No. of bitstreams: 0 Previous issue date: 2021-01-01 The present study investigated the serum microscopic agglutination test (MAT) among 203 bovine bulls with reproduction by natural means, without apparent signs of orchitis or inflammation of accessory reproductive glands. Simultaneously, the semen of all bulls was subjected to sperm viability analysis and PCR based on the 16S rRNA gene. PCR-positive results of semen samples were confirmed by sequencing. A modified seminal plasma agglutination (MSPA) test, replacing the blood serum of all bulls in the MAT with seminal plasma was performed as well. Eight (8/203 = 3.9%) semen samples from bulls were considered nonviable (necrospermia and azoospermia) without relation to the PCR diagnosis. No agglutinin titers were identified in MSPA test. A high frequency (132/203 = 65%) of leptospiral agglutinin titers was identified in the MAT, particularly for the Sejroe serogroup (Hardjo CTG, 100/203 = 49.3%; Wolffi 74/203 = 36.4%; Guaricura 72/203 = 35.5%; and Hardjoprajitno 56/203 = 27.6%). Three (3/203 = 1.5%) semen samples of bulls were positive in the PCR, but these results were not confirmed by sequencing. The high frequency of serovars from the Sejroe serogroup typically adapted to bovines indicates the need for measures for the prophylaxis/control of the pathogen on the sampled farms. Discrepancies among the MAT, sperm viability, and molecular detection of leptospires in semen highlight the need for a combination of methods to diagnose leptospirosis in bovine bulls. To our knowledge, modified seminal plasma agglutination is described for the first time here to investigate anti-Leptospira antibodies produced locally in the genital tract in the diagnosis of bovine leptospirosis among bulls that reproduce by natural means. Introdução: As verminoses impactam em sérios prejuízos para a ovinocultura. São enfermidades com ciclo oro-fecal, em que os animais infectados eliminam os ovos do parasito pelas fezes, e no meio ambiente eclodem, e dão origem, as larvas infectantes. Na espécie ovina são raros os estudos com o diagnóstico da oesofagostomose pela histopatologia. Em cabanhas endêmicas, identificam-se: animais com histórico de emagrecimento progressivo, queda da produção de lã, anorexia, diarreia verde escura, anemia, hipoproteinemia e morte. Os principais achados anatomopatológicos: formação de nódulos no intestino e reação do tecido linfóide regional. Na microscopia enterite linfoplasmocitária nodular. Objetivo: relatar os achados clínicos e anatomopatológicos da enterite linfoplasmocitária em ovino. Relato de caso: Foi realizada a necropsia de uma ovelha, fêmea, 4 anos, mestiça, com histórico de emagrecimento progressivo há 4 meses, sem resposta terapêutica a vermifugação. Caquexia, mucosas hipocoradas, palidez visceral. Conteúdo intestinal com aspecto esverdeado, presença de múltiplos nódulos calcificados na serosa do rúmen, retículo, omaso e abomaso. Áreas de congestão intestinal e linfadenomegalia (linfonodos mesentérios). Fragmentos de mucosa intestinal e linfonodos mesentéricos foram fixados em formal 10% e colorados em Hematoxilina e Eosina (H&E). O exame microscópico identificou enterite, hiperplasia linfocitária, com aumento dos folículos no tecido linfóide e edema nodal. Resultados: A oesofagostomose é uma verminose, subdiagnosticada em ovinos, causa enterite linfoplasmocitária e nódulos intestinais, o que, em algumas situações, inviabiliza a ação dos antiparasitários. É importante, suspeitar desta enfermidade, em ovelhas que apresentam este histórico. Os achados clínicos e anatomopatológicos possibilitaram diagnosticar enterite linfoplasmocitária, causada por Oesophagostomum spp. em ovelha. Conclusão: O exame anatomopatológico possibilitou estabelecer o diagnóstico de enterite linfoplasmocitária, ressalta-se a importância dos exames complementares, na elucidação dos óbitos em animais de produção.

We investigated the bacteriological and physical-chemical characteristics, as well as genotypic and phenotypic and profiles of Staphylococcus species isolated from 240 samples of breast milk from a bank in a teaching hospital. Dornic acidity of milk revealed that 95.4% (229/240) had acceptable limits (< 8.0 oD). Caloric intake showed a wide variation in cream content (4%), fat (4%) and energy values (559.81 Kcal/L). Staphylococcus (105/186 or 56.5%) and Enterobacter (25/186 or 13.4%) were the most prevalent genera, although other microorganisms were identified, including Klebsiella pneumoniae and Pseudomonas aeruginosa. Amoxicillin/clavulanic acid (125/157 or 79.6%), vancomycin (115/157 or 73.2%), and cephalexin (112/157 or 71.3%) were the most effective antimicrobials. High resistance rates of isolates were found to penicillin G (141/157 or 89.8%), ampicillin (135/157 or 86%), and oxacillin (118/157 or 75.2%). Multidrug resistance to ≥ 3 antimicrobials occurred in 66.2% (123/186) of the isolates. Residues of microbial multiplication inhibitory substances were found in 85% (204/240) of samples. Among the coagulase-positive-CPS and negative-CoNS staphylococci, the mecA gene was detected in 53.3% (8/15) and 75% (30/40), respectively. Genes sea, seb and sec were detected in 20% (3/15) of CPS, while tsst-1 was detected in 13.34% (2/15). In addition, 13.3% (2/15) of S. aureus were toxin-producers. Genes sea, seb and sec were detected in 90% (36/40), 5% (2/40) and 15% (6/40) CoNS, respectively. Enterotoxin production was identified in 5% (2/40) of CoNS. The identification of multidrug-resistant bacteria, staphylococci species toxin-producers harboring methicillin-resistance genes, and residues of microbial multiplication inhibitory substances reinforce the need for a continuous vigilance of milk quality offered to infant consumption by human banks. Etiology, multiple antibiotic resistance index, and acute-phase protein profile in the serum and synovial fluid of 44 horses presenting septic arthritis were investigated. Young animals up to 24 months of age not yet submitted to taming were sampled. Synovial samples from the horses were submitted to culture and bacterial isolates subjected to 18 antimicrobials to investigate the in vitro multidrug resistance pattern. Also, hematological aspects, fibrinogen, and serum levels of some acute-phase proteins (total protein, albumin, globulin, amyloid A, and C-reactive protein) were assessed. To statistical analysis, microbiological isolation were divided into four groups: G1 = animals with negative isolation, G2 = isolation of Gram-negative bacteria, G3 = isolation of Gram-positive bacteria), and G4 = isolation of fungi. Microbial isolation was obtained from 77.2% (32/44) of the synovial samples. Escherichia coli (7/44 = 15.9%), Staphylococcus aureus (5/44 = 11.4%), Streptococcus equi subsp. equi (3/44 = 6.8%), Aspergillus niger (3/44 = 6.8%), Pseudomonas aeruginosa (2/44 = 4.5%), Streptococcus equi subsp. zooepidemicus (1/44 = 2.3%), Trueperella pyogenes (1/44 = 2.3%), and Rhodococcus equi (1/44 = 2.3%) were prevalent bacterial agents. Aspergillus niger is reported by the first time as a primary agent of septic arthritis in horses. Gentamicin (84.9%), marbofloxacin (79.3%) and ceftriaxone (72.4%) were the most effective antimicrobials, whereas high resistance of the isolates (>50%) was found to erythromycin (75.9%), clarithromycin (75.9%), penicillin (69%), clindamycin (58.6%), and streptomycin (55.2%). Multiple antibiotic resistance index (≥ 0.3) was identified in 50% (16/32) of the isolates, with highest resistance in Gram-negative bacterial isolates. Serum levels of amyloid A, C-reactive protein, fibrinogen, globulin, and total protein, as well as the number of nucleated cells in the synovial fluid had values above the reference for horses, reinforcing acute-phase proteins as biomarkers in diagnosis of septic arthritis. We highlighted the diversity of microorganisms that may be involved in equine septic arthritis, the high resistance of bacterial isolates to conventional antimicrobials, the high lethality of young horses with septic arthritis, and importance of in vitro antimicrobial susceptibility pattern tests to therapeutic approaches in the disease. RESUMO: Foram investigados a etiologia, o índice de resistência múltipla aos antibióticos e o perfil de proteínas de fase aguda, no soro ou líquido sinovial, de 44 equinos com sinais clínicos compatíveis com artrite séptica. Foram amostrados animais até 24 meses de idade não submetidos a doma. As amostras de líquido sinovial dos 44 animais foram submetidas à cultura bacteriológica e fúngica, e os isolados bacterianos submetidos in vitro diante de 18 antimicrobianos visando investigar a multirresistência. Também foram avaliados os aspectos hematológicos e o fibrinogênio sérico, bem como os níveis séricos de certas proteínas de fase aguda (proteína total, albumina, globulina, amilóide sérico A e proteína C reativa). O isolamento microbiano das amostras foi dividido em quatro grupos: G1 = animais com isolamento negativo; G2 = isolamento de bactérias Gram-negativas; G3 = isolamento de bactérias Gram-positivas e G4 = isolamento fúngico. O isolamento microbiano foi obtido em 77,2% (32/44) das amostras de líquido sinovial. Aspergillus niger é relatado pela primeira vez como agente primário de artrite séptica em cavalos. Gentamicina (84,9%), marbofloxacino (79,3%) e ceftriaxona (72,4%) foram os antimicrobianos mais eficazes, enquanto elevada resistência dos isolados (>50%) foi encontrada para eritromicina (75,9%), claritromicina (75,9%), penicilina (69%), clindamicina (58,6%) e estreptomicina (55,2%). O índice de resistência múltipla aos antibióticos (≥ 0,3) foi identificado em 50% (16/32) dos isolados, com maior resistência obervada em bactérias Gram-negativas. Os níveis séricos de amilóide A, proteína C reativa, fibrinogênio, globulina e concentração total de proteínas, bem como o número de células nucleadas no líquido sinovial apresentaram valores acima da referência para equinos saudáveis, reforçando o uso destas proteínas de fase aguda como biomarcadores no diagnóstico da doença. O presente estudo enfatiza a diversidade de microrganismos que podem estar envolvidos na artrite séptica equina, a alta resistência dos isolados aos antimicrobianos convencionais, a alta mortalidade de equinos jovens com artrite séptica e a importância de realizar o tratamento da afecção com respaldo em testes de sensibilidade microbiana in vitro.

No. of bitstreams: 0 Previous issue date: 2021-03-01 Caseous lymphadenitis (CL) in sheep is a chronic contagious disease caused by Corynebacterium pseudotuberculosis, commonly characterized by abscess formation in peripheral lymph nodes and disseminated infections. Nonetheless, other microorganisms, including with zoonotic relevance, can be isolated from CL-resembling lymph nodes. Currently, mycobacteria have been reported in visceral granulomatous lesions in small ruminants, a fact that poses a public health issue, particularly in slaughtered sheep intended for human consumption. Cytology using fine needle aspiration and microbiological culturing are suitable tests for routine diagnostic, whereas present drawbacks and molecular methods have been confirmatory. Data about the occurrence of mycobacteria in both lymph nodes with aspect of CL and apparently healthy visceral nodes of sheep slaughtered for human consumption are scarce. In this study, 197 visceral lymph nodes of sheep showed lymphadenitis and 202 healthy visceral lymph nodes of slaughtered sheep intended for human consumption were submitted to conventional bacteriological diagnosis, mycobacteria culturing, and cytological evaluation. Compatible Corynebacterium isolates were subjected to multiplex PCR targeting 16S rRNA, rpoB, and pld genes to detect C. pseudotuberculosis. Based on microbiological identification, C. pseudotuberculosis (86/197; 43.7%), streptococci γ-hemolytic (17/197; 8.6%), and Trueperella pyogenes (12/197; 6.1%) were prevalent in lymph nodes with abscesses, as opposed to staphylococci (53/202; 26.2%) in apparently healthy lymph nodes. No mycobacteria were isolated. Cytology identified 49.2% (97/197) Gram-positive pleomorphic organisms (coryneform aspect). Multiplex PCR confirmed genetic material of C. pseudotuberculosis in 74.4% (64/86) of the samples with C. pseudotuberculosis isolation and 66% (64/97) samples with cytological coryneform aspect (κ = 86.78%; 95% CI = 79.87–93.68%). These findings emphasize the prevalence of C. pseudotuberculosis in abscess formation among peripheral lymph nodes of sheep. Other bacteria were also identified in lymph nodes sampled that resembling C. pseudotuberculosis–induced infections that may difficult the diagnosis. Multiplex PCR revealed a valuable assay to detect C. pseudotuberculosis, in addition to routine methods applied to CL-diagnosis. No mycobacteria were identified in lymph nodes sampled, with and without apparent lesions. Nonetheless, due to public health impacts, this pathogen should be considered as a differential diagnosis of C. pseudotuberculosis–induced infections during inspection procedures of slaughtered sheep intended for human consumption. Rhodococcus equi is a well-recognized Gram-positive intracellular facultative bacterium that is opportunistic in nature, which causes pyogranulomatous infections in humans and multiple host animals. The pathogenicity of the microorganism has been attributed to the presence of plasmid-encoded virulence-associated proteins (Vap). To date, three host-associated virulence plasmid types of R. equi have been identified as follows: the circular pVAPA and pVAPB, related, respectively, to equine and porcine isolates, and a recently described linear pVAPN plasmid associated with bovine strains, although these three types are found in human isolates. Recent phylogenomic studies support the evidence that human R. equi infection is zoonotically acquired. Nevertheless, data regarding distribution and prevalence of the host-adapted virulence plasmid types of R. equi isolated from meat animals are scarce or unnoticed. Here, the three host-associated virulence plasmid types (pVAPA, pVAPB, and pVAPN) were investigated in 154 R. equi isolates recovered from lymph nodes of cattle with lymphadenitis (n = 31), faeces of cattle without enteric signs (n = 49), as well as different clinical specimens from human patients (n = 74). The analysis of virulence profile of 74 R. equi from humans revealed six (8.1%) isolates pVAPB (type 8), two (2.7%) pVAPN, and one (1.3%) pVAPB (type 11), all of which were from lung samples from people living with HIV/AIDS. From the lymph node samples of cattle, 41.9% (13 of 31) isolates revealed pVAPN type, whereas all isolates from faecal samples were negative for three host-associated types. Here, recently described bovine-associated pVAPN type was detected in R. equi isolates recovered from the lungs of people living with HIV/AIDS and lymph nodes from slaughtered cattle intended for human consumption; a finding that represents a public health concern, mainly in countries where undercooked or raw meat are traditionally consumed.

Objetivou-se neste trabalho elaborar uma bebida fermentada de soja tipo ‘iogurte’ enriquecidacom inulina e saborizada com ameixa e avaliar sua qualidade microbiológica, sensorial e físicoquímica. Quatro tratamentos com adição de 0, 5, 10 e 15% de inulina foram produzidos. Asanalises microbiológicas foram de coliformes totais e termotolerantes, bolores e leveduras eSalmonella sp. A análise sensorial foi realizada por teste de ordenação e teste de aceitação como uso de escala hedônica. As análises físico-químicas (umidade, cinzas, proteína, lipídios,fibras, cor, viscosidade, pH, acidez e MEV) foram realizadas no produto que obteve maiorpreferência pelos consumidores no teste sensorial de ordenação. Na análise microbiológicatodos os tratamentos estavam dentro do padrão. O produto com 15% de adição de inulina obtevemaior preferência. Os principais componentes foram carboidratos 19,31%, proteínas 2,09%,com destaque para a fibra alimentar total 10,95%, sendo 9,49% para fibra alimentar solúvel e1,46% para fibra alimentar insolúvel. O pH diminuiu de 4,35 para 4,15 e a acidez aumentou de0,33 para 0,41. Conclui-se que a bebida fermentada de soja tem significativa quantidade defibras e é uma boa opção para pessoas com intolerância a lactose.

These animals share common pathogens with domestic swine (Sus scrofa); however, their role as potential carrier remains unclear. This study focused on detecting the prevalence of influenza A antibodies in Tayassu pecari and Pecari tajacu from commercial rearing farms from two states in Brazil. A set of 50 blood samples from Pecari tajacu and 55 from Tayassu pecari were analyzed using a commercial indirect ELISA in order to investigate anti influenza A antibodies. Pecari tajacu samples presented 22% (11/50) of seropositivity for the virus. Serological surveillance is an important tool to identify the presence and the spread of the influenza virus in feral pigs. A biotecnica de refrigeracao de semen equino para o trasporte de material genetico esta amplamente distribuida. Com a necessidade da manutencao da viabilidade espermatica e indispensavel a autilizacao e diluentes para este fim. O objetivo deste estudo foi avaliar se a adicao de quercetina, que e um flavonoide antioxidante, em diluentes espermaticos altera a viabilidade dos espermatozoides equinos refrigerados. Foram avaliados cinco ejaculados de um garanhao, por meio da analise de motilidade e vigor espermatico, em diferentes periodos de refrigeracao em quatro meios de diluicao: BotuSemen ® ; BotuSemen ® adicionado de 20 µg/mL de quercetina (SIGMA ® ); Solucao aquosa com 10% leite em po desnatado (Molico ® ) ou Solucao aquosa com 10% leite em po desnatado (Molico ® ) adicionado de 20 µg/mL de quercetina (SIGMA ® ). Por meio dos resultados obtidos no presente estudo, foi possivel verificar que nao ha diferenca estatistica entre a motilidade espermatica de semen de garanhao diluido em BotuSemen ® em relacao ao diluido solucao aquosa com 10% leite em po desnatado (Molico ® ) pelo periodo de refrigeracao avaliado (36h).

Coxiella burnetii is a zoonotic pathogen with a worldwide distribution that is responsible for Q fever in humans. It is a highly infectious bacterium that can be transmitted from cattle to humans through the consumption of unpasteurized milk. We report the molecular identification of C. burnetii in raw cow's milk being sold directly for human consumption in Brazil without official inspection or pasteurization. One hundred and twelve samples of raw milk were analysed by real-time quantitative PCR (qPCR), and C. burnetii was detected in 3.57% (4/112) of the samples at a concentration ranging from 125 to 404 bacteria per millilitre. The identification of this zoonotic pathogen in raw milk sold directly for human consumption is a public health concern since C. burnetii can be transmitted through the oral route. This result indicates that health education and other preventive measures should be officially implemented in Brazil to prevent the spread of infection. To our knowledge, this is the first qPCR-based detection of C. burnetii in raw milk samples from cows sold in Brazil that do not undergo official inspection or pasteurization.

Brucellosis remains as a public health concern worldwide. In domestic animals, the disease is characterized by reproductive disorders in male and female. Besides extensive use of serological tests and recent development of molecular biology techniques, microbiological culture of Brucella species is yet considered a “gold standard” method for diagnosis. Here, semen of 335 bovine bulls was subjected simultaneously to microbiological culture in Brucella agar, Farrell media, and CITA media to evaluate comparatively the best selective media for isolation of Brucella sp. Among all 335 samples, B. abortus B19 strain was isolated from semen of five (1.49%) bulls using the three selective media. However, Farrell media was considered the best selective media for microbiological diagnosis, because of allowed isolation of B. abortus B19 strain from bull semen without bacterial commensal or fungal contamination of plates. The aim of this study was to identify Mycobacterium species in milk samples from cows positive to the comparative intradermal test. Milk samples from 142 cows positive to the comparative intradermal test carried out in 4,766 animals were aseptically collected, cultivated on Lowenstein-Jensen and Stonebrink media and incubated for up to 90 days. Colonies compatible with mycobacteria were stained by Ziehl-Neelsen to detect acid-fast bacilli, while to confirm the Mycobacterium genus, conventional PCR was performed. Fourteen mycobacterial strains were isolated from 12 cows (8.4%). The hsp65 gene sequencing identified M. engbaekii (n=5), M. arupense (n=4), M. nonchromogenicum (n=3), and M. heraklionense (n=2) species belong to the Mycobacterium terrae complex. Despite the absence of M. tuberculosis complex species in the milk samples, identification of these mycobacteria highlights the risk of pathogen transmission from bovines to humans throughout milk or dairy products, since many of mycobacterial species described here have been reported in pulmonary and extrapulmonary diseases both in immunocompetent and immunocompromised people. Foram utilizados 60 cães apresentando quadro neurológico da cinomose com evolução de 10 dias. Os animais foram internados e receberam tratamento de suporte; foram avaliados diariamente e realizados hemograma, dosagem bioquímica e exame de urina tipo I. Os grupos 1 e 2 foram tratados com ribavirina e sua associação com DMSO; os grupos 3 e 4 com DMSO e prednisona e o grupos 5 com ribavirina e prednisona e o grupo 6 com ribavirina, prednisona e DMSO. Os animais foram anestesiados para a colheita de líquor, medula óssea e sangue, antes do tratamento para diagnóstico através da RT-PCR. As amostras negativas foram analisadas pela técnica de hn-PCR. Todos os animais apresentaram resultado positivo em pelo menos uma das duas reações. O efeito adverso da ribavirina e a sua associação com a prednisona foi a anemia hemolítica, que foi confirmada pela observação de bilirrubina na urina apenas dos cães tratados com ribavirina. The present study aims at the identification of undesirable effects of ribavirin, predinisone and DMSO in dogs naturally infected by canine distemper virus. The research analyzed 60 dogs with clinical neurological signs and 10 days of evolution. The animals were hospitalized for the appropriate support treatment; were daily observed, and complete blood cells count, biochemical analysis, and urine exam type I were conducted. Groups 1 and 2 were treated with ribavirin and its combination with DMSO; Groups 3 and 4 treated with prednisone and DMSO, Group 5 treated with ribavirin and prednisone, while Group 6 with ribavirin, prednisone and DMSO. Before the treatment, animals were anesthetized for the cerebrospinal fluid, bone marrow and blood samples collection for the diagnosis based on RT-PCR. The negative samples were analyzed using the hn-PCR technique. All the animals presented positive results in at least one of the 2 tests. The adverse result of ribavirin and its association with prednisone was characterized by haemolytic anemia, confirmed by the evaluation of bilirrubin occurrence only in the urine of dogs treated with ribavirin. B. abortus infections in bulls are related to reproductive tract infections, although infected animals show transient serological titers or nonreactor status. Thus, diagnosis of bovine brucellosis based exclusively on serological tests probably underestimates B. abortus infections in bulls. In this scenario, three hundred thirty-five serum samples from reproductively mature bovine bulls were subjected simultaneously to standard serodiagnosis using the rose Bengal test (RBT), 2-mercaptoethanol (2-ME), complement fixation (CFT), and fluorescence polarization assay (FPA). Furthermore, conventional semen plasma agglutination (SPA) and modified 2-ME, FC and, FPA were carried out in all bulls replaing serum by seminal plasma. Semen from all bulls was also analyzed for sperm viability, microbiological culture in Farrell media, and polymerase chain reaction (PCR). Only eight (2.38%) semen samples were considered improper for reproduction services (necrospermia and azoospermia), although none of these animals was positive in any of the diagnosis methods used. Five bulls (1.49%) were simultaneously positive in conventional RBT, 2-ME, SPA, modified 2-ME, microbiological culture in Farrell media, and in PCR for B. abortus strain 19. Two (1.67%) bulls were positive in PCR for B. abortus field strains and negative in all other tests, although semen was considered viable to reproduction service. The identification of B. abortus B19 strain in serum and semen of bulls occurred probably due to improper vaccination of males or infection by B19 strain shedding by vaccinated females that could to contaminated environment of farms. In addition, detection of B. abortus field strains only using PCR in bulls without sperm viability abnormalities indicate the need for including molecular methods to improve diagnosis of the disease in bovine bulls. The caudal vena cava thrombosis, or pulmonary thromboembolism, in cattle is correlated with lactic acidosis, caused by diets rich in grains and highly fermentable, associated or not to septic situations, used in feedlots of beef or high-producing dairy cattle. This paper reports an unusual caudal vena cava thrombosis in a cow, secondary to Trueperella (Arcanobacterium) pyogenes infection, resulting in reduced milk production, anorexia, pale mucous membranes, ruminal atony, sternal decubitus and autoauscultation position. The heart was enlarged at necropsy, presence of clots distributed along the thoracic cavity, adherence between lung and pleura, abscesses, emphysema, petechiae, suffusions and ecchymosis in lungs, thickening of the caudal vena cava wall, hepatomegaly with chronic passive congestion ("nutmeg" aspect), and rumenitis. In lab, the actinomycete Trueperella (Arcanobacterium) pyogenes was isolated from liver and lung samples, probably resulting through dissemination of the bacteria of the rumen content, what reaffirms the opportunistic behavior of this actinomycete. Resumo: A síndrome da veia cava caudal ou tromboembolismo pulmonar bovino está relacionada à acidose láctica causada por dietas ricas em grãos e altamente fermentáveis, associados ou não a quadros sépticos, usadas em confinamentos de bovinos de corte ou para vacas leiteiras de alta produção. O presente artigo reporta caso raro de trombose da veia cava caudal em uma vaca, secundária a infecção por Trueperella (Arcanobacterium) pyogenes, apresentando reduzida produção de leite, anorexia, palidez de mucosas, atonia ruminal, decúbito esternal e posição de autoauscultação. À necrópsia observou-se coração aumentado de tamanho, coágulos distribuídos por toda cavidade torácica, aderência entre os pulmões e pleura, abscessos, enfisema, petéquias, sufusões, equimoses nos pulmões, espessamento da parede da veia cava caudal com trombo, hepatomegalia com congestão passiva crônica (aspecto de "noz moscada"), e ruminite. Em laboratório isolou-se o actinomiceto Trueperella (Arcanobacterium) pyogenes a partir de amostras de fígado e pulmão, provavelmente resultando da disseminação da bactéria proveniente do conteúdo ruminal, e reafirma o comportamento oportunista deste actinomiceto.

Molecular identification classified the strains as Nocardia farcinica and Nocardia cyriacigeorgica. The thermorresistance to boiling of the isolates was carried out and was observed bacterial growth after boiling. Staphylococcus sp., Corynebacterium sp., Actinobacillus sp., Nocardia sp. e as enterobacterias os micro-organismos mais frequentemente envolvidos. A doença está relacionada a injúrias e traumas nas mamas e tetos. Edema e fibrose da glândula mamária, leite com aspecto viscoso ou muco-purulento são observados nos casos de mastite clínica. O diagnóstico é baseado no exame clínico da mama associado ao cultivo microbiológico do leite. Os métodos indiretos para o diagnóstico de mastite subclínica como California Mastits Test e Contagem de Células Somáticas não são padronizados para a espécie em questão. A terapia para a mastite equina é baseada no uso sistêmico de antimicrobianos, com base nos resultados obtidos no teste “in vitro” de sensibilidade microbiana. Não há medidas de controle específicas para a mastite equina. Objetivou-se neste estudo revisar os principais aspectos relacionados a mastite em éguas com ênfase na etiologia, epidemiologia, sinais clínicos, diagnóstico, tratamento, medidas de controle e profilaxia. Mastits is considered uncommon disease in mares. Streptococcus equi. Staphylococcus sp., Corynebacterium sp., Actinobacillus sp., Nocardia sp. and enterobacterias are major microorganisms involved in equine mammary infections. The disease is commonly related to traumatic lesions in mammary glands and teats. Edema, fibrosis, masses to palpation of glands, and viscous to seropurulent milk are mainly clinical signs observed in affected animals. The diagnosis is based on clinical exam of mammary glands and microbiological culture of the milk. There are no standard to use of indirect exams on diagnosis, including California Mastits Test and Somatic Cell Count. Systemic antimicrobials are recommended in therapy, based on previous “in vitro” susceptibility microbiological test. No specific control measures are indicated in equine mastits. The present study reviewed the mastits in mares, with emphasis to etiology, epidemiology findings, clinical manifestation, diagnosis, treatment and control aspects. RESUMO A terapia antimicrobiana ainda e a ferramenta de escolha para o controle de infeccoes causadas pelos generos Pasteurella e Mannheimia . O trabalho apresenta os resultados dos Testes de Sensibilidade a Antimicrobianos, realizados atraves do metodo de difusao pelo sistema de discos, de 71 amostras de Pasteurella multocida e 71 de Mannheimia haemolytica isoladas de varias especies animais, provenientes da regiao de Botucatu, Sao Paulo, Brasil, entre Janeiro de 2000 e Novembro de 2007. Os resultados demonstram grande diferenca de sensibilidade entre os isolados, variando de 74,54% (sulfa-trimetoprim) a 100,00% (ciprofloxacina e florfenicol) , em relacao a P. multocida e de 80,05% (gentamicina) a 100,00% (ampicilina), em relacao a M. haemolytica , alem da ocorrencia de resistencia multipla. Estes dados indicam a ocorrencia de selecao de cepas resistentes, devido ao uso indiscriminado de antimicrobianos, e a importância de testar a sensibilidade, in vitro , de cepas patogenicas envolvidas em casos clinicos, para definir a terapia antimicrobiana mais apropriada. Susceptibility Profile of Samples of Pasteurella multocida and Mannheimia haemolytica Isolated in the Botucatu’s Region, Sao Paulo, Brazil, in the Period from January 2000 to November 2007 ABSTRACT The antimicrobial therapy is still the tool of choice for control of infections caused by Pasteurella and Mannheimia genres. The paper presents the results of Antimicrobial Susceptibility Tests, performed by disk diffusion method, of 71 samples of Pasteurella multocida and 71 of Mannheimia haemolytica isolated from many species of animals coming from Botucatu’s region, Sao Paulo, Brazil, between January 2000 and November 2007. The results demonstrate large difference in susceptibility among the isolates, ranging from 74,54% (sulfa-trimethoprim) to 100,00% (ciprofloxacin and florfenicol), with regard to Pasteurella multocida , and from 80,05% (gentamicin) to 100,00% (ampicilin), with regard to Mannheimia haemolytica , in addition of the occurrence of multiple resistance. Avaliou-se a qualidade microbiológica, constituintes nutricionais e presença de substâncias inibidoras do crescimento bacteriano em 100 amostras informais de leite de vacas, comercializadas na região sudoeste do estado de São Paulo, Brasil. Verificou-se que 77% da Contagem de Células Somáticas (CCS) e 86% da Contagem Bacteriana Total (CBT) das amostras estavam em desacordo com os valores máximos exigidos pela Instrução Normativa 62 do MAPA. Identificaram-se teores em desacordo para sólidos totais (36%), proteína (23%), gordura (38%), extrato seco desengordurado (43%) e nitrogênio uréico (73%). Em 59% das amostras detectou-se a presença de resíduos de inibidores do crescimento bacteriano. Isolaram-se 240 linhagens de micro-organismos com predomínio de enterobactérias, estreptococos, estafilococos e fungos. O leite informal é comercializado na região sem as mínimas condições de higiene e oferece riscos a saúde pública, além de representar sério problema sócio-econômico para região. The informal milk is recognized as the product marketed without inspection. We evaluated the microbiological quality, nutritional ingredients and substances inhibiting bacterial growth in 100 informal samples from cows milk, marketed in the southwest region of the state of Sao Paulo, Brazil. It was found that 77% of Somatic Cell Count (SCC) and 86% of Total Bacterial Count (TBC) of the samples were at odds with the maximum values required by Instruction Rules 62 of the Ministry of Agriculture and Livestock Supply. It was identified to solids contents in disagreement (36%), protein (23%), fat (38%), nonfat dry extract (43%) and 73% urea nitrogen. In 59% of the samples was detected residues of the bacterial growth inhibitors. Were isolated 240 strains of micro-organisms with a prevalence of enterobacteria, streptococci, staphylococci and fungi. The informal milk is marketed in the region without the minimum hygienic conditions and present risks to public health, and represents serious socio-economic problem for the region. A ribavirina é uma droga antiviral inibidora da replicação de alguns vírus e é reconhecida a sua eficácia frente ao vírus da cinomose “in vitro”. Foram selecionados dez animais com sinais neurológicos e evolução clínica da enfermidade de até dez dias. Foi colhido o líquor de todos os animais e neste foram avaliadas principalmente as características de celularidade e proteinorraquia. Os animais incluídos na pesquisa receberam ribavirina por via oral, na dose de 30mg/kg, a cada 24 horas, durante 15 dias. No último dia de tratamento foi realizada nova colheita de líquor. Os parâmetros liquóricos foram avaliados pelo teste de Mc Nemar, para os seguintes achados: pleocitose, número de linfócitos acima de 60%, proteínas acima de 25 mg/dl e teste de Pandy. Não houve significância estatística entre as alterações da primeira colheita e a segunda. Apenas três animais vieram a óbito, com piora clínica progressiva; outros dois animais não obtiveram melhora aparente, porém sobreviveram ao período de tratamento e os demais apresentaram redução dos sinais. A ribavirina foi capaz de diminuir a gravidade das lesões na maioria dos animais tratados. Canine Distemper is an important infecto-contagious disease with no effective antiviral treatment. Ribavirin is an antiviral drug that inhibits some virus replication and well known for its effectiveness against Canine Distemper Virus “in vitro”. Ten dogs with neurological signs and 10 days of maximum clinical evolution were selected to the present study. The cerebrospinal fluid was collected from all the animals for protein compounds and cellularity characteristics evaluations. Animals were treated with ribavirin orally, with dose of 30 mg/kg, each 24 hours, for 15 days. On the 15th day of treatment, a second cerebrospinal fluid collection was made and the parameters were evaluated by Mc Nemar test to the following findings: pleocytosis, lymphocytes above 60%, protein up to 25 mg/dL and Pandy test. No statistical difference was found between the first and second moments. However only three animals died with progressive clinical worsening; other two animals did not show clinical improvement, but survived to the treatment and the other animals presented signs reduction. Ribavirin has proven effectiveness in decreasing the gravity of lesions in majority of treated animals. The epidemiological findings, clinical signs, microbiological exams, somatic cell count, in vitro susceptibility profile of strains, efficacy of intramammary treatment and control measures were discussed. Florfenicol (96.2%), cefoperazona (92.3%), cefaloxin (84.6%) and ceftiofur (84.6%) were the most effective antimicrobials, and neomicin (27.0%) and enrofloxacin (17.4%) the least effective antimicrobials. No. of bitstreams: 0 Previous issue date: 2013-04-24 Background: Mycobacterium spp. is one of the most important species of zoonotic pathogens that can be transmitted from cattle to humans. The presence of these opportunistic, pathogenic bacteria in bovine milk has emerged as a public-health concern, especially among individuals who consume raw milk and related dairy products. To address this concern, the Brazilian control and eradication program focusing on bovine tuberculosis, was established in 2001. However, bovine tuberculosis continues to afflict approximately 1,3 percent of the cattle in Brazil. In the present study, 300 samples of milk from bovine herds, obtained from both individual and collective bulk tanks and informal points of sale, were cultured on Löwenstein-Jensen and Stonebrink media. Polymerase chain reaction (PCR)-based tests and restriction-enzyme pattern analysis were then performed on the colonies exhibiting phenotypes suggestive of Mycobacterium spp., which were characterized as acid-fast bacilli.Results: Of the 300 bovine milk samples that were processed, 24 were positively identified as Mycobacterium spp.Molecular identification detected 15 unique mycobacterial species: Mycobacterium bovis, M. gordonae, M. fortuitum, M. intracellulare, M. flavescens, M. duvalii, M. haemophilum, M. immunogenum, M. lentiflavum, M. mucogenicum, M. novocastrense, M. parafortuitum, M. smegmatis, M. terrae and M. vaccae. The isolation of bacteria from the various locations occurred in the following proportions: 9 percent of the individual bulk-tank samples, 7 percent of the collective bulk-tank samples and 8 percent of the informal-trade samples. No statistically significant difference was observed between the presence of Mycobacterium spp. in the three types of samples collected, the milk production profiles, the presence of veterinary assistance and the reported concerns about bovine tuberculosis prevention in the herds.Conclusion: The microbiological cultures associated with PCR-based identification tests are possible tools for the investigation of the presence of Mycobacterium spp. in milk samples. Using these methods, we found that the Brazilian population may be regularly exposed to mycobacteria by consuming raw bovine milk and related dairy products. Few studies have investigated the etiological complexity of this disease, as well as multidrug resistance of isolates. In this study, 60 horses with arthritis had synovial fluid samples aseptically collected, and tested by microbiological culture and in vitro susceptibility test (disk diffusion) using nine antimicrobials belonging to six different pharmacological groups. Bacteria were isolated in 45 (75.0%) samples, as follows: Streptococcus equi subsp. equi (11=18.3%), Escherichia coli (9=15.0%), Staphylococcus aureus (6=10.0%), Streptococcus equi subsp. zooepidemicus (5=8.3%), Staphylococcus intermedius (2=3.3%), Proteus vulgaris (2=3.3%), Trueperella pyogenes (2=3.3%), Pseudomonas aeruginosa (2=3.3%), Klebsiella pneumoniae (1=1.7%), Rhodococcus equi (1=1.7%), Staphylococcus epidermidis (1=1.7%), Klebsiella oxytoca (1=1.7%), Nocardia asteroides (1=1.7%), and Enterobacter cloacae (1=1.7%). Ceftiofur was the most effective drug (> 70% efficacy) against the pathogens in the disk diffusion test. In contrast, high resistance rate (> 70% resistance) was observed to penicillin (42.2%), enrofloxacin (33.3%), and amikacin (31.2%). Eleven (24.4%) isolates were resistant to three or more different pharmacological groups and were considered multidrug resistant strains. The present study emphasizes the etiological complexity of equine septic arthritis, and highlights the need to institute treatment based on the in vitro susceptibility pattern, due to the multidrug resistance of isolates. According to the available literature, this is the first report in Brazil on the investigation of the etiology. of the septic arthritis in a great number of horses associated with multidrug resistance of the isolates.
