Abstract

Aerosols are being collected and analyzed for trace elements in two sites in Amazonia since January 2008. On eof the site, Manaus is located in a very pristine area in Central Amazonia. The site is nt affected directly by any urban plume for thousands of kilometers. A second site is located in Porto Velho, in a region with heavy land use change and deforestation. Optical properties (light scattering ad absorption) are also being measured in order to study the climatic impact of aerosols. It was observed a clear seasonal pattern for both sites, with higher concentrations in the dry season. But the difference in seasonal concentrations observed for Porto Velho is much larger due to stronger anthropogenic influences. In Manaus during the wet season, very low concentrations of heavy metals, maybe the smallest measured in continental regions are reported. Positive Matrix Factorization (PMF) was used to separate the different aerosol components. In general, for fine and coarse mode and wet and dry season, 3 aerosol components could be observed: 1) Natural biogenic aerosol; 2) biomass burning component; 3) Soil dust both locally and long range transported Sahara dust

Abstract

The oxidation of biogenic volatile organic compounds (BVOCs) gives a range of products, from semi-volatile to extremely low-volatility compounds. To treat the interaction of these secondary organic vapours with the particle phase, global aerosol microphysics models generally use either a thermodynamic partitioning approach (assuming instant equilibrium between semi-volatile oxidation products and the particle phase) or a kinetic approach (accounting for the size dependence of condensation). We show that model treatment of the partitioning of biogenic organic vapours into the particle phase, and consequent distribution of material across the size distribution, controls the magnitude of the first aerosol indirect effect (AIE) due to biogenic secondary organic aerosol (SOA). With a kinetic partitioning approach, SOA is distributed according to the existing condensation sink, enhancing the growth of the smallest particles, i.e. those in the nucleation mode. This process tends to increase cloud droplet number concentrations in the presence of biogenic SOA. By contrast, an approach that distributes SOA according to pre-existing organic mass restricts the growth of the smallest particles, limiting the number that are able to form cloud droplets. With an organically mediated new particle formation mechanism, applying a mass-based rather than a kinetic approach to partitioning reduces our calculated global mean AIE due to biogenic SOA by 24 %. Our results suggest that the mechanisms driving organic partitioning need to be fully understood in order to accurately describe the climatic effects of SOA.

Abstract

B10.A mice develop experimental autoimmune uveoretinitis after active immunization with the interphotoreceptor retinoid-binding protein (IRBP). CD4+ T cells play an important role in the development of the disease. In this study we have isolated and characterized a CD4+ T cell line and a T cell clone that induce experimental autoimmune uveoretinitis when transferred into naive B10.A mice. The cell line was isolated from draining lymph nodes of IRBP-immunized animals by repeated cycles of IRBP stimulation. The line was shown to be pathogenic after 4 rounds of in vitro stimulation with IRBP at 5 x 10(6) cells/mouse. A T cell clone derived from this line by limiting dilution was shown to be pathogenic when the same number of cells was injected; incidence and severity of disease, however, were much lower. After 16 rounds of IRBP-specific stimulation the cell line was pathogenic at 10(5) cells/mouse. Analysis of the V beta repertoire revealed that at this point the line was mostly composed of V beta 8.2- and V beta 6-positive cells (> 80% of the population). The uveitogenic clone expressed V beta 8.2. Both the T cell line and the clone elaborated an unrestricted lymphokine profile in vitro. However, when these cells were adoptively transferred into naive recipients, mRNA isolated from the uveitic retina showed only Th1 type cytokines. These data help to characterize the nature of pathogenic cells involved in ocular autoimmunity.

Abstract

The HadGEM2 Earth System climate model was used to assess the impact of biomass burning on surface ozone concentrations over the Amazon forest and its impact on vegetation. Simulated surface ozone concentration is evaluated against observations taken at two sites in the Brazilian Amazon forest. The model is able to reproduce the observed diurnal cycle of surface ozone mixing ratio at the two sites, but overestimates the magnitude of the monthly averaged hourly measurements by 5-15ppb for each available month at one of the sites. We vary biomass burning emissions over South America by ±20, 40, 60, 80 and 100% to quantify the modelled impact of biomass burning on surface ozone concentrations and ozone damage on vegetation productivity over the Amazon forest. Decreasing South American biomass burning emissions by 100% (i.e. to zero) reduces surface ozone concentrations and suggests a 15% increase in monthly mean net primary productivity averaged over the Amazon forest, with local increases up to 60%: this gives us an estimate of the effect of current biomass burning on plant productivity. When biomass burning emissions are increased by 100%, our model simulates a maximum impact of 10% reduction in monthly mean net plant productivity averaged over the Amazon forest, with local peaks of 50-60% reduction for the months of intense fire activity.
