Χρησιμοποιήθηκαν τρία στελέχη του πρωτόζωου λεϊσμάνια, δηλαδή ένα ευαίσθητο στη σινεφουνγίνη του είδους L. tropica και δυο φυσικώς ανθεκτικά των ειδών L. tropica και L. infantum. Η μελέτη έγινε με τη βοήθεια ραδιοσεσημασμένων ουσιών που ήταν η τριτιωμένη θυμιδίνη, η ουριδίνη και η λευκίνη. Τα μεγαλομορια του παρασίτου που είχαν σημανθεί με τα ως άνω ραδιενεργά στοιχεία βρίσκονταν σε δυο διαφορετικές φάσεις, στερεά και υγρή, και αναλύονταν με τη μέθοδο του Schneider. Οι μετρήσεις διενεργούνταν σε μετρητή υγροΰ σπινθηρισμοΰ (Beckman) και η ενσωμάτωση των ραδιοϊσοτόπων εκφραζόταν σε κρούσεις ανά λεπτό (cpm). Η συγκέντρωση των πρωτεϊνών των παρασίτων καθορίστηκε με τη μέθοδο Lowry. Η ενσωμάτωση των ραδιενεργών στοιχείων στα μεγαλομορια εκφράζεται σε cmp/mg πρωτεϊνών του πρωτόζωου. Παρατηρήθηκε σημαντικού βαθμού αναστολή της ενσωμάτωσης της θυμιδίνης που ήταν ανάλογη της συγκέντρωσης της σινεφουνγίνης και του χρόνου έκθεσης των καλλιεργειών των προμαστιγωτών μορφών του ευαίσθητου στελέχους L. tropica στο αντιβιοτικό. Η αναστολή της βιοσύνθεσης του DNA που επέφερε η σινεφουνγίνη σε συγκέντρωση 2,6 μΜ (υπερδεκαπλάσια της λεϊσμανιοστατικής και υποδεκαπλάσια της λεϊσμανιοκτόνου) στο ευαίσθητο στέλεχος L. tropica κυμάνθηκε από 25% έως 64% σε χρόνους δράσης της μιας και είκοσι τεσσάρων ωρών αντίστοιχα. Δε σημειώθηκε αναστολή της βιοσύνθεσης του RNA στο πρωτόζωο. Σχετικά με την ενσωμάτωση της λευκίνης παρατηρήθηκε μικρότερου βαθμού αναστολή στο ίδιο στέλεχος σε σχέση με την αντίστοιχη της θυμιδίνης που κυμάνθηκε από 10% έως 24%. Αντίθετα, η σύνθεση του DNA των ανθεκτικών στελεχών δεν επηρεάστηκε ακόμα και όταν οι χρησιμοποιούμενες συγκεντρώσεις της σινεφουνγίνης για έξι ώρες ήταν η λεϊσμανιοκτόνος 26 μΜ και πολλαπλάσιες αυτής. Η δράση του αντιβιοτικού σινεφουνγίνη στις βιοσυνθετικές λειτουργίες του πρωτόζωου λεϊσμάνια είναι ανεξάρτητη του είδους και συσχετίζεται με την έκφραση της ευαισθησίας ή ανθεκτικότητας του στελέχους. Η σινεφουνγίνη παρεμβαίνει στη σύνθεση του DNA των πρωτόζωων που έχουν υψηλό βαθμό ευαισθησίας σ' αυτήν., The aim of the work presented here was the in vitro study of the action of the antibiotic sinefungin on the macromolecules DNA, RNA and the proteins of different isolates of Leismannia spp. in relation to the resistance or to the sensitivity of the strains examined. The mode of action of sinefungin of the Leishmania strains was detected by incorporating radiolabeled thymidine, uridine and leucine into promastigote forms, to study the synthesis of DNA, RNA and proteins respectively. In the sensitive strain L. tropica a statistically significant reduction of the rate of incorporation of thymidine to the DNA of the parasite, resulted to a 64% inhibition of leishmanial DNA synthesis by sinefungin at a concentration of 2.6 mM, when the time of action of the drug was 24 hours. In the sensitive strain L. tropica the reduction of the rate of incorporation of thymidine was depended on the concentration and the time of action of the drug. No inhibition of leishmanial RNA synthesis was observed. Our research showed that the action of the antibiotic sinefungin to the macromolecular biosynthesis of the protozoan Leishmania is independent of the strain and is strongly related with the expression of the sensitivity and the resistance of the strain. The main target of the antibiotic examined was the DNA of the Leishmania strains sensitive to the drug.

Στην παρούσα ερευνητική εργασία μελετήθηκε η δράση του αντιβιοτικού σινεφουνγίνη στην προσκόλληση επτά στελεχών Leishmania των ειδών L. infantum, L. tropica και L. major στα μακροφάγα της κυτταρικής σειράς J774G8. Αναστολή της προσκόλλησης αρχικού σταδίου της φαγοκυττάρωσης παρατηρήθηκε μόνο για ευαίσθητα στελέχη έναντι της σινεφουνγίνη ς των ειδών L. tropica και L. major. Η ύπαρξη συσχέτισης της συμπεριφοράς του πρωτόζωου έναντι της σινεφουνγίνης στις καλλιέργειες των προμαστιγωτών μορφών και των αλληλεπιδράσεων τους με τα μακροφάγα της κυτταρικής σειράς J774G8 διαπιστώθηκε μόνο όταν η σινεφουνγίνη προστέθηκε στις καλλιέργειες των προμαστιγωτών μορφών (72 ώρες με συγκεντρώσεις σινεφουνγίνης 2,6 και 26 μΜ). Μετά από επώαση 2 ωρών των προμαστιγωτών μορφών του ευαίσθητου στελέχους L. tropica η αναστολή βρέθηκε 17 και 37% σε συγκεντρώσεις 2,6 και 26 μΜ του αντιβιοτικού αντίστοιχα. Μετά από επώαση 24 ωρών των προμαστιγωτών με τα μακροφάγα η αναστολή προσκόλλησης για το ευαίσθητο στέλεχος L. tropica βρέθηκε 52 και 77% και για το ευαίσθητο στέλεχος L. Major 73 και 86% σε συγκεντρώσεις 2,6 και 26 μΜ του αντιβιοτικού αντίστοιχα. Αποδείχθηκε η απουσία τοξικότητας της σινεφουνγίνη ς για τα μακροφάγα κύτταρα στα οποία δεν επέφερε μορφολογικές αλλοιώσεις και διαταραχή της ανάπτυξης τους. Η σινεφουνγίνη παρεμβαίνει αποτελεσματικά στην προσκόλληση προμαστιγωτών-μακροφάγων και η δράση της αυτή συσχετίζεται με την έκφραση ευαισθησίας ή ανθεκτικότητας του πρωτόζωου., The effect of Sinefungin on the attachment of the Leishmania strains L. infantum, L. tropica and L. major to the macrophages cell line J774G8 was studied. Inhibition ofthe attachment to the surface of macrophages was observed to the Sinefungin sensitive strains L. tropica and L. major. This inhibition was obtained only for Sinefungin pretreated promastigotes (72 hours with 2.6 and 26 Μμ Sinefungin) when incubated with macrophages for 2 and 24 hours. After 2 h incubation period of the promastigote forms of the sensitive L. tropica with macrophages, the inhibition of the attachment was 100%, whereas for the sensitive L. major strain the inhibition was 17 and 36% at a concentration of Sinefungin 2.6 and 26 μΜ respectively. The percentage of the inhibition 24 h post infection was in the range of 52 to 77% for the L. tropica strain and from 73 to 86% for the L. major strain for promastigote pretreated with 2.6 and 26 μΜ Sinefungin respectively. Controls showed that Sinefungin was not toxic for the macrophages when used for 3 days at the above mentioned concentrations. Inhibition of the protozoa - macrophage cell line J774G8 adherence was a common finding leading us to the conclusion that Sinefungin is involved actively into that process.

Iridium (Ir)-(COD)-pentamidine tetraphenylborate (CAS 225-75-4) was selected from a primary screening to be evaluated in vitro on three Leishmania (L.) strains comparatively to pentamidine used as reference compound. The IC50 values obtained from in vitro evaluation on promastigotes of L. major CRE 26, L. donovani DD8 and L. donovani LV9 were 3.9, 23.5, and 3.3 mumol/l for Ir-(COD)-pentamidine tetraphenylborate and 1.6, 7.7, and 3.9 mumol/l for pentamidine isethionate, respectively. Cytotoxicity on mouse peritoneal macrophages led to determine a chemotherapeutic index of 1.7 for Ir-(COD)-pentamidine tetraphenylborate and 4 for pentamidine. Considering L. donovani DD8, the uptake of iridium complex by the promastigotes was shown to be saturable with a Km value of 17.4 mumol/l and Vmax of 1.3 nmol/mg protein/2 h. After 2 and 4 h incubation of treated promastigotes in drug free medium the absence of Ir-complex efflux is in favour of intracellular drug binding. As a matter of fact iridium complex was shown to bind ribosomal subunits in vitro, with no effect on macromolecular biosynthesis.
