Ion bombardment during inductively coupled plasma reactive-ion etching and ion-implantation introduces irreparable crystalline damage to gallium nitride (GaN) power devices, leading to early breakdown and high leakage current. To circumvent this, a bi-layer selective area growth mask was engineered to grow up to 3.0 µm thick epitaxy of GaN using plasma-assisted molecular beam epitaxy as an ion-damage-free alternative to standard epitaxial processing routes. The masks and regrown architectures are characterized via SEM, conductive-atomic force microscopy (AFM), x-ray photo electron spectroscopy, Raman, and cathodoluminescence. Mask deposition conditions were varied to modulate and minimize the stress induced during thermal cycling. The resulting mesas exhibit low leakage, attributed to naturally terminated sidewalls as measured by an innovative perpendicular AFM measurement of the regrown sidewall. The regrown sidewall exhibited RMS (root mean square) roughness of 1.50 (±0.34) nm and defect density of 1.36 × 106 (±1.11 × 106) cm−2. This work provides a method to eliminate defect-inducing steps from GaN vertical epitaxial processing and stands to enhance GaN as a material platform for high-efficiency power devices.

The transformation of 2 ¯ 01 β - Ga 2 O 3 to h- GaN under exposure to RF nitrogen plasma was monitored in situ by reflection high-energy electron diffraction. Analysis of the reaction kinetics reveals that the nitridation process is initiated by the formation of an oxynitride phase and proceeds via two-dimensional nucleation and growth of wurtzite GaN grains. X-ray photoelectron spectra suggest a Ga − (N x O 1 − x) type configuration dominates the surface early in the nitridation process. The surface restructuring is followed by a diffusion-fed phase transformation, which propagates the wurzite GaN structure into the substrate upon reaching 70 % nitrogen anion site occupation, corresponding to the oxygen solubility in h - GaN. A direct correlation is observed between the nitridated film morphology and the epitaxial film crystallinity, demonstrating control of the residual strain, lateral coherence, and mosaicity in subsequent GaN epitaxy by the nitridation conditions. This study provides mechanistic details of the nitridation reaction of 2 ¯ 01 β - Ga 2 O 3 facilitating the optimization of the nitridation process toward improving GaN - 2 ¯ 01 β - Ga 2 O 3 heterojunctions. The lack of suitable p-type dopant for β-Ga2O3 remains a hurdle for vertical power device applications. Epitaxy of GaN on Ga2O3 substrates was demonstrated as an alternative. (–201)-oriented β-Ga2O3 was converted into (0001)-oriented hexagonal GaN via nitrogen plasma in a plasma-assisted molecular beam epitaxy chamber, as verified by XRD and RHEED. The resulting nitridated GaN layers were characterized by TEM, x-ray reflectivity, and AFM to relate the nitridation conditions to crystallinity, layer thickness, and surface roughness. The crystallinity of subsequently grown epitaxial GaN films was quantified via XRD rocking curves and related to the nitridation layer properties across varying nitridation conditions. Abtract Clinical circulating cell-free DNA (cfDNA) testing is now routine, however test accuracy remains limited. By understanding the life-cycle of cfDNA, we might identify opportunities to increase test performance. Here, we profile cfDNA release across a 24-cell line panel and utilize a cell-free CRISPR screen (cfCRISPR) to identify mediators of cfDNA release. Our panel outlines two distinct groups of cell lines: one which releases cfDNA fragmented similarly to clinical samples and purported as characteristic of apoptosis, and another which releases larger fragments associated with vesicular or necrotic DNA. Our cfCRISPR screens reveal that genes mediating cfDNA release are primarily involved with apoptosis, but also identify other subsets of genes such as RNA binding proteins as potential regulators of cfDNA release. We observe that both groups of cells lines identified primarily produce cfDNA through apoptosis. These results establish the utility of cfCRISPR, genetically validate apoptosis as a major mediator of DNA release in vitro, and implicate ways to improve cfDNA assays.

Diabetes mellitus (DM) is associated with a dysfunctional intestinal barrier and an increased risk for systemic infection and inflammation in people, though the pathogenic mechanisms leading to this are poorly understood. Using a canine model of DM, we showed that the peroxisomal proliferator-activated receptor-α agonist fenofibrate modulates plasma lipid profiles and markers of intestinal barrier function. A 3-week course of fenofibrate reduced fasting interstitial glucose and inflammatory cytokine IL-8 and TNF-α concentrations, which correlated with reduced triglyceride levels. The lipidomic profile exhibited significantly lower levels of triacylglycerols, phosphatidylethanolamines, diacylglycerols, and ceramides following fenofibrate administration. On histopathological analysis, we observed an aberrant amount of intraepithelial CD3+ T lymphocytes (IEL) in the small intestine of dogs with spontaneous and induced-DM. Fenofibrate reduced IEL density in the duodenum of dogs with DM and enhanced markers of intestinal barrier function in vivo and in vitro. There were minimal changes in the intestinal microbial composition following fenofibrate administration, suggesting that repair of intestinal barriers can be achieved independently of the resident microbiota. Our findings indicate that lipid metabolism is critical to functionality of the intestinal epithelium, which can be rescued by PPARα activation in dogs with DM.

This study surveyed freshwater mussels (family Unionidae) in 116 lakes and reservoirs east of the Missouri River in South Dakota, USA, during 2017. Using two-person–hour/site timed searches, evidence of a total of 1789 mussels, including 1053 live mussels, was obtained from 50 waters. Nine species, from two different orders, were found in lakes and reservoirs throughout five of the six major river drainages east of the Missouri River. The native species observed included Giant Floater Pyganodon grandis, Fatmucket Lampsilis siliquoidea, Threeridge Amblema plicata, White Heelsplitter Lasmigona complanata, Wabash Pigtoe Fusconaia flava, Deertoe Truncilla truncata, and Pink Heelsplitter Potamilus alatus. Giant Floater was the most widespread and abundant species observed, representing 63.3% of the live mussels sampled. Two non-native species, Zebra Mussel Dreissena polymorpha and Chinese Basket Clam Corbicula fluminea, were also documented from three water bodies in the lower Missouri River drainage. Overall, mussel abundance was negatively correlated with lake water conductivity and positively correlated with turbidity. No significant correlations were observed between species abundance and water temperature, pH, dissolved oxygen, or substrate particle size.

PurposeThe recent approval of anti-programmed death-ligand 1 immunotherapy in combination with nab-paclitaxel for metastatic triple-negative breast cancer (TNBC) highlights the need to understand the role of chemotherapy in modulating the tumor immune microenvironment (TIME).Experimental designWe examined immune-related gene expression patterns before and after neoadjuvant chemotherapy (NAC) in a series of 83 breast tumors, including 44 TNBCs, from patients with residual disease (RD). Changes in gene expression patterns in the TIME were tested for association with recurrence-free (RFS) and overall survival (OS). In addition, we sought to characterize the systemic effects of NAC through single-cell analysis (RNAseq and cytokine secretion) of programmed death-1-high (PD-1HI) CD8+ peripheral T cells and examination of a cytolytic gene signature in whole blood.ResultsIn non-TNBC, no change in expression of any single gene was associated with RFS or OS, while in TNBC upregulation of multiple immune-related genes and gene sets were associated with improved long-term outcome. High cytotoxic T-cell signatures present in the peripheral blood of patients with breast cancer at surgery were associated with persistent disease and recurrence, suggesting active antitumor immunity that may indicate ongoing disease burden.ConclusionsWe have characterized the effects of NAC on the TIME, finding that TNBC is uniquely sensitive to the immunologic effects of NAC, and local increases in immune genes/sets are associated with improved outcomes. However, expression of cytotoxic genes in the peripheral blood, as opposed to the TIME, may be a minimally invasive biomarker of persistent micrometastatic disease ultimately leading to recurrence.

BackgroundAtrial fibrillation (AF) adversely impacts health-related quality of life (hrQoL). While some patients demonstrate improvements in hrQoL, the factors associated with large improvements in hrQoL are not well described.MethodsWe assessed factors associated with a 1-year increase in the Atrial Fibrillation Effect on Quality-of-Life score of 1 SD (≥18 points; 3× clinically important difference), among outpatients in the Outcomes Registry for Better Informed Treatment of Atrial Fibrillation I registry.ResultsOverall, 28% (181/636) of patients had such a hrQoL improvement. Compared with patients not showing large hrQoL improvement, they were of similar age (median 73 versus 74, P=0.3), equally likely to be female (44% versus 48%, P=0.3), but more likely to have newly diagnosed AF at baseline (18% versus 8%; P=0.0004), prior antiarrhythmic drug use (52% versus 40%, P=0.005), baseline antiarrhythmic drug use (34.8% versus 26.8%, P=0.045), and more likely to undergo AF-related procedures during follow-up (AF ablation: 6.6% versus 2.0%, P=0.003; cardioversion: 12.2% versus 5.9%, P=0.008). In multivariable analysis, a history of alcohol abuse (adjusted OR, 2.41; P=0.01) and increased baseline diastolic blood pressure (adjusted OR, 1.23 per 10-point increase and >65 mm Hg; P=0.04) were associated with large improvements in hrQoL at 1 year, whereas patients with prior stroke/transient ischemic attack, chronic obstructive pulmonary disease, and peripheral arterial disease were less likely to improve (P

Nearshore coral reefs at Teahupo’o, Tahiti, are currently threatened by destruction from proposed plans to build a new judging tower in the reef lagoon for the 2024 Olympic surfing event. Local community members were trained to utilize 3D photogrammetry techniques to create high-resolution habitat maps of three sites that will be impacted by dredging and tower construction. The resulting orthomosaics were analyzed to quantify and characterize the coral community structure at each study site. Species diversity, coral colony count, coral colony size, and percent cover of live coral and living benthos were extracted from all survey plots. The resulting data show these sites support healthy and diverse coral communities that contribute to the ecological function of the larger reef system at Teahupo’o. The Hawai’i State Division of Aquatic Resources Penalty Matrix was used to estimate the USD value of the live corals and algae identified among the study sites and the total area that will be impacted by the planned development project. This study highlights the utility of 3D photogrammetry for effective citizen science as well as the large economic and ecological impacts that may occur if this proposed construction occurs.

Reactive oxygen species (ROS) are a set of oxygen- and nitrogen-containing radicals. They are produced from a wide range of sources. In biological contexts, cellular stress leads to an overproduction of ROS, which can lead to genetic damage and disease development. In industry, ROS are often productively used for water purification or for analyzing the possible toxicity of an industrial process. Because of their ubiquity, detection of ROS has been an analytical goal across a range of fields. To understand complicated systems and origins of ROS production, it is necessary to move from qualitative detection to quantitation. Analytical techniques that combine quantitation, high spatial and temporal resolution, and good specificity represent detection methods that can fill critical gaps in ROS research. Herein, we discuss the continued progress and limitations of fluorescence, electrochemical, and electron paramagnetic resonance detection of ROS over the last ten years, giving suggestions for the future of the field. In photovoltaic (PV) materials development, the complex relationship between device performance and underlying materials parameters obfuscates experimental feedback from current-voltage (J-V) characteristics alone. Here, we address this complexity by adding temperature and injection dependence and applying a Bayesian inference approach to extract multiple device-relevant materials parameters simultaneously. Our approach is an order of magnitude faster than the cumulative time of multiple individual spectroscopy techniques, with added advantages of using device-relevant materials stacks and interface conditions. We posit that this approach could be broadly applied to other semiconductor- and energy-device problems of similar complexity, accelerating the pace of experimental research. Photovoltaic (PV) research has historically taken decades to bring new materials to market, as the pace of development is often limited by our ability to identify the causes of underperformance. There are many ways to make an underperforming PV cell, and their signatures on device efficiency alone are not unique. This uncertainty plagues other energy-storage or -conversion devices with similarly complex combinations of materials but may be used to our advantage. By probing or biasing a PV cell in different operating conditions, the signature of underlying material properties becomes more unique. This connection between the operating conditions, the material properties, and the PV cell output may be solved through Bayesian inference algorithms. While computationally expensive, high-performance computing can enable such inference as a tool for experimentalists, a tool that could become increasingly valuable for accelerating the pace of materials research in PV and related fields. High-performance computing can greatly improve the workflow of experimentalists in energy materials, through the use of Bayesian inference. This allows us to solve the inverse problem of extracting underlying materials properties through the measurement of the electrical behavior of completed devices. Cheaper, faster measurements can be substituted for longer direct measurements of individual properties, without sacrificing accuracy or precision. We provide a general framework to apply this to other materials systems and devices.

Purpose The COVID-19 pandemic threatens our current ICU capabilities nationwide. As the number of COVID-19 positive patients across the nation continues to increase, the need for options to address ventilator shortages is inevitable. Multi-patient ventilation (MPV), in which more than one patient can use a single ventilator base unit, has been proposed as a potential solution to this problem. To our knowledge, this option has been discussed but remains untested in live patients with differing severity of lung pathology. Methods The objective of this study was to address ventilator shortages and patient stacking limitations by developing and validating a modified breathing circuit for two patients with differing lung compliances using simple, off-the-shelf components. A multi-patient ventilator circuit (MPVC) was simulated with a mathematical model and validated with four animal studies. Each animal study had two human-sized pigs: one healthy and one with lipopolysaccharide (LPS) induced ARDS. LPS was chosen because it lowers lung compliance similar to COVID-19. In a previous study, a control group of four pigs was given ARDS and placed on a single patient ventilation circuit (SPVC). The oxygenation of the MPVC ARDS animals was then compared to the oxygenation of the SPVC animals. Results Based on the comparisons, similar oxygenation and morbidity rates were observed between the MPVC ARDS animals and the SPVC animals.
