15. Wastewater, reclaimed water, and seawater utilization in the production of microalgae-based fuels

Abstract

Biofuel production from microalgae is affected by the cost of the cultivation of microalgal biomass. The cultivation of microalgae for third-generation biofuels in wastewater, reclaimed water, and seawater could be a cost-effective way to obtain microalgal biomass. This option would serve to reduce costs and treat wastewaters and reclaimed water at the same time. Autotrophic and mixotrophic cultivation make use of wastewater and nutrients, using light and CO2 to obtain high percentages of nitrogen and phosphorous removal. The advantage of microalgae grown in this way is twofold: wastewater treatment and microalgal production without the addition of water and nutrients. Biofuels from microalgae grown in this way would replace fossil fuels with renewable and carbon-neutral fuels in a sustainable way and contribute to the circular economy of water resources. In this review, an updated evaluation of microalgal cultivation in wastewater, reclaimed water, and seawater together with biofuel generation will be shown.

39. A Lux-like Quorum Sensing System in the Extreme Acidophile Acidithiobacillus ferrooxidans

Abstract

The genome of the acidophilic, proteobacterium Acidithiobacillus ferrooxidans, contains linked but divergently oriented genes, termed afeI and afeR, whose predicted protein products are significantly similar to the LuxI and LuxR families of proteins. A possible promoter and Lux box are predicted upstream of afeI. A cloned copy of afeI, expressed in E. coli, encodes an enzyme that catalyzes the production of a diffusible compound identified by gas chromatography and mass spectrometry as an unsubstituted N-acyl homoserine lactone (AHL) of chain length C14. This AHL can be detected by a reporter strain of Sinorhizobium meliloti Rm41 suggesting that it is biologically active. The reporter strain also responds to extracts of the supernatant of A. ferrooxidans grown to early stationary phase in sulfur medium indicating that a diffusible AHL is produced by this microorganism. Semi-quantitative RT-PCR experiments indicate that afeI and afeR are expressed maximally in early stationary phase and are more expressed when A. ferrooxidans is grown in sulfur- rather than iron-containing medium. Given the predicted amino acid sequence and functional properties of AfeI and AfeR it is proposed that A. ferrooxidans has a quorum sensing system similar to the LuxI-LuxR paradigm.

Abstract

Protein phosphatases are involved in many cellular processes. One of the most abundant and best studied members of this class is protein phosphatase type-2A (PP2A). In this study, PP2A was purified from the mussel Mytilus chilensis. Using both SDS-PAGE and size exclusion gel filtration under denaturant conditions, it was confirmed that the PP2A fraction was essentially pure. The isolated enzyme is a heterodimer and the molecular estimated masses of the subunits are 62 and 28 kDa. The isolated PP2A fraction has a notably high p-NPP phosphatase activity, which is inhibited by NaCl. The hydrolytic p-NPP phosphatase activity is independent of the MgCl2 concentration. The time courses of the inhibition of the PP2A activity of p-NPP hydrolysis by increasing concentrations of three phycotoxins that are specific inhibitors of PP2A are shown. Inhibitions caused by Okadaic acid, dinophysistoxin-1 (DTX1, 35-methylokadiac acid) and Microcystine L-R are dose-dependent with inhibition constants (Ki) of 1.68, 0.40 and 0.27 nM respectively. Microcystine L-R, the most potent phycotoxin inhibitor of PP2A isolated from Mytilus chilensis with an IC50 = 0.25 ng/ml, showed the highest specific inhibition effect an the p-NPP hydrolisis. The calculated IC50 for DTX1 and OA was 0.75 ng/ml and 1.8 ng/ml respectively.

45. FIRST REPORT OF DIARRHETIC SHELLFISH TOXINS IN MAGELLANIC FJORDS, SOUTHERN CHILE.

Abstract

Diarrhetic shellfish poisoning (DSP) is a gastrointestinal disease caused by polyether toxins produced by dinoflagellates and accumulated in shellfish. Until now, in Chile, harmful algal blooms associated with DSP have been confined to north of 46°00'S. Following a bloom of Dinophysis sp., in Estero Nuñez (53°19'S, 72°30'W) in March 1998, phytoplankton and mussel samples were collected for qualitative and quantitative analysis. Dinophysistoxin-1 (DTX-1), a diarrhetic shellfish toxin, was identified and quantified in extracts of Mytilus chilensis Hupe, a chilean native filter bivalve. DTX- 1 was measured using precolumn derivatization high-performance liquid chromatography method with fluorometric detection. The presence of DSP toxins was determined by a commercial colorimetric Protein Phosphatase assay (VDM-Test), which proved successful for the rapid screening of shellfish to detect DSP toxins. Okadaic acid was not detected in any mussel samples; the mussel shells showed only the presence of DTX- 1, ranging from 65.0-583.8 ng of DTX- 1 per gram of mussel digestive gland. This is the first report and quantitative analysis of DSP toxins in the Magellan region and extends 500 miles to the south, the known distribution of DSP toxins in Chilean coasts. The phytoplankton quantitative analysis showed Dinophysis acuminata (Clarapéde & Lachmann) as the most probable source of the DSP toxin. [ABSTRACT FROM AUTHOR]
