7. Modelling microtubule dynamics and microtubule-motor self-organisation

Abstract

The microtubule cytoskeleton is a self-organising system of microtubule filaments, molecular motors and regulatory proteins that plays a number of vital roles in all eukaryotic cells, acting as a scaffold to provide structural support to the cell and forming tracks for intracellular transport. Arguably one of its most important roles is in assembling the mitotic spindle, the highly dynamic force-generating structure that segregates chromosomes during mitosis. Critical to the spindle's dynamism is the dynamic instability of microtubules. At the heart of the long-standing GTP cap model of dynamic instability is the notion that stochastic fluctuations of the GTP cap drive microtubules to catastrophe, but a quantitative understanding of GTP cap fluctuations is missing. In Chapter 2 a simple mathematical model is developed that predicts microtubule fluctuation behaviour in the steady-state. The theory is validated by data, which suggests that microtubules are remarkably stable during most of their lifetime. How microtubules and motors self-organise into the complex, bipolar architecture of the spindle is not well understood. In Chapter 3 a microscopic computational model of microtubule-motor self-organisation is developed based on in vitro experiments. Two control parameters are identified capturing system composition and kinetics that predict the outcome of self-organisation and uncover generic, mechanistic rules governing active filament-motor networks. Design principles of spindle bipolarity are elucidated which highlight the combined effect of motor activity and filament dynamics on self-organisation. Computer simulations are an increasingly powerful tool for modelling the cytoskeleton, but existing models differ in key respects. In Chapter 4 the effects of spatial dimensionality and the inclusion/omission of steric interactions on simulated microtubule-motor networks is systematically evaluated. Spatial dimensionality and steric interactions are found to have a combined effect on the probability of microtubule crossings, which in turn play critical and distinct roles in several archetypal microtubule-motor networks. Guidelines for appropriate modelling choices are discussed.

Abstract

Achieving a net-zero carbon economy requires significant structural changes in the financial system, including a substantial shift in investment towards low-carbon assets. Through the alignment of expectations, promotion of herding behavior, utilization of public finance, reduction of capital costs and attainment of low-carbon investment thresholds in developing nations, and implementation of robust financial regulations and policies, the financial system can assume a central role in re-orienting economies onto a net-zero course. Taken together, such mechanisms highlight the positive tipping points that can be triggered within sustainable finance and emphasize the necessity of policy interventions to activate and capitalize on these dynamics. The identification and activation of critical and positive tipping points can lead to the amplification of sustainable investments and foster transformative changes in the practices of the financial sector. Equity demands that such finance be provided by developed to developing countries, according to their needs. Breaking investment lock-ins that exclude poor and vulnerable countries from international climate finance is necessary to ensure an equitable, timely transition.

Abstract

During cell division, mitotic motors organize microtubules in the bipolar spindle into either polar arrays at the spindle poles or a "nematic" network of aligned microtubules at the spindle center. The reasons for the distinct self-organizing capacities of dynamic microtubules and different motors are not understood. Using in vitro reconstitution experiments and computer simulations, we show that the human mitotic motors kinesin-5 KIF11 and kinesin-14 HSET, despite opposite directionalities, can both organize dynamic microtubules into either polar or nematic networks. We show that in addition to the motor properties the natural asymmetry between microtubule plus- and minus-end growth critically contributes to the organizational potential of the motors. We identify two control parameters that capture system composition and kinetic properties and predict the outcome of microtubule network organization. These results elucidate a fundamental design principle of spindle bipolarity and establish general rules for active filament network organization.
