Global biodiversity gradients are generally expected to reflect greater species replacement closer to the equator. However, empirical validation of global biodiversity gradients largely relies on vertebrates, plants, and other less diverse taxa. Here we assess the temporal and spatial dynamics of global arthropod biodiversity dynamics using a beta-diversity framework. Sampling includes 129 sampling sites whereby malaise traps are deployed to monitor temporal changes in arthropod communities. Overall, we encountered more than 150,000 unique barcode index numbers (BINs) (i.e. species proxies). We assess between site differences in community diversity using beta-diversity and the partitioned components of species replacement and richness difference. Global total beta-diversity (dissimilarity) increases with decreasing latitude, greater spatial distance and greater temporal distance. Species replacement and richness difference patterns vary across biogeographic regions. Our findings support long-standing, general expectations of global biodiversity patterns. However, we also show that the underlying processes driving patterns may be regionally linked.

Introduction: Behavioural and psychological symptoms of dementia (BPSDs) are a group of highly prevalent symptoms in residents with a neurocognitive disorder (NCD), including agitation and depressive symptoms. Caregiving staff often mention knowledge and skills deficits regarding optimal BPSD management. While several staff trainings on BPSDs management exist internationally, their transferability to other clinical contexts is limited, owing to methodological challenges. Therefore, to address this implementation gap, there is a strong need for trainings based on high-quality research to strengthen existing evidence, ensure feasibility and reproducibility. Methods: This qualitative study, part of a larger research project, occurred in 2022 on a long-term care (LTC) center unit and an alternate level of care (ALT) hospital unit located in the Province of Quebec, Canada. This study aimed to: 1) evaluate the needs and perceptions of staff caregivers regarding BPSD management, 2) identify the training content and modalities to prioritize according to experts, 3) develop an intradisciplinary training on BPSD management, and 4) pretest the preliminarily version of the training. Objectives one and two were evaluated using focus groups and objective four using individual cognitive interviews. Qualitative interview data was audio-recorded with participants’ consent, transcribed verbatim, and thematically content analyzed using an established method. Results: Overall, thirteen caregivers participated in objective one (8 from the LTC unit, and 6 from the ALT unit). The main staff training need identified on each site was the management of resident’s aggressive behaviours. Staff verbalized a preference for a virtual training. Objective 2 involved a panel of eight experts. Experts recommended the use of an online training platform, and certain training models and indicators. Based on caregivers and experts’ input, five interactive online staff training capsules lasting from 20-25 minutes each and an algorithm guiding the evaluation and management of BPSDs were created. The training capsules and the algorithm were iteratively improved following cognitive interviews with 4 caregivers from the participating sites. Conclusion: An interactive virtual staff training on BPSD management was created based on staff and expert consultation. The next step in investigation will be to evaluate the feasibility and acceptability of the training.

Background: Reported advantages to robotic thoracic surgery include shorter length of stay (LOS), improved lymphadenectomy, and decreased complications. It is uncertain if these benefits occur when introducing robotics into a well-established video-assisted thoracoscopy (VATS) practice. We compared the two approaches to investigate these advantages. Materials and methods: IRB approval was obtained for this project. Patients who underwent segmentectomy or lobectomy from May 2016–December 2018 were propensity-matched 2: 1 (VATS: robotic) and compared using weighted logistic regression with age, gender, Charlson Comorbidity Index, surgery type, stage, Exparel, and epidural as covariates. Complication rates, operation times, number of sampled lymph nodes, pain level, disposition, and LOS were compared using Wilcoxon rank-sum and with Rao-Scott Chi-squared tests. Results: 213 patients (142 VATS and 71 robot) were matched. Duration of robotic cases was longer than VATS (median 186 min (IQR 78) vs. 164 min (IQR 78.75); p 0.05). Conclusions: The robotic approach facilitates better lymph node sampling, even in an established VATS practice.

Une étude critique des méthodes de préparation de catalyseurs monométalliques permet de voir que les techniques impliquant un échange (ionique ou chimique) conduisent à des catalyseurs à forte dispersion avec des moyens relativement simples. Au niveau de la caractérisation des catalyseurs, il ne semble pas exister de méthodes exhaustives permettant d'étudier les phases métalliques contenant des cristallites de taille inférieure à 1. 0 nm. Les techniques de microscopie électronique à haute résolution, la spectroscopie XPS et l'EXAFS semblent par contre les plus prometteuses. L'influence du support joue à plusieurs niveaux, en particulier sur la morphologie, la structure et sur les propriétés électroniques de la phase métallique. L'intérêt particulier d'étudier les effets de support résulte dans la création de phase métallique plus active et sélective. The current aim of catalyst research is to develop more and more active and selective dispersed metallic phases for a given reaction as well as to improve their stability. Catalytic properties depend directly on the physicochemical characteristics of the metallic phase (size of crystallites, composition, electron structure, interactions with the support). This is why improving such catalysts requires mastering synthesis routes and the characterizing of small supported metallic particles. The aim of this critical study is to highlight both the preparation methods leading to well-controlled monometallic phases and the physical techniques suited for characterizing them. By considering the metal-support entity as a veritable catalytic space, we have examined the direct influence of the nature of the support on the end characteristics of the metallic particles (morphology, structure, electron properties). We have found that impregnation techniques with an interaction (physical or chemical) can lead to high-dispersion metallic phases associated with a narrow dispersion of crystallite size. Among other techniques used, note will be taken of ion implantation, vapodeposition and radiolytic synthesis. Among overall and controlled-atmosphere characterization techniques, XPS and EXAFS spectroscopy were used to characterize, respectively, the electron properties and the crystalline structure (chemical environment) of metallic particles. Small metallic particles (less than 1. 0 nm in size) are more difficult to characterize because of the limitations of the physical techniques normally used. High-resolution electron microscopy was used for the local characterization of these small particles. The present results mainly concern the size of these small aggregates, but this technique also gives information on their morphology (aggregates of atoms, individual atoms in the aggregate) and on their crystalline structure (microdiffraction). Likewise, there is an entire domain of indirectcharacterization that is widely used, in which especially the accessibility and degree of oxidation reduction are measured of the metals by means of reagents or probe molecules (H2, O2, CO). However, this very precious information in the field of catalysis must always be compared with the result of direct characterization (size, electron properties). Concerning the influence of the support, this review shows that the metal-support pair is a full-fledged entity and that it is almost impossible to characterize it from the individual properties of the metal and of the support. At present, the principal results have to do with semiempirical classifications concerning the stability of metallic phases during heat treatments and with the influence of the metal-support interaction force on the morphology and electron properties of particles (load transfer, modifications of the electron structure of the transition metal).

BackgroundIron accumulates in the brain during aging and is the focus of intensive research as an abnormal load, particularly in Deep Gray Matter (DGM), is related to neurodegeneration. Magnetic Resonance Imaging (MRI) metrics such as Quantitative Susceptibility Mapping (QSM) and apparent transverse relaxation rate R2* can be used to follow up iron in vivo. While the influence of age and sex on iron levels has already been reported, a careful consideration of neuronal risk factors, as well as for an enhanced sensitivity, is needed to define the normal evolution.MethodsQSM and R2* at ultra-high field MRI are used to study iron in DGM using a carefully-characterized cohort of the healthy aging brain (SENIOR). Seventy-seven cognitively healthy elders (from 54 to 78 y/o) with clinical, biology, genetics, and cardiovascular risk factors careful evaluation. Differences linked with age, sex, cardiovascular risk factors and weight are studied.ResultsAge and sex have an influence on the brain iron deposition measured by QSM and R2* in a context of normal aging, without appearance of a pathological neurodegenerative process. Iron deposition shows higher values in the caudate and the putamen in older participants. Female participants present a higher level of iron in the amygdala, and males in the thalamus. Female participants also present differences in the accumbens, caudate and hippocampus when evaluating the joint age and sex effect. Participants with higher cardiovascular risk factors showed higher values of the iron, even without any impairment in their cognitive capability. An overweight is related with a higher iron load in the putamen for QSM and R2* in female participants. We controlled that these modifications of iron deposition are not related to a specific profile in the genotype of ApoE loci.ConclusionsEstablishing baseline values of QSM and R2* as iron probes in the context of aging is essential to determine differences in the process of neurodegeneration. Age and sex of participants are important factors that affect brain iron normal values. On the other hand, the presence of cardiovascular risk factors, which can be associated with age related diseases, can also potentially be linked with the iron deposition in the brain.

Summary: Reactive changes of glial cells during neuroinflammation impact brain disorders and disease progression. Elucidating the mechanisms that control reactive gliosis may help us to understand brain pathophysiology and improve outcomes. Here, we report that adult ablation of autism spectrum disorder (ASD)-associated CHD8 in astrocytes attenuates reactive gliosis via remodeling chromatin accessibility, changing gene expression. Conditional Chd8 deletion in astrocytes, but not microglia, suppresses reactive gliosis by impeding astrocyte proliferation and morphological elaboration. Astrocyte Chd8 ablation alleviates lipopolysaccharide-induced neuroinflammation and septic-associated hypothermia in mice. Astrocytic CHD8 plays an important role in neuroinflammation by altering the chromatin landscape, regulating metabolic and lipid-associated pathways, and astrocyte-microglia crosstalk. Moreover, we show that reactive gliosis can be directly mitigated in vivo using an adeno-associated virus (AAV)-mediated Chd8 gene editing strategy. These findings uncover a role of ASD-associated CHD8 in the adult brain, which may warrant future exploration of targeting chromatin remodelers in reactive gliosis and neuroinflammation in injury and neurological diseases.

Key Clinical Message The purpose of this article is to highlight that oral Kaposi's disease can be indicative of a high viral load of HIV, either in the case of primary infection or therapeutic failure. Abstract We report two cases of Kaposi Disease associated with HIV. The first case was a 30‐year‐old patient who unaware of her HIV‐positive status, and who was diagnosed with AIDS stage because of the biopsy revealed a gingival location of Kaposi disease. The second case was a 34‐year‐old patient who was referred to our department with a history of palatal lesion and claimed at first having no previous known medical conditions although his overall health condition seemed deteriorated. Our clinical examination was evocative of Kaposi Disease, which was confirmed by an emergency blood assessment and histological examination. Our diagnosis led us to disclose the HIV‐positive status of the patient and identify a progression to the AIDS stage, which allowed us to reintroduce the patient in the hospital framework. This case emphasizes the role of the oral surgeon as a key actor thanks to their knowledge of the clinical buccal manifestations of sexually transmitted infections (STI), in an era of resurgence of those conditions in vulnerable key populations.

Mammals have evolved sex-specific adaptations to reduce energy usage in times of food scarcity. These adaptations are well described for peripheral tissue, though much less is known about how the energy-expensive brain adapts to food restriction, and how such adaptations differ across the sexes. Here, we examined how food restriction impacts energy usage and function in the primary visual cortex (V1) of adult male and female mice. Molecular analysis and RNA sequencing in V1 revealed that in males, but not in females, food restriction significantly modulated canonical, energy-regulating pathways, including pathways associated waith AMP-activated protein kinase, peroxisome proliferator-activated receptor alpha, mammalian target of rapamycin, and oxidative phosphorylation. Moreover, we found that in contrast to males, food restriction in females did not significantly affect V1 ATP usage or visual coding precision (assessed by orientation selectivity). Decreased serum leptin is known to be necessary for triggering energy-saving changes in V1 during food restriction. Consistent with this, we found significantly decreased serum leptin in food-restricted males but no significant change in food-restricted females. Collectively, our findings demonstrate that cortical function and energy usage in female mice are more resilient to food restriction than in males. The neocortex, therefore, contributes to sex-specific, energy-saving adaptations in response to food restriction.

Monthly variation in accumulation and release of phenolics in bryophytes, and their allelopathic effects are little known despite their ecological significance. We conducted a field transplant experiment to investigate Sphagnum growth and extracellular enzymes that modulate phenolic contents and release along water table level (WTL) gradient. Specifically, we assessed monthly changes in height increment, phenolic release, and allelopathic effect during the growing season, as well as extracellular enzymes at the end of the experiment, for the two Sphagnum species: Sphagnum angustifolium C.E.O. Jensen, 1896, a hollow dwelling species, and Sphagnum magellanicum Bridel, 1798, a hummock species. Both Sphagnum species showed season- and WTL-dependent height increment and released phenolics. Sphagnum angustifolium exhibited negative allelopathy regardless of WTL, while S. magellanicum demonstrated mainly positive allelopathy at high WTL and negative allelopathy at low WTL. The degree of allelopathy was not contingent on phenolic release, but phenolic production was negatively correlated with height growth of the two species at their atypical habitats. Moreover, phenoloxidase or peroxidase did not show high activity at low WTL compared to high WTL. Our findings demonstrate for the first time that the strength and direction of allelopathy in Sphagnum may vary depending on the growing precipitation and temperature, WTL, and inter- and intraspecific trait variability. Longer term experiments may further elucidate the seasonal dynamics and mechanism of Sphagnum allelopathy. Abstract The moss layer transfer technique has been developed to restore the carbon sequestration function and typical vegetation of Sphagnum-dominated peatlands after peat extraction in North America. However, the technique does not lead to successful bryophyte establishment when applied to peatlands with a richer residual fen peat. Therefore, we evaluated an alternative method of active rewetting and passive vegetation establishment using vegetation surveys and carbon dioxide and methane (CH4) flux measurements at a post-extracted fen in southern Manitoba, Canada. After one growing season post-rewetting, wetland vegetation established and the site was a net carbon sink over the growing season. However, high abundance of Carex lasiocarpa 10 years post-treatment led to higher CH4 emissions than the reference ecosystem. Successful establishment of wetland vegetation is attributed to the area being surrounded by undisturbed fens that can provide a local source of plant propagules. Bryophyte expansion was less successful than vascular plants, likely due to episodic flooding and shading from the sedge communities. Therefore, careful management of water levels to just below the peat surface is needed if reference vegetation community recovery is the goal of restoration. Water level management will also play a key role in controlling CH4 emissions to maximize carbon sequestration potential.

Introduction: Cervical cancer, the second leading cause of cancer-related death for women worldwide, remains a preventable yet persistent disease that disproportionately affects women in low and middle-income countries (LMICs). While existing therapies for treating cervical dysplasia are effective, they are often inaccessible in LMICs. Ethanol ablation is an alternative low-cost, accessible therapy that we previously enhanced into ethyl cellulose (EC)-ethanol gel formulation to improve efficacy.Methods: To evaluate the efficacy of EC-ethanol, in this study, we developed a 3D in vitro model of cervical dysplasia featuring a central lesion of cervical cancer cells surrounded by fibroblasts and keratinocytes. Using a GelMA hydrogel formulation (8.7% w/v), we successfully built a 3D model that captured the architectural complexity of cervical dysplasia. We evaluated changes in cell coverage and cell viability. Then, we compared the viscoelastic properties of the GelMA hydrogels to human cervical tissue and using micro-CT imaging, we assessed EC-ethanol injection deposition in the hydrogel, revealing retention of virtually the entire injected volume near the injection site. Finally, we measured changes in cell viability and cell coverage after the EC-ethanol injection.Results: The developed 3D in vitro model successfully replicated the architectural complexity of cervical dysplasia, demonstrating high cell viability and capturing cell responses effectively. The GelMA hydrogel formulation (8.7% w/v) exhibited viscoelastic properties akin to human cervical tissue. Micro-CT imaging revealed efficient deposition of EC-ethanol within the hydrogel, with retention of the injected volume near the injection site. Furthermore, the EC-ethanol injection significantly reduced cervical cancer cell viability and cell coverage while preserving healthy cells within the model.Conclusion: Our findings indicate that our 3D in vitro model mirrored the architecture of cervical dysplasia and demonstrated the potential of EC-ethanol for localized treatment of cervical dysplasia.

Thousands of endoparasitoid wasp species in the families Braconidae and Ichneumonidae harbor "domesticated endogenous viruses" (DEVs) in their genomes. This study focuses on ichneumonid DEVs, named ichnoviruses (IVs). Large quantities of DNA-containing IV virions are produced in ovary calyx cells during the pupal and adult stages of female wasps. Females parasitize host insects by injecting eggs and virions into the body cavity. After injection, virions rapidly infect host cells which is followed by expression of IV genes that promote the successful development of wasp offspring. IV genomes consist of two components: proviral segment loci that serve as templates for circular dsDNAs that are packaged into capsids, and genes from an ancestral virus that produce virions. In this study, we generated a chromosome-scale genome assembly for Hyposoter didymator that harbors H. didymator ichnovirus (HdIV). We identified a total of 67 HdIV loci that are amplified in calyx cells during the wasp pupal stage. We then focused on an HdIV gene, U16, which is transcribed in calyx cells during the initial stages of replication. Sequence analysis indicated that U16 contains a conserved domain in primases from select other viruses. Knockdown of U16 by RNA interference inhibited virion morphogenesis in calyx cells. Genome-wide analysis indicated U16 knockdown also inhibited amplification of HdIV loci in calyx cells. Altogether, our results identified several previously unknown HdIV loci, demonstrated that all HdIV loci are amplified in calyx cells during the pupal stage, and showed that U16 is required for amplification and virion morphogenesis.

Abstract Arctic ecosystems are undergoing rapid changes, including increasing disturbance by herbivore populations, which can affect plant species coexistence and community assemblages. Although the significance of mosses in Arctic wetlands is well recognized, the long‐term influence of medium‐sized herbivores on the composition of moss communities has received limited attention. We used data from a long‐term (25 years) Greater Snow Goose (Anser caerulescens atlanticus) exclusion experiment in Arctic tundra wetlands to assess changes in the composition of moss communities at multiple spatial scales (cell, 4 cm2; quadrat, 100 cm2; exclosure, 16 m2). We investigated how snow goose grazing and grubbing can alter the composition of the moss community by measuring changes in alpha and beta diversity, as well as in the strength of plant interspecific interactions between moss species. Our results indicate that goose foraging significantly increased species diversity (richness, evenness, and inverse Simpson index) of moss communities at the cell and quadrat scales but not the exclosure scale. Goose foraging reduced the dissimilarity (beta diversity) of moss communities at all three scales, mainly due to decreased species turnover. Furthermore, goose foraging increased positive interaction between moss species pairs. These findings emphasize the critical role of geese in promoting moss species coexistence and increasing homogeneity in Arctic wetlands. This study illustrates how top‐down regulation by herbivores can alter plant communities in Arctic wetlands and highlights the importance of considering herbivores when examining the response of Arctic plant biodiversity to future climate change.
