Sex ratio (ratio between the number of male and female individuals in a population) is an important factor contributing to the survival rates of the population in wild. Imbalanced sex ratio is often the cause of diminished survival rates of endangered species. The analysis of DNA collected through noninvasive methodologies effectively allows to monitor populations of rare species or species that are difficult to observe in the wild. Such method was used to determine the sex ratio in populations of the capercaillie (Tetrao urogallus) and black grouse (Lyrurus tetrix) in the Tatra National Park. We collected over 250 samples (mostly excrement) in the area of the Tatra National Park and identified species via microsatellite genotyping and determined the sex using fragments of the CHD gene. We found that the majority of the samples belonged to male specimens irrespective of the species suggesting a large deviation from an equal sex ratio in wild populations of capercaillie and black grouse in the Tatra National Park. Our results point out the fundamental contribution of genetic assessment of sex ration to monitor the viability of both species in the Tatra National Park.

Purpose We aimed to study brain tissue oxygenation during the period of controlled reduction of arterial blood pressure – a maneuver often used in extended endoscopic skull base surgery for bloodless operative field. Methods Intracranial pressure, arterial blood pressure and the resultant cerebral perfusion pressure were measured during extended endoscopic skull base surgery in 5 patients with diagnosed tumors of the skull base and arterial hypertension. Simultaneously, in those patients, we measured partial pressure of oxygen in the brain parenchyma (PbtO2). Results Values of PbtO2 lower than 15 mm Hg (risk of brain ischemia) were observed in 3 patients for periods of 40 min, 110 min and 123 min, respectively. In 2 of these patients, no hypotension (mean arterial pressure Conclusion Moderate reduction of arterial pressure, often used to obtain bloodless operative field during extended endoscopic skull base surgery, may in patients with the medical history of arterial hypertension be associated with critically low values of partial oxygen pressure in brain tissue.

Recent increase in incidence of meningiomas suggests the need to search for new risk factors. Leptin, a potentially pro-angiogenic and proliferative agent, could be a candidate for this role, as its expression correlates with body mass index (BMI). Because development of meningioma has also been linked to sex hormones, bisphenol A (BPA), a known xenoestrogen, can also be taken into consideration as a potential risk factor. The aim of this study was to determine plasma concentrations of both substances in patients with meningiomas and to match it to patients with gliomas - a group of brain tumors less hormone- and BMI-dependent.Concentrations of BPA and leptin were measured in plasma of 24 patients with low grade meningioma and in 29 patients with glioma, using gas chromatography-mass spectrometry (GC-MS) and ELISA kits, respectively. The concentrations of both substances in patients with neoplasms were interpreted in relation to their concentration in healthy population, published in recent reports.Free and conjugated BPA were present in both meningioma and glioma patients. Moreover, their concentrations far exceeded those reported in the healthy population. Nevertheless, the level of leptin revealed to be significantly higher in meningioma patients than in glioma patients.Occurrence of both meningioma and glioma may be accompanied by increased concentrations of leptin and BPA. Further large-scale studies are needed to clarify whether the presence of both substances may play a role in pathogenesis or influence clinical course in patients with brain neoplasms.

This study focuses on morphological and genetic differences between two central European subspecies of the noctuid moth Noctua interjecta Hübner: N. interjecta interjecta Hübner, 1803 from Transcarpathia in Ukraine and N. interjecta caliginosa (Schawerda, 1919) from Poland. While the morphological differences between these taxa are fairly obvious, individuals from these two populations were found to differ also genetically, as indicated by the cytochrome c oxidase I (COI) gene sequences. The Transcarpathian population exhibits solely the NImizH3 haplotype, but the Polish one the relatively distant NImizH1 and NImizH2 haplotypes. A Neighbour-Joining tree and a network of haplotypes separated the two haplotypes found in Poland (N. i. caliginosa) from those derived from NImizH3 (N. i. interjecta). The divergence time for these two groups was estimated as 400–600 thousand years before present, coinciding with the maximum extent of the Pleistocene ice-sheet covering Europe.

There are several genetic studies of the grayling (Thymallus thymallus) in Central Europe, however, the phylogeography and genetic structure of the populations from some areas are still poorly recognized. The aim of this study was to determine the distribution of genetic diversity and the history of the postglacial migration of the species in Poland. Biological samples were collected in seven rivers and one fish farm. All specimens were genotyped at 12 microsatellite loci. Moreover, the sequence of ND5 and control region mtDNA gene was determined. We found clear differences in genetic diversity among the investigated population, with the lowest values found for the fish farm (Rutki) and intensively exploited river (Plytnica), and the highest for coastal and submontane rivers (Vistula, Wislok, San). Phylogenetic analysis based on mtDNA suggests the presence of two postglacial colonization waves of grayling in Poland: (I) from the refugium of the Danube; (II) the Atlantic Basin. Currently, managing the populations on the European scale should involve stocking populations with the materials obtained from fish caught in the same rivers taking into account both environmental factors and rational angling. Stocking plans should consider local populations whose mixing will reduce the heterozygosity of the population without affecting the genetic pool.

Population size and effective population size are important factors affecting probability of extinction of small, isolated population. Hence, from conservation perspective, it is recommended to monitor changes in population size of endangered species. Genetic methods, based on genetic profiling of non-invasive samples of biological material, despite some limitations, were proved to be efficient method in tracking individuals in the field and estimate populations' parameters. We used this strategy to investigate isolated population of the capercaillie (Tetrao urogallus) in the Gorce Mountains. In two study periods (2012–2013 and 2017–2018) almost 400 faeces and feathers were collected. Microsatellite genotyping was performed to identify individuals and estimate genetic diversity. We found that population is stable in terms of size and genetic indices, although allelic richness has significantly increased between 2012–2013 and 2017–2018. In the overall study period (2012–2018) there were 52 individuals identified. However, only 10 birds were found in both study periods. This suggests low survival in the population. Moreover, genetic data indicated low effective population size of the capercaillie in the Gorce Mts. Thus, we suggest that monitoring, either genetic and based on field-surveys, should be implemented in the management and protection of this population.

BACKGROUND Intracranial aneurysms are common, occurring in about 1-2% of the population. Saccular aneurysm is a pouch-like pathological dilatation of an intracranial artery that develops when the cerebral artery wall becomes too weak to resist hemodynamic pressure and distends. OBJECTIVES The aim of this study was to determine whether the development of intracranial aneurysms and subarachnoid hemorrhage (SAH) affects neuronal phospholipid metabolism, and what influence different invasive treatments have on brain free radical phospholipid metabolism. MATERIAL AND METHODS The level of polyunsaturated fatty acid (PUFA) cyclization products - F2-isoprostanes and F4-neuroprostanes - was examined using liquid chromatography - mass spectrometry (LC-MS) in the plasma of patients with brain aneurysm and resulting subarachnoid hemorrhage. RESULTS It was revealed that an aneurysm leads to the enhancement of lipid peroxidation with a significant increase in plasma F2-isoprostanes and F4-neuroprostanes (more than 3-fold and 11-fold, respectively) in comparison to healthy subjects. The rupture of an aneurysm results in hemorrhage and an additional increase in examined prostaglandin derivatives. The embolization and clipping of aneurysms contribute to a gradual restoration of metabolic homeostasis in brain cells, which is visible in the decrease in PUFA cyclization products. CONCLUSIONS The results indicate that aneurysm development is associated with enhanced inflammation and oxidative stress, factors which favor lipid peroxidation, particularly in neurons, whose membranes are rich in docosahexaenoic acid, a precursor of F4-neuroprostanes.

To analyze the expression of β-catenin and N-cadherin in large series of meningioma cases and to investigate their correlation with peritumoral brain edema (PTBE).Study group consists of 154 patients diagnosed with intracranial meningioma divided into: low-grade (G1) and high-grade (G2 or G3) group. PTBE was graded into four groups (0, I, II, III) using Steinhoff classification. The expression of N-cadherin, β-catenin was analyzed and graded based on the positive ratio of immunoreactivity. The results were analyzed statistically.104 cases were low-grade and 50 high-grade meningiomas. PTBE was observed in 103(66.8 %) cases: 57 grade I, 44 grade II and 2 grade III. Positive N-cadherin expression was found only in the membrane of the neoplastic cells in 50(48.1%) cases of low-grade, and in 34(68%) of high-grade group. In low-grade meningioma, β-catenin expression was observed within the cytoplasm and nucleus in 54(51.9%) cases. In high-grade meningiomas, β-catenin expression was observed in 33(66%) tumors only within the nucleus. N-cadherin expression was observed in 36 cases with PTBE grade I, 28 with grade II and 2 with grade III. β-catenin expression was observed in 40 cases with PTBE grade I, 24 with grade II and 2 with grade III. The results were statistically significant.Significant N-cadherin expression especially in high-grade meningioma group was found. β-catenin expression was the most evident in the nucleus rather than in cytoplasm. The degree of PTBE correlated with the N-cadherin and β-catenin expression and was the most prominent in high-grade meningioma group.

Human-induced changes in the natural environment impact upon a wide variety of genetic processes at all levels of population structure. Insight into these processes is now achieved by analyses of genetic diversity allowing the past and present condition of populations to be assessed and compared. Given its severe fragmentation and small size, the population of the Capercaillie Tetrao urogallus in Poland can be regarded as especially prone to gene-pool alteration. Moreover, this is a region of Europe in which different genetic lineages of the Capercaillie may be present, ensuring that certain conservation decisions may be even more difficult than they otherwise would be. Bearing the above in mind, we decided to study polymorphism of the mitochondrial DNA Control Region (mtDNA CR) in remnant and extinct populations of the Capercaillie in Poland. The extinct population was represented by samples from birds hunted in the Lower Silesian Forest (SW Poland) between 1951–1962. We compared sequences obtained with data from the large, continuous population present in Russia and Scandinavia. Analysing 331 bp of mtDNA CR for 168 individuals, we identified 24 haplotypes, among which only 6 had not been reported previously in the species according to GenBank. Our data confirmed a previous suggestion regarding the genetic structure of the species in Poland, i.e. that birds from Augustow Forest (NE Poland) are related to Russian and Scandinavian populations, constituting a part of the lowland group within the boreal lineage. The Carpathian population is in turn confirmed as an independent genetic group within the boreal lineage, while specimens from the Solska Forest (SE Poland) prove to be highly differentiated from birds in other Polish populations; most likely because they derive from — or have been connected via gene flow with — birds present in the Balkans. Further genetic analysis is necessary to resolve this evolutionary relationship between Capercaillies from south-eastern Poland and those of the Balkans. Where practice is concerned, the overall advice is that three Conservation Units are to be recognised in Poland. In addition to those, the extinct population from the Lower Silesian Forest has been deemed part of the lowland group within the boreal lineage, albeit a distinctive one, given the possibility of gene exchange with Carpathian birds. We further suggest that some aspect of this extinct population's genetic diversity reflects an uncontrolled process of reintroduction taking place in the past.

Urbanised areas are capable of exerting a strong impact on the distribution of genetic diversity within populations of animals. Urban invertebrate species are currently either relicts from pre-urban ecosystems, or have immigrated during or following urbanisation. We analysed 10 microsatellite loci in 196 specimens of the harlequin ladybird (Harmonia axyridis), an invasive species in Poland sampled in three of the country's cities. Of Asiatic origin, this ladybird has been spreading through other continents, including Europe. Results showed that the Polish cities are being invaded by harlequin ladybirds that are uniform in terms of their genetic pool, with no significant genetic differentiation present between the urban populations investigated. Rapid spread and what are probably the large numbers of individuals colonising new areas allow this species to maintain high genetic diversity and avoid bottleneck effects. However, we suggest that urban populations differ in terms of genetic diversity. The highest genetic diversity characterises the most recently invaded area of Olsztyn in the north of Poland. Genetic data further suggest that this population could be still in an expansion phase. The earliest colonising population in Wroclaw exhibits signs of a Wahlund effect, suggesting that gene flow among local groups within this urban area could be disturbed or impaired. We conclude that, in the case of an urban population of the harlequin ladybird, successful colonisation is followed by the onset of a decline in genetic diversity, with isolation between local sampling sites appearing. Further studies are required, however, if this process is to be elucidated.

We examined the relationship between individual heterozygosity of male Red-breasted Flycatchers (Ficedula parva; a small long-distance migratory, socially monogamous bird species) and their mating success, arrival time and age. Using eight polymorphic microsatellite loci, we found that male heterozygosity is related to both mating success and arrival time, but not to age. Mated and earlier arriving males had higher heterozygosity than later arrivals and bachelors, but we did not find a relationship between age and individual heterozygosity of males. To our knowledge, this is the first study to provide evidence about the relationship between individual genetic diversity and arrival time, thus arrival time could be used as a signal of individual heterozygosity and quality.

Knowledge about population size is of high importance for conservationists. We used non-invasively collected samples and microsatellite genotyping to estimate the size of the Tatra National Park population of the endangered capercaillie Tetrao urogallus. This population is one of the most important strongholds of the species in Poland. In 2016 over 150 samples (faeces and feathers) of the capercaillie were collected throughout area of the Tatra National Park. Then, DNA was extracted and genetic profiles were evaluated, using nine microsatellite markers. We obtained 81 reliable genotypes. Among them, 34 unique genotypes were found, corresponding to Minimum Number of individuals Alive in the investigated population. Application of capture-recapture models in the R package Capwire indicated, that the area was inhabited by approx. 54 birds, whereas regression model suggested presence of 36–64 individuals. Previous field surveys suggested that the number of birds in the Tatra National Park is about 50. Hence, we assumed that genetic tagging of non-invasive samples performs well in estimating the abundance of the capercaillie in the investigated population.

Introduction: Alcohol dehydrogenase (ADH) isoenzymes and aldehyde dehydrogenase (ALDH) exist in the brain. Alcohol dehydrogenase and ALDH are also present in brain tumor cells. Moreover, the activity of class I isoenzymes was significantly higher in cancer than healthy brain cells. The activity of these enzymes in tumor tissue is reflected in the serum and could thus be helpful for diagnostics of brain neoplasms. The aim of this study was to investigate the potential role of ADH and ALDH as markers for brain tumors. Material and methods: Serum samples were taken for routine biochemical investigation from 115 patients suffering from brain tumors (65 glioblastomas, 50 meningiomas). For the measurement of the activity of class I and II ADH isoenzymes and ALDH activity, fluorometric methods were used. The total ADH activity and activity of class III and IV isoenzymes were measured by the photometric method. Results : There was a significant increase in the activity of ADH I isoenzyme and ADH total in the sera of brain tumor patients compared to the controls. The diagnostic sensitivity for ADH I was 78%, specificity 85%, and positive and negative predictive values were 86% and 76% respectively. The sensitivity and specificity of ADH I increased with the stage of the carcinoma. Area under receiver-operating characteristic curve for ADH I was 0.71. Conclusions : The results suggest a potential role for ADH I as a marker for brain tumor.

We analyse the genetic variability in the yellow-necked mouse (Apodemus flavicollis) population in the city of Warsaw, Poland, and its surroundings - a species that has begun to occupy the city only in the last 30 years. We also compare the genetic variability of this species with corresponding data collected in the same time and areas for another species - the striped field mouse (Apodemus agrarius). The results indicate a gradual decrease in genetic diversity and increase in relatedness in the population of A. flavicollis from non-urban locations towards sites with the highest anthropopressure. The genetic structure was more pronounced in the 'recent invader' (A. flavicollis) than in the 'permanent inhabitant' (A. agrarius), which has a much longer city colonization history (more than 100 years). In general, FST was higher in A. flavicollis, which may indicate different and independent ways of city colonization by the species. The process by which urban areas are settled by a new, typically forest-dwelling species such as A. flavicollis, more 'sensitive' to the conditions of life in a city, probably includes not only successful events of penetration of the city by small groups of individuals (the founder effect), but also temporary extinctions of local urban populations of A. flavicollis or at least marked fluctuations in species population numbers. Suitably planned areas at the city borders could play an important role as 'gateways' through which individuals from non-urban populations could migrate into the city and join urban populations.

Purpose Gliomas are the most common primary brain tumors. The etiology is still unclear and the progression from low to high-grade gliomas is frequent. The molecular mechanisms are quite established, however the heterogeneity of glioblastomas force the scientist to look for the new therapeutic targets. The aim of the study was to evaluate the caspase-3 and survivin expression in correlation with MIB-1 expression in gliomas of various grade to assess the apoptosis in gliomas and to determinate new possible targets for the future therapy. Material and methods We identified 131 patients with a histopathological diagnosis of astrocytic tumors (diffuse astrocytoma, anaplastic astrocytoma and glioblastoma). The evaluation of caspase-3, survivin and MIB-1 expression was done using immunohistochemical methods. Results Caspase-3 and survivin expression was observed both in low- and high-grade astrocytomas. The differences in expression were the most evident in glioblastoma group. All primary glioblastomas (31 cases) expressed caspase-3. In secondary glioblastoma group only 17 out of 30 specimens were positive for caspase-3. Survivin expression was observed in 80.6% primary glioblastomas and in all examined secondary glioblastomas and the staining was strong and diffuse in all cases. MIB-1 expression was low in diffuse astrocytomas (DA) and ranged between 1 and 5%. In anaplastic astrocytoma group it was ranged between 5 and 10% and the highest percentage of the positive cells was observed in glioblastoma cases and ranged from 10% even to 30%. The most evident MIB-1 expression was observed in the cells surrounding the pathological blood vessels and necrosis. Conclusions The high incidence of survivin and caspase-3 expression in diffuse and anaplastic astrocytoma cases may suggest, that the regulation between pro- and antiapoptotic proteins may play an important role in tumor growth and progression. The overexpression of survivin and MIB-1 expression in glioblastoma cases also may confirm the theory about the important role of anti-apoptotic and proliferation processes in glioblastoma progression and as such may be potential therapeutic targets.

While the hazel grouse Tetrastes bonasia declined in the twentieth century to the point where certain local subpopulations in Poland disappeared altogether, the overall population of the species in the country now seems to be back on the increase. To consider the remaining genetic diversity in that population, analyses of a fragment of the hypervariable domain I of the mitochondrial control region were performed on faecal samples from 251 of the birds. This work provided confirmation of conclusions drawn from previous observations that birds in the populations present in smaller and more isolated forests were characterised by more limited genetic diversity than those occurring in continuous, less-fragmented forests. The level of genetic diversity among birds from central Poland is also found to be high, most likely because recent recolonisation there has involved individuals from both the north and south of the country, as is also suggested when migration rates are estimated. Analysis further indicated a pronounced genetic structure, with a range of processes seen to affect genetic differentiation from one region of Poland to another. Analysis of molecular variance (AMOVA) provided for an assignment of populations to groups corresponding well with the geographical distribution of forests in Poland. Gene flow among the populations is shown to be very low, with no more than 1.5 female migrants per generation coming in. Both high nucleotide diversity and mismatch distribution, and the AMOVA and nested clade analysis (NCA) suggest that birds following at least two different post-glacial recolonisation routes encountered one another in north-eastern Poland. Equally, there are clear indications that the north-eastern and southern parts of the country were recolonised in different periods. The genetic results presented here underline the importance of gene flow in maintaining genetic variability within populations of the hazel grouse, at the same time suggesting that the country’s southern and north-eastern populations should be managed independently where conservation efforts are concerned.

Small and isolated populations are prone to future extinctions and thus perceived as ‘living dead’. Although generally considered to be of low conservation value, their existence can still enhance species survival at the landscape scale through improving the connectivity of other populations and facilitating some (even if little) gene flow. We investigated the demography and genetic status of a tiny and highly isolated local population of Maculinea (= Phengaris) alcon near its distribution margin with the aim of identifying the features that allow it to persist. The study comprised intensive mark-recapture, surveys of Gentiana pneumonanthe foodplants and butterfly eggs laid on them, as well as genetic analyses. The population has been found to be characterised by low genetic diversity and estimated at only a few tens of individuals. The foodplant availability turned out to be the most obvious factor limiting M. alcon abundance. Nevertheless, the life expectancy of adult butterflies is fairly long, and their flight period very short, implying that most individuals occur within the same time window. Together with the relatively little protandry observed, i.e. almost synchronous emergence of males and females, this increases the chances of random mating among the individuals. Moreover, the butterflies move freely across the core habitat fragment. All things concerned, the effective population size is presumably not much lower than the recorded population size. Our findings provide guidelines for pinpointing those among ‘living dead’ populations that are likely to be the most persistent and thus worth conservation efforts aimed at preserving them.

The Montagu’s harrier (Circus pygargus) is a long-distance migratory raptor, breeding in Europe and Western Asia and wintering in sub-Saharan Africa. The population of the species has declined in Europe during the twentieth century, and Montagu’s harrier is red-listed in many European countries as declining or threatened. The main aims of the study were to evaluate the genetic diversity of European breeding populations and estimate the genetic differentiation among them, using polymorphism in the hypervariable domain of the mitochondrial control region. We analysed 158 individuals from central Spain, Germany, the Czech Republic and Poland. The results indicated high genetic diversity in the European breeding population, probably reflecting the large population size of the species. However, we found decreased genetic variability in the breeding population of Germany. Among the 18 identified haplotypes, 2 were of high frequency. We examined changes in genetic diversity in populations of two small mammal species inhabiting islands on a lake. We hypothesized that a less mobile species, such as the bank vole, would lose genetic diversity quicker than the yellow-necked mouse, which can more easily cross habitat barriers. In contrast to this we found that the effects of isolation were much more prominent in the case of the mouse than the vole. In the vole population, on the larger island, genetic diversity remained constant over subsequent years in spite of marked isolation. On the smaller island, we noted an increase in genetic diversity which was probably caused by immigration of a small group of individuals. Genetic diversity in the mouse population decreased markedly over the course of the study. Recent avian studies suggest that offspring sex ratios can vary with environmental or parental characteristics. One example is that females may adjust brood sex ratio of fledglings in response to the sexual attractiveness of their mate and are expected to produce more sons when mated with older or more ornamented males. We determined offspring sex ratios at fledging (158 nestlings from 33 broods) in a population of migratory bird species, Red-breasted Flycatchers (Ficedula parva) that were breeding on the western edge of their geographic range, in the strictly protected part of the Bialowieza Forest (north-eastern Poland). The ratio of individual broods ranged from all male to all female, but the average offspring sex ratio at the population level did not deviate significantly from the expected 50:50 sex ratio. Meningiomas constitute up to 13% of all intracranial tumors. The predictive factors for meningioma have not been unambiguously defined; however some limited data suggest that the expression of matrix metalloproteinases (MMPs) and vascular endothelial growth factor (VEGF) may be associated with the presence of peritumoral brain edema (PTBE) and worse clinical outcome. The aim of this study was to analyze the expressions of MMP-9 and VEGF in a group of meningiomas of various grades and to study associations between these two markers and PTBE. The study included patients with supratentorial meningiomas. The patients were divided into low- (G1) and high-grade meningiomas (G2 and G3). PTBE was assessed on MRI. The expressions of VEGF and MMP-9 were determined immunohistochemically. The expression of MMP-9 was observed significantly more often in G3 meningiomas than in lower grade tumors. The presence of stage II or III PTBE was associated with a significant increase in MMP-9 expression. The expression of VEGF did not differ across the PTBE stages. Our findings point to a significant role of MMP-9 and VEGF in the pathogenesis of peritumoral brain edema in low- and high-grade meningiomas.

We have attempted to describe the genetic structure and variability of the striped field mouse population inhabiting 17 locations in and around Warsaw, Poland, within a gradient of anthropopressure and discuss the possible factors that could potentially form the observed pattern. Ecological characteristics of the striped field mouse prevent decreases in genetic variability within local urban populations. High population density, ability to cross environmental barriers and to use “green corridors” allows this species to maintain gene flow in fragmented urban landscape. However, genetic variability and gene flow were efficiently reduced in the central part of the city. The results indicated that the degree of human pressure, defined based on the level of vegetation cover, is a good indicator of isolation. In the studied striped field mouse population, genetic interactions among particular locations (local populations) are modified in comparison with populations inhabiting natural areas, by replacement of the isolation-by-distance pattern of differentiation with the “isolation-by-infrastructure” pattern. As indicated by Bayesian analysis, the urban population from the right side of the Vistula river form common genetic cluster with ex-urban population located northward from the city, while the population from the left side of the river probably exchanges genes with neighbouring northern and southern ex-urban populations. However local populations at locations within the highest zone of anthropopressure are clearly isolated, and presently constitute separate genetic units.

Human brain tissue contains various alcohol dehydrogenase (ADH) isoenzymes and possess also aldehyde dehydrogenase (ALDH) activity. In our last experiments we have shown that ADH and ALDH are present also in the brain tumour cells. Moreover the activities of total ADH and class I isoenzymes were significantly higher in cancer tissue than healthy cells. It can suggests that these changes may be reflected by enzyme activity in the serum of patients with brain cancer. Serum samples were taken for routine biochemical investigation from 62 patients suffering from brain cancer (36 glioblastoma, 26 meningioma). For the measurement of the activity of class I and II ADH isoenzymes and ALDH activity, the fluorometric methods were used. The total ADH activity and activity of class III and IV isoenzymes were measured by the photometric method. A statistically significant increase of class I alcohol dehydrogenase isoenzymes was found in the sera of patients with brain cancer. The median activity of this class isoenzyme in the patients group increased about 24 % in the comparison to the control level. The total alcohol dehydrogenase activity was also significantly higher (26 %) among patients with brain tumour than healthy ones. The activities of other tested ADH isoenzymes and total ALDH were unchanged. The increase of the activity of total ADH and class I alcohol dehydrogenase isoenzyme in the sera of patients with brain cancer seems to be caused by the release of this isoenzyme from tumour’s cells.

The aim of our study was to find suitable molecular markers for genetic studies of the population of Montagu's harrier Circus pygargus. We used the crossspecies amplification strategy to test the usefulness of 24 primer pairs, amplifying the microsatellite loci of several other members of Accipitridae. The analysis was performed on 139 Montagu's harriers from breeding populations in Spain and Poland. We found an amplification success of 50%; however, the level of polymorphism in cross-amplified microsatellites was low, especially in terms of heterozygosity. We did not find significant differences in genetic variability, estimated based on microsatellite markers, between breeding populations from Spain and Poland. The level of genetic differentiation between these two populations was low (FST = 0.016), although significant. An analysis of genotypes of nestlings in 10 nests suggested one case of extra-pair paternity.

Background and purpose To assess blood flow velocity in the middle cerebral artery (MCA) during transnasal endoscopic procedures performed with decreased hemodynamic parameters. Materials and methods In 40 patients who underwent endoscopic skull base surgery in controlled hypotension (studied group) and in 13 patients operated without reduction of hemodynamic parameters (control group), blood flow velocity in MCA was assessed with transcranial color Doppler sonography. Results Blood flow velocity in MCA remained within the range of age-specific reference values in all patients before operation. It decreased significantly in both groups after induction of anesthesia and then dropped even further in studied group of patients when hemodynamic parameters were reduced; the systolic velocity fell below the normal reference values in 25% of patients, the mean velocity in 50% and the diastolic velocity in 57% of patients. The diastolic velocity was much more heavily influenced by diminished hemodynamic parameters than systolic velocity in the studied group as opposed to the control group where reduction of blood flow velocity pertained equally systolic and diastolic velocity. Conclusion During transnasal endoscopic procedures performed in moderate hypotension, in addition to significant drop of blood flow velocity to values well below the normal reference range, a divergent reduction of systolic and diastolic velocity was detected. Since divergent systolic and diastolic velocity may indicate an early phase of cerebral autoregulation compromise, and the decrease of mean blood flow velocity in MCA corresponds with a decrease of cerebral blood flow, further investigations in this field seem warranted.

Despite advances in knowledge, in many bird species, the true mating system is unknown, and especially so for birds breeding at low densities under natural conditions. We studied paternity at the population and breeding pair level in the Red-breasted Flycatcher ( Ficedula parva), a small, migrant passerine breeding in old-growth Białowieża Forest. Over seven breeding seasons we genotyped a total of 159 nestlings and 25 adult males as fathers from 33 broods of the Red-breasted Flycatcher. Extra-pair young were present in eight broods (24.2%). Twelve (7.5%) chicks mismatched with the ‘social’ male in several loci. Both low breeding density and synchrony in breeding phenology can explain such findings. Also of interest was the share of extra-pair paternity at the individual level, in relation to the age/ornamentation of the extra-pair male. Male Red-breasted Flycatchers exhibit delayed plumage maturation. Young males (second year) are without an orange badge on the throat and breast, while older males (after-second year) are ornamented and have an orange badge. The extra-pair nestlings were found in eight broods, four with young males as a fathers (36.4%) and four in broods of old males (18.2%). However, in all seven cases when the age of the extra-pair partner was known, irrespective of the age of the social father, the extra-pair partner was a male with an orange patch on the breast. This indicated female preference for ornamented males as partners for extra-pair copulation.

The two-spot ladybird Adalia bipunctata (Linnaeus, 1758) (Coleoptera: Coccinellidae) is a widely distributed generalist, feeding preferably on aphids. The species inhabits various environments, including anthropogenic landscape, where pollution and microclimate enables to attain very high numbers of favorite prey. Recently, the abundance of the two-spot ladybird has declined, mainly as an after-effect of invasion of the competing species — the harlequin ladybird Harmonia axyridis (Pallas, 1773). High phenotypic polymorphisms made the two-spot ladybird classic model for population genetic studies of Coccinellidea. However, studies investigating diversity of the species at DNA level have Coccinellidae been scarce. In our investigation we used microsatellite markers to characterize genetic diversity in the two-spot ladybird populations from different environments. We made an attempt to amplify 13 loci, identified previously in the genome of the species, however due to lack of amplification, monomorphisms and presence of the null alleles, the final set of six markers useful for analysis on a population level was obtained. Analyzing genotypes of 124 individuals, we confirmed high genetic diversity of the species, suggested previously by studies, applying allozymes and mitochondrial DNA. Also, microsatellites revealed weakly pronounced genetic structure in a large scale (localities separated by a distance of 500 kilometers). However, we found some indications (signs of the Wahlund effect) that gene flow could be disturbed among sites in diverse environments (rural, suburban and urban). Our data suggested that invasion of the harlequin ladybird has not yet affected populations of the two-spot in such extent to alter genetic diversity or genetic structure.

The Black Grouse Tetrao tetrix populations in Europe have become fragmented and reduced in numbers. These processes affected genetic diversity of the species, altering patterns of gene flow and genetic structure. In Poland, the Black Grouse is one of the most endangered bird species, however very little is known about diversity of this native population, bordering on to the area of Europe in which the species maintains a continuous range. To complete the knowledge of genetic structure of European populations, we analysed microsatellite polymorphism in 97 native Polish birds, along with 39 introduced individuals, originating in Belarus and at a breeding centre in Poland, regarded as representing the gene pool from the north-eastern part of the country. The results confirmed that isolation of populations and recent decreases in their sizes have reduced genetic diversity in Poland's populations of the Black Grouse. The results also indicated the presence of two genetic groups in Poland, involving birds of the north-eastern part of the country and the Carpathians Foothills in a first group, and the population from the Sudetes (Izerskie and Karkonosze Mountains) in a second. We suggest that the conservation effort should treat these two groups as independent units. We also analysed a fragment of the mitochondrial Control Region (CRmtDNA). The comparisons of sequences obtained with data from other Eurasian populations indicate that populations of the Black Grouse in Poland should be included within a large Conservation Unit — the northern tetrix — which comprises birds from Eastern and Northern Europe.

We tested 24 microsatellite markers for the red-breasted flycatcher, Ficedula parva, using the primers for the PCR reaction described for other species from genus Ficedula. The amplification efficiency, specificity of the products, and polymorphism of cross-amplified microsatellites were determined based on the genotypes of 65 adult males from a natural population breeding in Bialowieza Forest (Eastern Poland). Based on obtained results, we identified 9 highly polymorphic microsatellites, consistently amplifying in majority of individuals. Among those markers between 4 and 26 alleles per locus (mean 15.4) were found and the observed heterozygosity ranged from 0.393 to 0.939. The majority of loci (except for Fhy350 and Fhy458) were in Hardy-Weinberg equilibrium. Accordingly, the values of FIS did not significantly differ from zero 0, except for the locus Fhy350. Introduction: While chronic cigarette smoking can lead to increased risk of stroke, the acute effects of smoking have not been established. We studied the changes in blood flow parameters in the major cerebral arteries caused by smoking one cigarette. Method: Using transcranial color-coded sonography (TCCS), we studied the anterior cerebral artery (ACA), middle cerebral artery (MCA), posterior cerebral artery (PCA), and the internal carotid artery (ICA) of 36 healthy male volunteers before and after they smoked one cigarette. Results: Blood flow velocity increased to a different degree in all but one of the arteries examined after participants smoked a single cigarette: The end diastolic velocity increased significantly by 7.8% in the PCA, 8% in the ACA, and 14.4% in the MCA. The peak systolic velocity increased significantly by 7.5% in the MCA. Blood flow velocity remained unchanged in the ICA only. Blood pressure and heart rate increased as did the flow velocity ratio for the MCA/ICA. The pulsatility index decreased after smoking from 0.92 ± 0.13 to 0.87 ± 0.14 in the MCA, 0.93 ± 0.15 to 0.87 ± 0.13 in the ACA, and 0.95 ± 0.17 to 0.89 ± 0.16 in the PCA. Conclusions: The results suggest that the direct effect of smoking on cerebral circulation includes peripheral vasodilatation with possible constriction of the main trunk of the basal cerebral arteries.

The serotine bat, Eptesicus serotinus is the most frequently rabies-infected (European bat lyssavirus 1-type, EBLV-1) bat species in Europe. To confirm Lyssavirus infection of this bat in Poland, we tested for the presence of rabies virus RNA from oropharyngeal swabs using RT-PCR. There was a 0.9% (two out of 212 individuals) level of infection within the overall population of serotine bats studied. However, an appreciation of the potential for pathogen transmission and disease risk requires an understanding of the dispersal of the primary host, and any large-scale geographic barriers that may impede gene flow. Thus, we also studied the patterns of bat dispersal via population genetics using nuclear (seven microsatellite loci) and mitochondrial (mtDNA control region) markers, examined in 12 subpopulations distributed across the country. The aims of this study were (i) to determine the genetic variability of the striped field mouse (Apodemus agrarius Pallas, 1771) population in the area of the Warsaw conurbation and the genetic differentiation among local populations of this species within the city, and (ii) to examine the influence of the degree of isolation on the appearance of such genetic structure. Comparison of the genetic structure of the urban population with that of populations inhabiting rural areas was intended to shed light on the depth of genetic changes in this species caused by life in an urban environment. Eight microsatellite loci were used for genotyping and these molecular analyses were successfully performed for 269 individuals. Genetic differentiation (FST) was much higher in the case of the urban population. STRUCTURE analysis indicated that this population was subdivided into several local populations, whereas for the rural locations, the highest likelihood was for a single genetic group (one genetically unstructured population). Moreover, in contrast to the rural population, we found no isolation by distance in the urban population. Despite clear subdivision of the urban population, the level of genetic variability was very similar in both analyzed groups. Our findings indicate the occurrence of an advanced process of differentiation of the urban population by microevolution, whereas populations from rural locations displayed very small and mainly insignificant genetic differentiation. Urban local populations, situated near the city borders or close to the natural ecological corridor (banks of the Vistula River), showed higher genetic variability and were less differentiated from populations inhabiting rural sites than from local populations in the city centre. These results provide support for the importance of ecological corridors in preserving the genetic variability of the urban striped field mouse population.

Recent advances in surgical endoscopy have made it possible to reach nearly the whole cranial base through a transnasal approach. These ‘expanded approaches’ lead to the frontal sinuses, the cribriform plate and planum sphenoidale, the suprasellar space, the clivus, odontoid and atlas. By pointing the endoscope laterally, the surgeon can explore structures in the coronal plane such as the cavernous sinuses, the pyramid and Meckel cave, the sphenopalatine and subtemporal fossae, and even the middle fossa and the orbit. The authors of this contribution use most of these approaches in their endoscopic skull base surgery. The purpose of this contribution is to review the hitherto established endoscopic approaches to the skull base and to illustrate them with photographs obtained during self-performed procedures and/or cadaver studies.

The capercaillie (Tetrao urogallus) is one of Poland's most endangered bird species, with an estimated population of 380–500 individuals in four isolated areas. To study these natural populations in Poland further, more than 900 non-invasive genetic samples were collected, along with samples from 59 birds representing large, continuous populations in Sweden and Russia; and from two centres in Poland breeding capercaillie. Microsatellite polymorphism at nine loci was then analysed to estimate within-population genetic diversity and genetic differentiation among populations. The results confirmed that isolation of populations and recent decreases in their sizes have reduced genetic diversity among capercaillie in Poland, with all the country’s natural populations found to be experiencing the genetic after-effects of demographic bottlenecks. The results of analyses of genetic differentiation and structure further suggest the presence of a 'lowland' cluster (encompassing birds of the Augustowska and Solska Primaeval Forests in Poland, and of Sweden and Russia), and a Carpathian cluster. Capercaillie from Sweden and Russia are also found to differ markedly. The Polish lowland populations seem more closely related to birds from Scandinavia. Our genetic analysis also indicates that the stocks at breeding centres are of a high genetic diversity effectively reflecting the origins of founder individuals, though identification of ancestry requires further study in the case of some birds. Overall, the results sustain the conclusion that the Polish populations of capercaillie from the Carpathians and the lowlands should be treated as independent Management Units (MUs). This is to say that the breeding lines associated with these two sources should be maintained separately at breeding centres. The high level of genetic differentiation of birds from the Solska Primaeval Forest suggests that this population should also be assigned the status of independent MU.

Despite a severe decrease in the number of hazel hens during the 20th century, nowadays this grouse species is rather common in the forests of Northeastern and Southern Poland. We used mitochondrial control region and microsatellite markers to examine the genetic variability of Polish populations of hazel hens. We used non-invasively collected faeces to estimate genetic variability within populations, genetic differentiation among populations as well as genetic differentiation between two regions inhabited by two different subspecies of hazel hens. Our results confirm the usefulness of DNA from faeces to obtain reliable information on the population genetics of hazel hens. We found a rather high level of genetic variability in the Polish population. Genetic variability was higher in birds from continuous forests in the South of the country than in birds from fragmented habitats in the Northeast. Genetic differentiation was higher among subpopulations from Northeastern Poland. Additionally, both classes of molecular markers suggested the presence of two distinct genetic groups of birds, corresponding to previously described subspecies. We conclude that the genetic variability of the Polish hazel hen population has been influenced by habitat fragmentation and the history of the population during its post-glacial colonization of Poland from different glacial refugia.

Plebejus argus is a model species for studying the biology, population ecology and genetics of butterflies. It is patchily distributed throughout most of its European range and considered to be sedentary. Habitats of the butterfly have to encompass two vital larval-resources, i.e. specific food plants and ants, since caterpillars are obligatorily myrmecophilous. The genetic structure of nine P. argus populations (85 individuals) was studied at an intermediate geographical scale (Eastern Poland, diameter of about 400 km) using two kinds of molecular markers i.e. COI (mtDNA) and EF-1α (nuclear gene). Both markers were highly variable with as many as 16 haplotypes and 39 alleles, respectively. Great genetic differentiation in the COI gene was detected (overall FST = 0.411, P < 0.001) but little genetic differentiation in the EF-1α gene (FST = 0.021, P < 0.001). The number of COI haplotypes (ranging from one to seven) and their distribution varied considerably among P. argus populations. The possibility that this heterogeneity was related to Wolbachia was excluded as this endoparasitic bacterium was not detected in samples from any of the populations studied. PCA and SAMOVA analyses divided the sampled populations into two or three groups, which could indicate different colonization routes. Moreover, the differences in genetic differentiation with respect to mtDNA and nuclear markers may suggest male-biased dispersal of P. argus at a larger scale. The hypothesis that females are philopatric is consistent with direct observations of the restricted colonization abilities of the butterfly, while the relatively homogeneous genetic structure revealed by previous allozyme studies in some areas might be explained by the possible higher mobility of males.

1. The endangered butterfly Phengaris alcon exists in two ecotypes (P. ‘alcon’ and P. ‘rebeli’), which inhabit contrasting biotopes (wet and warm/dry grasslands respectively) and use different larval food plants. The initially flower-bud-feeding caterpillars complete their development as social parasites of Myrmica ants, and the specificity of these relationships shows geographical variation. 2. We studied the genetic structure of 16 populations (365 individuals) of both ecotypes in eastern Europe, sampling P. ‘rebeli’ in two disjunct areas in Lithuania and southern Poland, and P. ‘alcon’ on Polish localities between them. We analysed the cytochrome c oxidase subunit I (COI) mitochondrial gene, the EF1-α nuclear gene and five polymorphic microsatellite loci. 3. All individuals shared an identical COI haplotype, which we hypothesise may be linked to a selective sweep associated with the presence of the Wolbachia B strain in all populations. 4. For nuclear markers, we did not find a clear pattern reflecting division into two putative ecotypes. However, ecotypes differed significantly in their genetic variability, i.e., the P. ‘rebeli’ ecotype was less polymorphic, and its populations were much more differentiated (FST: 0.632 for EF1-α and 0.504 for microsatellites) than the P. ‘alcon’ ecotype (0.177 and 0.082, respectively). 5. Our microsatellite data suggest that all populations of P. ‘alcon’ form a single clade but that P ‘rebeli’ can be split into either six or two clades. The former model would indicate many independent origins, especially in the mountainous areas of southern Poland. The latter, not mutually exclusive, grouping clearly reflects the use of different host ants.

Genetic variation of the globally threatened obligatorily myrmecophilous Large Blue butterfly Phengaris (Maculinea) arion (Lepidoptera) was studied, using six microsatellite markers, in a country where its decline is dramatic (Poland). Material was collected on 13 sites showing considerable ecological variation as far as biotope, larval food plant and host ants of the butterfly were concerned. Genetic variability, estimated in terms of number of alleles and heterozygosity, was the lowest in the most isolated populations. However on sites localized in areas where suitable biotopes were extensive and interconnected, P. arion still held relatively high genetic diversity. Pairwise FST values indicated small and moderate differentiation among samples (FST = 0.01–0.15), with the exceptions of two isolated localities (0.20). We did not find clear evidence of isolation by distance. The presence of four or five genetic clusters was indicated. Analysis of the membership of each individual to each cluster showed that the vast majority of individuals from three isolated populations were clustered in three separate genetic groups. The most distinct population was the one, which had been found to be specialized towards Myrmica lobicornis in previous studies. Individuals from the remaining populations could not be clustered in separate genetic groups, however some dominance of different clusters in geographical regions was observed. Some portion of the population’s genetic variability could be explained by geographical distribution, however the percentage of variation, explaining the differences between two main regions (S and NE Poland), was very low. We conclude that the main factor shaping the current genetic structure of P. arion in Poland is the recent isolation of populations related to habitat fragmentation but local ecological specializations may be also a potential factor. Therefore the necessity of activities aiming to halt the further reduction of genetic variability, as well as the monitoring of priority populations (e.g. those belonging to unique host races), should be emphasized in future action plans in Central Europe.
