironment: basophils, eosinophils, and immature, intermediate, and mature neutrophils. In a series of 267 newly diagnosed MM patients (GEM2012MENOS65 trial), only the frequency of mature neutrophils at diagnosis was significantly associated with patient outcome, and a high mature neutrophil/T-cell ratio resulted in inferior progression-free survival (P < .001). Upon fluorescence-activated cell sorting of each neutrophil subset, T-cell proliferation decreased in the presence of mature neutrophils (0.5-fold; P = .016), and the cytotoxic potential of T cells engaged by a BCMA×CD3-bispecific antibody increased notably with the depletion of mature neutrophils (fourfold; P = .0007). Most interestingly, RNA sequencing of the 3 subsets revealed that G-MDSC–related genes were specifically upregulated in mature neutrophils from MM patients vs controls because of differential chromatin accessibility. Taken together, our results establish a correlation between the clinical significance, immunosuppressive potential, and transcriptional network of well-defined neutrophil subsets, providing for the first time a set of optimal markers (CD11b/CD13/CD16) for accurate monitoring of G-MDSCs in MM.
