Abstract

In recent years, zebrafish have become commonly used as a model for studying human traits and disorders. Their small size, high fecundity, and rapid development allow for more high-throughput experiments compared to other vertebrate models. Given that zebrafish share >70% gene homologs with humans and their genomes can be readily edited using highly efficient CRISPR methods, we are now able to rapidly generate mutations impacting practically any gene of interest. Unfortunately, our ability to phenotype mutant larvae has not kept pace. To address this challenge, we have developed a protocol that obtains multiple phenotypic measurements from individual zebrafish larvae in an automated and parallel fashion, including morphological features (i.e., body length, eye area, and head size) and movement/behavior. By assaying wild-type zebrafish in a variety of conditions, we determined optimal parameters that avoid significant developmental defects or physical damage; these include morphological imaging of larvae at two time points [3 days post fertilization (dpf) and 5 dpf] coupled with motion tracking of behavior at 5 dpf. As a proof-of-principle, we tested our approach on two novel CRISPR-generated mutant zebrafish lines carrying predicted null-alleles of syngap1b and slc7a5, orthologs to two human genes implicated in autism-spectrum disorder, intellectual disability, and epilepsy. Using our optimized high-throughput phenotyping protocol, we recapitulated previously published results from mouse and zebrafish models of these candidate genes. In summary, we describe a rapid parallel pipeline to characterize morphological and behavioral features of individual larvae in a robust and consistent fashion, thereby improving our ability to better identify genes important in human traits and disorders.

Abstract

Background: Regular physical activity plays a crucial role in modulating cellular metabolism and mitigating the progression of neurodegenerative diseases such as Alzheimer's, Parkinson's, and Multiple Sclerosis. Objective: The objective of this review is to evaluate the molecular mechanisms by which exercise influences cellular metabolism, with a focus on its potential as a therapeutic intervention for neurological disorders. Methods: A comprehensive literature review was conducted using peer-reviewed scientific articles, with a focus on the period between 2015 and 2024, to analyze the effects of exercise on mitochondrial function, oxidative stress, and metabolic health. Results: The findings indicate that exercise promotes mitochondrial biogenesis, enhances oxidative phosphorylation, and reduces reactive oxygen species, contributing to improved energy production and cellular resilience. These metabolic adaptations are associated with delayed disease progression and reduced symptoms in patients with neurodegenerative conditions. Additionally, integrating exercise with nutritional strategies may further enhance therapeutic outcomes by addressing metabolic disturbances comprehensively. Conclusions: This review concludes that personalized exercise protocols should be developed to optimize metabolic benefits for patients with neurological diseases, while future research should focus on biomarker development for individualized treatment approaches. Abstract

Objectives: This study aimed to investigate the effects of external (EXT) versus internal (INT) focus of attention on acquiring and transferring simple and complex motor skills in novice female volleyball players. Methods: Fifty-seven participants were assigned to one of the three groups: The external focus (EXT), the internal focus (INT), and the control group (CON). Over a 6-week intervention, participants practiced a simple skill (volleyball passing) and a complex skill (overhead tennis serve). Pre-tests, post-tests, and transfer tests (conducted two weeks later) assessed movement form and accuracy. Results: The results showed a statistically significant interaction among focus methods, skill complexity, and testing periods for movement form and performance outcome. The EXT group showed higher movement form and performance outcome improvements during the simple skill (passing) compared to the INT and CON groups. For the complex skill (serving), the EXT group outperformed the INT and CON groups regarding movement outcome; however, no differences in movement form were observed between the EXT and INT groups. Post hoc Tukey tests indicated that the EXT group scored higher than the INT and CON groups in the post-test and transfer test for the simple skill, with movement form improving from 1.68 ± 0.48 to 2.32 ± 0.58. In the complex skill, the EXT group's movement outcome improved from 11.32 ± 6.83 to 44.47 ± 16.57. Conclusions: External focus significantly enhances movement form and outcome in simple tasks and performance outcomes in more complex skills among novice athletes. [ABSTRACT FROM AUTHOR]

Abstract

The current study examined the changes in muscle oxygenation values and swimming performance after six sessions of sprint interval training during a three-week period in untrained swimmers. Twelve swimmers of both genders (age: 23.5 ± 5.6yrs) executed the twice-weekly experimental training protocol (EXP, n = 12), consisting of a 4 × 50 m front-crawl swimming (repeated sprint training—RST) with maximal intensity, and 2 min of passive recovery in between, after a short in-water warm-up. The control group (CON, n = 9) performed a continuous swimming set (200 m) at 120 b pm−1, with the same weekly frequency. Performance times in two maximum swim trials (400 m: T400 and 50 m: T50), muscle oxygenation of the deltoid muscle (SmO2) immediately after T400 and T50, 1-min heart rate recovery (HRR1) after T400, T50, and swim strokes during both swim trials (S/T400, S/T50) were assessed. For the EXP group, T400 improved by 2.4 (p = 0.011). In contrast, T50 presented no significant improvement (1%, p > 0.05). SmO2 decreased at T400 (5.5%, p = 0.017) and increased at T50 (3.7%, p = 0.030). HRR1 improved after T400 (7.9%, p = 0.002), T50 (4.6%, p = 0.005) and RST (9.6%, p = 0.002). S/T400 and S/T50 remained relatively unchanged (p > 0.05). The CON group presented no significant changes in any of the variables examined. In conclusion, six sprint interval training sessions can improve aerobic capacity over a 3-week training period, as indicated by the enhanced T400 performance and the reduced HRR1 values, in previously trained swimmers. Finally, the sensitivity of the near-infrared spectroscopy method to detect short-term training-induced changes is highlighted. [ABSTRACT FROM AUTHOR]

Abstract

The increasing ubiquity of digital devices in childhood had outpaced the understanding of their effects on cognitive development, creating a significant research gap regarding their long-term impact. Objective: The present narrative overview explored the complex relationship between digital device usage and cognitive development in childhood. Methods: We conducted a comprehensive literature search across multiple databases, including PubMed, Embase, Scopus, and Web of Science, to critically assess cognitive domains such as attention, memory, executive functions, problem-solving skills, and social cognition. Incorporating over 157 peer-reviewed studies published between 2001 and 2024, we used strict inclusion and exclusion criteria to ensure scientific rigor. Results: The review integrated empirical findings with established theoretical frameworks, particularly from cognitive development and media psychology, to highlight both the advantages and risks of early, frequent exposure to technology. The potential for digital devices to enhance cognitive skills, such as multitasking and information processing, was weighed against risks such as cognitive overload, diminished attention spans, and impaired social skills. We also examined psychological and behavioral outcomes, including identity formation, emotional regulation, and maladaptive behaviors associated with excessive screen time. Additionally, we identified strategies to mitigate negative effects, emphasizing structured digital engagement and parental involvement to support healthy cognitive and psychological growth. Our findings provided actionable recommendations for parents, educators, and policymakers, promoting optimal digital practices that enhanced cognitive development while safeguarding against potential harms. Conclusions: The review offered essential insights for stakeholders in child development, education, and policy-making, highlighting the need for balanced integration of digital tools in childhood learning environments. Abstract

The present study addressed the effect of the cognitive stimulation program Lumosity, in the stimulation of the cognitive functions of memory, selective attention, visual attention, processing speed, problem solving and cognitive flexibility related to learning disorders. Methodology with quantitative, cross-sectional approach has a correlational scope and a pre-experimental, pretest-posttest design of a single group. For the sample, a non-probabilistic purposive sampling was applied for a total of five school children between 8 and 11 years of age. The cognitive processes were evaluated by means of pretest and posttest with the test (ENI), correlating them with the results obtained during each intervention session, the sessions were applied twice a week, during eight weeks, giving a total of sixteen sessions, at the beginning of each session physiological, emotional and psychological aspects were registered, As for the results, based on the weekly averages of the stimulation program, an ascending curve was observed in 80% of the participants, as for the application of the ENI scale, significant changes were evidenced and others were linear, the highest ascending learning score was 71.4% (five areas) and the lowest was 71.4% (five areas). The highest ascending learning score was 71.4% (five areas) and the lowest was 28.5% (two areas)., El presente estudio abordo el efecto del programa de estimulación cognitiva Lumosity, en la estimulación de las funciones cognitivas de memoria, atención selectiva, atención visual, velocidad de procesamiento, resolución de problemas y flexibilidad cognitiva relacionadas con los trastornos de aprendizaje. Metodología con enfoque cuantitativo, transversal tiene un alcance correlacional y un diseño preexperimental, pretest-postest de un solo grupo. Para la muestra se aplicó, un muestreo no probabilístico de tipo intencionado para un total de cinco niños escolares entre los 8 y 11 años. Los procesos cognitivos fueron evaluados por medio de pretest y postest con la prueba (ENI), correlacionándolos con los resultados obtenidos durante cada sesión de intervención, las sesiones se aplicaron dos veces a la semana, durante ocho semanas, dando un total de dieciséis sesiones, al inicio de cada sesión se registraban aspectos fisiológicos, emocionales y psicológicos En cuanto a los resultados, con base en los promedios semanales del programa de estimulación, se observó una curva ascendente en el 80% de los participantes, en cuanto a la aplicación de la escala ENI, se lograron evidenciar cambios significativos y otras quedaron lineales, el puntaje de aprendizaje ascendente mayor fue de 71.4% (cinco áreas) y el menor de 28.5% (dos áreas).

Abstract

Plant–soil biodiversity interactions are fundamental for the functioning of terrestrial ecosystems. Yet, the existence of a set of globally distributed topsoil microbial and small invertebrate organisms consistently associated with land plants (i.e., their consistent soil-borne microbiome), together with the environmental preferences and functional capabilities of these organisms, remains unknown. We conducted a standardized field survey under 150 species of land plants, including 58 species of bryophytes and 92 of vascular plants, across 124 locations from all continents. We found that, despite the immense biodiversity of soil organisms, the land plants evaluated only shared a small fraction (less than 1%) of all microbial and invertebrate taxa that were present across contrasting climatic and soil conditions and vegetation types. These consistent taxa were dominated by generalist decomposers and phagotrophs and their presence was positively correlated with the abundance of functional genes linked to mineralization. Finally, we showed that crossing environmental thresholds in aridity (aridity index of 0.65, i.e., the transition from mesic to dry ecosystems), soil pH (5.5; i.e., the transition from acidic to strongly acidic soils), and carbon (less than 2%, the lower limit of fertile soils) can result in drastic disruptions in the associations between land plants and soil organisms, with potential implications for the delivery of soil ecosystem processes under ongoing global environmental change.

Abstract

Mineral-associated organic carbon (MAOC) constitutes a major fraction of global soil carbon and is assumed less sensitive to climate than particulate organic carbon (POC) due to protection by minerals. Despite its importance for long-term carbon storage, the response of MAOC to changing climates in drylands, which cover more than 40% of the global land area, remains unexplored. Here we assess topsoil organic carbon fractions across global drylands using a standardized field survey in 326 plots from 25 countries and 6 continents. We find that soil biogeochemistry explained the majority of variation in both MAOC and POC. Both carbon fractions decreased with increases in mean annual temperature and reductions in precipitation, with MAOC responding similarly to POC. Therefore, our results suggest that ongoing climate warming and aridification may result in unforeseen carbon losses across global drylands, and that the protective role of minerals may not dampen these effects.

We conducted a global field survey in urban greenspaces and neighboring natural ecosystems across 56 cities from six continents, and found that urban soils are important hotspots for soil bacterial, protist and functional gene diversity, but support highly homogenized microbial communities worldwide. Urban greenspaces had a greater proportion of fast-growing bacteria, algae, amoebae, and fungal pathogens, but a lower proportion of ectomycorrhizal fungi than natural ecosystems. These urban ecosystems also showed higher proportions of genes associated with human pathogens, greenhouse gas emissions, faster nutrient cycling, and more intense abiotic stress than natural environments. City affluence, management practices, and climate were fundamental drivers of urban soil communities. Soils are one of Earth’s largest reservoirs of ARGs, which are integral for soil microbial competition, and have potential implications for plant and human health. Yet, their diversity and global patterns remain poorly described. Here, we analyzed 285 ARGs in soils from 1012 sites across all continents and created the first global atlas with the distributions of topsoil ARGs. Results We show that ARGs peaked in high latitude cold and boreal forests. Climatic seasonality and mobile genetic elements, associated with the transmission of antibiotic resistance, were also key drivers of their global distribution. Dominant ARGs were mainly related to multidrug resistance genes and efflux pump machineries. We further pinpointed the global hotspots of the diversity and proportions of soil ARGs. Abstract

It is shown that the spanning set for L^2([0, 1]) provided by the eigenfunctions {sqrt{2} sin(n\pi x)}_{n=1}^{\infty} of the particle-in-a-box in quantum mechanics provide a very effective variational basis for more general problems. The basis is scaled to [a,b], where a and b are then used as variational parameters. What is perhaps a natural basis for quantum systems confined to a spherical box in R^d, turns out to be appropriate also for problems that are softly confined by U-shaped potentials, including those with strong singularities at r=0. This narrative review examines the interplay among body image perceptions, social media influence, physical measurements, and their impact on psychological well-being, focusing on the roles of cultural and gender differences and the need to understand the research methodologies employed in this field. In the age of digital proliferation, platforms like Instagram and Facebook have reshaped body image concerns, often leading to increased dissatisfaction and psychological distress due to constant exposure to idealized images and a culture of social comparison. Physical attributes such as weight, height, and BMI are scrutinized under societal standards of health and attractiveness, contributing to a spectrum of mental health issues including low self-esteem, depression, and eating disorders. This examination reveals how cultural norms and gender expectations further complicate body image perceptions, affecting individuals differently based on societal and personal ideals. It synthesizes current research and types of methods to illuminate how these factors together influence mental health and self-esteem, advocating for comprehensive interventions and policy measures aimed at mitigating body dissatisfaction and promoting a healthier, more inclusive understanding of body image. Abstract

Background/Objectives: The purpose of this study was to investigate whether or not there are notable differences between male and female vocational training students in terms of academic, behavioral, and psychophysiological characteristics. Methods: A total of 411 vocational training students, 210 of whom were male and 201 of whom were female, participated in an online survey that assessed academic, behavioral, and psychophysiological patterns. Results: In terms of extraversion and openness to experience, our research revealed that no statistically significant differences were noticed. When it came to agreeableness, however, significant differences were seen, with female students getting higher marks than male students. Furthermore, we found that there were significant gender differences in terms of neuroticism and conscientiousness, with female students achieving higher scores than male students. When compared to their male counterparts, females displayed higher performance in terms of academic success. They also demonstrated superior values in terms of reported stress, anxiety, and cognitive inflexibility. Conclusions: The findings of this study provide a more in-depth understanding of the numerous factors that have an impact on students engaging in vocational training.
