- Abstract
Debido al cambio climático, en los últimos años se han observado cambios en la composición de la uva y un incremento en el contenido en azúcares y del pH, produciéndose vinos con mayor graduación alcohólica y con menor acidez. Una estrategia biotecnológica prometedora para la reducción del contenido inicial de glucosa en los mostos es el empleo de las enzimas glucosa oxidasa (GOX) y catalasa (CAT). Su uso permite disminuir el grado alcohólico final, sin embargo, durante este tratamiento se ha observado un incremento notable en el contenido de ácido glucónico. Por ello, la capacidad que poseen algunas cepas de la levadura Schizosaccharomyces pombe para reducir la concentración de ácido glucónico es de gran interés enológico. En el presente trabajo, se evalúa el uso de diferentes cepas de Schizosaccharomyces spp. en la fermentación de un mosto Verdejo tratado enzimáticamente con GOX/CAT. Los resultados obtenidos sugieren que el uso de un tratamiento enzimático junto con el empleo de Schizosaccharomyces spp. puede ser eficaz para la reducción del contenido alcohólico final de los vinos., Grado en Enología

Natural events present multiple types of sensory cues, each detected by a specialized sensory modality. Combining information from several modalities is essential for the selection of appropriate actions. Key to understanding multimodal computations is determining the structural patterns of multimodal convergence and how these patterns contribute to behaviour. Modalities could converge early, late or at multiple levels in the sensory processing hierarchy. Here we show that combining mechanosensory and nociceptive cues synergistically enhances the selection of the fastest mode of escape locomotion in Drosophila larvae. In an electron microscopy volume that spans the entire insect nervous system, we reconstructed the multisensory circuit supporting the synergy, spanning multiple levels of the sensory processing hierarchy. The wiring diagram revealed a complex multilevel multimodal convergence architecture. Using behavioural and physiological studies, we identified functionally connected circuit nodes that trigger the fastest locomotor mode, and others that facilitate it, and we provide evidence that multiple levels of multimodal integration contribute to escape mode selection. We propose that the multilevel multimodal convergence architecture may be a general feature of multisensory circuits enabling complex input-output functions and selective tuning to ecologically relevant combinations of cues.

Objectives: CARB-ES-19 is a comprehensive, multicenter, nationwide study integrating whole-genome sequencing (WGS) in the surveillance of carbapenemase-producing K. pneumoniae (CP-Kpn) and E. coli (CP-Eco) to determine their incidence, geographical distribution, phylogeny, and resistance mechanisms in Spain. Methods: In total, 71 hospitals, representing all 50 Spanish provinces, collected the first 10 isolates per hospital (February to May 2019); CPE isolates were first identified according to EUCAST (meropenem MIC > 0.12 mg/L with immunochromatography, colorimetric tests, carbapenem inactivation, or carbapenem hydrolysis with MALDI-TOF). Prevalence and incidence were calculated according to population denominators. Antibiotic susceptibility testing was performed using the microdilution method (EUCAST). All 403 isolates collected were sequenced for high-resolution single-nucleotide polymorphism (SNP) typing, core genome multilocus sequence typing (cgMLST), and resistome analysis. Results: In total, 377 (93.5%) CP-Kpn and 26 (6.5%) CP-Eco isolates were collected from 62 (87.3%) hospitals in 46 (92%) provinces. CP-Kpn was more prevalent in the blood (5.8%, 50/853) than in the urine (1.4%, 201/14,464). The cumulative incidence for both CP-Kpn and CP-Eco was 0.05 per 100 admitted patients. The main carbapenemase genes identified in CP-Kpn were blaOXA−48 (263/377), blaKPC−3 (62/377), blaVIM−1 (28/377), and blaNDM−1 (12/377). All isolates were susceptible to at least two antibiotics. Interregional dissemination of eight high-risk CP-Kpn clones was detected, mainly ST307/OXA-48 (16.4%), ST11/OXA-48 (16.4%), and ST512-ST258/KPC (13.8%). ST512/KPC and ST15/OXA-48 were the most frequent bacteremia-causative clones. The average number of acquired resistance genes was higher in CP-Kpn (7.9) than in CP-Eco (5.5). Conclusion: This study serves as a first step toward WGS integration in the surveillance of carbapenemase-producing Enterobacterales in Spain. We detected important epidemiological changes, including increased CP-Kpn and CP-Eco prevalence and incidence compared to previous studies, wide interregional dissemination, and increased dissemination of high-risk clones, such as ST307/OXA-48 and ST512/KPC-3. - Abstract
[Introduction] There is no consensus regarding optimal duration of antibiotic therapy for Pseudomonas aeruginosa bacteremia. We aimed to evaluate the impact of short antibiotic course., [Methods] We present a retrospective multicenter study including patients with P. aeruginosa bacteremia during 2009–2015. We evaluated outcomes of patients treated with short (6–10 days) versus long (11–15 days) antibiotic courses. The primary outcome was a composite of 30-day mortality or bacteremia recurrence and/or persistence. Univariate and inverse probability treatment-weighted (IPTW) adjusted multivariate analysis for the primary outcome was performed. To avoid immortal time bias, the landmark method was used., [Results] We included 657 patients; 273 received a short antibiotic course and 384 a long course. There was no significant difference in baseline characteristics of patients. The composite primary outcome occurred in 61/384 patients in the long-treatment group (16%) versus 32/273 in the short-treatment group (12%) (p = 0.131). Mortality accounted for 41/384 (11%) versus 25/273 (9%) of cases, respectively. Length of hospital stay was significantly shorter in the short group [median 13 days, interquartile range (IQR) 9–21 days, versus median 15 days, IQR 11–26 days, p = 0.002]. Ten patients in the long group discontinued antibiotic therapy owing to adverse events, compared with none in the short group. On univariate and multivariate analyses, duration of therapy was not associated with the primary outcome., [Conclusions] In this retrospective study, 6–10 days of antibiotic course for P. aeruginosa bacteremia were as effective as longer courses in terms of survival and recurrence. Shorter therapy was associated with reduced length of stay and less drug discontinuation.

- Abstract
An analysis of perceived altruism was conducted within the framework of Kelley's (1973) attributional model. In a 2 by 2 factorial design, the consistency and distinctiveness of a donor's prosocial history were manipulated. Subjects were provided with written scenarios depicting the work of a voluntary welfare organization aiding the elderly. Subsequent evaluations of the donor-actors on the altruism dimension revealed that there is a direct relationship of consistency and an inverse relationship of distinctiveness with the attribution of altruism. Esta investigación está diseñada para probar qué tan efectivos son estos videos en la clase de inglés, principalmente en el desarrollo de la habilidad de hablar realizado en pequeños grupos. Este estudio involucró a 18 estudiantes de 10th calificación. El estudio investigó si la aplicación de videoclips con grupos pequeños a lo largo con actividades individuales de enseñanza-aprendizaje mejoraron las habilidades orales de los jóvenes de inglés como lengua extranjera los alumnos más. Los resultados sugirieron que enseñar a hablar a estudiantes de EFL con videoclips usando Las técnicas y actividades para grupos reducidos podrían ser una de las mejores alternativas para mejorar representaciones orales de jóvenes estudiantes. El estudio mostró que el uso del aprendizaje cooperativo mejoró el habla de los estudiantes. habilidades. Los alumnos mostraron mejora en algunos aspectos como la pronunciación, dominio del vocabulario y confianza.

Introduction: Healthcare-associated (HCA) infections represent a growing public health problem. The aim of this study was to compare community-onset healthcare associated (CO-HCA) bacteremic urinary tract infections (BUTI) and hospital-acquired (HA)-BUTI with special focus on multidrug resistances (MDR) and outcomes. Methods: ITUBRAS-project is a prospective multicenter cohort study of patients with HCA-BUTI. All consecutive hospitalized adult patients with CO-HCA-BUTI or HA-BUTI episode were included in the study. Exclusion criteria were: patients \ 18 years old, non-hospitalized patients, bacteremia from another source or primary bacteremia, non-healthcare related infections and infections caused by unusual pathogens of the urinary tract. Th main outcome variable was 30-day all-cause mortality with day 1 as the first day of positive blood culture. Logistic regression was used to analyze factors associated with clinical cure at hospital discharge and with receiving inappropriate initial antibiotic treatment. Cox regression was used to evaluate 30-day all-cause mortality. Results: Four hundred forty-three episodes were included, 223 CO-HCA-BUTI. Patients with CO-HCA-BUTI were older (p \ 0.001) and had more underlying diseases (p = 0.029) than those with HA-BUTI. The severity of the acute illness (Pitt score) was also higher in CO-HCABUTI (p = 0.026). Overall, a very high rate of MDR profiles (271/443, 61.2%) was observed, with no statistical differences between groups. In multivariable analysis, inadequate empirical treatment was associated with MDR profile (aOR 3.35; 95% CI 1.77?6.35), Pseudomonas aeruginosa (aOR 2.86; 95% CI 1.27?6.44) and Charlson index (aOR 1.11; 95% CI 1.01?1.23). Mortality was not associated with the site of acquisition of the infection or the presence of MDR profile. However, in the logistic regression analyses patients with CO-HCA-BUTI (aOR 0.61; 95% CI 0.40?0.93) were less likely to present clinical cure. Conclusion: The rate of MDR infections was worryingly high in our study. No differences in MDR rates were found between CO-HCA-BUTI and HA-BUTI, in the probability of receiving inappropriate empirical treatment or in 30-day mortality. Surgical antimicrobial prophylaxis (SAP) is important for the prevention of prosthetic joint infections (PJIs) and must be effective against the microorganisms most likely to contaminate the surgical site. Our aim was to compare different SAP regimens (cefazolin, cefuroxime, or vancomycin, alone or combined with gentamicin) in patients undergoing total knee (TKA) and hip (THA) arthroplasty. In this preclinical exploratory analysis, we analyzed the results of intraoperative sample cultures, the ratio of plasma antibiotic levels to the minimum inhibitory concentrations (MICs) for bacteria isolated at the surgical wound and ATCC strains, and serum bactericidal titers (SBT) against the same microorganisms. A total of 132 surgical procedures (68 TKA, 64 THA) in 128 patients were included. Cultures were positive in 57 (43.2%) procedures (mostly for coagulase-negative staphylococci and Cutibacterium spp.), the rate was lower in the group of patients receiving combination SAP (adjusted OR 0.475, CI95% 0.229&ndash, 0.987). The SAP regimens evaluated achieved plasma levels above the MICs in almost all of intraoperative isolates (93/94, 98.9%) and showed bactericidal activity against all of them (SBT range 1:8&ndash, 1:1024), although SBTs were higher in patients receiving cefazolin and gentamicin-containing regimens. The potential clinical relevance of these findings in the prevention of PJIs remains to be determined.

Globant, empresa Argentina de desarrollo tecnológico con más de 1,000 proyectos a nivel mundial e internos para soporta el crecimiento que tiene cada año con sus empleados de planta. El proyecto que estamos analizando es BW4, su principal objetivo es integrar las bases de datos de 3 áreas (M&A, Accounting y Planning) que tiene Globant a nivel interno con el fin de centralizar la información y obtener las métricas necesarias para el seguimiento estratégico de los proyectos de la compañía. Dentro de este proyecto, se está implementando la metodología ágil Scrum, para el mejoramiento y desarrollo, uno de los principales problemas que se encontró fue que no se tener un aprovechamiento óptimo de los recursos humanos que se poseía. La investigación de la metodología hace referencia al uso de la misma teniendo en cuenta que el proyecto se encuentra en ejecución en su séptimo mes. Para el análisis y evaluación de la metodología Scrum frente al proyecto y el impacto se obtuvieron las siguientes conclusiones: • Al principio del proyecto se obtuvo poco conocimiento de la metodología y de las tecnologías frente al proyecto, esto se ve reflejado en la velocidad (Velocity). • La planning que se realiza en cada Sprint no se termina de comprometer ya que se evidencia que durante el Sprint pueden llegar nuevos requerimientos o prioridades que no se tenían en el Sprint. • El trabajo de equipo y el acompañamiento por parte del Scrum Master es uno de los puntos más fuertes según reveló las encuestas y estudio realizado. Otro punto fuerte del equipo es la maduración que se va obteniendo durante el Sprint, las métricas de puntos alcanzados vs los puntos comprometidos muestran una curva de aprendizaje rápida en la metodología y proyecto. • Una debilidad que se evidenció en la encuesta realizada es que el PO no tiene total autonomía por el producto, ya que la organización en ocasiones no respeta la autonomía brindada al inicio del proyecto, esto puede ocasionar retrasos en la claridad del producto y por ende perdida de presupuesto. • Se evidencia una oportunidad de mejora en el detalle de las historias de usuario que el equipo debe trabajar cada Sprint, este es el punto más débil de acuerdo al análisis realizado por el equipo de trabajo. En conclusión, de acuerdo a nuestro análisis las personas que están trabajando dentro del proyecto tienen una curva de aprendizaje rápida lo que hace un rápido crecimiento frente al valor entregado, por otro lado, se debe analizar y mejorar las prácticas de la metodología para mejorar la velocidad de entrega de las historias de usuario por Sprint. Se recomienda que el PO, Scrum Master y equipo trabajen en la autonomía del producto para obtener claridad dentro del backlog y así aumentar la productividad de cada recurso. Globant, an Argentine technology development company with more than 1000 projects worldwide and internally to support the growth it has every year. The project that we are analyzing is BW4, its main objective is to integrate the databases of 3 areas (M&A, Accounting and Planning) that Globant has internally in order to centralize the information and obtain the necessary metrics for the strategic monitoring of the company's projects. Within this project, the agile Scrum methodology is implemented, one of the main problems in the analysis of the methodology is that you don’t have an optimal use of the human resources that you have. The research of the methodology refers to the use of it, taking into account that the project is in execution in its seventh month. For the analysis and evaluation of the Scrum methodology regarding the project and the impact, the following conclusions were obtained: • At the beginning of the project, little knowledge of the methodology and technologies was obtained, this is reflected in the speed (Velocity) during the first sprints. • The planning that is carried out for each Sprint is not fully compromised since it is evident that during the Sprint new requirements or priorities may arrive that were not had in the Sprint. • The teamwork and the commitment by the Scrum Master is one of the strongest points as revealed by the surveys and study carried out. • Other strong point of the team is the maturation that is being obtained during the Sprints, the metrics of points achieved vs points committed shows a rapid learning curve in the methodology and project. A weakness that was evidenced in the survey carried out is that the PO does not have total autonomy of the product, since the organization sometimes does not respect the autonomy provided at the beginning of the project, this can cause delays in the clarity of the product and therefore loss budget. • An opportunity for improvement is in the detail of the user stories that the team must work on each Sprint, this is the weakest point according to the analysis carried out by the work team. In conclusion, according to our analysis, the people that are working on the project have a rapid learning curve, which makes quick growth compared to the value delivered, on the other hand, the practices of the methodology must be analyzed and improved in order to get better speed of delivery of user stories for each Sprint. 44. Combination versus monotherapy as definitive treatment for Pseudomonas aeruginosa bacteraemia: a multicentre retrospective observational cohort study

Objectives: CARB-ES-19 is a comprehensive, multicenter, nationwide study integrating whole-genome sequencing (WGS) in the surveillance of carbapenemase-producing K. pneumoniae (CP-Kpn) and E. coli (CP-Eco) to determine their incidence, geographical distribution, phylogeny, and resistance mechanisms in Spain. Methods: In total, 71 hospitals, representing all 50 Spanish provinces, collected the first 10 isolates per hospital (February to May 2019); CPE isolates were first identified according to EUCAST (meropenem MIC > 0.12 mg/L with immunochromatography, colorimetric tests, carbapenem inactivation, or carbapenem hydrolysis with MALDI-TOF). Prevalence and incidence were calculated according to population denominators. Antibiotic susceptibility testing was performed using the microdilution method (EUCAST). All 403 isolates collected were sequenced for high-resolution single-nucleotide polymorphism (SNP) typing, core genome multilocus sequence typing (cgMLST), and resistome analysis. Results: In total, 377 (93.5%) CP-Kpn and 26 (6.5%) CP-Eco isolates were collected from 62 (87.3%) hospitals in 46 (92%) provinces. CP-Kpn was more prevalent in the blood (5.8%, 50/853) than in the urine (1.4%, 201/14,464). The cumulative incidence for both CP-Kpn and CP-Eco was 0.05 per 100 admitted patients. OXA–48 (263/377), blaKPC–3 (62/377), blaVIM–1 (28/377), and blaNDM–1 (12/377). All isolates were susceptible to at least two antibiotics. Interregional dissemination of eight high-risk CP-Kpn clones was detected, mainly ST307/OXA-48 (16.4%), ST11/OXA-48 (16.4%), and ST512-ST258/KPC (13.8%). ST512/KPC and ST15/OXA-48 were the most frequent bacteremia-causative clones. The average number of acquired resistance genes was higher in CP-Kpn (7.9) than in CP-Eco (5.5). Conclusion: This study serves as a first step toward WGS integration in the surveillance of carbapenemase-producing Enterobacterales in Spain. We detected important epidemiological changes, including increased CP-Kpn and CP-Eco prevalence and incidence compared to previous studies, wide interregional dissemination, and increased dissemination of high-risk clones, such as ST307/OXA-48 and ST512/KPC-3.
