13. Assessing supernumerary root occurrence as a possible adaptation enhancing teeth performance in Mediterranean deer populations

Teeth root morphology and integrity are essential to provide appropriate attachment, allowing for continuous and functional movement, with implications for adequate food processing, animal performance, and longevity. We studied the occurrence of supernumerary roots in mandibular molariform teeth of red deer (Cervus elaphus Linnaeus, 1758) from seven separate populations spanning a range of latitudes in the Iberian Peninsula. We analyzed the influence of several factors, including sex, origin (native vs. reintroduced), lineage, and habitat, to assess extra root prevalence variations. The highest prevalence in deciduous teeth was found in [pm.sub.3] (14%) and in permanent teeth in [M.sub.1] (3%). We found significant differences between areas, lineages, and soil type; however, no significant relationships were found with the origin or the sex of individuals. We speculate that the high prevalence of supernumerary roots in [M.sub.1] might be related to increased wear in grazers. Furthermore, we suggest that this high prevalence in deciduous teeth might be associated with a hard diet, dry climatic conditions, and a harsh weaning period, which indicate that supernumerary roots might have an adaptive value. Nous étudions la présence de racines surnuméraires de dents molariformes mandibulaires chez des cerfs élaphes (Cervus elaphus Linnaeus, 1758) de sept populations distinctes couvrant une fourchette de latitudes dans la péninsule ibère. Nous analysons l'influence de plusieurs facteurs, dont le sexe, l'origine (indigène ou réintroduite), l'ascendance et l'habitat afin d'évaluer les variations de la prévalence de racines surnuméraires. Introduction: Bacteremia is a life-threatening condition that can progress to sepsis and septic shock, leading to significant mortality in the emergency department (ED). The standard diagnostic method, blood culture, is time-consuming and prone to false positives and false negatives. Although not widely accepted, several clinical and artificial intelligence-based algorithms have been recently developed to predict bacteremia. However, these strategies require further identification of new variables to improve their diagnostic accuracy. This study proposes a novel strategy to predict positive blood cultures by assessing sepsis-induced immunosuppression status through endotoxin tolerance assessment. Methods: Optimal assay conditions have been explored and tested in sepsis-suspected patients meeting the Sepsis-3 criteria. Blood samples were collected at ED admission, and endotoxin (lipopolysaccharide, LPS) challenge was performed to evaluate the innate immune response through cytokine profiling. Results: Clinical variables, immune cell population biomarkers, and cytokine levels (tumor necrosis factor [TNFα], IL-1β, IL-6, IL-8, and IL-10) were measured. Patients with positive blood cultures exhibited significantly lower TNFα production after LPS challenge than did those with negative blood cultures. The study also included a validation cohort to confirm that the response was consistent. Discussion: The results of this study highlight the innate immune system immunosuppression state as a critical parameter for sepsis diagnosis. Notably, the present study identified a reduction in monocyte populations and specific cytokine profiles as potential predictive markers. This study showed that the LPS challenge can be used to effectively distinguish between patients with bloodstream infection leading to sepsis and those whose blood cultures are negative, providinga rapid and reliable diagnostic tool to predict positive blood cultures. The potential applicability of these findings could enhance clinical practice in terms of the accuracy and promptness of sepsis diagnosis in the ED, improving patient outcomes through timely and appropriate treatment. Key Points: Question: Does the use of a nonselective extracorporeal blood purification (EBP) device connected to the cardiopulmonary bypass circuit reduce the incidence of acute kidney injury (AKI) in high-risk patients undergoing cardiothoracic surgery? Findings: In this randomized clinical trial that included 343 adults, a significant decrease in cardiac surgery–associated AKI was observed in those treated with EBP compared with those who were not (28.4% vs 39.7%). Meaning: In high-risk patients undergoing cardiac surgery, the use of a nonselective EBP device was associated with a significant reduction in AKI in the first 7 days after surgery. Importance: Cardiac surgery–associated acute kidney injury (CSA-AKI) remains a significant problem following cardiopulmonary bypass (CPB). Various strategies are proposed to attenuate CSA-AKI, including extracorporeal blood purification (EBP), but little is known about the effect of EBP through an acrylonitrile-sodium methallylsulfonate/polyethyleneimine membrane during CPB. Objective: To determine whether the use of an EBP device in a nonemergent cardiac surgery population reduces CSA-AKI after CPB. Design, Setting, and Participants: This double-blind, randomized clinical trial was conducted in 2 tertiary hospitals in Spain. Patients 18 years or older undergoing nonemergent cardiac surgery who were at high risk for CSA-AKI were enrolled from June 15, 2016, through November 5, 2021, with follow-up data through February 5, 2022. Of 1156 patients assessed, 343 patients were randomized (1:1) to either receive EBP or standard care. Intervention: Nonselective EBP device connected to the CPB circuit. Main Outcomes and Measures: The primary outcome was the rate of CSA-AKI in the 7 days after randomization. Results: Among 343 patients randomized (169 to receive EBP and 174 to receive usual care), the mean (SD) age was 69 (9) years and 119 were females. The rate of CSA-AKI was 28.4% (95% CI, 21.7%-35.8%) in the EBP group vs 39.7% (95% CI, 32.3%-47.3%) in the standard care group (P =.03), with an adjusted difference of 10.4% (95% CI, 2.3%-18.5%) using a log-binomial model (P =.01). No significant differences (P >.05) were observed in most of the predefined clinical secondary end points or post hoc exploratory end points. In a sensitivity analysis, EBP was found to be more effective in terms of CSA-AKI reduction in patients with chronic kidney disease, diabetes, hypertension, low left ventricular ejection fraction (<40%), and lower body mass index (<30). No differences were observed between the groups in adverse events tracking. Conclusions and Relevance: The use of a nonselective EBP device connected to the CPB circuit in a nonemergent population of patients undergoing cardiac surgery was associated with a significant reduction of CSA-AKI in the first 7 days after surgery. The present study aimed to evaluate the potential hazardous effects of NO
3 − concentration in drinking water on health, feed intake, rumen fermentation and microbiota, and nitrogen excretion of Holstein heifers fed a high-concentrate diet for a sustainable water use. Twenty-four Holstein heifers were individually allocated and assigned to one of four treatments with increasing drinking water NO3 − concentration: CTR, without NO3 − ; LOW, with 44 mg NO3 − /L; MOD, with 110 mg NO3 − /L; and HIGH, with 220 mg NO3 − /L. The entire study lasted 168 days. Fortnightly water NO3 − concentration and daily feed and water intake were recorded. Blood parameters, rumen pH, volatile fatty acids, NO3 − and NO2 − concentration, microbiota, and apparent total tract digestibility were determined at the beginning and at the end of the study. Most of the analyzed parameters were similar among treatments. Denitrifying bacteria population, estimated as nosZ gene copies, were greater in HIGH animals than in CTR animals at the end of the study. In conclusion, drinking water NO3 − concentration up to 220 mg/L has no detrimental effect on health, feed intake, rumen fermentation, nor N excretion in dairy beef cattle for periods up to 168 days; moreover, denitrifying bacteria population increased, which are related with the neutralization of the greenhouse gas N2 O. To identify and mitigate biodiversity decline, it is necessary to develop and implement appropriate ecological assessment methods. This particularly relevant in ecosystems under strong human pressure, such as riverine environments, in which ecological assessment requires several standardized methods useful in a wide range of riverine characteristics and habitats. In this work, we related otter diet composition with a set of environmental variables in representative river reaches of two contiguous river basins with a high diversity of habitats and varying degrees of human alteration. Our goal was to determine to what extent otter prey communities were sensitive to habitat characteristics and human pressures. Our results suggest that otter prey community in heavily anthropized rivers vary significantly across three gradients: habitat productivity, habitat structure and habitat stability. We also compared the results obtained from the otter diet with those obtained from electrofishing. Overall, the otter diet changed spatially in a similar way to the fish community sampled by electrofishing, but both methods showed disagreements that suggest different biases in some habitats and for some species. We conclude that otter diet analysis is a potential supplementary method for assessing river ecological status and that human alterations related to habitat structural simplification and water extraction can prevent freshwater biodiversity recovery in human‐dominated landscapes despite improvements in biochemical water quality. 28. Surveillance and prevention of healthcare-associated infections: best practices to prevent ventilator-associated events.

- Abstract
Ventilator associated pneumonia (VAP) leads to an increase in morbidity, mortality, and healthcare costs. In addition to increased evidence from the latest European and American guidelines (published in 2017 and 2022, respectively), in the last two years, several important clinical experiences have added new prevention tools to be included to improve the management of VAP. This paper is a narrative review of new evidence on VAP prevention. We divided VAP prevention measures into pharmacological, non-pharmacological, and ventilator care bundles. Most of the effective strategies that have been shown to decrease the incidence of complications are easy to implement and inexpensive. The implementation of care bundles, accompanied by educational measures and a multidisciplinary team should be part of optimal management. In addition to ventilator care bundles for the prevention of VAP, it could possibly be beneficial to use ventilator care bundles for the prevention of noninfectious ventilator associated events. A través de boies emergents de deriva anomenades pop-up buoys, es pretén ampliar les capacitats comunicatives d’aquests observatoris. El present TFG es divideix en dues parts degut a la gran quantitat d’objectius que hem d’assolir, la primera part la farà en Joan Armajach Riera i la segona part en Daniel Felguera Calleja. La primera part tindrà com objectiu principal integrar els diferents elements hardware necessaris per a la localització i la transmissió de dades en les pop-up buoys emergents de deriva a partir d’una connexió de satèl·lits Argos. El dispositiu que anirà integrat haurà de ser compacte i de mida reduïda. A més de posseir una gran autonomia ja que haurà d’estar llargs períodes de temps sense necessitat de recarregar-ho. En la segona part del treball implementarem un programari per la descàrrega, emmagatzematge i tractament de les dades rebudes de la plataforma Argos i enviades des de les boies emergents de deriva. També s'implementarà una interfície per a la representació de les dades obtingudes en un mapa on es pugui veure la trajectòria de les boies així com les marques temporals, la validesa de cada missatge i la velocitat de desplaçament de la nostre boia., La monitorización del fondo marino a través de plataformas autónomas resulta complicada por la falta de un canal de comunicación entre el observatorio y el usuario que permita saber el estado del sistema de monitorización y el acceso a sus datos. A través de boyas emergentes de deriva llamadas pop-up buoys, se pretende ampliar las capacidades comunicativas de estos observatorios. El presente TFG se divide en dos partes debido a la gran cantidad de objetivos que debemos alcanzar, la primera parte la realizará Joan Armajach Riera y la segunda parte Daniel Felguera Calleja. La primera parte tendrá como objetivo principal integrar los distintos elementos hardware necesarios para la localización y la transmisión de datos en las pop-up buoys emergentes de deriva a partir de una conexión de satélites Argos. También se implementará una interfaz para la representación de los datos obtenidos en un mapa donde pueda verse la trayectoria de las boyas así como las marcas temporales, la validez de cada mensaje y la velocidad de desplazamiento de nuestra boya., Monitoring the seabed through autonomous platforms is complicated by the lack of a communication channel between the observatory and the user that allows them to know the status of the monitoring system and access to their data. By means of pop-up buoys, the aim is to expand the communicative capabilities of these observatories. This TFG is divided into two parts due to the large number of objectives to be achieved. The first part will be carried out by Joan Armajach Riera and the second part by Daniel Felguera Calleja. The first part will have as its main objective to integrate the different hardware elements necessary for the localisation and transmission of data in the pop-up buoys emerging from a connection of Argos satellites. The device to be integrated must be compact and small in size. In addition to having a great autonomy, as it will have to remain there for long periods of time without needing to be recharged. In the second part of the work we will implement a program for downloading, packaging and processing the data collected from the Argos platform and sent from the emerging drifting buoys. We will also implement an interface for the representation of the data obtained on a map where the trajectory of the boies can be seen as well as the time stamps, the validity of each message and the speed of displacement of our buoy.

- Abstract
[Background] Integrative studies of animals and associated microbial assemblages (i.e., the holobiont) are rapidly changing our perspectives on organismal ecology and evolution. Insular vertebrates provide ideal natural systems to understand patterns of host-gut microbiota coevolution, the resilience and plasticity these microbial communities over temporal and spatial scales, and ultimately their role in the host ecological adaptation., [Methods] Here we used the endemic Balearic wall lizard Podarcis lilfordi to dissect the drivers of the microbial diversity within and across host allopatric populations/islets. By focusing on three extensively studied populations/islets of Mallorca (Spain) and fecal sampling from individually identified lizards along two years (both in spring and autumn), we sorted out the effect of islet, sex, life stage, year and season on the microbiota composition. We further related microbiota diversity to host genetics, trophic ecology and expected annual metabolic changes., [Results] All the three populations showed a remarkable conservation of the major microbial taxonomic profile, while carrying their unique microbial signature at finer level of taxonomic resolution (Amplicon Sequence Variants (ASVs)). Microbiota distances across populations were compatible with both host genetics (based on microsatellites) and trophic niche distances (based on stable isotopes and fecal content). Within populations, a large proportion of ASVs (30–50%) were recurrently found along the four sampling dates. The microbial diversity was strongly marked by seasonality, with no sex effect and a marginal life stage and annual effect. The microbiota showed seasonal fluctuations along the two sampled years, primarily due to changes in the relative abundances of fermentative bacteria (mostly families Lachnospiraceae and Ruminococcaceae), without any major compositional turnover., [Conclusions] These results support a large resilience of the major compositional aspects of the P. lilfordi gut microbiota over the short-term evolutionary divergence of their host allopatric populations (<10,000 years), but also indicate an undergoing process of parallel diversification of the both host and associated gut microbes. Predictable seasonal dynamics in microbiota diversity suggests a role of microbiota plasticity in the lizards’ metabolic adaptation to their resource-constrained insular environments. Overall, our study supports the need for longitudinal and integrative studies of host and associated microbes in natural systems.

- Abstract
During the last two decades, several Marine Heatwaves (MHWs) have affected coralligenous assemblages in the Mediterranean Sea, causing catastrophic mass mortalities of several habitat-forming species such as gorgonians, corals, and sponges. Even though Marine Protected Areas (MPAs) are contributing to effectively protect marine ecosystems, the impacts associated to extreme climatic events within MPAs are jeopardizing their protective role. Therefore, minimizing local stressors within MPAs is crucial to minimize interactive effects with global, more difficult to manage, stressors. To address this, we assessed to what extent the regulation of diving frequentation can support more effective protection to climate change, focusing on the case study of the Medes Islands, which has recently suffered the impacts of different global stressors and is one of the most visited MPAs in the Mediterranean Sea. We combined 6 years of demographic data of the red gorgonian Paramuricea clavata with population modelling tools, to explore the long-term viability of this species to different managing schemes and mass mortality events scenarios. Overall, our results show that climate-adaptive management of the recreational diving activity under climate change can enhance the long-term viability of this key Mediterranean habitat-forming octocoral, which is otherwise predicted to go locally extinct at shallow depths (<25 m) within the next 20 years. This study provides one of the few attempts to quantify to what extent an adaptive management scheme may help delay climate change impacts in a Marine Protected Area
