This study investigates the effects on hard-bottom macro-zoobenthic communities of converting a mariculture plant into an Integrated Multi-Trophic Aquaculture (IMTA) system. This study was conducted from 2018 to 2021 in the semi-enclosed Mar Grande basin of Taranto (northern Ionian Sea), on a facility located 600 m off the coastline, with a production capacity of 100 tons per year of seabass (Dicentrarchus labrax) and seabream (Sparus aurata). The results from seasonal sampling performed in a treatment site, where bioremediators (filter-feeding invertebrates such as sponges, polychaetes, mussels, and macroalgae) were deployed, and a control site without bioremediators were compared. Before the IMTA installation, the hard substrates under the cages were sparsely inhabited, with significant sediment coverage. By 2021, the treatment site exhibited revitalized and more diverse macro-zoobenthic communities, with species richness increasing from 83 taxa in 2018 to 104 taxa, including notable growth in sponges, annelids, mollusks, and bryozoans. In contrast, the control site showed no substantial changes in biodiversity over the same period. Biodiversity indices, including Shannon and Margalef indices, improved significantly at the treatment site, particularly during summer months, highlighting a more resilient and balanced benthic environment. Taxonomic distinctness (delta+) and multivariate analyses (PERMANOVA, PCO) confirmed significant spatial and temporal shifts in community structure at the treatment site, driven by IMTA implementation. SIMPER analysis identified key taxa contributing to these changes, which played a pivotal role in structuring the community. The emergence of filter feeders, predators, and omnivores at the treatment site suggests enhanced nutrient cycling and trophic complexity, while the decline in opportunistic species further supports improved environmental conditions. These findings demonstrate the potential of IMTA to promote recovery and sustainable mariculture practices, also offering a comprehensive understanding of its positive effects on hard-bottom benthic community dynamics.

Abstract Background The reproductive cycle of the recently described sponge Tethya meloni was investigated for a period of 15 months (September 2018 – November 2019) in the Mar Piccolo of Taranto (Southern Italy) and was compared with data previously collected for the other two sympatric species of the same genus known for Mediterranean Sea, T. citrina and T. aurantium. Results T. meloni is a gonochoric species with a sex ratio strongly shifted towards females. Asexual budding was a seasonal process, limited to few specimens. In a specimen collected in September 2018 both oocytes and buds occurred, suggesting that in T. meloni the sexual and asexual phases may coexist both at the population and individual levels. Conclusions The data obtained from this research compared with the available literature confirm the high temporal variability of the reproductive cycles in the Mediterranean species of Tethya, but with common general characteristics. In sexual reproduction, the oocyte production period lasts several months, with a peak between summer and autumn while spermatogenesis, shorter but with greater reproductive effort, follows the onset of oogenesis. The asexual reproduction phase of T. meloni, on the other hand, occurs in a short period and seems to have less importance in the overall reproductive process.

Starting from D-xylonolactone and D-ribonolactone, several five-membered bromolactones, related to the C1–C5 portion of mycalin A lactone, have been synthesized. The bromination of D-ribonolactone with HBr/AcOH, without a subsequent transesterification step, has been studied for the first time, giving us most of the acetylated lactones investigated in the present study. For each compound, where possible, both the C-3 alcohol and the corresponding acetate were prepared. Evaluation of their anti-tumor activity showed that all the acetates possess a good cytotoxicity towards human melanoma (A375), human cervical adenocarcinoma (HeLa) and human metastatic melanoma (WM266) cancer cells, comparable or even higher than that displayed by the original mycalin A lactone. Lactone acetates derived from D-ribonolactone showed the higher selectivity of action, exhibiting a strong cytotoxicity on all the tested tumor cells but only a limited toxicity on healthy human dermal fibroblast (HDF) cells, used as a control. Wound healing assays showed that two of these substances inhibit the migration of the WM266 cells.

The biodiversity of macrobenthic invertebrates of two artificial hard substrates close to a mariculture plant was assessed in order to understand the effect on the fouling community of an innovative Integrated Multi-Trophic Aquaculture (IMTA) system. The examined hard substrates were (i) vertical bare collectors that are placed around the cages as new colonizable substrates, which were investigated from the early colonization and (ii) artificial hard substrates already present under the cages analyzed over time to observe changes due to the action on the water column by filter feeder organisms colonizing the above vertical collectors. Overall, 186 taxa were collected (both the substrates), of which 99 as sessile habitat-former or structuring macroinvertebrates and 87 as associated fauna, mostly vagile forms. On the vertical collectors 121 taxa were collected, among which 44 sessile structuring species and 77 vagile-associated taxa; on the artificial hard substrates under the cages, 124 taxa were identified, 95 belong to the first category and 29 as associated taxa. The two analyzed substrates shared 43% of sessile species (40) and 22% of associated species (19). At the end of the first year of experimentation, the study revealed Sabella spallanzanii and mussels as the most abundant taxa. Lastly, the communities under the cages showed an increase in biodiversity after the placement of collectors. The changes were attributed to the decrease in particulate matter originating as wastes from the breeding cage, which was intercepted by the filter feeder community developed on the vertical collectors.

The use of marine sponges dates back thousands of years, and interest in these animals is increasing as new applications are discovered. Their potential is extensive, both in their ancient and still popular use as bath sponges for cosmetics and regarding the more recent discovery of bioactive secondary metabolites mainly of interest for the pharmaceutical industry and the less developed aquariology. Despite their proven biofiltration and ecosystem restoration ability and the biomass supply problem for the interested industries, few integrated multi-trophic aquaculture (IMTA) systems incorporate these invertebrates in their facilities. Therefore, in this brief review, the benefits that marine sponges could bring to rapidly growing IMTA systems are summarized, highlighting their suitability for a circular blue economy.

Poriferans, as sessile organisms without rigid external covering, use secondary metabolites for protection from predators and fouling organisms. The present study tested the antifouling activity of ethanolic extract of the Mediterranean alien calcareous sponge Paraleucilla magna towards juvenile mussels Mytilus galloprovincialis. Furthermore, toxicity tests on nauplii of brine shrimp Artemia salina and two microalgae strains, Nannochloropsis sp. and Tetraselmis suecica, were also conducted. A total attachment inhibition of M. galloprovincialis was achieved at a concentration of 400 µg/mL of sponge extract. The 50% mortality of A. salina nauplii was recorded at a concentration of 500 µg/mL of ethanolic extract. The growth inhibitory effect on both marine microalgae strains has been registered at a concentration of 300 µg/mL. Our results suggest promising natural antifouling activity and low toxicity of the ethanolic extract of P. magna that could be used as antifouling compound.

The expansion of aquaculture practices in coastal areas can alter the balance of microbial communities in nearby marine ecosystems with negative impacts on both farmed and natural species, as well as on human health through their consumption. Among marine filter-feeder invertebrates, poriferans are known as effective microbial bioremediators, even though they are currently still underutilized in association with fish mariculture plants. In this study, we investigate the microbial bioremediation capability of the demosponge Hymeniacidon perlevis in an experimental land-based fish farm where this species occurred consistently in the drainage conduit of the wastewater. Microbiological analyses of cultivable vibrios, total culturable bacteria (37 °C), fecal and total coliforms, and fecal enterococci were carried out on the fish farm wastewater in two sampling periods: autumn and spring. The results showed that H. perlevis is able to filter and remove all the considered bacterial groups from the wastewater, including human potential pathogens, in both sampling periods. This finding sustains the hypothesis of H. perlevis use as a bioremediator in land-based aquaculture plants as well.

The results of an ex-ante survey aiming to assess the impact of a fish farm in the Mar Grande of Taranto (southern Italy, Mediterranean Sea) on the surrounding environment are reported. There, the implementation of an innovative IMTA plant was planned, with the goals of environment bioremediation and commercially exploitable biomass production. Analyses were conducted in February and July 2018. Both seawater and sediments were sampled at the four corners of the fish farm to detect the existing biological and physico-chemical features. The investigation was performed to identify the best area of the farming plant for positioning the bioremediating system, but also to obtain a data baseline, to compare to the environmental status after the bioremediating action. Data were also analyzed by canonical analysis of principal coordinates (CAP). All the measurements, in particular, microbiology and macrobenthic community characterization using AZTI’s Marine Biotic Index (AMBI) and the Multivariate-AMBI (M-AMBI) indices, suggest that the effect of fish farm waste was concentrated and limited to a small portion of the investigated area in relation to the direction of the main current. A site named A3, which was found to be the most impacted by the aquaculture activities, especially during the summer season, was chosen to place the bioremediation system.

Sponges are an important constituent of filter-feeder benthic communities, characterized by high ecological plasticity and abundance. Free bacteria constitute an important quota of their diet, making them excellent candidates in aquaculture microbial bioremediation, where bacteria can be a serious problem. Although there are studies on this topic, certain promising species are still under investigation. Here we report applied microbiological research on the filtering activity of Sarcotragus spinosulus on two different concentrations of the pathogenic bacterium Vibrio parahaemolyticus in a laboratory experiment. To evaluate the effects of the filtration on the surrounding nutrient load, the release of ammonium, nitrate, and phosphate was also measured. The results obtained showed the efficient filtration capability of S. spinosulus as able to reduce the Vibrio load with a maximum retention efficiency of 99.72% and 99.35% at higher and lower Vibrio concentrations, respectively, and remarkable values of clearance rates (average maximum value 45.0 ± 4.1 mL h−1 g DW−1) at the highest Vibrio concentration tested. The nutrient release measured showed low values for each considered nutrient category at less than 1 mg L−1 for ammonium and phosphate and less than 5 mg L−1 for nitrate. The filtering activity and nutrient release by S. spinosulus suggest that this species represents a promising candidate in microbial bioremediation, showing an efficient capability in removing V. parahaemolyticus from seawater with a contribution to the nutrient load.

In this paper, we report data from the first year of rearing of a set of filter feeder bioremediator organisms: macrobenthic invertebrates (sabellid polychaetes and sponges), coupled with macroalgae, realized in a mariculture fish farm. This innovative integrated multi-trophic aquaculture (IMTA) system was realized at a preindustrial level in the Gulf of Taranto (southern Italy, northern Ionian Sea), within the framework of the EU Remedia Life project. Long lines containing different collector typologies were placed around the fish breeding cages. Vertical collectors were utilized for both polychaetes and sponges, whilst macroalgae were cultivated in horizontal collectors. Data on the growth and mortality of the target species after the first year of rearing and cultivation are given together with their biomass estimation. Polychaete biomass was obtained from natural settlement on ropes previously hung in the system, while sponges and macroalgae were derived from explants and/or inocules inserted in the collectors. The description of the successional pattern occurring on collectors used for settling until reaching a “stable” point is also described, with indications of additional filter feeder macroinvertebrates other than polychaetes and sponges that are easily obtainable and useful in the system as bioremediators as well. The results demonstrate an easy, natural obtaining of large biomass of sabellid polychaetes settling especially from about a 4 to 10 m depth. Sponges and macroalgae need to be periodically cleaned from the fouling covering. The macroalgae cycle was different from that of invertebrates and requires the cultivation of two different species with about a 6-month cycle for each one. The present study represents one of the first attempts at IMTA in the Mediterranean area where invertebrates and macroalgae are co-cultured in an inshore fish farm. Possible utilization of the produced biomass is also suggested.

Soil nitrogen abundance, as well as nitrogen use efficiency (NUE), significantly affect the crop yield and grain protein content (GPC). Depending on the genotype, a negative correlation between the yield and GPC can occur. The aim of the study was to assess the agronomic performance, and to explore physiological pathways for the efficient use of N fertilizer for two durum wheat cultivars, “Aureo” and “Vespucci”. After fertilization, the nitrogen content and values of some of the agronomic parameters and yield-related traits increased in both cultivars; nevertheless, a simultaneous rise in both the yield and GPC occurred only in Aureo. The biochemical parameters, analyzed at tillering, confirm the genotypic specificity of nitrogen use. In Vespucci’s roots, the nitrogen supply did not affect the nitrate reductase (NR), but greatly increased the amino acids and proteins, suggesting that ammonium is preferentially assimilated. In Aureo, nitrate is in part assimilated by the roots, as suggested by the ammonium increase and NR enhancement. In the leaves of both cultivars, organic nitrogen significantly increased after fertilization; however, the rise in amino acids, as well as in NR activity, was higher in Aureo than in Vespucci. These results indicate that the different nitrogen use, and in particular the diverse NR behavior, at tillering, are in part responsible of the cultivar differences in grain yield and GPC.
