International audience; Egypt has no domestic silver ore sources and silver is rarely found in the Egyptian archaeological record until the Middle Bronze Age. Bracelets found in the tomb of queen Hetepheres I, mother of pyramid builder king Khufu (date of reign c. 2589-2566 BC), form the largest and most famous collection of silver artefacts from early Egypt, but they have not been analysed for decades. We analysed samples from the collection in the Museum of Fine Arts, Boston using bulk XRF, micro-XRF, SEM-EDS, X-ray diffractometry and MC-ICP-MS to obtain elemental and mineralogical compositions and lead isotope ratios, to understand the nature and metallurgical treatment of the metal and identify the possible ore source. We found that the pieces consist of silver with trace copper, gold, lead and other elements. The minerals are silver, silver chloride and a possible trace of copper chloride. Surprisingly, the lead isotope ratios are consistent with ores from the Cyclades (Aegean islands, Greece), and to a lesser extent from Lavrion (Attica, Greece), and not partitioned from gold or electrum as previously surmised. Sources in Anatolia (Western Asia) can be excluded with a high degree of confidence. Imaging of a cross-section of a bracelet fragment reveals that the metal was repeatedly annealed and cold-hammered during creation of the artefacts. The results provide new information about silver ore sources, commodity exchange networks and metallurgy in Egypt during the Early Bronze Age.

The theme of ports and shipping provides many overlapping and complementary interests for maritime and industrial archaeologists. This chapter aims to present the areas of shared theoretical interest and to introduce sites and features of particular relevance to industrial archaeological research. It examines terrestrial infrastructure, ships, and shipwrecks as industrial archaeological sites within a global context, primarily focused on the 17th to 20th centuries. The theme is explored under topic headings which cover ships as individual industrial sites, landing sites lacking infrastructure, small-scale coastal infrastructure, enclosed harbours, docks, naval dockyards and graving docks. Having dealt with major port infrastructure the chapter examines port related industries and the influence of ports on urban development. Ports and shipping are established as suitable topics for study by industrial archaeologists bringing greater understanding of ports as centres and distributors of technological innovation. An industrial archaeological approach also enables a better understanding of ports and shipping as components in networks facilitating not only trade but the globalization of technologies, and the exchange of social and economic ideas and practices.

Prostate cancer (PCa) is one of the most frequent tumor types in the male Western population. Early‐stage PCa and late‐stage PCa are dependent on androgen signaling, and inhibitors of the androgen receptor (AR) axis represent the standard therapy. Here, we studied in detail the global impact of darolutamide, a newly approved AR antagonist, on the transcriptome and AR‐bound cistrome in two PCa cell models. Darolutamide strongly depleted the AR from gene regulatory regions and abolished AR‐driven transcriptional signaling. Enhancer activation was blocked at the chromatin level as evaluated by H3K27 acetylation (H3K27ac), H3K4 monomethylation (H3K4me1), and FOXA1, MED1, and BRD4 binding. We identified genomic regions with high affinities for the AR in androgen‐stimulated, but also in androgen‐depleted conditions. A similar AR affinity pattern was observed in healthy and PCa tissue samples. High FOXA1, BRD4, H3K27ac, and H3K4me1 levels were found to mark regions showing AR binding in the hormone‐depleted setting. Conversely, low FOXA1, BRD4, and H3K27ac levels were observed at regulatory sites that responded strongly to androgen stimulation, and AR interactions at these sites were blocked by darolutamide. Beside marked loss of AR occupancy, FOXA1 recruitment to chromatin was also clearly reduced after darolutamide treatment. We furthermore identified numerous androgen‐regulated super‐enhancers (SEs) that were associated with hallmark androgen and cell proliferation‐associated gene sets. Importantly, these SEs are also active in PCa tissues and sensitive to darolutamide treatment in our models. Our findings demonstrate that darolutamide is a potent AR antagonist blocking genome‐wide AR enhancer and SE activation, and downstream transcription. We also show the existence of a dynamic AR cistrome that depends on the androgen levels and on high AR affinity regions present in PCa cell lines and also in tissue samples., Schematic model showing how darolutamide blocks androgen (R1881)‐mediated AR signaling by inhibiting the function of normal enhancers and super‐enhancers, and impairing downstream gene transcription.

The mammary intraductal xenografting technique has been established to inject cells or other substances directly into the mammary ducts of female mice. Using this refined xenografting method provides the possibility of mimicking the normal microenvironment of preinvasive breast lesions including, ductal carcinoma in situ (DCIS), to study of the progression of DCIS to invasive breast cancer in a more relevant manner than with other mammary xenografting methods. Xenografting into the mammary fat pad delivers cells directly into the stroma and bypasses the occurrence of invasive transition, during which cells invade through the basement membrane. Either breast cancer cell lines or patient-derived breast cancer cells can be injected into the mammary duct using this protocol to model breast cancer progression. This protocol will cover the procedures required to perform this technique.

We describe a school science outreach initiative that introduced learners to applied nuclear physics research by means of a two-day workshop that involved learners and teachers from 5 schools in the Western Cape province of South Africa. During this workshop, the participants were introduced to the naturally occurring, inert, colorless, and tasteless radioactive gas radon (222Rn). During the first day of the workshop, the participants were informed about the detrimental health impacts of inhaling radon and its daughter radionuclides and were shown how indoor radon activity concentrations can be measured using the electret ion chamber (EIC) technology. The learners were then each supplied with a short-term electret (E-PERM, Radelec, Frederick, MD, USA) and associated ion chamber to enable them to make radon measurements in their homes. The teachers in turn were supplied with EICs to enable them make radon measurements in their schools. The participants returned the EICs on the second day of the workshop, one week later. Here, the drop in the potential difference across each electret was measured in order to calculate the average indoor radon activity concentration. A total of 49 indoor radon concentrations were measured. The average indoor radon concentrations were 36 ± 26 Bqm−3 in homes and 41 ± 36 Bqm−3 in schools, while the highest concentration was found to be 144 Bqm−3. These levels were compared to predictions from a model that uses input information about the uranium content associated with the surface geology at each measurement location. The predictions compared well with the measured values.

This paper reviews eleven ceramic objects in the Museum of Fine Arts, Boston (MFA) that have been attributed variously over the past 150 years to the French ceramicist Bernard Palissy (1510-1590), his studio, workshop, school, followers, circle of, or imitators. It reviews the attributions of these objects at the time of their loan or acquisition by the Museum, and examines how they fit into current research about his work. Each of the works was analyzed with non destructive, qualitative XRF and four pieces were sampled for study with SEM/EDS. These samples were further analyzed with particle-induced gamma ray spectroscopy (PIGE) at C2RMF. The analysis of one rustic platter is discussed in depth.

Little is known about the role of Sox11 in the regulation of mammary progenitor cells. Sox11 is expressed by mammary bud epithelial cells during embryonic mammary gland development and is not detected in mammary epithelial cells after birth. As Sox11 is an oncofetal gene, we investigated the effects of reducing Sox11 levels in embryonic mammary progenitor cells and found that Sox11 regulates proliferative state, stem cell activity and lineage marker expression. We also investigated the effect of reducing Sox11 levels in two transplantable Brca1-deficient oestrogen receptor-negative mouse mammary tumour cell lines, to assess whether Sox11 regulates similar functions in tumour progenitor cells. When Sox11 levels were reduced in one Brca1-deficient mammary tumour cell line that expressed both epithelial and mesenchymal markers, similar effects on proliferation, stem cell activity and expression of lineage markers to those seen in the embryonic mammary progenitor cells were observed. Orthotopic grafting of mammary tumour cells with reduced Sox11 levels led to alterations in tumour-initiating capacity, latency, expression of lineage markers and metastatic burden. Our results support a model in which tumours expressing higher levels of Sox11 have more stem and tumour-initiating cells, and are less proliferative, whereas tumours expressing lower levels of Sox11 become more proliferative and capable of morphogenetic/metastatic growth, similar to what occurs during embryonic mammary developmental progression., Summary: Brca1−/− mammary tumours expressing Sox11 at high levels have more stem- and tumour-initiating cells, and are less proliferative, whereas tumours expressing Sox11 at lower levels become more proliferative and are capable of morphogenetic/metastatic growth.

Models for iron innovation in Eurasia are predicated on understanding the relationship between the bronze and iron industries. In eastern Anatolia, the South Caucasus, and Iran, the absence of scientific analyses of metallurgical debris has obscured the relative chronology, spatial organization, and economic context of early iron and contemporary copper-alloy industries. Survey and excavation at Mtsvane Gora, a fortified hilltop site close to major polymetallic ore sources in the Lesser Caucasus range, recovered metallurgical debris dating to the 8th-6th centuries BC. Optical microscopy, scanning electron microscopy, and energy and wavelength dispersive spectrometry revealed evidence for both iron and copper-alloy metallurgy, including smithing and alloying. Metal particles trapped within clear iron smithing slags were contaminated with copper, arsenic, and tin, suggesting that iron and copper-alloy working took place in the same hearths. The discovery of a small fragment of unprocessed material consisting of pyrite and jarosite, minerals typical of major nearby polymetallic ore deposits, links the secondary smithing and alloying at Mtsvane Gora with nearby mining activities, though the nature of those connections remains unclear. While the earliest iron in the region probably predates the Mtsvane Gora assemblage, the remains date to a period when iron use was still expanding, and they are at present the earliest analytically confirmed, radiocarbon-dated iron metallurgical debris in the Caucasus. The remains are therefore significant for understanding the spread of iron innovation eastward from Anatolia and the Levant. When considered in light of evidence from other Near Eastern sites, the results support a model for innovation in which early iron manufacturing was at least partially integrated with the copper-alloy metallurgical economy.

Before the start of its restoration in 2007, the Salvator Mundi was thought to be one of a number of copies of a long-lost Leonardo da Vinci painting, depicting Christ giving a blessing with his right hand while holding a crystal orb in his left. During the restoration treatment, a scientific examination of the painting was carried out to elucidate the painting’s materials and techniques. Microscopic sampling of the painting was necessarily limited, and nine out of ten samples were prepared and analyzed as cross-sections. A number of analytical methods were employed selectively, including stereomicroscopy, visible and fluorescent light microscopy, scanning electron microscopy with energy dispersive spectroscopy, Raman microspectroscopy, Fourier transform infrared microspectroscopy in transmission mode and with attenuated total reflection. The pigments characterized were lead white, vermilion, red iron oxide earth, red lake, natural ultramarine, lead–tin yellow, umber, and charcoal, carbon and bone blacks. Manganese-containing soda-lime glass was detected in the ground, imprimitura and paint layers, and a walnut oil medium was identified by pyrolysis gas chromatography-mass spectrometry. Cross-section studies revealed aspects of the painting’s stratigraphy: a size layer, white ground and off-white imprimitura, followed by a complex sequence of paint layers applied by the artist to achieve sophisticated visual effects.

Paid area link 6 (Pal6) underpass, a crucial tunnel connection of London Underground's Victoria station upgrade project, was engineered within notoriously difficult sub-artesian granular River Terrace Deposits (RTD) immediately below the Victorian-age District and Circle line masonry tunnel, a structure highly sensitive to ground displacements. Due to the proximity of the overlying operational railway, the consequences of ground movement were of very high risk. Historic tunnel construction in the heterogeneous RTD had limited success with permeation grouting, and face losses and instabilities occurred. For the case of Pal6, the railway tunnel overhead magnified the difficulties. Permeation grouting still remained the most feasible stabilising option, but only with significant improvements in both grouting technique and grout types. A low-risk tunnelled solution, using a bespoke and sophisticated construction approach, enabled safe continuous excavation within the severely restricted structural constraints. Successful tunnelling demanded: complete sequence recovery of the RTD for accurate soil characterisation for design of the grouting system; caution with permeation grouting; appropriate grout selection to promote permeation without claquage (uncontrolled fracturing by grout with its consequent irregular distribution), thus precisely targeting discrete zones; uniform treatment; and observational control by monitoring real-time ground movements.
