Abstract

Background and importanceAtypical teratoid rhabdoid tumors (ATRTs) are typically aggressive pediatric tumors with a median survival of 11 months. Due to the paucity of cases in adults, the clinical behavior of these pathologies is not well understood. Here we present the case of a 41-year-old female patient with postoperative hyperprogression of a sellar ATRT and provide a detailed description of the molecular composition of this tumor, the protocol used to treat this patient, and the ultimate outcome of this patient.Clinical presentationThe patient is a 41-year-old woman who presented with headaches and double vision. MRI revealed a sellar/suprasellar mass with involvement of bilateral cavernous sinuses. Following the quick symptom progression, resection of the tumor with exploration of the bilateral cavernous sinuses was performed, with a final pathologic diagnosis of ATRT-MYC, a known subtype of ATRT. The tumor recurred within 1 month of surgery, attaining a size equivalent to its preoperative state. Postoperatively, the patient received craniospinal radiation and adjuvant chemotherapy with an excellent response but had a recurrence of the tumor in the brainstem 1 year after her diagnosis and died 13 months after presentation.DiscussionSellar ATRT in adults is an exceedingly rare entity. The detailed description of our case highlights the aggressiveness of these tumors and the utility of postoperative chemotherapy and radiation, but also the inevitable progression of these tumors along the craniospinal axis.ConclusionSellar ATRTs should be considered in the differential diagnosis of a sellar/suprasellar mass, especially in women in their 40s. Emphasis should be placed on accurate diagnosis and quick postoperative recovery with early initiation of adjuvant radiation and chemotherapy.

Abstract

Die Forderungen und Ansprüche des Handels und der Konsumenten nach Pflanzen ohne Besatz durch Schaderreger und/oder von ihnen induzierten Symptomen führen im Gartenbau zu häufig wiederholten Bekämpfungsmaßnahmen in engen Applikationsfolgen. Die Umsetzung der zum Schutz des Verbrauchers und der Umwelt geforderten Reduzierung der Risiken von zugelassenen Pflanzenschutzmitteln (PSM) führte dazu, dass in der Praxis oft keine ausreichend weite Wirkstoffpalette für einen Wirkstoffwechsel vorhanden ist. Selektion auf Resistenzen von Schaderregern und Minderwirkungen sind die Folge. Im Gartenbau wird zunehmend über Minderwirkungen von Insektiziden und Akariziden berichtet, ohne dass ein zweifelsfreier Nachweis der Ursachen erfolgte. In einem Konsortium aus Vertretern mehrerer öffentlicher und privater Einrichtungen wurden deshalb Methoden zur Bestimmung der Sensitivität/Resistenz etabliert, um das Wissen über Resistenzen gegen Insektizide und Akarizide im deutschen Gartenbau zu verbessern. Die von der IRAC (Insecticide Resistance Action Committee) empfohlenen und damit international anerkannten Methoden wurden ebenfalls berücksichtigt. In diesem Beitrag wird berichtet, wieviel Wirkstoff bei verschiedenen, auch international validierten Resistenztests im Labor (Biotests) auf den behandelten Oberflächen tatsächlich abgelagert wird. Die mit der Kryo-Rasterelektronenmikroskopie (Kryo-REM) sichtbar gemachte Diversität der Mikromorphologie von Pflanzenoberflächen gibt Anlass das zu überprüfen. Am Beispiel von Pflanzenschutzmittelapplikationen in Rosenkulturen wird gezeigt, dass auch die Evaluation der Wirkstoffbeläge in der gartenbaulichen Praxis ein wesentlicher Baustein für die Beurteilung der Sensitivität der Schaderreger ist. Die Analyse der Wirkstoffe ermöglicht die Validierung der Applikation von PSM in Vorbereitung der Biotests und die Bewertung der Wirkungen von PSM-Anwendungen im Labor und in der Praxis. Ein zweiter Beitrag, der sich mit der Sensitivität ausgewählter Schaderreger des Gartenbaus gegenüber verschiedenen PSM beschäftigt, wird an anderer Stelle veröffentlicht werden.

3. Giant Faraday rotation in atomically thin semiconductors

Abstract

Abstract Faraday rotation is a fundamental effect in the magneto-optical response of solids, liquids and gases. Materials with a large Verdet constant find applications in optical modulators, sensors and non-reciprocal devices, such as optical isolators. Here, we demonstrate that the plane of polarization of light exhibits a giant Faraday rotation of several degrees around the A exciton transition in hBN-encapsulated monolayers of WSe2 and MoSe2 under moderate magnetic fields. This results in the highest known Verdet constant of -1.9 × 107 deg T−1 cm−1 for any material in the visible regime. Additionally, interlayer excitons in hBN-encapsulated bilayer MoS2 exhibit a large Verdet constant (V IL ≈ +2 × 105 deg T−1 cm−2) of opposite sign compared to A excitons in monolayers. The giant Faraday rotation is due to the giant oscillator strength and high g-factor of the excitons in atomically thin semiconducting transition metal dichalcogenides. We deduce the complete in-plane complex dielectric tensor of hBN-encapsulated WSe2 and MoSe2 monolayers, which is vital for the prediction of Kerr, Faraday and magneto-circular dichroism spectra of 2D heterostructures. Our results pose a crucial advance in the potential usage of two-dimensional materials in ultrathin optical polarization devices.

Abstract

Summary: Background: While vaccines have established utility against COVID-19, phase 3 efficacy studies have generally not comprehensively evaluated protection provided by previous infection or hybrid immunity (previous infection plus vaccination). Individual patient data from US government-supported harmonized vaccine trials provide an unprecedented sample population to address this issue. We characterized the protective efficacy of previous SARS-CoV-2 infection and hybrid immunity against COVID-19 early in the pandemic over three-to six-month follow-up and compared with vaccine-associated protection. Methods: In this post-hoc cross-protocol analysis of the Moderna, AstraZeneca, Janssen, and Novavax COVID-19 vaccine clinical trials, we allocated participants into four groups based on previous-infection status at enrolment and treatment: no previous infection/placebo; previous infection/placebo; no previous infection/vaccine; and previous infection/vaccine. The main outcome was RT-PCR-confirmed COVID-19 >7–15 days (per original protocols) after final study injection. We calculated crude and adjusted efficacy measures. Findings: Previous infection/placebo participants had a 92% decreased risk of future COVID-19 compared to no previous infection/placebo participants (overall hazard ratio [HR] ratio: 0.08; 95% CI: 0.05–0.13). Among single-dose Janssen participants, hybrid immunity conferred greater protection than vaccine alone (HR: 0.03; 95% CI: 0.01–0.10). Too few infections were observed to draw statistical inferences comparing hybrid immunity to vaccine alone for other trials. Vaccination, previous infection, and hybrid immunity all provided near-complete protection against severe disease. Interpretation: Previous infection, any hybrid immunity, and two-dose vaccination all provided substantial protection against symptomatic and severe COVID-19 through the early Delta period. Thus, as a surrogate for natural infection, vaccination remains the safest approach to protection. Funding: National Institutes of Health.

Abstract

Midfrontal theta increases during scenarios when conflicts are successfully resolved. Often considered a generic signal of cognitive control, its temporal nature has hardly been investigated. Using advanced spatiotemporal techniques, we uncover that midfrontal theta occurs as a transient oscillation or “event” at single trials with their timing reflecting computationally distinct modes. Single-trial analyses of electrophysiological data from participants performing the Flanker (N = 24) and Simon task (N = 15) were used to probe the relationship between theta and metrics of stimulus-response conflict. We specifically investigated “partial errors”, in which a small burst of muscle activity in the incorrect response effector occurred, quickly followed by a correction. We found that transient theta events in single trials could be categorized into two distinct theta modes based on their relative timing to different task events. Theta events from the first mode occurred briefly after the task stimulus and might reflect conflict-related processing of the stimulus. In contrast, theta events from the second mode were more likely to occur around the time partial errors were committed, suggesting they were elicited by a potential upcoming error. Importantly, in trials in which a full error was committed, this “error-related theta” occurred too late with respect to the onset of the erroneous muscle response, supporting the role of theta also in error correction. We conclude that different modes of transient midfrontal theta can be adopted in single trials not only to process stimulus-response conflict, but also to correct erroneous responses.

Abstract

Heat shock protein 70 (Hsp70) is frequently overexpressed in many different tumor types. However, Hsp70 has also been shown to be selectively presented on the plasma membrane of tumor cells, but not normal cells, and this membrane form of Hsp70 (mHsp70) could be considered a universal tumor biomarker. Since viable, mHsp70-positive tumor cells actively release Hsp70 in lipid micro-vesicles, we investigated the utility of Hsp70 in circulation as a universal tumor biomarker and its potential as an early predictive marker of therapeutic failure. We have also evaluated mHsp70 as a target for the isolation and enumeration of circulating tumor cells (CTCs) in patients with different tumor entities. Circulating vesicular Hsp70 levels were measured in the peripheral blood of tumor patients with the compHsp70 ELISA. CTCs were isolated using cmHsp70.1 and EpCAM monoclonal antibody (mAb)-based bead approaches and characterized by immunohistochemistry using cytokeratin and CD45-specific antibodies. In two out of 35 patients exhibiting therapeutic failure two years after initial diagnosis of non-metastatic breast cancer, progressively increasing levels of circulating Hsp70 had already been observed during therapy, whereas levels in patients without subsequent recurrence remained unaltered. With regards to CTC isolation from patients with different tumors, an Hsp70 mAb-based selection system appears superior to an EpCAM mAb-based approach. Extracellular and mHsp70 can therefore serve as a predictive biomarker for therapeutic failure in early-stage tumors and as a target for the isolation of CTCs in various tumor diseases.

Abstract

Action-stopping in humans involves bursts of beta oscillations in prefrontal-basal ganglia regions. To determine the functional role of these beta bursts we took advantage of the Race Model framework describing action-stopping. We incorporated beta bursts in three race model variants, each implementing a different functional contribution of beta to action-stopping. In these variants, we hypothesized that a transient increase in beta could (1) modulate decision thresholds, (2) change stop accumulation rates, or (3) promote the interaction between the Stop and the Go process. We then tested the model predictions using EEG recordings in humans performing a Stop-signal task. We found that the model variant in which beta increased decision thresholds for a brief period of time best explained the empirical data. The model parameters fitted to the empirical data indicated that beta bursts involve a stronger decision threshold modulation for the Go process than for the Stop process. This suggests that prefrontal beta influences stopping by temporarily holding the response from execution. Our study further suggests that human action-stopping could be multi-staged with the beta acting as a pause, increasing the response threshold for the Stop process to modulate behavior successfully. Overall, our approach of introducing transient oscillations into the race model and testing against human electrophysiological data provides a novel account of the puzzle of prefrontal beta in executive control.

Abstract

For the paradigmatic three-disk scattering system, we confirm a recent conjecture for open chaotic systems, which claims that resonance states are composed of two factors. In particular, we demonstrate that one factor is given by universal exponentially distributed intensity fluctuations. The other factor, supposed to be a classical density depending on the lifetime of the resonance state, is found to be very well described by a classical construction. Furthermore, ray-segment scars, recently observed in dielectric cavities, dominate every resonance state at small wavelengths also in the three-disk scattering system. We introduce a new numerical method for computing resonances, which allows for going much further into the semiclassical limit. As a consequence we are able to confirm the fractal Weyl law over a correspondingly large range.

Abstract

Cortico-basal ganglia beta oscillations (13–30 Hz) are assumed to be involved in motor impairments in Parkinson’s Disease (PD), especially in bradykinesia and rigidity. Various studies have utilized the unilateral 6-hydroxydopamine (6-OHDA) rat PD model to further investigate PD and test novel treatments. However, a detailed behavioral and electrophysiological characterization of the model, including analyses of popular PD treatments such as DBS, has not been documented in the literature. We hence challenged the 6-OHDA rat hemi-PD model with a series of experiments (i.e., cylinder test, open field test, and rotarod test) aimed at assessing the motor impairments, analyzing the effects of Deep Brain Stimulation (DBS), and identifying under which conditions excessive beta oscillations occur. We found that 6-OHDA hemi-PD rats presented an impaired performance in all experiments compared to the sham group, and DBS could improve their overall performance. Across all the experiments and behaviors, the power in the high beta band was observed to be an important biomarker for PD as it showed differences between healthy and lesioned hemispheres and between 6-OHDA-lesioned and sham rats. This all shows that the 6-OHDA hemi-PD model accurately represents many of the motor and electrophysiological symptoms of PD and makes it a useful tool for the pre-clinical testing of new treatments when low β (13–21 Hz) and high β (21–30 Hz) frequency bands are considered separately.

ZusammenfassungDer wichtigste Grundsatz der Integrativen Medizin ist die Verbindung der konventionellen ärztlichen Medizin und der ärztlichen Komplementärmedizin zu einem sinnvollen Gesamtkonzept. Ziel ist es, die individuell beste Therapie für die Patient*innen zu finden und Nebenwirkungen so weit wie möglich zu reduzieren. Entscheidender Faktor sind hier auch explizit die Patient*innen, die nach ausführlicher Aufklärung über alle zur Verfügung stehenden Therapieoptionen in die Lage versetzt werden sollen, aktiv und mitverantwortlich über ein medizinisches Gesamtkonzept für ihre Behandlung mitzuentscheiden. Der Beitrag stellt mit dem Krankenhaus für Naturheilweisen (KfN) in München und der Liste Integrative Medizin zwei Beispiele vor, die sich für die Verbindung von konventioneller und Komplementärmedizin in der Öffentlichkeit und in der Praxis starkmachen.

Abstract

Die Verteilung eines Pflanzenschutzmittels im Pflanzenbestand ist ganz wesentlich von der Verfügbarkeit und Verwendung einer geeigneten Applikationstechnik abhängig. Nur eine gute Verteilung gewährleistet den sicheren Kontakt der Zielorganismen mit dem Wirkstoff. Das gilt insbesondere für Schädlinge die sich auf der Blattunterseite aufhalten. Die Auswirkung des Düsentyps sowie die Verwendung eines Netzmittels auf die Wirkstoffanlagerung an die Blattober- und -unterseite und die resultierende Wirksamkeit sind Gegenstand dieser Untersuchungen. Für die Ermittlung der Wirkstoffanlagerung wurde ein künstliches Blatt eingesetzt, bei dem die daran befestigten Glasfaserfilter als Blattober- bzw. -unterseite getrennt analysiert werden konnten. Die Verwendung einer Hohlkegeldüse erhöhte im Vergleich zu einer Flachstrahldüse die Anlagerung auf der Oberseite der künstlichen Blätter in schmalblättrigen Brachyscome multifida. Die Doppelflachstrahldüse unterschied sich nicht von den anderen Düsentypen. In breitblättrigen Pelargonium-Zonale-Hybriden und Fuchsia-Hybriden wurden keine Unterschiede zwischen den Düsentypen festgestellt. Die Wirkstoffanlagerung auf der Blattunterseite der künstlichen Blätter an der Fuchsia-Hybride und die Wirksamkeit auf Trialeurodes vaporariorum war unabhängig vom Düsentyp gering.

Abstract

Flexible behavior requires restraint of actions that are no longer appropriate. This behavioral inhibition critically relies on frontal cortex - basal ganglia circuits. Within the basal ganglia, the globus pallidus pars externa (GPe) has been hypothesized to mediate selective proactive inhibition: being prepared to stop a specific action, if needed. Here we investigate population dynamics of rat GPe neurons during preparation-to-stop, stopping, and going. Rats selectively engaged proactive inhibition towards specific actions, as shown by slowed reaction times (RTs). Under proactive inhibition, GPe population activity occupied state-space locations farther from the trajectory followed during normal movement initiation. Furthermore, the state-space locations were predictive of distinct types of errors: failures-to-stop, failures-to-go, and incorrect choices. Slowed RTs on correct proactive trials reflected starting bias towards the alternative action, which was overcome before progressing towards action initiation. Our results demonstrate that rats can exert cognitive control via strategic adjustments to their GPe network state.
