The objectives of this study were: 1- to evaluate the association of Bovine Viral Diarrhea Virus (BVDV), Bovine Herpes Virus 1 (BoHV-1), and Neospora caninum (N. caninum) with the risk for Late Embryonic Loss (LEL) in grazing dairy cows, 2- to evaluate blood progesterone concentration at the time of LEL occurrence, and 3- to describe a novel ultrasound-guided technique for conceptus sampling. We run a prospective cohort study involving 92 cows (46 LEL and 46 NLEL). An LEL cow was that having an embryo with no heartbeat, detached membranes, or floating structures, including embryo remnants detected at pregnancy check by ultrasonography (US) 28–42 days post-AI, whereas an NLEL cow was that with embryo heartbeats detectable by US at pregnancy check 28–42 d post-IA. We took two blood samples from every cow at pregnancy check by US (the day of LEL detection) and 28 d later to perform serological diagnosis of BVDV, BoHV-1, and N. caninum ; and to measure blood progesterone concentration at pregnancy check (28–42 d post-AI). We also sampled the conceptus from all the LEL cows. We performed PCR to detect BVDV, BoHV-1, and N. caninum in sampled conceptuses from LEL cows. Finally, we evaluated the associations of risk factors (serological titers, seroconversion, and progesterone) with LEL odds with logistic models. The risk for LEL was associated with serological titers to BVDV (P = 0.03) and tended to be associated with seroconversion to BVDV, given that 19.6% (9/46) in LEL and 6.5% (3/46) in NLEL cows seroconverted to BVDV (P = 0.09). In addition, BVDV was detected in conceptuses from LEL cows that seroconverted to BVDV but not in LEL cows that did not seroconvert. Conversely, the risk for LEL was not associated with the titers or seroconversion to BoHV-1 and N. caninum. BoHV-1 and N. caninum were not identified in any of the conceptuses. Finally, blood progesterone concentration was similar in LEL and NLEL cows, and it was not associated with the risk for LEL (P = 0.54). Background: there is consensus that reproductive efficiency plays a crucial role in the profitability of modern goat herds. Objective: to evaluate the effect of collection on semen quality in Santandereana goat breed. Materials and Methods: a prospective study was conducted on a commercial farm in Piedecuesta, Santander, Colombia where males (n = 10) were enrolled. Semen was collected from males with ages ranging from 2 to 5 years; an artificial vagina and electroejaculator were used. The first ejaculate was day zero and the second ejaculate was day 9. Motility analysis was performed with the help of a computer-assisted semen analysis. An analysis of repeated measures over time was performed with the SAS academics statistical package, using the Proc Glimmix method. Results: the artificial vagina showed the highest parameters for sperm vitality (81.03 ± 4.2 %) vs. (65.04 ± 4.7 %), motility (77.03 ± 4.1 %) vs. (54.1 ± 4.5) and progressive motility (34.5 ± 4.6) vs. (21.9 ± 4.4) (p < 0.05). There were not statistically significant (p > 0.05) in total motility, sperm vitality, and progressive motility between the first and second ejaculates. The sperm variables were not associated with progressive movement and speed (ALH, VAP, LIN) (p > 0.05). Conclusions: the method of semen collection with an artificial vagina was the most practice, easy, and showed better motility, sperm vitality, and progressive motility compared to the electroejaculator method. In the last 30 years, the scientific community has developed and proposed different models and numerical approaches for the study of vibrations induced by railway traffic. Most of them are formulated in the frequency/wave number domain and with a 2.5D approach. Three-dimensional numerical models formulated in the time/space domain are less frequently used, mainly due to their high computational cost. Notwithstanding, these models present very attractive characteristics, such as the possibility of considering nonlinear behaviors or the modelling of excess pore pressure and non-homogeneous and non-periodic geometries in the longitudinal direction of the track. In this study, two 3D numerical approaches formulated in the time/space domain are compared and experimentally validated. The first one consists of a finite element approach and the second one of a finite difference approach. The experimental validation in an actual case situated in Carregado (Portugal) shows an acceptable fitting between the numerical results and the actual measurements for both models. The main objective of this study was to evaluate the risk factors for late embryonic loss (LEL) in supplemented grazing dairy cows. Additional objectives were to assess the incidence of LEL and its association with the reproductive performance of cows. A data set containing productive, reproductive, and health records of 13,551 lactations was used. A retrospective case-control study involving 631 cows with LEL (cases) and 2,524 controls (4 controls per case within each study year) was run. A case of LEL was defined when the embryo had no heartbeat or there was evidence of detached membranes or floating structures including embryo remnants by ultrasonography (US) at 28 to 42 d post-artificial insemination (AI), whereas a non-case was defined as a cow diagnosed with positive pregnancy by US 28 to 42 d post-AI and reconfirmed as pregnant 90 ± 7 d post-AI. Four controls per case were randomly selected from the non-cases with a temporal matching criterion (±3 d around the date of the fecundating AI of the case). Multivariable logistic models were offered with the following predictors: year of LEL (2011 through 2015), season of LEL (summer vs. fall vs. winter vs. spring), parity (1 vs. 2 vs. ≥3), uterine disease (UD), non-uterine disease (NUD), body condition score at parturition, body condition score at 28 to 42 d post-AI (BCS-LEL), days in milk (DIM), and daily milk yield (MY). Statistical significance was set at P < 0.05 and a tendency was set at P ≤ 0.10. We found that 4.7, 22, and 23% of cows had LEL, UD, and NUD, respectively. Cases tended to have higher daily MY than controls (32.5 vs. 31.8 kg); also, cases had much longer calving to pregnancy interval (226 vs. 118 d), lower hazard of pregnancy [hazard ratio = 0.39, 95% confidence interval (CI) = 0.35–0.43], and higher odds for non-pregnancy [odds ratio (OR) = 2.89, 95% CI = 2.37–3.54] than controls. We found that the odds for LEL increased with parity number (OR = 2.48, 95% CI = 1.99–3.08 for parity ≥3) and with BCS-LEL <2.50 (OR = 1.81, 95% CI = 1.33–2.47). Conversely, the odds for LEL decreased with BCS-LEL >3.00 (OR = 0.70, 95% CI = 0.53–0.91). The odds for LEL increased with UD (OR = 1.23, 95% CI = 1.01–1.49), NUD (OR = 1.24, 95% CI = 1.01–1.54), DIM (OR = 1.03, 95% CI = 1.00–1.05), and daily MY (OR = 1.14, 95% CI = 1.04–1.25) in univariable models only. Finally, the odds for LEL were not associated with year, season, DIM, and body condition score at parturition. los procesos de obtención de SCP han sido desarrollados por diferentes investigadores tanto a nivel nacional como internacional. Objetivo: definir los pará- metros críticos del proceso de obtención de SCP, para incrementar el rendimiento y la calidad de este producto y sus derivados. Métodos: se utilizó el método de purificación de Pichansky, con algunas modificaciones en las siguientes variables: relación masa/volumen, concentración del etanol y el tiempo de agitación. Los materiales utilizados están avalados por el Sistema de Gestión de Calidad del Centro. Resultados: las variantes #2 y #4 del proceso de extracción, utilizadas a escala de laboratorio, son las de mejores resultados, ya que se obtuvo un rango de porcentaje sólidos totales entre un 12,4 y 14,3 %, con un rendimiento entre un 48,5 y 83,0 %; siendo seleccionada la variante #4 para elaborar los lotes experimentales a escala de reactor. Conclusiones: las variables estudiadas: tiempo de agitación, concentración de etanol y relación masa/volumen, así como el porcentaje de los sólidos totales, el porcentajedel rendimiento y las propiedades organolépticas, respondieron positivamente en las variantes #2 y #4, las cuales permitieron cumplir con los objetivos propuestos en esta investigación, y pueden emplearse en la elaboración de los lotes productivos dependiendo de la concentración de las soluciones alcohólicas de propóleos que se quieran producir. Dry wastes (dw) generated in processing mangoes, composed (in dry weight) mainly of soluble carbohydrates (71 ± 2%) and fiber (16 ± 1%), were evaluated as substrates in a "home-made" solid-state fermentation (using polyurethane foam as inert support matrix, various C:N ratios, moisture contents, and incubation periods) of Trichoderma asperellum T8a, a promising biological control agent against the mango pathogen Colletotrichum gloeosporioides (causal agent of anthracnose). Highest spore production (2.5 x 106 up to 76 ± 3 x 108 spores g-1 dw) occurred after 8 days of incubation [at 28 ± 1 °C, relative humidity of 85 ± 5%, photoperiod of 12h (540 Lux) - 12h (20 Lux)] at a C:N ratio of 26, and a moisture content of 78%. Scanning electron microscopy showed that T. asperellum T8a was able to grow on mango industrial wastes and into polyurethane foam. The extensive growth can be related to cellulases secreted by this fungus, liberating glucose from these wastes to its growth. Most (94 ± 1%) of the spores grown on mango industrial wastes survived storage at 4 °C for 7 days and were equally effective as those grown on potato dextrose agar medium (86 ± 4% viable) in biological control tests against C. gloeosporioides ATCC MYA 456. Results indicate the potential use of mango industrial wastes as substrates to produce T. asperellum T8a spores in situ (mango orchards) under a cheap "home-made" solid-state fermentation, reducing problems associated with wastes disposal and permitting the production of a biological control agent against C. gloeosporioides. Two extension projects have been carried out recently on the Madrid Metro, including 97 km of large-diameter (9.38 m) tunnels. Some technical specifications for the shields (thrust and torque) needed to excavate the tunnels have been analyzed in this work. The soil stability problem at the tunnel face has also been studied. Numerical tools based on a discrete method (particulate materials) have been used for these purposes. Results from the models are compared with real data and show the promising possibilities of the method to design both tunnels and tunneling machines. [ABSTRACT FROM AUTHOR]

Abstract: This work shows that Plasmodium falciparum merozoite surface protein-6 (MSP-6) peptides specifically bind to membrane surface receptor on human erythrocytes. Three high activity binding peptides (HABPs) were found: peptides 31175 (41MYNNDKILSKNEVDTNIESN60) and 31178 (101YDIQATYQFPSTSGGNNVIP120) in the amino terminal region and 31191 (361EIDSTINNLVQEMIHLFSNNY380) at the carboxy terminal. Their binding to erythrocytes was saturable. HABPs 31191 and 31178 recognized 56 and 26kDa receptors on erythrocyte membrane and inhibited in vitro Plasmodium falciparum merozoite invasion of erythrocytes by between 27% and 46% at 200μgml−1 concentration, suggesting that these MSP-6 protein peptides play a possible role in the invasion process. Abstract: Tryptophan–threonine-rich antigen (TryThrA) is a Plasmodium falciparum homologue of Plasmodium yoelii-infected erythrocyte membrane pypAg-1 antigen. pypAg-1 binds to the surface of uninfected mouse erythrocytes and has been used successfully in vaccine studies. The two antigens are characterized by an unusual tryptophan-rich domain, suggesting similar biological properties. Using synthetic peptides spanning the TryThrA sequence and human erythrocyte we have done binding assays to identify possible TryThrA functional regions. We describe four peptides outside the tryptophan-rich domain having high activity binding to normal human erythrocytes. The peptides termed HABPs (high activity binding peptides) are 30884 (61LKEKKKKVLEFFENLVLNKKY80) located at the N-terminal and 30901 (401RKSLEQQFGDNMDKMNKLKKY420), 30902 (421KKILKFFPLFNYKSDLESIM440) and 30913 (641DLESTAEQKAEKKGGKAKAKY660) located at the C-terminal. Studies with polyclonal goat antiserum against synthetic peptides chosen to represent the whole length of the protein showed that TryThrA has fluorescence pattern similar to PypAg-1 of P. yoelii. All HABPs inhibited merozoite in vitro invasion, suggesting that TryThrA protein may be participating in merozoite–erythrocyte interaction during invasion. [Copyright &y& Elsevier]

Solenodons are insectivores found only in Hispaniola and Cuba, with a Mesozoic divergence date versus extant mainland mammals. Solenodons are the oldest lineage of living eutherian mammal for which a mitogenome sequence has not been reported. We determined complete mitogenome sequences for six Hispaniolan solenodons (Solenodon paradoxus) using next-generation sequencing. The solenodon mitogenomes were 16,454–16,457 bp long and carried the expected repertoire of genes. A mitogenomic phylogeny confirmed the basal position of solenodons relative to shrews and moles, with solenodon mitogenomes estimated to have diverged from those of other mammals ca. 78 Mya. Control region sequences of solenodons from the northern (n = 3) and southern (n = 5) Dominican Republic grouped separately in a network, withFST= 0.72 (p = 0.036) between north and south. This regional genetic divergence supports previous morphological and genetic reports recognizing northern (S. p. paradoxus) and southern (S. p. woodi) subspecies in need of separate conservation plans. In this paper, a three-dimensional numerical model is applied for studying railway vibrations. The numerical model is based on finite element method formulated in the time domain with an implicit scheme of integration. In the first part of the paper the numerical approach is briefly described. An uncoupled scheme is applied: (i) firstly the train loads are calculated taking into account dynamic excitation with a 2D vehicle-track model; (ii) the computed loads on each sleeper are introduced into a 3D numerical model, developed in the commercial code Plaxis, which is used to simulate the wave propagation in the ground. In the second part of the paper a real case study, located in Portugal, is presented, being used for the experimental validation of the proposed model. The numerical results of ground vibrations show an acceptable agreement with real measurements. Therefore, the proposed approach can be used as a reliable prediction tool based on PLAXIS software, which allows simulating railway vibrations in some specific cases such as soft soils, inhomogeneous geometries in the longitudinal direction of the track and transition zones. Abstract: The process of Mycobacterium tuberculosis infection of the macrophage implies a very little-known initial recognition and adherence step, important for mycobacterial survival; many proteins even remain like hypothetical. The Rv1510c gene, encoding a putatively conserved membrane protein, was investigated by analysing the M. tuberculosis genome sequence data reported by Cole et al. and a previous report that used PCR assays to show that the Rv1510 gene was only present in M. tuberculosis. This article confirmed all the above and identified the transcribed gene in M. tuberculosis, Mycobacterium africanum, and in M. tuberculosis clinical isolates. Antibodies raised against peptides from this protein recognised a 44kDa band, corresponding to Rv1510c theoretical mass (44,294Da). Assays involving synthetic peptides covering the whole protein binding to U937 and A549 cell lines led to recognising five high activity binding peptides in the Rv1510 protein: 11094, 11095, 11105, 11108, and 11111. Their affinity constants and Hill coefficients were determined by using U937 cells. Cross-linking assays performed with some of these HABPs showed that they specifically bound to a U937 cell line 51kDa protein, but not to Hep G2 or red blood cell proteins, showing this interaction’s specificity. MAEBL is an erythrocyte binding protein located in the rhoptries and on the surface of mature merozoites, being expressed at the beginning of schizogony. The structure of MAEBL originally isolated from rodent malaria parasites suggested a molecule likely to be involved in invasion. We thus became interested in identifying possible MAEBL functional regions. Synthetic peptides spanning the MAEBL sequence were tested in erythrocyte binding assays to identify such possible MAEBL functional regions. Nine high activity binding peptides (HABPs) were identified: two were found in the M1 domain, one was found between the M1 and M2 regions, five in the erythrocyte binding domain (M2), and one in the protein’s repeat region. The results showed that peptide binding was saturable; some HABPs inhibited in vitro merozoite invasion and specifically bound to a 33 kDa protein on red blood cell membrane. Rhoptry-associated protein 1 (RAP1) is a merozoite antigen within Plasmodium falciparum rhoptries as yet having no specific function described for it. Synthetic peptides spanning the RAP1 sequence were tested in erythrocyte binding assays to identify possible RAP1 functional regions. Five high activity binding peptides (HABPs) were identified; 26201, 26202, 26203 and 26204 spanned residues 461C–K540 within RAP1 Cys region, whilst 26188 (201T–Y220) was located in p67 amino terminal. The results showed that peptide binding was saturable, some HABPs inhibited in vitro merozoite invasion and specifically bound to a 72 kDa protein in red blood cell membrane. Plasmodium falciparum rhoptry-associated proteins 1 (RAP1) and 2 (RAP2) are antigens presenting themselves as candidates for a subunit malaria vaccine. RAP2 protein, non-overlapping, consecutive peptides were synthesised and tested in red blood cell (RBC) binding assays to identify their receptor–ligand interaction in recognising RAP2 regions involved in the in vitro merozoite invasion process. Four high activity binding peptides (HABPs) were identified in the resulting 20 peptides. Peptides 26220 (61NHFSSADELIKYLEKTNINT80), 26225 (161IKKNPFLRVLNKASTTTHAT180) and 26229 (241RSVNNVISKNKTLGLRKRSS260) were located in the amino terminal and central part of the protein and HABP 26235 (361FLAEDFVELFDVTMDCYSRQ380) was located at the carboxy terminal. All these HABPs showed saturable binding and presented dissociation constants between 500 and 950 nM; the number of binding sites per RBC ranged from 48 000 to 160 000. High binding peptides’ critical amino acids involved in RBC binding were determined by competition binding assays; their amino acids appear in bold in the sequences shown above. SDS-PAGE results showed that peptides 26220, 26225 and 26229 had at least two different sets of 62 and 42 kDa HABP receptors on RBCs and that peptide 26235 had at least two different sets of 77 and 62 kDa. HABPs inhibited in vitro merozoite invasion by between 54% and 94% at 200 μM, suggesting that these RAP2 peptides are involved in the in vitro P. falciparum invasion process. [Copyright &y& Elsevier]
