The concept of dominance is frequently used to describe changes in rapidly reconfiguring ecosystems, but the definition of dominance can vary widely among studies. Using coral reefs as a model, we use extensive benthic composition data to explore how variability in applying dominance concepts can shape perceptions. We reveal that coral dominance is sensitive to the exclusion of key algal groups and the categorization of other benthic groups, with ramifications for detecting an ecosystem phase shift. For example, ignoring algal turf inflates the dominance of hard and soft corals in the benthic habitats underpinning reef ecosystems. We need a consensus on how dominance concepts are applied so that we can build a more comprehensive understanding of ecosystem shifts across a broad range of aquatic and terrestrial settings. For reefs, we highlight the benefits of comprehensive and inclusive surveys for evaluating and managing the altered ecosystem states that are emerging in the Anthropocene.

Environmental disturbances and fishing are well known drivers of coral reef fish population size, length-frequency, and assemblage structure. However, few studies have partitioned the spatial and temporal impacts of multiple disturbance events and long-term no-take marine reserve (NTMR) protection on the biomass of juvenile and adult reef-fishes based on the known size of sexual maturity. Here, we document responses in the biomass of juvenile and adult wrasses (Labridae) Hemigymnus melapterus, H. fasciatus, Cheilinus fasciatus, and Oxycheilinus digramma, to environmental disturbance events, NTMR protection, and predator density on inshore fringing coral reefs at the Palm and Whitsunday Island groups, Great Barrier Reef (GBR), Australia from 2007 to 2018 (12 years). The biomass of juvenile and adult wrasses on inshore GBR reefs were driven predominantly by benthic habitat associations, rather than by NTMR protection or density of wrasse predators (Plectropomus spp.). Despite similar species-specific associations of juvenile and adult wrasses with benthic cover, juvenile wrasse biomass consistently declined following coral bleaching and cyclone events. Conversely, adult wrasses had variable responses to disturbance events, including some increases in biomass. Disturbance-mediated declines in the biomass of juvenile wrasses are likely to generate ongoing reductions in the abundance of these species on inshore GBR reefs. Our findings provide further evidence that habitat loss impacts a range of coral reef fishes beyond those that are directly reliant upon live coral. Shifts in assemblage structure, loss of biodiversity, and reductions in fishery productivity will become increasingly apparent in coral reef ecosystems if anthropogenic global warming continues unabated.

Western Australia’s coral reefs have largely escaped the chronic pressures affecting other reefs around the world, but are regularly affected by seasonal storms and cyclones, and increasingly by heat stress and coral bleaching. Reef systems north of 18°S have been impacted by heat stress and coral bleaching during strong El Nino phases and those further south during strong La Nina phases. Cumulative heat stress and the extent of bleaching throughout the northern reefs in 2016 were higher than at any other time on record. To assess the changing regime of disturbance to reef systems across Western Australia (WA), we linked their site-specific exposure to damaging waves and heat stress since 1990 with mean changes in coral cover. Since 2010, there has been a noticeable increase in heat stress and coral bleaching across WA. Over half the reef systems have been severely impacted by coral bleaching since 2010, which was further compounded by cyclones at some reefs. For most (75%) reef systems with long-term data (5–26 yrs), mean coral cover is currently at (or near) the lowest on record and a full recovery is unlikely if disturbances continue to intensify with climate change. However, some reefs have not yet experienced severe bleaching and their coral cover has remained relatively stable or increased in recent years. Additionally, within all reef systems the condition of communities and their exposure to disturbances varied spatially. Identifying the communities least susceptible to future disturbances and linking them through networks of protected areas, based on patterns of larval connectivity, are important research and management priorities in coming years while the causes of climate change are addressed.

Understanding ecological processes that shape contemporary and future communities facilitates knowledge-based environmental management. In marine ecosystems, one of the most important processes is the supply of new recruits into a population. Here, we investigated spatiotemporal variability in coral recruitment at 15 reefs throughout the Dampier Archipelago, north-western Australia between 2015 and 2017 and identified the best environmental predictors for coral recruitment patterns over this period. Large differences in recruitment were observed among years with the average density of recruits increasing by 375% from 0.017 recruits cm−2 in 2015 to 0.059 recruits cm−2 in 2017. Despite differences in recruitment among years, the rank order of coral recruit density among reefs remained similar among years, suggesting that spatial variation in recruitment within the Dampier Archipelago is partly deterministic and predictable. The density of coral recruits was best explained by percent cover of live corals at both local (within 5 m) and meso-scales (within 15 km), water turbidity and an oceanographic model that predicted larval dispersal. The highest density of coral recruits (~0.13 recruits cm−2 or 37 recruits per tile) occurred on reefs within sub-regions (15 km) with greater than 35% coral cover, low to moderate turbidity (KD490 < 0.2) and moderate to high modelled predictions of larval dispersal. Our results demonstrate that broad-scale larval dispersal models, when combined with local metrics of percent hard coral cover and water turbidity, can reliably predict the relative abundance of coral recruits over large geographical areas and thus can identify hotspots of recruit abundance and potential recovery following environmental disturbances; information that is essential for effective management of coral reefs.

Coral reefs provide critical ecological and geomorphic (e.g. sediment production for reef-fronted shoreline maintenance) services, which interact in complex and dynamic ways. These services are under threat from climate change, requiring dynamic modelling approaches that predict how reef systems will respond to different future climate scenarios. Carbonate budgets, which estimate net reef calcium carbonate production, provide a comprehensive ‘snap-shot’ assessment of reef accretionary potential and reef stability. These budgets, however, were not intended to account for the full suite of processes that maintain coral reef services or to provide predictive capacity on longer timescales (decadal to centennial). To respond to the dual challenges of enhancing carbonate budget assessments and advancing their predictive capacity, we applied a novel model elicitation and review method to create a qualitative geo-ecological carbonate reef system model that links geomorphic, ecological and physical processes. Our approach conceptualizes relationships between net carbonate production, sediment transport and landform stability, and rates knowledge confidence to reveal major knowledge gaps and critical future research pathways. The model provides a blueprint for future coral reef research that aims to quantify net carbonate production and sediment dynamics, improving our capacity to predict responses of reefs and reef-fronted shorelines to future climate change.

Canopy-forming macroalgae can construct extensive meadow habitats in tropical seascapes occupied by fishes that span a diversity of taxa, life-history stages and ecological roles. Our synthesis assessed whether these tropical macroalgal habitats have unique fish assemblages, provide fish nurseries and support local fisheries. We also applied a meta-analysis of independent surveys across 23 tropical reef locations in 11 countries to examine how macroalgal canopy condition is related to the abundance of macroalgal-associated fishes. Over 627 fish species were documented in tropical macroalgal meadows, with 218 of these taxa exhibiting higher local abundance within this habitat (cf. nearby coral reef) during at least one life-history stage. Major overlap (40%-43%) in local fish species richness among macroalgal and seagrass or coral reef habitats suggest macroalgal meadows may provide an important habitat refuge. Moreover, the prominence of juvenile fishes suggests macroalgal meadows facilitate the triphasic life cycle of many fishes occupying diverse tropical seascapes. Correlations between macroalgal canopy structure and juvenile abundance suggests macroalgal habitat condition can influence levels of replenishment in tropical fish populations, including the majority of macroalgal-associated fishes that are targeted by commercial, subsistence or recreational fisheries. While many macroalgal-associated fishery species are of minor commercial value, their local importance for food and livelihood security can be substantial (e.g. up to 60% of landings in Kenyan reef fisheries). Given that macroalgal canopy condition can vary substantially with sea temperature, there is a high likelihood that climate change will impact macroalgal-associated fish and fisheries.

Genetic diversity and connectivity are key factors in determining a population’s resilience to future disturbance. This is especially relevant to corals, which are in global decline due to increasing frequency and strength of thermal anomalies and severe tropical cyclones. While many studies have investigated genetic diversity and population structure in corals, they focused on species being removed at the greatest rate from coral reefs, acroporids and pocilloporids, and it is unclear whether the patterns observed in these species reflects those found in more resilient species. Here, we use microsatellite markers and two Lagrangian models with differing resolutions, to investigate population structure in a stress-tolerant coral survivor Cyphastrea microphthalma, Family Merulinidae, along the north-western Australian coastline. We found evidence of four genetic clusters with some level of admixture among them. However, while there was evidence of population structure within the intensively sampled Pilbara region, the patterns of connectivity differed to those reported previously. WA populations of C. microphthalma were also characterised by lower levels of genetic diversity at higher latitudes. High- and moderate-resolution Lagrangian models did not significantly predict regional-scale genetic connectivity across the Pilbara and Ningaloo (500 km). Although the high-resolution model explained an order of magnitude more genetic variation, suggesting model resolution and resolving coastal processes are important. Over broad spatial scales (nearly 2000 km), all moderate-resolution model particle release durations significantly predicted the genetic differentiation, although over-water distance best-predicted the genetic distance across this spatial scale. This study improves the understanding of connectivity in this region by focusing on a stress-tolerant species incorporating a spatially more intensive sampling effort than previous coral studies. It also shows that further development of Lagrangian models is required, such as inclusion of multi-generational stepwise models and larval behaviour, to improve predictions of connectivity for this coral species in this region.

Dredging can have significant impacts on aquatic environments, but the direct effects on fish have not been critically evaluated. Here, a meta-analysis following a conservative approach is used to understand how dredging-related stressors, including suspended sediment, contaminated sediment, hydraulic entrainment and underwater noise, directly influence the effect size and the response elicited in fish across all aquatic ecosystems and all life-history stages. This is followed by an in-depth review summarizing the effects of each dredging-related stressor on fish. Across all dredging-related stressors, studies that reported fish mortality had significantly higher effect sizes than those that describe physiological responses, although indicators of dredge impacts should endeavour to detect effects before excessive mortality occurs. Studies examining the effects of contaminated sediment also had significantly higher effect sizes than studies on clean sediment alone or noise, suggesting additive or synergistic impacts from dredging-related stressors. The early life stages such as eggs and larvae were most likely to suffer lethal impacts, while behavioural effects were more likely to occur in adult catadromous fishes. Both suspended sediment concentration and duration of exposure greatly influenced the type of fish response observed, with both higher concentrations and longer exposure durations associated with fish mortality. The review highlights the need for in situ studies on the effects of dredging on fish which consider the interactive effects of multiple dredging-related stressors and their impact on sensitive species of ecological and fisheries value. This information will improve the management of dredging projects and ultimately minimize their impacts on fish.

Quantifying the role of biophysical and anthropogenic drivers of coral reef ecosystem processes can inform management strategies that aim to maintain or restore ecosystem structure and productivity. However, few studies have examined the combined effects of multiple drivers, partitioned their impacts, or established threshold values that may trigger shifts in benthic cover. Inshore fringing reefs of the Great Barrier Reef Marine Park (GBRMP) occur in high-sediment, high-nutrient environments and are under increasing pressure from multiple acute and chronic stressors. Despite world-leading management, including networks of no-take marine reserves, relative declines in hard coral cover of 40–50% have occurred in recent years, with localized but persistent shifts from coral to macroalgal dominance on some reefs. Here we use boosted regression tree analyses to test the relative importance of multiple biophysical drivers on coral and macroalgal cover using a long-term (12–18 yr) data set collected from reefs at four island groups. Coral and macroalgal cover were negatively correlated at all island groups, and particularly when macroalgal cover was above 20%. Although reefs at each island group had different disturbance-and-recovery histories, degree heating weeks (DHW) and routine wave exposure consistently emerged as common drivers of coral and macroalgal cover. In addition, different combinations of sea-surface temperature, nutrient and turbidity parameters, exposure to high turbidity (primary) floodwater, depth, grazing fish density, farming damselfish density, and management zoning variously contributed to changes in coral and macroalgal cover at each island group. Clear threshold values were apparent for multiple drivers including wave exposure, depth, and degree heating weeks for coral cover, and depth, degree heating weeks, chlorophyll a, and cyclone exposure for macroalgal cover, however, all threshold values were variable among island groups. Our findings demonstrate that inshore coral reef communities are typically structured by broadscale climatic perturbations, superimposed upon unique sets of local-scale drivers. Although rapidly escalating climate change impacts are the largest threat to coral reefs of the GBRMP and globally, our findings suggest that proactive management actions that effectively reduce chronic stressors at local scales should contribute to improved reef resistance and recovery potential following acute climatic disturbances.

The worlds' coral reefs are declining due to the combined effects of natural disturbances and anthropogenic pressures including thermal coral bleaching associated with global climate change. Nearshore corals are receiving increased anthropogenic stress from coastal development and nutrient run-off. Considering forecast increases in global temperatures, greater understanding of drivers of recovery on nearshore coral reefs following widespread bleaching events is required to inform management of local stressors. The west Pilbara coral reefs, with cross-shelf turbidity gradients coupled with a large nearby dredging program and recent history of repeated coral bleaching due to heat stress, represent an opportune location to study recovery from multiple disturbances. Mean coral cover at west Pilbara reefs was monitored from 2009 to 2018 and declined from 45% in 2009 to 5% in 2014 following three heat waves. Recruitment and juvenile abundance of corals were monitored from 2014 to 2018 and were combined with biological and physical data to identify which variables enhanced or hindered early-stage coral recovery of all hard corals and separately for the acroporids, the genera principally responsible for recovery in the short-term (

Coral reefs are periodically impacted by disturbance events that reduce live coral cover and habitat complexity, with concomitant effects on fish assemblage structure. While the density of some fish species may increase following coral loss, most species decline. Determining which species are ‘winners’ and ‘losers’ following disturbances is fundamental to inform projections of future reef community structure, biodiversity, and productivity. Here, we analyse a long-term (2006–2018), spatially extensive (≈ 700 km) ‘natural experiment’ in which the responses of 11 wrasse taxa to acute disturbance events and no-take marine reserve (NTMR) protection were quantified on fringing coral reefs in the Palm (18°34′ S, 146°29′ E), Whitsunday (20°08′ S, 148°56′ E), and Keppel Island (23°10′ S, 150°57′ E) groups, Great Barrier Reef, Australia. The responses of wrasse densities to benthic habitat change were taxa specific and temporally consistent. Disturbance-mediated reductions in live hard coral cover and/or habitat complexity resulted in density declines for Hemigymnus melapterus, Hemigymnus fasciatus, Cheilinus fasciatus, Labroides spp., Oxycheilinus digramma, and Thalassoma spp. Conversely, Halichoeres spp. densities correlated positively with increased relative cover of sand and rubble, while Stethojulis spp., Anampses spp., Epibulus insidiator, and Bodianus spp. displayed variable responses to habitat changes. No wrasses exhibited an NTMR effect and predator density, irrespective of NTMR status, only influenced five taxa across all island groups. The lack of NTMR effects and variable top-down predator effects suggest that taxa-specific benthic habitat associations were the predominant drivers of wrasse densities on inshore GBR reefs.

High densities of the corallivorous crown-of-thorns starfish (CoTS; Acanthaster sp.) have occurred throughout the Indo-Pacific often resulting in widespread coral loss. Whilst CoTS have previously been recorded at Barrow and the Montebello Islands, in the Pilbara offshore bioregion of northwestern Australia, their densities were relatively low. Outbreak densities of CoTS have been described as the level at which the rate of coral consumption by the starfish is equal to or greater than rate at which the coral grows. In 2014, we recorded densities as high as 320 ± 58 CoTS ha−1 in the region which is well above recognised outbreak densities. Whilst there is little terrestrial runoff and agriculture in the Pilbara region, both temperature and chlorophyll-α levels appear to be sufficient to allow a high degree of CoTS larval success in most years. The region was subjected to anomalously high water temperatures during the summers of 2010–2011 and particularly 2012–2013 which resulted in the mortality of almost 70% of live coral. We hypothesise that the high densities of CoTS observed are a result of CoTS responding to a reduced food supply and aggregating around the remaining live coral resulting in outbreak densities rather than a significant increase in the number of CoTS in the area. The small amount of remaining live coral is concentrated in a few areas and this, combined with high densities of CoTS in these areas, suggest that CoTS represent a significant threat to the recovery of the coral communities of the region.

Regulation of organelle transport by the cytoskeleton is fundamental for eukaryotic survival. Cytoskeleton motors are typically modular proteins with conserved motor and diverse cargo binding domains. Motor:cargo interactions are often indirect and mediated by adaptor proteins e.g. Rab GTPases. Rab27a, via effector melanophilin (Mlph), recruits myosin-Va to melanosomes and thereby disperses them into melanocytes dendrites. To better understand how adaptors regulate motor:cargo interaction we used single melanosome fluorescence recovery after photo-bleaching (smFRAP) to characterise the association kinetics between myosin-Va, its adaptors and melanosomes. We found that myosin-Va and Mlph rapidly recovered after photo-bleaching, while Rab27a did not, indicating that myosin-Va and Mlph dynamically associate with melanosomes and Rab27a does not. This suggests that dynamic Rab27a:effector interaction rather than Rab27a melanosome:cytosol cycling regulates myosin-Va:melanosome association. Accordingly a Mlph-Rab27a fusion protein reduced myosin-Va smFRAP, indicating that it stabilised melanosomal myosin-Va. Finally, we tested the functional importance of dynamic myosin-Va:melanosome interaction. We found that while a myosin-Va-Rab27a fusion protein dispersed melanosomes in myosin-Va deficient cells, dendrites were significantly less elongated than in wild-type cells. Given that dendrites are the prime sites of melanosome transfer from melanocytes to keratinocytes we suggest that dynamic myosin-Va:melanosome interaction is important for pigmentation in vivo. Seagrasses are important habitats providing many ecological services. Most species have broad distributions with maximum dispersal distances of 100's of kms, however there is limited understanding of dispersal distances of colonising species like Halodule uninervis. It commonly grows in disturbed environments and could disperse to other meadows via clonal fragments. Effective conservation management requires greater understanding of genetic structure, dispersal barriers, and connectivity timescales to predict recovery following disturbance. Despite fragment viability of up to 28 days in a congenera, this theory remains untested in situ. Using 80 neutral single nucleotide polymorphisms, we investigated genetic diversity, gene flow patterns and structure among 15 populations of H. uninervis along 2000 km of Western Australian coastline. These data were combined with a multi-generational oceanographic dispersal model and a barrier dispersal analysis to identify dispersal barriers and determine which fragment dispersal duration (FDD) and timescale over which stepping-stone connectivity occurred, best matched the observed genetic structure. The 2-7 day FDD best matched the genetic structure with 4–12 clusters, with barriers to dispersal that persisted for up to 100 years. Modelling suggested greater fragmentation of metapopulations towards the southern edge of the species distribution, but genetic diversity did not decline. Several long-term boundaries were identified even with fragment viability of up to 28 days. This suggests H. uninervis dispersal is spatially limited by factors like oceanographic features and habitat continuity which may limit dispersal of this species. This study reiterates that potential dispersal does not equal realised dispersal, and management scales of 10's of kilometers are required to maintain existing meadows. Recruitment from distances further than this scale are unlikely to aid recovery after extreme disturbance events, particularly towards the range edge of H. uninervis distribution.

Metro market real estate cycles for office, industrial, retail, apartment, and hotel properties may be specified as first order Markov chains, which allow analysts to use a well-developed application, ¡§staying time¡¨. Anticipations for time spent at each cycle point are consistent with the perception of analysts that these cycle changes speed up, slow down, and pause over time. We find that these five different property types in U.S. markets appear to have different first order Markov chain specifications, with different staying time characteristics. Each of the five property types have their longest mean staying time at the troughs of recessions. Moreover, industrial and office markets have much longer mean staying times in very poor trough conditions. Most of the shortest mean staying times are in hyper supply and recession phases, with the range across property types being narrow in these cycle points. Analysts and investors should be able to use this research to better estimate future occupancy and rent estimates in their discounted cash flow (DCF) models.

Pocillopora damicornis is one of the most extensively studied coral species globally, but high levels of phenotypic plasticity within the genus make species identification based on morphology alone unreliable. As a result, there is a compelling need to develop cheap and time-effective molecular techniques capable of accurately distinguishing P. damicornis from other congeneric species. Here, we develop a fluorescence-based quantitative real-time PCR (qPCR) assay to genotype a single nucleotide polymorphism that accurately distinguishes P. damicornis from other morphologically similar Pocillopora species. We trial the assay across colonies representing multiple Pocillopora species and then apply the assay to screen samples of Pocillopora spp. collected at regional scales along the coastline of Western Australia. This assay offers a cheap and time-effective alternative to Sanger sequencing and has broad applications including studies on gene flow, dispersal, recruitment and physiological thresholds of P. damicornis.

Coral reefs have been heavily affected by elevated sea-surface temperature (SST) and coral bleaching since the late 1980s; however, until recently coastal reefs of north-western Australia have been relatively unaffected compared to Timor Sea and eastern Australian reefs. We compare SST time series with changes in coral cover spanning a period of up to 36 years to describe temporal and spatial variability in bleaching and associated coral mortality throughout the Pilbara–Ningaloo region. Declines in coral cover ranged from 12.5 to 51.3%, with relative declines ranging from 38 to 92%. Since 2013, coral cover throughout the region has declined to historically low levels at four of five subregions, with impaired recovery occurring at two subregions. Observations are consistent with global trends of repeated severe heat waves, coral bleaching and acute declines in coral cover. Locations within this study region have already experienced multiple coral-bleaching events within a period of less than 5 years. There is a high likelihood that reefs in the western Pilbara and northern Ningaloo regions will experience more frequent marine heatwaves, coral bleaching and mortality events in the future. Action, therefore, needs to be taken now to support the resilience of coral reef ecosystems in the region, which is arguably the most important coral-reef province on Australia’s western coast.

Hypoglycemia is a common iatrogenic consequence of type 1 diabetes therapy that can lead to central nervous system injury and even death if untreated. In the absence of clinically effective neuroprotective drugs we sought to quantify the putative neuroprotective effects of imposing hypothermia during the reperfusion phase following aglycemic exposure to central white matter. Mouse optic nerves (MONs), central white matter tracts, were superfused with oxygenated artificial cerebrospinal fluid (aCSF) containing 10 mmol/L glucose at 37°C. The supramaximal compound action potential (CAP) was evoked and axon conduction was assessed as the CAP area. Extracellular lactate was measured using an enzyme biosensor. Exposure to aglycemia, simulated by omitting glucose from the aCSF, resulted in axon injury, quantified by electrophysiological recordings, electron microscopic analysis confirming axon damage, the extent of which was determined by the duration of aglycemia exposure. Hypothermia attenuated injury. Exposing MONs to hypothermia during reperfusion resulted in improved CAP recovery compared with control recovery measured at 37°C, an effect attenuated in alkaline aCSF. Hypothermia decreases pH implying that the hypothermic neuroprotection derives from interstitial acidification. These results have important clinical implications demonstrating that hypothermic intervention during reperfusion can improve recovery in central white matter following aglycemia.

Understanding the spatial and temporal distribution of coral assemblages and the processes structuring those patterns is fundamental to managing reef assemblages. Cross-shelf marine systems exhibit pronounced and persistent gradients in environmental conditions, however, these gradients are not always reliable predictors of coral distribution or the degree of stress that corals are experiencing. This study used information from government, industry and scientific datasets spanning 1980&ndash, 2017, to explore temporal trends in coral cover in the geographically complex system of the Dampier Archipelago, northwest Australia. Coral composition at 15 sites surveyed in 2017 was also modelled against environmental and spatial variables (including turbidity, degree heat weeks, wave exposure, and distance to land/mainland/isobath) to assess their relative importance in structuring coral assemblages. High spatial and temporal heterogeneity was observed in coral cover and recovery trajectories, with reefs located an intermediate distance from the shore maintaining high cover over the past 20 years. The abundance of some prominent genera in 2017 (Acropora, Porites, and Turbinaria spp.) decreased with the distance from the mainland, suggesting that inshore processes play an important role in dictating the distribution of these genera. The atypical distributions of these key reef-building corals and spatial heterogeneity of historical recovery trajectories highlight the risks in making assumptions regarding cross-shelf patterns in geographically complex systems.

Executive Summary Adding a stochastic element to a well-understood real estate cycle model offers opportunities like those seen in earlier such syntheses of real estate analysis and statistics. The discrete real estate cycle points in the model require a discrete probability model, here a first order Markov chain. Many statistical applications flow from the combined model. Three Markov chain count variables have obvious real estate cycle appeal. Staying time, first recurrence time, and first passage time already exist in the Markov chain literature but only staying time is in the real estate cycle literature. The most fundamental innovation is in probabilistic forecasting. Being able to describe real estate cycle risk, cycle point by cycle point many quarters ahead, could improve evaluation of prospects for property disposal. It is also a simple spreadsheet application to describe real estate cycle risks that influence cash flows from operations across four-quarter spans.

Executive Summary. Relatively low-level Markov chain methods and widely available information allow this extension of real estate cycle risk analysis to office portfolios across cities initially in different cycle conditions. Examples include evaluation of cycle conditions at the end of a holding period and for cash flows from operations across a span of quarters. Standard spreadsheet functions serve to provide examples of changes in real estate cycle prospects, including before/after changes in portfolio weights, applying mean-variance dominance, mean-semivariance dominance, and stochastic dominance analysis.

Understanding the drivers of dispersal among populations is a central topic in marine ecology and fundamental for spatially explicit management of marine resources. The extensive coast of Northwestern Australia provides an emerging frontier for implementing new genomic tools to comparatively identify patterns of dispersal across diverse and extreme environmental conditions. Here, we focused on the stripey snapper (Lutjanus carponotatus), which is important to recreational, charter-based and customary fishers throughout the Indo-West Pacific. We collected 1,016 L. carponotatus samples at 51 locations in the coastal waters of Northwestern Australia ranging from the Northern Territory to Shark Bay and adopted a genotype-by-sequencing approach to test whether realized connectivity (via larval dispersal) was related to extreme gradients in coastal hydrodynamics. Hydrodynamic simulations using CONNIE and a more detailed treatment in the Kimberley Bioregion provided null models for comparison. Based on 4,402 polymorphic single nucleotide polymorphism loci shared across all individuals, we demonstrated significant genetic subdivision between the Shark Bay Bioregion in the south and all locations within the remaining, more northern bioregions. More importantly, we identified a zone of admixture spanning a distance of 180 km at the border of the Kimberley and Canning bioregions, including the Buccaneer Archipelago and adjacent waters, which collectively experiences the largest tropical tidal range and some of the fastest tidal currents in the world. Further testing of the generality of this admixture zone in other shallow water species across broader geographic ranges will be critical for our understanding of the population dynamics and genetic structure of marine taxa in our tropical oceans.

Objective: Hypoglycemia is a common adverse event and can injure central nervous system (CNS) white matter (WM). We determined if glutamate receptors were involved in hypoglycemic WM injury. Methods: Mouse optic nerves (MON), CNS WM tracts, were maintained at 37°C with oxygenated artificial cerebrospinal fluid (ACSF) containing 10 mM glucose. Aglycemia was produced by switching to 0 glucose ACSF. Supra-maximal compound action potentials (CAPs) were elicited using suction electrodes and axon function was quantified as the area under the CAP. Amino acid release was measured using HPLC. Extracellular [lactate] was measured using an enzyme electrode. Results: About 50% of MON axons were injured after 60 min of aglycemia (90% after 90 min); injury was not affected by animal age. Blockade of NMDA-type glutamate receptors improved recovery after 90 min of aglycemia by 250%. Aglycemic injury was increased by reducing [Mg2+]o or increasing [glycine]o, and decreased by lowering pHo, expected results for NMDA receptor-mediated injury. Extracellular pH increased during aglycemia, due to a drop in [lactate-]o. Aglycemic injury was dramatically reduced in the absence of [Ca2+]o. Extracellular aspartate, a selective NMDA receptor agonist, increased during aglycemia. Interpretation: Aglycemia injured WM by a unique excitotoxic mechanism involving NMDA receptors (located primarily on oligodendrocytes). During WM aglycemia, the selective NMDA agonist, aspartate, is released, probably from astrocytes. Injury is mediated by Ca2+ influx through aspartate-activated NMDA receptors made permeable by an accompanying alkaline shift in pHo caused by a fall in [lactate-]o. These insights have important clinical implications.

Severe climatic disturbance events often have major impacts on coral reef communities, generating cycles of decline and recovery, and in some extreme cases, community-level phase shifts from coral- to algal-dominated states. Benthic habitat changes directly affect reef fish communities, with low coral cover usually associated with low fish diversity and abundance. No-take marine reserves (NTRs) are widely advocated for conserving biodiversity and enhancing the sustainability of exploited fish populations. Numerous studies have documented positive ecological and socio-economic benefits of NTRs; however, the ability of NTRs to ameliorate the effects of acute disturbances on coral reefs has seldom been investigated. Here, we test these factors by tracking the dynamics of benthic and fish communities, including the important fishery species, coral trout (Plectropomus spp.), over 8 years in both NTRs and fished areas in the Keppel Island group, Great Barrier Reef, Australia. Two major disturbances impacted the reefs during the monitoring period, a coral bleaching event in 2006 and a freshwater flood plume in 2011. Both disturbances generated significant declines in coral cover and habitat complexity, with subsequent declines in fish abundance and diversity, and pronounced shifts in fish assemblage structure. Coral trout density also declined in response to the loss of live coral, however, the approximately 2:1 density ratio between NTRs and fished zones was maintained over time. The only post-disturbance refuges for coral trout spawning stocks were within the NTRs that escaped the worst effects of the disturbances. Although NTRs had little discernible effect on the temporal dynamics of benthic or fish communities, it was evident that the post-disturbance refuges for coral trout spawning stocks within some NTRs may be critically important to regional-scale population persistence and recovery.

Summary In animal cells, microtubule and actin tracks and their associated motors (dynein, kinesin, and myosin) are thought to regulate long- and short-range transport, respectively [1–8]. Consistent with this, microtubules extend from the perinuclear centrosome to the plasma membrane and allow bidirectional cargo transport over long distances (>1 μm). In contrast, actin often comprises a complex network of short randomly oriented filaments, suggesting that myosin motors move cargo short distances. These observations underpin the “highways and local roads” model for transport along microtubule and actin tracks [2]. The “cooperative capture” model exemplifies this view and suggests that melanosome distribution in melanocyte dendrites is maintained by long-range transport on microtubules followed by actin/myosin-Va-dependent tethering [5, 9]. In this study, we used cell normalization technology to quantitatively examine the contribution of microtubules and actin/myosin-Va to organelle distribution in melanocytes. Surprisingly, our results indicate that microtubules are essential for centripetal, but not centrifugal, transport. Instead, we find that microtubules retard a centrifugal transport process that is dependent on myosin-Va and a population of dynamic F-actin. Functional analysis of mutant proteins indicates that myosin-Va works as a transporter dispersing melanosomes along actin tracks whose +/barbed ends are oriented toward the plasma membrane. Overall, our data highlight the role of myosin-Va and actin in transport, and not tethering, and suggest a new model in which organelle distribution is determined by the balance between microtubule-dependent centripetal and myosin-Va/actin-dependent centrifugal transport. Macroalgal fields are a feature of the shallow tropical benthos, yet their importance for coral reef fish population dynamics remains poorly understood. The abundance of fish recruits was recorded using underwater visual census at six macroalgal and 11 coral reef sites in the Montebello and Barrow Islands. Surveys identified 6,935 individual recruit fish from 105 species, 54 genera and 20 families. Of these, 1,401 recruits from 48 species, 31 genera and 14 families were observed in macroalgal sites. Sixteen of the 105 recruit species (15.2 %) were observed exclusively at macroalgal sites. Forty-two (87.5 %) of these species have been observed as adults on adjacent coral reefs. Species composition of fish recruits differed significantly between the two habitats. Corallivore, small omnivore and zooplanktivore recruits had significantly higher numbers in the coral sites, while the results clearly demonstrate that juveniles, within the genera Lethrinus and Choerodon, as well as large algal croppers, are predominantly found at macroalgal (74–100 %) rather than coral-dominated sites. High-canopy macroalgae cover was positively correlated with abundance of these taxa, particularly Lethrinids (r 2 = 0.40). This study is the first to highlight the important attributes of tropical macroalgal fields and suggests that they have a similar role to seagrass meadows as essential juvenile habitat, thus warranting greater attention in conservation planning and ecological studies.

No-take marine reserves are widely advocated as a means to conserve biodiversity and sustain fisheries. Within adequately protected reserves, the abundance, mean size and age of targeted fish and invertebrate species are often increased. However, the magnitude and rate of recovery within reserves varies among locations and species, and increases in abundance in reserves are not universal. One potential contributing factor to variability in reserve performance is spatial variability in the supply of recruits. Many reef fish species exhibit disproportionate levels of recruitment in relatively small areas (so-called "recruitment hotspots"). Here we tested the hypothesis that the presence of recruitment hotspots enhances the effectiveness of reserves for two important fishery species, coral trout (Plectropomus maculatus) and stripey snapper (Lutjanus carponotatus), on coral reefs of the Keppel Island group, Great Barrier Reef. To do this we compared fish densities in areas that did or did not contain previously identified recruitment hotspots, both inside and outside reserves. For both species, the mean densities of adult fish above the minimum legal size were approximately three times as high in reserves with hotspots than in reserves without hotspots. Furthermore, the mean densities in reserves without hotspots were similar to those in nearby areas that were open to fishing. In contrast, the densities of sub-legal size individuals of both species were primarily explained by the presence of recruitment hotspots, irrespective of reserve protection. Our results suggest that identifying and incorporating recruitment hotspots into the selection of sites for reserves could enhance both conservation and fisheries objectives. © 2013 Elsevier Ltd.

No-take marine reserves (NTMRs) are being implemented for various conservation and fishery management objectives. Well-designed ecological monitoring programs should accompany the implementation of NTMRs to assess their performance against stated objectives. However, monitoring programs often have limited funding and limited access to experienced (“expert”) underwater visual census (UVC) observers. Consequently, use of non-expert, volunteer observers to collect marine ecological data has increased. The need to assess the quality and utility of non-expert data has been acknowledged. Few studies have directly compared expert and non-expert assessments, and these are generally one-point-in-time comparisons. The geographically isolated, environmentally unique and biologically diverse coral reefs of north-west Australia are under threat from climate change and other localised human impacts. Similar to many other regions around the world, effective mitigation of these threats through ecosystem-based marine spatial planning is currently constrained by a lack of knowledge of the extent and direction of dispersal of eggs, larvae, recruits, juveniles or adults among the coral reefs (termed population connectivity). Here, we present the outcomes of a series of consultations between Marine Protected Area planners, managers and scientists aimed at facilitating the integration of this connectivity knowledge into effective management guidelines and policy in north-west Australia. Through this process, we first synthesised current knowledge of population connectivity in the fields of oceanography, larval biology and population genetics, and then identified and answered to the best of our ability the most useful questions for the spatial planning of Marine Protected Area networks. Key findings indicate that these systems are likely ecologically independent for many coral reef organisms, with hard corals exhibiting the most localised dispersal of species studied so far. Thus, given that hard corals are also the habitat forming species, and that more widely dispersing species such as fish are likely less vulnerable to small scale disturbance, we propose that no-take areas that facilitate resilience of hard coral populations should maximise biodiversity more generally. This means no-take areas that are large enough to encompass routine dispersal distances of corals (10–20 km), and are spaced at similar distances, will not only maintain self-replenishment, but also aid recovery after disturbance through connectivity between no-take areas. The results can be applied in a regional and wider context, and provide a valuable template for transfer of scientific knowledge into effective policy.

We studied the roles of glycogen in axonal pathways of the central nervous system (CNS) and peripheral nervous system (PNS). By using electrophysiological recordings, in combination with biochemical glycogen assay, it was possible to determine whether glycogen was crucial to axon function under different conditions. Glycogen was present both in mouse optic nerve (MON) and in mouse sciatic nerve (MSN). Aglycemia caused loss of the compound action potential (CAP) in both pathways after a latency of 15 min (MON) and 120 min for myelinated axons (A fibers) in the MSN. With the exception of unmyelinated axons (C fibers) in the MSN, CAP decline began when usable glycogen was exhausted. Glycogen was located in astrocytes in the MON and in myelinating Schwann cells in the MSN; it was absent from the Schwann cells surrounding unmyelinated C fibers. In MON, astrocytic glycogen is metabolized to lactate and “shuttled” to axons to support metabolism. The ability of lactate to support A fiber conduction in the absence of glucose suggests a common pathway in both the CNS and the PNS. Lactate is released from MON and MSN in substantial quantities. That lactate levels fall in MSN in the presence of diaminobenzidine, which inhibits glycogen phosphorylase, strongly suggests that glycogen metabolism contributes to lactate release under resting conditions. Glycogen is a “backup” energy substrate in both the CNS and the PNS and, beyond sustaining excitability during glucose deprivation, has the capacity to subsidize the axonal energy demands during times of intense activity in the presence of glucose. © 2013 Wiley Periodicals, Inc.

Microtubules and F-actin, and associated motor proteins, are considered to play complementary roles in long- and short-range organelle transport. However, there is growing appreciation that myosin/F-actin networks can drive long-range transport. In melanocytes myosin-Va and kinesin-1 have both been proposed as long-range centrifugal melanosome transporters. Here we investigated the role of kinesin-1 heavy chain (Kif5b) and its suggested targeting factor Rab1a in transport. Using confocal microscopy and sub-cellular fractionation we did not detect Kif5b or Rab1a on melanosomes. Meanwhile functional studies, using siRNA knockdown and dominant negative mutants, did not support a role for Kif5b or Rab1a in melanosome transport. To probe the potential of Kif5b to function in transport we generated fusion proteins that target active Kif5b to melanosomes and tested their ability to rescue perinuclear clustering in myosin-Va deficient cells. Expression of these chimeras, but not full length Kif5b, dispersed melanosomes with similar efficiency to myosin-Va. Our data indicate that kinesin/MT can compensate for defects in myosin-Va/actin-based transport in mammals but that endogenous Kif5b plays little role in transport in melanocytes due to its inefficient recruitment to melanosomes.

Global climate change poses a serious threat to the future health of coral reef ecosystems. This calls for management strategies that are focused on maximizing the evolutionary potential of coral reefs. Fundamental to this is an accurate understanding of the spatial genetic structure in dominant reef-building coral species. In this study, we apply a genotyping-by-sequencing approach to investigate genome-wide patterns of genetic diversity, gene flow, and local adaptation in a reef-building coral, Pocillopora damicornis, across 10 degrees of latitude and a transition from temperate to tropical waters. We identified strong patterns of differentiation and reduced genetic diversity in high-latitude populations. In addition, genome-wide scans for selection identified a number of outlier loci putatively under directional selection with homology to proteins previously known to be involved in heat tolerance in corals and associated with processes such as photoprotection, protein degradation, and immunity. This study provides genomic evidence for both restricted gene flow and local adaptation in a widely distributed coral species, and highlights the potential vulnerability of leading-edge populations to rapid environmental change as they are locally adapted, reproductively isolated, and have reduced levels of genetic diversity.

Marine reserves, areas closed to all forms of fishing, continue to be advocated and implemented to supplement fisheries and conserve populations [ [1], [2], [3] and [4]]. However, although the reproductive potential of important fishery species can dramatically increase inside reserves [ [5], [6], [7] and [8]], the extent to which larval offspring are exported and the relative contribution of reserves to recruitment in fished and protected populations are unknown [ [4], [9], [10] and [11]]. Using genetic parentage analyses, we resolve patterns of larval dispersal for two species of exploited coral reef fish within a network of marine reserves on the Great Barrier Reef. In a 1,000 km2 study area, populations resident in three reserves exported 83% (coral trout, Plectropomus maculatus) and 55% (stripey snapper, Lutjanus carponotatus) of assigned offspring to fished reefs, with the remainder having recruited to natal reserves or other reserves in the region. We estimate that reserves, which account for just 28% of the local reef area, produced approximately half of all juvenile recruitment to both reserve and fished reefs within 30 km. Our results provide compelling evidence that adequately protected reserve networks can make a significant contribution to the replenishment of populations on both reserve and fished reefs at a scale that benefits local stakeholders.

With rapidly increasing rates of contemporary extinction, predicting extinction vulnerability and identifying how multiple stressors drive non-random species loss have become key challenges in ecology. These assessments are crucial for avoiding the loss of key functional groups that sustain ecosystem processes and services. We developed a novel predictive framework of species extinction vulnerability and applied it to coral reef fishes. Although relatively few coral reef fishes are at risk of global extinction from climate disturbances, a negative convex relationship between fish species locally vulnerable to climate change vs. fisheries exploitation indicates that the entire community is vulnerable on the many reefs where both stressors co-occur. Fishes involved in maintaining key ecosystem functions are more at risk from fishing than climate disturbances. This finding is encouraging as local and regional commitment to fisheries management action can maintain reef ecosystem functions pending progress towards the more complex global problem of stabilizing the climate.

Lutjanus carponotatus ranges from the Indian Ocean to northern Australia and Papua New Guinea and is an important species for both commercial and recreational fisheries. Due to its life history characteristics and fishing pressures, L. carponotatus is vulnerable to overfishing and suitable management is essential. This phylogenetic and population genetic study assessed the population genetic structure of L. carponotatus at seven locations in three bioregions in Western Australia (WA) using the mitochondrial DNA control region. Analysis of molecular variance (AMOVA) indicated no genetic differentiation among locations within (oCT = 0.051) and among bioregions (oSC = −0.026). Phylogenetic analyses indicated no geographic partitioning within WA, though two distinct lineages were found. Within these two lineages there was no significant spatial partitioning, suggesting a panmictic population that can, under current conditions, be managed as a single stock. WA and Great Barrier Reef (GBR) population genetic data were then combined to determine the broad-scale connectivity between the east and west coast of Australia. L. carponotatus from the GBR did differ significantly (FST = 0.64, p = < 0.001) from the WA population, indicating the two populations do not exchange any migrants and should be managed as discrete stocks. As an indicator of population resilience, genetic variability was compared among WA and GBR populations of L. carponotatus and we show that the WA population has almost three times the genetic variability of the GBR population, suggesting that it will be better able to adapt to environmental change expected during the 21st century.

Accelerating coastal development and shipping activities dictate that dredging operations will intensify, increasing potential impacts to fishes. Coastal fishes have high economic, ecological, and conservation significance and there is a need for evidence-based, quantitative guidelines on how to mitigate the impacts of dredging activities. We assess the potential risk from dredging to coastal fish and fisheries on a global scale. We then develop quantitative guidelines for two management strategies: threshold reference values and seasonal restrictions. Globally, threatened species and nearshore fisheries occur within close proximity to ports. We find that maintaining suspended sediment concentrations below 44 mg/L (15–121 bootstrapped CI) and for less than 24 hours would protect 95% of fishes from dredging-induced mortality. Implementation of seasonal restrictions during peak periods of reproduction and recruitment could further protect species from dredging impacts. This study details the first evidence-based defensible approach to minimize impacts to coastal fishes from dredging activities.

Partnerships between scientists and local communities can increase research capacity and data delivery while improving management effectiveness through enhanced community participation. To encourage such collaboration, this study demonstrates how these partnerships can be formed, drawing on two case studies in coral reef ecosystems in very different social settings (Papua New Guinea and Australia). In each case, steps towards successfully engaging communities in research were similar. These included: (1) early engagement by collaborating organizations to build trust, (2) ensuring scientific questions have direct relevance to the community, (3) providing appropriate incentives for participation, and (4) clear and open communication. Community participants engaged in a variety of research activities, including locating and capturing fishes, collecting and recording data (weight, length and sex), applying external tags, and removing otoliths (ear bones) for ageing and elemental analysis. Research partnerships with communities enhanced research capacity, reduced costs and, perhaps more importantly, improved the likelihood of long-term community support for marine protected areas (MPAs).

The stimulus-evoked compound action potential (CAP) recorded from rodent optic nerve is polyphasic in profile, with total area under the CAP an index of nerve function. A decrease in CAP area signifies a decrease in the number of axons contributing to the CAP. A disadvantage of considering the CAP a uniform entity is that any distinctions between the axon-subpopulations contributing to the individual peaks go undetected. We illustrate two instances that demonstrate the advantages of resolving the CAP into its constituent peaks. The individual peaks of the CAP were quantified using curve-fitting procedures to describe the CAP as the sum of multiple Gaussian functions. The first example illustrates that the individual peaks comprising the CAP exhibit differential sensitivities to glucopenia, suggesting that the axons within the nerve cannot be considered a homogeneous population with regard to their metabolic characteristics. The second example illustrates that the complex waveform of the CAP recorded from the optic nerve of the hypomyelinated rumpshaker mutant mouse, comprising both positive and negative overlapping peaks, can be resolved into five individual peaks. In conclusion we show that assessing each individual peak of the CAP is a powerful analytic tool for identifying heterogeneous profiles of axon-subpopulations in the rodent optic nerve.

The Great Barrier Reef (GBR) provides a globally significant demonstration of the effectiveness of large-scale networks of marine reserves in contributing to integrated, adaptive management. Comprehensive review of available evidence shows major, rapid benefits of no-take areas for targeted fish and sharks, in both reef and nonreef habitats, with potential benefits for fisheries as well as biodiversity conservation. Large, mobile species like sharks benefit less than smaller, site-attached fish. Critically, reserves also appear to benefit overall ecosystem health and resilience: outbreaks of coral-eating, crown-of-thorns starfish appear less frequent on no-take reefs, which consequently have higher abundance of coral, the very foundation of reef ecosystems. Effective marine reserves require regular review of compliance: fish abundances in no-entry zones suggest that even no-take zones may be significantly depleted due to poaching. Spatial analyses comparing zoning with seabed biodiversity or dugong distributions illustrate significant benefits from application of best-practice conservation principles in data-poor situations. Increases in the marine reserve network in 2004 affected fishers, but preliminary economic analysis suggests considerable net benefits, in terms of protecting environmental and tourism values. Relative to the revenue generated by reef tourism, current expenditure on protection is minor. Recent implementation of an Outlook Report provides regular, formal review of environmental condition and management and links to policy responses, key aspects of adaptive management. Given the major threat posed by climate change, the expanded network of marine reserves provides a critical and cost-effective contribution to enhancing the resilience of the Great Barrier Reef.

The density and biomass of two coral reef fish species targeted by line fishers, Plectropomus maculatus and Lutjanus carponotatus, have increased within no-take marine protected areas on inshore reefs of the Great Barrier Reef (GBR). This phylogenetic and population genetic study, using mitochondrial DNA (mt DNA) from the control region, is a broad scale analysis of the genetic connectivity of P. maculatus (n = 164) and L. carponotatus (n = 188) within and between inshore islands of the GBR (latitudinal gradient 800 km) that contain the aforementioned no-take areas. Analysis of molecular variance (AMOVA) indicated high levels of gene exchange between locations within (P. maculatus oCT = −0.011; L. carponotatus oCT = 0.004) and between each island group (P. maculatus oSC = 0.003; L. carponotatus oSC = 0.003). Phylogenetic analysis showed no geographic partitioning but identified two distinct lineages for both species that were distributed throughout the sampled range, suggesting that both species are admixtures of differentiated lineages rather than stable populations. This study shows that populations of both species within the sampling range are panmictic and under current conditions, they may be managed as a single stock across the entire GBR. Thus, the study suggests that the island groups and the no-take marine protected areas within them are connected by larval transport to each other and to fished areas. This study cannot confirm if this connectivity is historical (evolutionary) or contemporary (ecological).

This study tested the efficiency of an underwater biopsy probe designed to remove tissue samples for molecular analysis from small- to medium-sized (15 to 70 cm) marine teleost fish in the field. The probe was fired at lower and higher power from a 76 cm rubber-propelled spear gun. Two species were targeted: Plectropomus maculatus and Lutjanus carponotatus. The results were highly successful for P. maculatus, with >80% of hits at lower and higher power resulting in sufficient tissue extraction for molecular analysis. The results were less consistent for L. carponotatus at lower power (45%) due to the relatively large scales of the fish. At higher power the success rate increased to 73%. In 97% of cases, fish swam away rapidly with only minor lesions and showing no signs of incapacitation. Furthermore, aquarium trials showed no post-impact mortality after a 2 wk period. Therefore, the biopsy probe tested in this study is considered successful when the spear gun is fired at the correct power for the target species. This will allow for easier tissue sample collection from small- to medium-sized teleost fish for population and phylogenetic studies than previous methods of killing or capturing individuals.

This study investigated body size to fecundity relationships of a reef fish species targeted by line fishing, and examines the potential benefits of increased batch fecundity in no-take reserves compared to fished areas around the Palm, Whitsunday and Keppel Island Groups, Great Barrier Reef, Australia. Lutjanus carponotatus batch fecundity increased with fork length in a non-linear relationship that was best described by a power function. Batch fecundity differed by more than 100-fold among individuals, with a range from 7,074 to 748,957 eggs in fish ranging from 184 to 305 mm fork length. Furthermore, egg diameter increased with fish size. Based on underwater visual census, the potential batch fecundity per unit area in all three island groups ranged from 1.0 to 4.2 times greater in the no-take reserves than in the fished areas between 2001 and 2004. In 2002, a mean 2.3-fold difference in biomass between no-take reserves and fished areas converted to a mean 2.5-fold difference in batch fecundity per unit area. Greater batch fecundity, longer spawning seasons and potentially greater larval survival due to larger egg size from bigger individuals might significantly enhance the potential benefits of no-take marine reserves on the Great Barrier Reef.
