Abstract Objectives: The objective of the present work was to evaluate the ultrasonic agitation, time and vehicle (propylene glycol or distilled water) on the antimicrobial potential and penetrability of calcium hydroxide pastes on infected dentin by means of Confocal Laser Scanning Microscopy (CLSM) and microbiological culture (MC). Materials and methods: Dentin specimens were infected with Enterococcus faecalis using a new contamination protocol of 5 days. The specimens were divided into eight groups and dressed with the pastes for 7 or 15 days: G1) calcium hydroxide (CH) + propylene glycol (prop)/7 days (d), G2) CH + prop/7d + ultrasonic agitation (U), G3) CH + distilled water (dw)/7d, G4) CH + dw/7d + U, G5) CH + prop/15d, G6) CH + prop/15d + U, G7) CH + dw/15d, G8) CH + dw/15d + U. The ultrasonic activation was made for 1 min in both directions with a plain point insert. After medications removal, the images obtained by CLSM showed the viable (green) and dead (red) bacteria with Live and Dead dye. By the MC, the dentinal wall debris obtained by burs were collected for colony counts. For the penetration test, the Rodamine B dye was added to the CH pastes and analyzed by CLSM. Results: The 7 and 15-days CH + prop+U pastes performed better antimicrobial efficacy, followed by the CH + dw+U/15d paste. Conclusions: All pastes demonstrated better penetration and antimicrobial activity against E. faecalis when agitated with ultrasound, even in periods of up to seven days. The propylene glycol vehicle showed better results. Clinical relevance: Agitation of the dressing that remains for less time inside the root canal can optimize the decontamination of endodontic treatment.

Low-carbon energy transitions taking place worldwide are primarily driven by the integration of renewable energy sources such as wind and solar power. These variable renewable energy (VRE) sources require energy storage options to match energy demand reliably at different time scales. This article suggests using a gravitational-based energy storage method by making use of decommissioned underground mines as storage reservoirs, using a vertical shaft and electric motor/generators for lifting and dumping large volumes of sand. The proposed technology, called Underground Gravity Energy Storage (UGES), can discharge electricity by lowering large volumes of sand into an underground mine through the mine shaft. When there is excess electrical energy in the grid, UGES can store electricity by elevating sand from the mine and depositing it in upper storage sites on top of the mine. Unlike battery energy storage, the energy storage medium of UGES is sand, which means the self-discharge rate of the system is zero, enabling ultra-long energy storage times. Furthermore, the use of sand as storage media alleviates any risk for contaminating underground water resources as opposed to an underground pumped hydro storage alternative. UGES offers weekly to pluriannual energy storage cycles with energy storage investment costs of about 1 to 10 USD/kWh. The technology is estimated to have a global energy storage potential of 7 to 70 TWh and can support sustainable development, mainly by providing seasonal energy storage services.

Neste artigo é mostrado o desenvolvimento de um banco de dados geográfico para subsidiar a análise da distribuição espacial da incidência da Leshimaniose Visceral no município de Cametá, no estado do Pará. Para tal objetivo desenvolveu-se um banco de dados geográfico com dados ecoepidemiológicos utilizando o PostGis. Após o processo do projeto lógico e físico da base de dados foi elaborado “queries” de consultas espaciais, conectadas ao PostGis, para a extração de informações. Posteriormente, utilizou-se o SIG OpenJump para a elaboração dos mapas temáticos. Como resultado observou-se que com as consultas espaciais, obtêm um rápido resultado sobre quais possíveis criadouros são propícios a proliferação da doença, utilizando as tecnologias PostGis e OpenJump, possibilitando expressar visualmente a incidência da doença de Leishimaniose Visceral no município estudado.

The aim of this study was to evaluate the subcutaneous tissue response in rats and the antimicrobial activity of intracanal calcium hydroxide dressings mixed with different substances against E. faecalis. Fifty four rats were divided into three experimental groups according to the vehicle in the calcium hydroxide treatment: 0.4% chlorohexidine in propylene glycol (PG),Casearia sylvestris Sw in PG and calcium hydroxide+PG (control group). The pastes were placed into polyethylene tubes and implanted into the subcutaneous tissue. After 7, 14 and 30 days, the samples were processed and histologically evaluated (hematoxylin and eosin). The tissue surface in contact with the material was analyzed, and the quantitative analysis determined the volume density occupied by the inflammatory infiltrate (giant cells, polymorphonuclear cells and mononuclear cells), fibroblasts, collagen fibers and blood vessels. For the antimicrobial analysis, 20 dentin blocks infected with E. faecalis were treated with calcium hydroxide pastes in different vehicles; 0.4% chlorhexidine in PG, PG, extract fromCasearia sylvestris Sw in PG and a positive control (infection and without medication) for 7 days. The efficiency of the pastes was evaluated by the live/dead technique and confocal microscopy. The results showed that 0.4% chlorhexidine induced a higher inflammatory response than the other groups. The Casearia sylvestris Sw extract showed satisfactory results in relation to the intensity of the inflammatory response. In the microbiological test, there were no statistical differences between the evaluated intracanal dressings and the percentage of bacterial viability was between 33 and 42%. The control group showed an 86% viability. Antimicrobial components such as chlorhexidine or Casearia sylvestris Sw did not improve the antimicrobial activity against E. faecalis in comparison to the calcium hydroxide+PG treatment. In addition, the incorporation of chlorhexidine in the calcium hydroxide paste promoted the highest inflammatory response.

Objective: To evaluate the response of rat subcutaneous tissue in implanted polyethylene tubes that were filled with GMTA Angelus and Portland cements containing different arsenic concentrations. Material and Methods: Atomic absorption spectrophotometry was utilized to obtain the values of the arsenic concentration in the materials. Thirty-six rats were divided into 3 groups of 12 animals for each experimental period. Each animal received two implants of polyethylene tubes filled with different test cements and the lateral of the tubes was used as a control group. Objectives: To investigate if there is a relation between the increase of bismuth oxide and the decrease of pH levels and an intensification of toxicity in the Portland cement. Material and Methods: White Portland cement (WPC) was mixed with 0, 15, 20, 30 and 50% bismuth oxide, in weight. For the pH level test, polyethylene tubes were filled with the cements and immersed in Milli-Q water for 15, 30 and 60 days. After each period, the increase of the pH level was assessed. For the biocompatibility, two polyethylene tubes filled with the cements were implanted in ninety albino rats (n=6). The analysis of the intensity of the inflammatory infiltrate was performed after 15, 30 and 60 days. The statistical analysis was performed using the Kruskal-Wallis, Dunn and Friedman tests for the pH level and the Kruskal-Wallis and Dunn tests for the biological analysis (p0.05). For the inflammatory infiltrates, no significant statistical differences were found among the groups in each period (p>0.05). ObjectiveTo evaluate the biocompatibility and the setting time of Portland cement clinker with or without 2% or 5% calcium sulfate and MTA-CPM.Material and MethodsTwenty-four mice (Rattus norvegicus) received subcutaneously polyethylene tubes filled with Portland cement clinker with or without 2% or 5% calcium sulfate and MTA. After 15, 30 and 60 days of implantation, the animals were killed and specimens were prepared for microscopic analysis. For evaluation of the setting time, each material was analyzed using Gilmore needles weighing 113.5 g and 456.5 g, according to the ASTM specification Number C266-08 guideline. Data were analyzed by ANOVA and Tukey's test for setting time and Kruskal-Wallis and Dunn test for biocompatibility at 5% significance level.ResultsHistologic observation showed no statistically significant difference of biocompatibility (p>0.05) among the materials in the subcutaneous tissues. For the setting time, clinker without calcium sulfate showed the shortest initial and final setting times (6.18 s/21.48 s), followed by clinker with 2% calcium sulfate (9.22 s/25.33 s), clinker with 5% calcium sulfate (10.06 s/42.46 s) and MTA (15.01 s/42.46 s).ConclusionsAll the tested materials showed biocompatibility and the calcium sulfate absence shortened the initial and final setting times of the white Portland cement clinker.

OBJECTIVES: To compare, in vivo, the accuracy of conventional and digital radiographic methods in determining root canal working length. MATERIAL AND METHODS: Twenty-five maxillary incisor or canine teeth from 22 patients were used in this study. Considering the preoperative radiographs as the baseline, a 25 K file was inserted into the root canal to the point where the Root ZX electronic apex locator indicated the APEX measurement in the screen. From this measurement, 1 mm was subtracted for positioning the file. The radiographic measurements were made using a digital sensor (Digora 1.51) or conventional type-E films, size 2, following the paralleling technique, to determine the distance of the file tip and the radiographic apex. OBJECTIVE: This study compared the influence of three epoxy resin-based sealers with distinct radiopacities on the observers' ability to detect root canal filling voids during radiographic analysis. MATERIAL AND METHODS: The root canals of 48 extracted maxillary canines were prepared and divided into three groups. Each group was laterally condensed with one sealer (AH Plus®, Acroseal® or a non-radiopaque sealer), and a longitudinal void was simulated in half of the specimens from each group (n=8). Buccolingual radiographs were obtained and randomly interpreted for voids by a radiologist and an endodontist in a blinded fashion. OBJETIVES: The aim of this study was to investigate through scanning electron microscopy (SEM) the cleaning of root canal walls after the use of experimental propolis or calcium hydroxide root canal dressings. MATERIAL AND METHODS: Twenty single-rooted teeth were used. After conventional cleaning and shaping procedures and removal of the smear layer with 17% EDTA, the teeth were divided into four groups according to the medication used (N=5): Group I (control) - No drug, Group II - Calcium hydroxide dressing, Group III - Propolis paste A70D and Group IV - Propolis paste D70D. The medications were introduced into the root canals and maintained for 7 days, then removed with a K-file and 5 mL of 1% sodium hypochlorite irrigation. This ex vivo study evaluated the quality of digital radiographic images obtained with the photostimulable phosphor plate system (Digora) according to the processing delay and maintenance of optical plates in either opaque (supplied with the system) or transparent protective plastic cases during this period. Five radiographs were obtained from the mandibular molar region of a dry human mandible using optical plates. These plates were placed in the protective plastic cases before obtaining the radiographs and were processed immediately or after processing delays of 5, 60 and 120 min, when the case was removed. The results revealed a reduction in image quality when processing was delay 120 min compared to the other times. The opaque case provided better protection to the sensor than the transparent case. In conclusion, a 120-min processing delay for the Digora system caused a reduction in image quality, yet without interfering with the quality of diagnosis. The opaque case supplied by the system's manufacturer provided better protection to the optical plate than the transparent case.

This study investigated the solubility of mineral trioxide aggregate (MTA) and Portland cement since its mixture until 672 hours, by means of two complimentary methods. Metal ring molds filled with the cements were covered with distilled water and, at each experimental time (3, 24, 72, 168, 336 and 672 hours), were weighed as soon as the plates in which the samples have been placed. Empty rings served as the control group (n=8). Mean weight gain and loss was determined and analyzed statistically by two-way ANOVA and Tukey's test for all pairwise comparisons. Only Portland cement showed less than 3% weight loss through 24 hours. Detached MTA residues were heavier than those of Portland cement over the 3 to 168 hours. The weight of MTA rings increased more than that of Portland rings within 672 hours (p=0.05). The findings of the present study indicate that, in an aqueous environment MTA is more soluble than Portland cement and exceeds the maximum weight loss considered acceptable by ISO 6876 standard (2001).

The aims of this study were to evaluate the quality of indirect digitized radiographic images taken during endodontic procedures and to compare the measurements recorded with this technique to those obtained from conventional radiographs. Two-hundred conventional periapical radiographs taken at the undergraduate Endodontics Clinic of the Dental School of Bauru were digitized. The conventional and indirect digitized images were compared by three examiners as to the quality and accuracy of the measurements recorded during endodontic treatment, in canal length determination, gutta-percha adaptation, lateral condensation and final obturation. The conventional radiographs were observed on a film viewer, surrounded by a dark card, and measured with magnifying glass and a millimeter ruler; the indirect digitized images were evaluated on the Digora® for Windows software, with free utilization of the bright/contrast tool. This study evaluated the sealing ability of apical plugs fabricated with gray MTA AngelusTM sealer, CPM TM sealer and MBPc sealer. The root canals of 98 extracted single-rooted human teeth were instrumented with #5 to #1 Gates Glidden drills according to the crown-down technique until the #1 drill could pass through the apical foramen. The specimens were then prepared with K-files, starting with an ISO 50 until an ISO 90 could be visualized 1 mm beyond the apex. After root canal preparation, the external surface of each root was rendered impermeable and roots were assigned to 3 experimental groups (n = 30), which received a 5-mm thick apical plug of gray MTA AngelusTM, CPM TM and MBPc, and two control groups (n=4). The remaining portion of the canal in the experimental groups was filled by the lateral condensation technique. The teeth of each group, properly identified, were fixed on utility wax by their crowns and were placed in plastic flasks, leaving the apex free and facing upward. The flasks were filled with 0.2% Rhodamine B solution, pH 7.0, so as to completely cover the root apex of all teeth. The sealing ability was analyzed by measuring 0.2% Rhodamine B leakage after all groups had been maintained in this solution for 48 hours. The aim of this study was to evaluate histologically, three types of calcium sulfate - Merck (Brazil), Surgiplaster (Italy) and Capset (USA) - in surgically created defects on rabbit femurs. Twenty male New Zealand rabbits were used. Two surgical bone defects (5 mm diameter x 8 mm depth) were created on each distal epiphysis using a #3 Dentoflex trephine bur. Defects were filled with: group 1 - di-hydrated calcium sulfate (Merck); group 2 - Capset (Lifecore-USA); group 3 - Surgiplaster (Classimport-Italy); group 4 - control (blood clot). The animals were sacrificed 30, 60, 90 and 180 days postoperatively. Semi-serial 6-mm-thick sections were obtained, stained with hematoxylin and eosin and examined under light microscopy. Bone defects treated with calcium sulfate exhibited new bone formation regardless of the product trademark.

This study aimed to compare the apical sealing of root-end fillings performed with Lysanda (zinc oxide-eugenol paste) with radiopacifiers (iodoform or zinc oxide) and calcium hydroxide. Root-end cavities were prepared and filled with different materials, as follows: Group I - Lysanda paste with iodoform; Group II - Lysanda paste with iodoform and calcium hydroxide; Group III - Lysanda paste with iodoform and zinc oxide; Group IV - Lysanda paste with zinc oxide; Group V - mineral trioxide aggregate (MTA). After filling, the teeth were immersed in 2% methylene blue for analysis of marginal leakage. It was observed that marginal leakage occurred in all groups. Lysanda paste with iodoform showed the lowest leakage, with no statistically significant difference compared to the other groups. All materials can be considered as good options for root-end filling.

This study evaluated the sealing ability of apical plugs made of white and gray MTA-Angelus® and white Portland cement placed via the root canal and having different thicknesses (2, 5 and 7 mm). Ninety extracted human single-rooted teeth were instrumented using a size 40 K-file to standardize the foraminal opening by the stepback technique. The teeth were assigned to 3 groups (n=30), according to the material used for fabrication of the apical plugs: A = gray MTA; B = white MTA; C = white Portland cement. The groups were subdivided into groups of 10 teeth each according to the apical plug thickness (2, 5 and 7 mm). MATERIAL AND METHODS: Forty human mandibular left and right central incisors, lateral incisors and premolars were used. The pulp tissue was removed and the root canals were filled with red die. Teeth were instrumented using three techniques: (i) K3 and ProTaper rotary systems; (ii) ultrasonic crown-down technique; and (iii) progressive manual technique. The purpose of this study was to test a gallium nitrate solution, a resorption inhibitor, employed as a root canal dressing in teeth submitted to late replantation. Thirty maxillary right central incisors of rats were avulsed and kept dry for thirty minutes. The teeth were instrumented and the root surfaces were treated with 1% hypochlorite solution followed by application of 2% sodium fluoride. Thereafter, the teeth were divided into two groups according to the root canal dressing: Group I, solution of gallium nitrate; and Group II, calcium hydroxide paste. The teeth were then replanted in their respective sockets. The animals were killed at 15, 30 and 60 days after replantation and the samples were processed for morphometric and microscopic analysis. The results demonstrated that the gallium nitrate solution and the calcium hydroxide paste limited the root resorption, yet they did not impair its occurrence. It may be concluded that gallium nitrate solution and calcium hydroxide paste demonstrated similar performance.Este trabalho tem como objetivo testar a solução de nitrato de gálio, um inibidor da reabsorção, como curativo intracanal para dentes reimplantados tardiamente. Trinta incisivos centrais superiores direitos de ratos foram avulsionados e mantidos a seco por trinta minutos. Todos os dentes tiveram os canais radiculares instrumentados e a superfície radicular tratada com hipoclorito de sódio a 1%, seguido de fluoreto de sódio a 2%. Após isso, os dentes foram divididos em dois grupos de acordo com a medicação intracanal: no grupo I, foi usada solução de nitrato de gálio e grupo II, pasta de hidróxido de cálcio. Todos os dentes foram, então, reimplantados em seus respectivos alvéolos. Passados 15, 30 e 60 dias do reimplante, os animais foram sacrificados e as peças obtidas, processadas em laboratório para análise microscópica e morfométrica. Os resultados mostraram que a solução de nitrato de gálio e a pasta de hidróxido de cálcio limitaram a reabsorção radicular, mas não impediram a sua ocorrência. Conclui-se que ambos os medicamentos têm ação semelhante.

Fourteen root perforations were performed for microscopic evaluation of the repair of interradicular tissue in dogs' teeth. These perforations were accomplished at low-speed with a STP 58 bur at the cervical third of the mesial root toward the furcation under irrigation with saline solution, followed by immediate sealing with ProRoot MTA, MTA-Angelus and white Portland cement. The dogs were killed after 90 days, revealing good results. The Kruskal-Wallis test did not demonstrate any statistically significant difference. It was concluded that the three materials showed good sealing in mineralized tissue, with complete closure, and they were free of inflammation in most teeth.Avaliou-se o reparo de perfurações em dentes de cães, tratadas com ProRoot MTA, MTA Angelus e cimento Portland branco. As perfurações foram feitas na região de furca de premolares, superiores e inferiores, com broca STP 58 sob refrigeração com soro fisiólogico. Os animais foram mortos após 90 dias e os dentes foram preparados para análise microscópica pela coloração da hematoxilina e eosina. Os três materiais propiciaram o selamento da perfuração com tecido mineralizado e o teste de Kruskal-Wallis demostrou não haver diferença estatística entre eles.

This research evaluated the sealer ability of 2 temporary filling materials (white Cimpat and IRM) and 1 restorative cement (glass ionomer), in canals prepared for root posts. Sixty human palatal roots of maxillary first molars were used. They were divided into 3 groups, according to the cements used: Group I (Cimpat), Group II (IRM) and Group III (glass ionomer). The roots were rendered impermeable, filled with the respective cements and soon after immersed into 0.2% Rhodamine B dye and maintained for 72 hours in an oven for 37°C. Microleakage was measured with a light microscope, cutting the roots longitudinally in buccolingual direction. The results showed that Group I presented significantly more leakage than Groups II and III, which were not significantly different from each other.Esta pesquisa avaliou a capacidade seladora de dois cimentos temporários (Cimpat branco e IRM) e um cimento restaurador em canais preparados para pinos intra-radiculares. Foram usadas 60 raízes palatinas de primeiros molares superiores humanos. Elas foram divididas em 3 grupos, conforme os cimentos usados: grupo I (Cimpat), grupo II (IRM) e grupo III (ionômero de vidro). As raízes foram impermeabilizadas, preenchidas com os respectivos cimentos e em seguida imersas em tintura de Rhodamina B a 0,2% e mantidas por 72 horas em uma estufa a 37ºC. A micro infiltração foi mensurada com um microscópio de luz nas raízes cortadas longitudinalmente, na direção vestíbubo-lingual. Os resultados mostraram que o grupo I apresentou, de modo significante, mais infiltração do que os grupos II e III, enquanto que entre estes não houve diferença significante.

The bone tissue has a great regenerative potential, with ability to completely restore its structure and original functions. In some situations, though, bone defects cannot be self-repaired, thus requiring the use of grafts for a correct treatment and good prognosis. This work aimed at microscopically analyzing the effect of the particle size of demineralized bovine cancellous bone matrix in micro and macrogranular forms on the repair of bone defects in femurs of rabbits, with blood clot used as control. At 1, 3 and 6 months after implantation of the materials, the animals were killed and the anatomic specimens were removed. A foreign body-type granulomatous reaction containing macrophages and multinucleated giant cells in contact with the implanted particles was observed. These results suggest a failure in demineralization and/or interruption of the antigenic potential during production of the biomaterial. It is concluded that the size of the particles did not influence the evolution of the repair process of bone defects, acting only as bone-filler substances, and that the material implanted should be improved by quality control during production, since it may represent a good alternative for bone graft.O tecido ósseo possui grande potencial regenerativo com capacidade para restaurar completamente sua estrutura e função originais. Há situações em que os defeitos ósseos não conseguem por si só obter o reparo, casos em que se fazem necessários o uso de enxertos, para um correto tratamento e bom prognóstico. Este experimento teve o propósito de analisar microscopicamente o efeito do tamanho das partículas de matriz de osso medular bovino desmineralizado, nas formas micro e macrogranular, na reparação de defeito ósseo em fêmures de coelhos, tendo como controle o coágulo sanguíneo. Após 1,3 e 6 meses da implantação dos materiais, os animais foram mortos e as peças anatômicas removidas. Uma reação granulomatosa tipo corpo estranho, envolvendo as partículas implantadas, foi observada. Estes resultados sugerem falhas na desmineralização e/ou na retirada de potenciais antigênicos durante a produção do biomaterial. Podemos concluir que o tamanho das partículas não influenciou na evolução do processo reparativo dos defeitos ósseos, atuando apenas como substâncias osteopreenchedoras e que o material implantado deverá sofrer um aprimoramento no controle de qualidade na linha de produção, uma vez que poderá representar uma boa alternativa para enxertos ósseos.

Considering previous studies on the similarity between the chemical composition of the mineral trioxide aggregate and the Portland cement, the purpose of this study was to investigate the pulp response of dog's teeth after pulpotomy and direct pulp protection with MTA Angelus and white Portland cement. Thirty eight pulp remnants were protected with these materials. One hundred and twenty days after treatment, the animals were sacrificed and the specimens removed and prepared for histological analysis. Both materials demonstrated the same results when used as pulp capping materials, inducing hard tissue bridge formation and maintaining pulp vitality in all specimens. The MTA Angelus and the white Portland cement showed to be effective as pulp protection materials following pulpotomy.Considerando estudos anteriores sobre a similaridade entre a composição química do agregado de trióxido mineral e o cimento Portland, o objetivo deste estudo foi investigar a resposta pulpar de dentes de cães após pulpotomia e proteção pulpar direta com MTA Angelus e cimento Portland branco. Trinta e oito remanescentes pulpares foram recobertos com esses materiais. Cento e vinte dias após o tratamento, os animais foram sacrificados e os espécimes removidos e preparados para análise histológica. Ambos os materiais demonstraram os mesmos resultados quando utilizados como materiais de capeamento pulpar, induzindo a formação de ponte de tecido mineralizado e mantendo a vitalidade pulpar em todos os espécimes. Ambos matérias se mostraram efetivos como protetores pulpares após pulpotomia em dentes de cães.
