7. A translational paradigm to dtudy the rffects of uncontrollable stress in humans

Contains fulltext : 221795.pdf (publisher's version ) (Open Access), Theories on the aetiology of depression in humans are intimately linked to animal research on stressor controllability effects. However, explicit translations of established animal designs are lacking. In two consecutive studies, we developed a translational paradigm to study stressor controllability effects in humans. In the first study, we compared three groups of participants, one exposed to escapable stress, one yoked inescapable stress group, and a control group not exposed to stress. Although group differences indicated successful stress induction, the manipulation failed to differentiate groups according to controllability. In the second study, we employed an improved paradigm and contrasted only an escapable stress group to a yoked inescapable stress group. The final design successfully induced differential effects on self-reported perceived control, exhaustion, helplessness, and behavioural indices of adaptation to stress. The latter were examined in a new escape behaviour test which was modelled after the classic shuttle box animal paradigm. Contrary to the learned helplessness literature, exposure to uncontrollable stress led to more activity and exploration; however, these behaviours were ultimately not adaptive. We discuss the results and possible applications in light of the findings on learning and agency beliefs, inter-individual differences, and interventions aimed at improving resilience to stress-induced mental dysfunction.

28. Actions of norepinephrine in the rat hippocampus

Abstract

Acting at postsynaptic alpha 1- and beta 1-receptors, norepinephrine (NE) exerts a complex action in rat hippocampus. It is currently believed that beta 1-receptor activation enhances excitability of recorded neurons, whereas alpha 1 activation suppresses reactivity to afferent stimulation. These reported effects of alpha-agonists are not consistent with alpha 1 effects found elsewhere in the brain. We have conducted experiments in the anesthetized rat and found that an amphetamine-induced increase in the dentate gyrus population spike can be blocked by a beta-antagonist but also by an alpha 1-antagonist. We have conducted experiments in the brain slide preparation and found that an alpha-agonist, phenylephrine (PHE), selectively enhances responses to N-methyl-D-aspartate (NMDA) but not to quisqualate. We propose that the product of activation of both alpha- and beta-receptor types will enhance reactivity of hippocampal cells to afferent stimulation.

Abstract

Objectives. This study assessed the interactive effect of two risk factors: 'Juvenile stress' and sex in the long-term consequences of 'Juvenile stress' in male and female rats. Methods. Rats were exposed to 'Juvenile stress' and to additional stress in adulthood. Measurements of anxiety and depressive-like behaviours were assessed in relation to each stress exposure and 'Sex-specific' sets of criteria in order to characterize individual profiles of altered behaviours. Results. While both male and female rats were affected by exposure to 'Juvenile stress', sex difference were evident in saccharine preference, coping with the stressful challenge of the two-way shuttle avoidance task, and on 'Adult stress' induced changes in saccharine preference. 'Profiling' altered behaviours revealed sex differences also in the prevalence of rats exhibiting different categories of 'Affected' behaviours, indicating that female rats are more susceptible to the long-term effects of 'Juvenile stress' and to the immediate effects of 'Adulthood stress'. Additionally, the prevalence of 'Affected' animals among 'Juvenile+ Adulthood stress' was similar, yet the profile of altered behaviours was significantly different. Conclusions. The 'Behavioural Profiling' approach presented here is of importance to understanding gender differences in the aetiology of predisposition to stress-related disorders, and of gender symptomatology differences in stress-related disorders.
