The aim was to evaluate the technological properties of films based on cassava starch/carboxymethyl cellulose added to soy lecithin and tocopherol mix. The films were prepared by casting from a cassava starch/carboxymethyl cellulose mixture and identified as control B (no soy lecithin and no tocopherol mix), L (with soy lecithin and no tocopherol mix) and LT and LT2 (with soy lecithin and tocopherol mix). Thickness, water solubility, water content, film weight, contact angle, permeability test, mechanical tests, antioxidant activity, biodegradability, color, thermogravimetry, and FTIR tests were performed. The results showed that the films containing soy lecithin (L, LT, and LT2) were more hydrophilic and had a denser structure than film B, but the LT and LT2 films were slightly yellow-green, malleable and showed few bubbles or cracks. In addition, all the films developed are biodegradable. It was also found that the antioxidant remained in the films to which it was added (LT and LT2) and that the LT2 film showed the highest antioxidant activity. Hydrogels consist of a network of highly porous polymeric chains with the potential for use as a wound dressing. Propolis is a natural product with several biological properties including anti-inflammatory, antibacterial and antioxidant activities. This study was aimed at synthesizing and characterizing a polyacrylamide/methylcellulose hydrogel containing propolis as an active ingredient, to serve as a wound dressing alternative, for the treatment of skin lesions. The hydrogels were prepared using free radical polymerization, and were characterized using scanning electron microscopy, infrared spectroscopy, thermogravimetry, differential scanning calorimetry, swelling capacity, mechanical and rheological properties, UV-Vis spectroscopy, antioxidant activity by the DPPH, ABTS and FRAP assays and biocompatibility determined in Vero cells and J774 macrophages by the MTT assay. Hydrogels showed a porous and foliaceous structure with a well-defined network, a good ability to absorb water and aqueous solutions simulating body fluids as well as desirable mechanical properties and pseudoplastic behavior. In hydrogels containing 1.0 and 2.5% propolis, the contents of total polyphenols were 24.74 ± 1.71 mg GAE/g and 32.10 ± 1.01 mg GAE/g and those of total flavonoids 8.01 ± 0.99 mg QE/g and 13.81 ± 0.71 mg QE/g, respectively, in addition to good antioxidant activity determined with all three methods used. Therefore, hydrogels containing propolis extract, may serve as a promising alternative wound dressing for the treatment of skin lesions, due to their anti-oxidant properties, low cost and availability. Species belonging to the Bauhinia genus, usually known as "pata-de-vaca", are popularly used to treat diabetes. Bauhinia ungulata var. obtusifolia (Ducke) Vaz is among them, of which the leaves are used as a tea for medicinal purposes in the Amazon region. A microencapsulation study of lyophilized aqueous extract from Bauhinia ungulata leaves, which contain phenolic compounds, using five different wall materials (maltodextrin DE 4-7, maltodextrin DE 11-14; β-cyclodextrin; pectin and sodium carboxymethylcellulose) is described in this paper. The microstructure, particle size distribution, thermal behavior, yield, and encapsulation efficiency were investigated and compared using different techniques. Using high-performance liquid chromatography, phenolics, and flavonoids were detected and quantified in the microparticles. The microparticles obtained with a yield and phenolics encapsulation efficiency ranging within 60–83% and 35–57%, respectively, showed a particle size distribution between 1.15 and 5.54 µm, spherical morphology, and a wrinkled surface. Among them, those prepared with sodium carboxymethylcellulose or pectin proved to be the most thermally stable. They had the highest flavonoid content (23.07 and 21.73 mg RUTE/g Extract) and total antioxidant activity by both the DPPH (376.55 and 367.86 µM TEq/g Extract) and ABTS (1085.72 and 1062.32 µM TEq/g Extract) assays. The chromatographic analyses allowed for quantification of the following substances retained by the microparticles, chlorogenic acid (1.74–1.98 mg/g Extract), p-coumaric acid (0.06–0.08 mg/g Extract), rutin (11.2–12.9 mg/g Extract), and isoquercitrin (0.49–0.53 mg/g Extract), compounds which considered to responsible for the antidiabetic property attributed to the species. Organogels have importance for topical applications because they can be used to deliver drugs in a controlled and prolonged fashion. These are materials consisting of a three-dimensional network of organic molecules dispersed in a solvent. Recent studies have demonstrated that the solvent could be replaced by oils from non-conventional biologic sources. There is a diversity of not-explored species in the Amazon that are promising sources of vegetable oils with a promising composition. This study developed an organogel with buriti (Mauritia flexuosa L.f) and cacay (Caryodendron orinocense Karst.) oils, using cetostearyl alcohol as an organogelator due to its compatibility, stability, security, affordability, and it is readily available. The oils were characterized, and the organogels were synthesized by studying their crystal evolution and oil-binding capacity. The microstructure was evaluated with polarized light microscopy, fractal dimension, FTIR spectroscopy, XRD, and thermal and rheological analyses. It was found that the critical gelation concentration was higher for cacay oil as it possessed a higher amount of polyunsaturated triacylglycerols. The crystals of the buriti organogel had a smaller lamellar shape, a greater surface area, and physical and thermal stability; although, it presented a slower crystal evolution due to the low number of minor compounds and a greater number of saturated triacylglycerols. The polar fraction of the organogelators as well as triacylglycerol and minor polar compounds are important in forming crystallization nuclei. Hydrogel is a biomaterial widely used in several areas of industry due to its great biocompatibility and adaptability to biological tissues. In Brazil, the Calendula plant is approved by the Ministry of Health as a medicinal herb. It was chosen to be incorporated in the hydrogel formulation because of its anti-inflammatory, antiseptic and healing effects. This study synthesized polyacrylamide hydrogel containing calendula extract and evaluated its efficiency as a bandage for wound healing. The hydrogels were prepared using free radical polymerization and characterized by Scanning Electron Microscopy, swelling analysis and mechanical properties by texturometer. The morphology of the matrices showed large pores and foliaceous structure. In vivo testing, as well as the evaluation of acute dermal toxicity, was conducted using male Wistar rats. The tests indicated efficient collagen fiber production, improved skin repair and no signs of dermal toxicity. The industrial processing of fruits in the Amazon region, such as tucuma, generates a large amount of coproducts with great nutritional potential. In this work, phenolic compounds from tucuma coproduct almonds were extracted and microencapsulated by spray drying using maltodextrin as the encapsulating agent and verified its antioxidant activity. Phenolic compounds were determined by UV spectroscopy and identified by Ultraefficiency Liquid Chromatography. Antioxidant activity was measured by ABTS and DPPH assay. Thermogravimetric techniques, infrared spectroscopy, scanning electron microscopy, moisture content and water activity were applied to characterize the microparticle. The crude extract and microparticle had total polyphenols of 135.1 mg/g ± 0.078 and 130.5 mg/g ± 0.024, respectively. Caffeic and gallic acids were identified. The crude extract and the microparticle showed good antioxidant activity by ABTS and DPPH assay, justified by the presence of the phenolic compounds found. The microparticle showed spherical and heterogeneous structures and good encapsulation efficiency from the spray drying process using maltodextrin. The results show that the extract of the tucuma almond coproduct can be used as a phenolic compound-rich source and microencapsulated with possible application for functional food production. New strategies for the delivery of bioactives in the deeper layers of the skin have been studied in recent years, using mainly natural ingredients. Among the strategies are organogels as a promising tool to load bioactives with different physicochemical characteristics, using vegetable oils. Studies have shown satisfactory skin permeation, good physicochemical stability mainly due to its three-dimensional structure, and controlled release using vegetable oils and low-molecular-weight organogelators. Within the universe of natural ingredients, vegetable oils, especially those from the Amazon, have a series of benefits and characteristics that make them unique compared to conventional oils. Several studies have shown that the use of Amazonian oils brings a series of benefits to the skin, among which are an emollient, moisturizing, and nourishing effect. This work shows a compilation of the main Amazonian oils and their nutraceutical and physicochemical characteristics together with the minority polar components, related to health benefits, and their possible effects on the synthesis of organogels for cosmetic purposes. Native Amazon palm trees have been target of research and product development especially because of their oil and fat. Tucumã-do-Pará (Astrocaryum vulgare) is a palm tree from which is possible to extract fruit seed fat. This article studied its physico-chemical characterization and analyzed its feasibility of use as raw material for nanostructured lipid nanoparticle (NLC) in pharmaceuticals. Through acidity, refraction and iodine indexes it was possible to determine that the fat has good quality and is in accordance with the literature. Ultraviolet spectrum scanning did not show peaks in region of dienes and trienes, compounds which appear when fat is decomposing. Infrared spectra and nuclear magnetic resonance determined spectral identity of the fat, confirming it is composed mainly by saturated groups. Fatty acid profile by gas chromatography coupled to mass spectrometer evidenced components in greater quantity: lauric and myristic acids in saturated part and oleic acid in unsaturated part. These acids have pharmacotechnical importance as permeation agents of active substances. Tucumã fat has thermal stability up to 350 °C and melting point of 35 °C. To reach nanostructured lipid carriers preparation it was necessary to increase its melting point by mixing it with carnauba wax. Through X-ray diffraction in crystallized tucumã fat/carnauba wax mixture it was possible to identify stable β′-form which is the ideal form for production of stable nanostructured lipid carriers. From this lipidic mixture of tucumã fat/carnauba wax in 1:1 ratio, 9 formulations of nanostructures lipid carriers were prepared, from which 3 formulations were distinguished by their excelente particle size and polydispersity index values. Cupuassu (Theobroma grandiflorum Schum.) is a typical Amazonian fruit, whose seed is used as raw material to produce cupulate. The by-product of its seeds, cupuassu seed by-product (CSB), is often discarded by the cosmetic and food industry, but studies showed that it has a high content of phenolic compounds with exploitable antioxidant activity. This study aimed to identify the classes of chemical compounds present in the extract of CSB by HPTLC, to confirm them by RMN 1H spectroscopy and to analyze the extract thermal analytical behavior in different processing stages. After elution of the extract in an isocratic system, TLC plates were developed with selective reagents for several classes of chemical compounds and antioxidant activity by the DPPH· method. Butter, CSB and crude extract presented mass losses characteristic of lipid decomposition reactions (92.63, 14.80, and 15.42%, respectively). Thermal degradation of bioactive and nutritional compounds (mainly polyunsaturated fatty acids) present in both CSB and crude extract has occurred at temperatures above 189 °C. Microencapsulation of the CBS extract by spray drying (MECoC) using maltodextrin as coating agent was shown to effectively protect the phenolic compounds. RMN 1H spectra confirmed HPTLC and FTIR results, which allowed us to conclude that the crude extract of the cupuassu by-product was mainly composed of terpenes, steroids and phenolic compounds, mainly flavonoids. These analytical techniques have proven to be effective tools for assessing the bioactive compounds incorporation into the polymeric matrix expected in the pre-formulation stage. Nanoemulsion‐based acai oil was obtained by the phase inversion method using two nonionic surfactants, Ceteareth‐20 (Brij™ CS20) and polyoxyethylene‐hydrogenated castor oil (Croduret™ 50), with the concentration of each surfactant varying from 7 to 10 %. The formulation with 10 % Brij CS20 presented the best values for droplet size, zeta potential, and polydispersity index (PDI). Ketoconazole was incorporated into this formulation, with an encapsulation efficiency of 98.31 % and equally good zeta potential, droplet size, and PDI, and spherical shape when observed by transmission electron microscopy. Molecular dynamics (MD) simulations of chitosan (CS) and albendazole (ABZ) were performed in the presence (BW1 BW2 and BW3) and absence (B1, B2 and B3) of water to evaluate CS/ABZ interactions in synthesis and solubility through solvation effect of nanoencapsulated ABZ. Molecular chemical reactivity was investigated through DFT-based reactivity descriptors, electrophilicity (ɷ), chemical potential (μ) and chemical hardness (η) as well as values of boundary molecular orbitals HOMO and LUMO that revealed interactions of the amino and hydroxyl groups of CS with groups of nitrogen, sulfur and ABZ ester bond. MD showed BW1 and BW2 with best Radial Distribution (RDF) profiles in aqueous medium as well as solvation energy indicating spontaneous formation of BW1 and BW2 nanoparticles. MD simulations demonstrate CS as a carrier capable of spontaneously encapsulating hydrophobic ABZ molecules in water-containing environments. Copaifera reticulata Ducke is a medicinal plants greatly employed in northern and northeastern Brazil. However, skin penetration is difficult when it is applied in natura. However, permeability increases through nanoformulations and phonophoresis and low frequency ultrasound that increases skin permeability of compounds. The effect of oil-resin of copaiba, in natura and in gel (nano-emulsion), associated to phonophoresis, was analyzed and compared for muscle repair in Wistar rats. Copaiba nano-emulsion was formulated by phase inversion. It was incorporated to hydrogel Carbopol® (20% nano-emulsion, 80% Carbopol®). Male animals (n=24) were divided into six groups and later underwent traumatic lesion of the gastrocnemius muscle. Muscle repair was analyzed by plasmatic dosage of aspartate aminotransferase (AST) and kinase creatine (CK). To better understand the biological properties of cupuassu seed residue resulting from fruit industrial processing, its antioxidant activity was determined by both the 2,2‐diphenyl‐1‐picrylhydrazyl and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) assays. Molecular modeling was used to correlate the antiradical power of eleven known cupuassu phenolic compounds with their structures. The extract antioxidant activity by both methods was very high compared with those of similar residues. The properties of the highest occupied and lowest unoccupied molecular orbitals indicate that the most energetically stable compounds possess the highest antioxidant activity. This was confirmed through the molecular electrostatic potential map by the nucleophilic surfaces on the oxygen atoms of carbonyl and hydroxyl groups. The concentrated extract of the industrial residue of cupuassu seeds, a popular Amazonian fruit, exhibits high antioxidant activity and antiradical power. Cocoa (Theobroma cacao L.) and its by-products are widely used by the pharmaceutical, cosmetics and food industries because they contain secondary metabolites with high levels of antioxidants. The objective of this study was to evaluate the thermal profile of cocoa by-products extract and its microparticles, obtained by spray drying to preserve its biological activity. The thermal analyses showed that the events of greater mass loss occurred in the temperature range of 407-455 °C (oil), 370-429 °C (cocoa waste), 175-302 °C (cocoa extract), 299-314 °C (microparticles) and 239-329 °C (microencapsulated extract). Through differential scanning calorimetry analysis, it was possible to detect a positive effect on microencapsulation of the extract, an increase in the stability of the active principles and similar bands both for the spectra of the extract and microparticles. The extract obtained, despite being derived from the solid residue (waste), showed high values of polyphenols content and antioxidant activity. The cocoa extract (Theobroma cacaoL.) has a significant amount of polyphenols (TP) with potent antioxidant activity (AA). This study aims to optimise microencapsulation of the extract of cocoa waste using chitosan and maltodextrin. Microencapsulation tests were performed according to a Box–Behnken factorial design, and the results were evaluated by response surface methodology with temperature, maltodextrin concentration (MD) and extract flowrate (EF) as independent variables, and the fraction of encapsulated TP, TP encapsulation yield,AA, yield of drying and solubility index as responses. The optimum conditions were: inlet temperature of 170 °C,MDof 5% andEFof 2.5 mL/min. HPLC analysis identified epicatechin as the major component of both the extract and microparticles. TP release was faster at pH 3.5 than in water. Hydrogels are three-dimensional hydrophilic polymeric networks, capable of absorbing large quantities of water and biological fluids and simulating biological tissue when swollen. Hydrogels are frequently explored for use in numerous biological and biomedical applications due to their desirable properties. Hydrogels are characterized as either synthetic, natural or hybrid, based on the nature of their constituent polymers. The use of natural polymers in hydrogels for biomedical applications is advantageous due to their biocompatibility, biodegradability and non-toxicity, whereas, synthetic polymers are hydrophobic, possessing strong covalent bonds within their matrix, which improves the mechanical strength, service life and absorbability of the gels. Their polymeric crosslinking structure defines their physical or chemical nature, while their polymeric composition indicates whether they are homopolymeric, copolymeric or multipolymeric. All the classifying properties of hydrogels affect their applicability and types of areas in which they can be incorporated. Chitosan has attracted much interest due to its special physical and chemical properties related to drug administration. Nanoparticles delivery systems from Vi Antigen are a promising approach in the struggle against typhoid fever. In this paper, we reported the obtainment and the characterization of Vi Antigen by Infrared spectroscopy as well as Molecular Modeling and Computational Chemistry studies of the Chitosan-Vi Antigen interaction through theoretical models. The results of the theoretical and experimental Infrared spectroscopy showed important bands related to N -Acetyl and O -Acetyl groups present in Vi Antigen. Important interactions related to its adsorption were observed through three-dimensional optimized structures. Two models were proposed for the Chitosan-Vi Antigen in adsorption system, one as a monomer and another as an optimized tetrasaccharide antigen. The Molecular Modeling studies presented the best conformation and binding site on the nanoparticle Chitosan-Vi Antigen in models proposed. The present study aimed at characterizing the oil extracted from Bertholletia excelsa H.B.K. almond, a native species from the Amazon region. Analytical methods used for oils and fats were employed through pharmacopoeia assays, AOCS (American Oil Chemists Society) standard methods as well as those recommended by ANVISA (National Health Surveillance Agency) such as acidity, peroxide value, saponification index, iodine value and refractive index, pH and relative density, and also thermoanalytical analyses (thermogravimetry, differential thermogravimetry and differential thermal analysis) as well as chromatographic analysis (gas chromatography). The characterization assessments of B. excelsa oil showed results indicating that the oil contains polyunsaturated fatty acids in large proportion. The termoanalytical tests indicated that B.excelsa oil showed thermal stability up to 220 °C, These results showed that the oil extracted from B. excelsa has acceptable characteristics and is of good quality. Background: Calendula officinalis L. (Asteraceae) is a Mediterranean specie, but in Europe and America it is cultivated for ornamental or medicinal purposes. This species is widely used for presenting activities, antiinflammatory antibacterial and antioxidant. However the therapeutic action is linked to the amount of assets of the extracted raw material. The extraction method of bioactive compounds is an important step in the manufacturing of herbal medicines, because secondary metabolites with therapeutic potential are usually found in small quantities in plant materials. Objective: Due the medical and commercial importance of C. officinalis, this study aimed to evaluate the impact of the extraction method on the quality of herbal extract and optimize the extraction of rutin from C. officinalis. Materials and Methods: The extraction of rutin was performed by ultrasound and shaker and the optimized conditions were determined by response surface methodology. Results: The results of ultrasound extraction assisted (UEA) and maceration dynamic (MD) showed that rutin yield ranged from 0.218 to 2.28% (w/w) when extract by ultrasound and 0.1-1.44% by MD. The optimal extraction condition for rutin (2.48% to UEA or 1.46% to MD) from C. officinalis by UEA or MD were a 19-22 min extraction, ethanol: water ratio of 35-40% and 0.05-0.056 mg/mL to raw material: solvent ratio. The industrial processing amazon fruits, like tucuma, generates a large amount of coproducts with large nutritional potential. Thus, this work obtained the oily extract of the tucuma almonds coproducts by green extraction using palm oil by the ultrasound method and then microencapsulated by atomization and verification of its antioxidant activity. Thermogravimetric techniques, infrared spectroscopy, scanning electron microscopy, moisture content, water activity were applied to characterize the microparticles. Total carotenoids were determined by UV spectroscopy and antioxidant activity was measured by 2,2′-azino-di-(3-ethylbenzthiazoline sulfonic acid and co-oxidation in the system β-carotene/linoleic acid. The oily extract and microparticle had total carotenoid contents of 3.305 mg/100 g ± 0.01 and 2.559 mg/100 g ± 0.01, respectively. The antioxidant activity assessed through the 2,2′-azino-di-(3-ethylbenzthiazoline sulfonic acid value was 584.75 μM/trolox ± 0.01 (oily extract) and 537.12 μM/trolox ± 0.01 (microparticle) were determined. In the system β-carotene/linoleic acid showed oxidation of 49.9% ± 1.8 lipophilic extract and 43.3% ± 2.3 microparticle. The results showed that the oily extract of the tucuma almond coproduct can be used as a carotenoid-rich source and microencapsuled with possible application for functional foods production. • Uxi (Endopleura uchi) is a native species from the Amazon region. • The fruits have potential for edible vegetable oil production, due to their chemical composition. • Uxi oil was obtained with high yields at temperatures of 40 and 60 °C and pressures of 300 and 400 bar. • The functional quality indices express that uxi oil can be consumed in the human diet as table oil, similarly to olive oil. In this research, uxi (Endopleura uchi) oil was obtained via supercritical CO 2 extraction in order to promote the valorization of the species in the industrial scenario, showing the potential of its oil as a functional food. The oil was extracted at temperatures of 40 and 60 °C and pressures of 200−400 bar. The highest yield was obtained at 60 °C / 400 bar (24.48 ± 0.20 %) in db. The oil showed a composition with predominance of oleic acid (n-9), as well as a stable thermal behavior up to 300 °C. Chronic wounds are severe breaks in the skin barrier that fail to heal in an acceptable time-frame, thus preventing the complete restoration of the tissue's anatomical and functional integrity, increasing the likelihood of infections and apoptosis. Hydrogels are known as a drug delivery system and have the potential to cover wounds and burns on the skin. Aloe barbadensis contains over 75 different bioactive compounds which are responsible for its anti-inflammatory and antimicrobial properties. In this study, the polyacrylamide-co-methylcellulose hydrogel containing Aloe barbadensis were developed. The extract was prepared from lyophilized Aloe barbadensis, using methanolic extraction, characterized by high performance liquid chromatography and incorporated into the hydrogels. These Aloe barbadensis hydrogels were characterized by degree of swelling, Fourier-transform infrared spectroscopy, scanning electron microscopy, and thermal profiling using thermogravimetric analysis. The minimum inhibitory concentration test was done on the Aloe barbadensis extract to evaluate its antibacterial and antifungal activity in vitro. The Aloe barbadensis hydrogels and were shown to swell to almost 2000% of their original sizes. The Fourier-transform infrared spectroscopy indicated the presence of bands characteristic of Aloe barbadensis and hydrogel polymers. The basic hydrogel showed greater thermal stability than the hydrogels with Aloe barbadensis. The minimum inhibitory concentration showed inhibition of the growth of S. aureus and Salmonella spp. at specific concentrations. The hydrogel therefore presents itself as an excellent potential curative cover of cutaneous lesions. [ABSTRACT FROM AUTHOR]
