The SPI-2 type III secretion system (T3SS) of intracellular Salmonella enterica translocates effector proteins into mammalian cells. Infection of antigen-presenting cells results in SPI-2 T3SS-dependent ubiquitination and reduction of surface-localized mature MHC class II (mMHCII). We identify the effector SteD as required and sufficient for this process. In Mel Juso cells, SteD localized to the Golgi network and vesicles containing the E3 ubiquitin ligase MARCH8 and mMHCII. SteD caused MARCH8-dependent ubiquitination and depletion of surface mMHCII. One of two transmembrane domains and the C-terminal cytoplasmic region of SteD mediated binding to MARCH8 and mMHCII, respectively. Infection of dendritic cells resulted in SteD-dependent depletion of surface MHCII, the co-stimulatory molecule B7.2, and suppression of T cell activation. SteD also accounted for suppression of T cell activation during Salmonella infection of mice. We propose that SteD is an adaptor, forcing inappropriate ubiquitination of mMHCII by MARCH8 and thereby suppressing T cell activation.

Coloro i quali hanno maturato una esperienza, anche piccola, presso una struttura di assistenza sociale e di cura, sanno quanto sia importante che gli operatori che la compongono siano dotati di alcune competenze che caratterizzano tale mestiere. Ci riferiamo alle cosiddette competenze sociali e relazionali, che unite alle conoscenze teoriche ed applicative completano il quadro delle richieste dell’ambiente lavorativo in questione. Il possesso di tali competenze consente il doppio vantaggio di agire correttamente con la propria utenza nell’erogare l’aiuto di cui necessita, e allo stesso tempo di preservare il lavoratore stesso da stati di forte stress legati ad una mansione che, senza gli “attrezzi del mestiere”, può essere coinvolgente, logorante ed invasiva. Il presente lavoro ha la finalità di presentare uno strumento di esplorazione della fiducia nelle proprie competenze sociali, relazionali e tecnico-organizzative. Lo strumento è stato somministrato a 281 operatori socio-assistenziali con differente livello di formazione, inseriti presso strutture residenziali operanti in Sicilia. Attraverso delle analisi fattoriali (utilizzando il Principal Axis Factoring), abbiamo isolato tre aree di competenza socio-relazionale (assertività emotiva, comprensione empatica e resilienza), e tre di tipo tecnico-organizzativo (pianificazione e relazione, competenze tecnico-professionali, capacità di operare in rete). Le scale ad esse collegate hanno mostrato buoni livelli di validità. Lo strumento che presentiamo, pur necessitando una estensione del campione normativo, potrà essere utile sia per finalità di ricerca che per la analisi dei fabbisogni formativi all’interno delle strutture socio-assistenziali e di cura.

Xanthomonas citri ssp. citri (Xcc) is the causal agent of citrus canker. This bacterium develops a characteristic biofilm on both biotic and abiotic surfaces. A biofilm-deficient mutant was identified in a screening of a transposon mutagenesis library of the Xcc 306 strain constructed using the commercial Tn5 transposon EZ-Tn5 < KAN-2 > Tnp Transposome (Epicentre). Sequence analysis of a mutant obtained in the screening revealed that a single copy of the EZ-Tn5 was inserted at position 446 of hrpM, a gene encoding a putative enzyme involved in glucan synthesis. We demonstrate for the first time that the product encoded by the hrpM gene is involved in beta-1,2-glucan synthesis in Xcc. A mutation in hrpM resulted in no disease symptoms after 4 weeks of inoculation in lemon and grapefruit plants. The mutant also showed reduced ability to swim in soft agar and decreased resistance to H 2 O 2 in comparison with the wild-type strain. All defective phenotypes were restored to wild-type levels by complementation with the plasmid pBBR1-MCS containing an intact copy of the hrpM gene and its promoter. These results indicate that the hrpM gene contributes to Xcc growth and adaptation in its host plant.

Since it has been found that all subjects with contact allergy to cocamidopropylbetaine (CAPB) have positive reactions to 3-dimetylaminopropylamine (DMAPA), and reports have appeared in literature of the sensitizing action of amidoamine in products containing CAPB, we aimed to verify the possibility that pure amidoamine may have a sensitizing role in subjects with positive reactions to CAPB. To this end, in 10 patients with contact allergy to a commercial CAPB, we tested DMAPA 1% aq. and a pure amidoamine in concentrations ranging from 0.5% aq. to 0.1% aq. The study showed that all patients with positive reactions to DMAPA reacted to amidoamine at 0.5% and 0.25% aq., while 4 of the 10 also had positive reactions to amidoamine at 0.1% aq. We consider that simultaneous allergic reaction to DMAPA and amidoamine represents cross-reactivity and hypotesize that DMAPA is in fact the true sensitizing substance, while amidoamine, which may in any case release DMAPA in vivo as a result of enzymatic hydrolysis, may favour the transepidermal penetration of the sensitizing agent. In addition, we advise that testing of CAPB be suspended, because, as suggested by chemico-structural analyses and demonstrated in vivo, when thoroughly purified, it no longer has a sensitizing action.

Topical corticosteroids (CS) are widely used in dermatology because of their anti-inflammatory, anti-proliferative and immuno-suppressive properties. On the other hand, the prolonged application of corticosteroids may induce adverse reactions such as allergic contact dermatitis (ACD). Patch testing CS often poses methodological issues correlated to their drug properties that may hide the manifestations of a positive reaction. Furthermore, the ideal concentration to patch test corticosteroid is still a matter of study and some vehicles have some well-known limitations. This article is divided into two parts: the first one investigated vehicles that may efficiently dissolve the corticosteroids, according to the polarity of the latter; the second part compared the results of the patch tests with hydrocortistone-17-butyrate using two different vehicles: ethanol, which is the standard one, and another vehicle selected as suitable from our CS solubility test.

The increasing concern about the environmental impact of ingredients largely used as sunscreens today is pushing research towards new frontiers for the synthesis and degradation potential of innovative sun protection structures. This study deals with the design, synthesis, efficacy evaluation, and environmental impact of new photo-stable molecules at extended electronic conjugation and which have the &lsquo, charge-transfer&rsquo, type of optical transition (push and pull). They efficiently perform absorption of electro-magnetic radiation in the UVB and UVA regions. A preliminary screening, dealing with a whole series of aromatic donors, led to the selection of a key substrate derived from waste material, provided with low environmental impact and largely available on the market. As far as the acceptor was concerned, an ester with cyan-acrylic structure and functionalized with aliphatic chains emerged as the most suitable. This allowed a shift of the &lambda, of absorption in the range of the ultraviolet wavelengths. The synthesis procedures include an acid-catalyst-fitted esterification, condensation, and cold crystallization. It was continuously monitored with suitable characterization instruments (GC-MS, 1H NMR, 13C NMR), in order to avoid by-products and parasitic reactions. The resulting structures (patent pending) are composed by a furan core condensed with cyano-acrylic esters, derivatized with branched chains of hydrocarbons of medium length. The obtained structures are capable of good absorption in the range of wavelengths between 300 and 400 nm. Because of their structures, the new molecules are largely soluble in many common oils and cosmetic ingredients. The new molecules have been successively evaluated in terms of photo-stability, biodegradability, compatibility, and UVB-UVA protection capability, both in-vivo and in-vitro, in comparison with existing sunscreens.

Patch testing is the standard procedure used to diagnose allergic contact dermatitis. It is an in-vivo test, which reproduces the reaction to a contact allergen. This in-vivo test aims to reproduce the elicitation phase of allergic contact dermatitis and is performed applying allergens under occlusion on the skin under standardized conditions. These guidelines for the best practice in performing patch test have been developed by an Italian group of experts taking in account the Italian legislation and local pharmacological governance. Guidelines are adapted from the original article under the guidance of the European Society of Contact Dermatitis (ESCD) and on the basis of the SIDAPA guidelines.
