Abstract Here, we report the effect of newly regenerated axons via scaffolds on reorganization of spinal circuitry and restoration of motor functions with epidural electrical stimulation (EES). Motor recovery was evaluated for 7 weeks after spinal transection and following implantation with scaffolds seeded with neurotrophin producing Schwann cell and with rapamycin microspheres. Combined treatment with scaffolds and EES-enabled stepping led to functional improvement compared to groups with scaffold or EES, although, the number of axons across scaffolds was not different between groups. Re-transection through the scaffold at week 6 reduced EES-enabled stepping, still demonstrating better performance compared to the other groups. Greater synaptic reorganization in the presence of regenerated axons was found in group with combined therapy. These findings suggest that newly regenerated axons through cell-containing scaffolds with EES-enabled motor training reorganize the sub-lesional circuitry improving motor recovery, demonstrating that neuroregenerative and neuromodulatory therapies cumulatively enhancing motor function after complete SCI.

In this study functional ultrasound (fUS) imaging has been implemented to explore the local hemodynamics response induced by electrical epidural stimulation and to study real-time in vivo functional changes of the spinal cord, taking advantage of the superior spatiotemporal resolution provided by fUS. By quantifying the hemodynamics and electromyographic response features, we tested the hypothesis that the temporal hemodynamics response of the spinal cord to electrical epidural stimulation could reflect modulation of the spinal circuitry and accordingly respond to the changes in parameters of electrical stimulation. The results of this study for the first time demonstrate that the hemodynamics response to electrical stimulation could reflect a neural-vascular coupling of the spinal cord. Response in the dorsal areas to epidural stimulation was significantly higher and faster compared to the response in ventral spinal cord. Positive relation between the hemodynamics and the EMG responses was observed at the lower frequencies of epidural stimulation (20 and 40 Hz), which according to our previous findings can facilitate spinal circuitry after spinal cord injury, compared to higher frequencies (200 and 500 Hz). These findings suggest that different mechanisms could be involved in spinal cord hemodynamics changes during different parameters of electrical stimulation and for the first time provide the evidence that neural-vascular coupling of the spinal cord circuitry could be related to specific organization of spinal cord vasculature and hemodynamics.

This study presents the first implementation of functional ultrasound (fUS) imaging of the spinal cord to monitor local hemodynamic response to epidural electrical spinal cord stimulation (SCS) on two small and large animal models. SCS has been successfully applied to control chronic refractory pain and recently was evolved to alleviate motor impairment in Parkinson's disease and after spinal cord injury. At present, however, the mechanisms underlying SCS remain unclear, and current methods for monitoring SCS are limited in their capacity to provide the required sensitivity and spatiotemporal resolutions to evaluate functional changes in response to SCS. fUS is an emerging technology that has recently shown promising results in monitoring a variety of neural activities associated with the brain. Here we demonstrated the feasibility of performing fUS on two animal models during SCS. We showed in vivo spinal cord hemodynamic responses measured by fUS evoked by different SCS parameters. We also demonstrated that fUS has a higher sensitivity in monitoring spinal cord response than electromyography. The high spatial and temporal resolutions of fUS were demonstrated by localized measurements of hemodynamic responses at different spinal cord segments, and by reliable tracking of spinal cord responses to patterned electrical stimulations, respectively. Finally, we proposed optimized fUS imaging and post-processing methods for spinal cord. These results support feasibility of fUS imaging of the spinal cord and could pave the way for future systematic studies to investigate spinal cord functional organization and the mechanisms of spinal cord neuromodulation in vivo.

Abstract Jute endophyte Staphylococcus hominis strain MBL_AB63 was reported to produce a novel antimicrobial peptide, ‘homicorcin’. This exhibits potential activity against a broad spectrum of Gram-positive bacteria. Eight genes were predicted to be involved in the sequential maturation of this peptide antibiotic, which includes structural (homA), dehydratase (homB), cyclase (homC), peptidase (homP), immunity (homI), oxidoreductase (homO), ATP-binding cassette transporter (homT1), and permease (homT2), respectively. Among the modification enzymes, HomB, HomC, and HomP exhibit sequence similarities with class I lantibiotic dehydratase, cyclase, and leader peptidase, respectively. The current study investigated the sequential modifications and secretion of homicorcin by constructing robust computational protein models and analyzing their interaction patterns using protein-protein docking techniques. To enhance comprehension of the protein arrangement, their subcellular localization was also extrapolated. The findings demonstrate a network of proteins that works in a synchronized manner, where HomC functions as an intermediary between HomB and the transporter (HomT). Following its dehydration by HomB and cyclization by HomC, the pro-homicorcin is taken out of the cell by the transporter and processed by HomP, resulting in the production of matured, processed homicorcin. This biosynthesis model for homicorcin will lay the groundwork for the sustainable and efficient production of this peptide antibiotic.

In this study, the neuroanatomy of the swine lumbar spinal cord, particularly the spatial orientation of dorsal roots was correlated to the anatomical landmarks of the lumbar spine and to the magnitude of motor evoked potentials during epidural electrical stimulation (EES). We found that the proximity of the stimulating electrode to the dorsal roots entry zone across spinal segments was a critical factor to evoke higher peak-to-peak motor responses. Positioning the electrode close to the dorsal roots produced a significantly higher impact on motor evoked responses than rostro-caudal shift of electrode from segment to segment. Based on anatomical measurements of the lumbar spine and spinal cord, significant differences were found between L1-L4 to L5-L6 segments in terms of spinal cord gross anatomy, dorsal roots and spine landmarks. Linear regression analysis between intersegmental landmarks was performed and L2 intervertebral spinous process length was selected as the anatomical reference in order to correlate vertebral landmarks and the spinal cord structures. These findings present for the first time, the influence of spinal cord anatomy on the effects of epidural stimulation and the role of specific orientation of electrodes on the dorsal surface of the dura mater in relation to the dorsal roots. These results are critical to consider as spinal cord neuromodulation strategies continue to evolve and novel spinal interfaces translate into clinical practice.

This study discusses the feasibility of diversifying the scope of application of lead-free Sr3ZBr3 (Z = As, Sb) as promising materials for their enhanced electronic, mechanical, optical, thermodynamic, and thermoelectric properties using first-principles calculation based on Density Functional Theory (DFT). Both compounds have cubic crystal structures, and both materials' dynamic stability is validated by the lack of negative frequencies in their phonon dispersion spectra. Besides ground state physical characteristics, we also examined the impact of hydrostatic pressure (0–21 GPa) on electronic, mechanical, and optical properties. The lattice parameters exhibit a linear decrease from 6.27 to 5.61 Å for Sr3AsBr3 and 6.45 to 5.71 Å for Sr3SbBr3 under increasing pressure, attributed to bond length reduction, which alters their physical properties. Initially observed direct band gap (Γ-Γ) of Sr3AsBr3 and Sr3SbBr3 were 2.35 and 2.30 eV using modified Becke-Johnson (mBJ) functional which reduces to 1.15 and 0.82 eV as the compounds go from 0 to 21 GPa pressure. This study reveals enhanced optical properties under pressure, with broader spectral response, increased sensitivity, and improved dielectric constant. Optical absorption and conductivity also shift toward lower-energy photons. The investigation further shows that the compounds show brittle to ductile transition under high pressure. Their ductility and anisotropy nature are further enhanced with pressure along with other mechanical features. The thermodynamic properties indicate their ability to withstand and perform well at high temperatures and pressures. Lastly, the thermoelectric properties of Sr3ZBr3 (Z = As, Sb) were assessed through electrical and thermal conductivity, Seebeck coefficient, power factor (PF), and figure of merit (ZT) as a function of temperature. Both compounds exhibit high PF and near-unity ZT. These findings underscore their potential as strong candidates for optoelectronic and thermoelectric devices, owing to their direct bandgap and exceptional properties.

A wireless body area network (WBAN) allows the integration of low power, invasive or non-invasive miniaturized sensors around a human body. Each intelligent sensor has enough capability to analyze and process the physiological parameters and to forward all the information to a central intelligent node for disease management, diagnosis and prescription. The data transmission rate of various biosensors is heterogeneous. Furthermore, the limited energy resources and computational power of these sensors have urged the development of low power energy efficient medium access control (MAC) protocol. This paper studies the performance of Preamble-Based time division multiple access (PB-TDMA) protocol for a heterogeneous non-invasive WBAN. Simulation results show that the performance of PB-TDMA protocol outperforms S-MAC and IEEE 802.11 DCF in terms of throughput and power consumption.

A comprehensive analysis of in vitro pumps used in cardiovascular research is provided in this review, with a focus on the characteristics of generated flows and principles of flow generations. The cardiovascular system, vital for nutrient circulation and waste removal, generates complex hemodynamics critical for endothelial cell function. Cardiovascular diseases (CVDs) could be caused by the disturbances in these flows, including aneurysms, atherosclerosis, and heart defects. In vitro systems simulate hemodynamic conditions on cultured cells in the laboratory to study and evaluate these diseases to advance therapies. Pumps used in these systems can be classified into contact and non-contact types. Contact pumps, such as piston and gear pumps, can generate higher flow rates, but they have a higher risk of contamination due to the direct interaction of pump with the fluid. Non-contact pumps, such as peristaltic and lab-on-disk centrifugal pumps, minimize contamination risks, but they are limited to lower flow rates. Advanced pumps including piezoelectric and I-Cor diagonal pumps are focused on improving the accuracy of flow replication and long-term stability. The operational principles, advantages, and some disadvantages of these pump categories are evaluated in this review, while providing insights for optimizing in vitro cardiovascular models and advancing therapeutic strategies against CVDs. The outcomes of the review elaborate the importance of selecting an appropriate pump system, to accurately replicate cardiovascular flow patterns.

Jute (Corchorus sp.), a commercially important and eco-friendly crop, is widely cultivated in Bangladesh, India, and China. Some varieties of this tropical plant such as the Corchorus olitorius. Variety accession no. 2015 (acc. 2015) has been found to be low-temperature tolerant. The current study was designed to explore the genome-wide variations present in the tolerant plant acc. 2015 in comparison to the sensitive farmer popular variety Corchorus olitorius var. O9897 using the whole genome resequencing technique. Among different variations, intergenic Single Nucleotide Polymorphism (SNPs) and Insertion-Deletion (InDels) were found in the highest percentage whereas approximately 3% SNPs and 2% InDels were found in exonic regions in both plants. Gene enrichment analysis indicated the presence of acc. 2015 specific SNPs in the genes encoding peroxidase, ER lumen protein retaining receptor, and hexosyltransferase involved in stress response (GO:0006950) which were not present in sensitive variety O9897. Besides, distinctive copy number variation regions (CNVRs) comprising 120 gene loci were found in acc. 2015 with a gain of function from multiple copy numbers but absent in O9897. Gene ontology analysis revealed these gene loci to possess different receptors like kinases, helicases, phosphatases, transcription factors especially Myb transcription factors, regulatory proteins containing different binding domains, annexin, laccase, acyl carrier protein, potassium transporter, and vesicular transporter proteins that are responsible for low temperature induced adaptation pathways in plants. This work of identifying genomic variations linked to cold stress tolerance traits will help to develop successful markers that will pave the way to develop genetically modified cold-resistant jute lines for year-round cultivation to meet the demand for a sustainable fiber crop economy.

The main focus of this research was to explore the present status of pineapple cultivation in Madhupur tract of Bangladesh. Primary data were collected using a pre-designed and pre-tested questionnaire by face-to-face interviews from a sample of 300 pineapple growers and two focus group discussions were also carried out from March to May 2022. According to the study findings majority (51%) of the respondents cultivate pineapple in their own land having medium experience (53.3%), followed mixed cropping pattern (89%) where 25.33 percent respondent cultivate banana as companion crop. ‘Calendar’ is the top most cultivated variety of pineapple in Madhupur tract. Majority of the respondent use balanced pesticide and ripening agent due to increasing demand of chemical free farm fresh pineapple among consumers. Higher profit compared to other crops (82.00 %) was the main reason for producing pineapple in the study area. Majority (26.67%) of the respondent followed (Grower- Faria- Bepari- Aratdar- Wholesaler- Retailer- Consumers) channel among the six dominant marketing channels to market their produced pineapple. The results of SWOT analysis revealed that weakness of pineapple cultivation in Madhupur tract have the potential to improve. If the government and other policy making organizations come forward to solve the problems arising in pineapple cultivation, then the position of pineapple as fruit will be consolidated soon in home and abroad.

Turmeric, a globally cultivated spice, holds significance in medicine, and cosmetics, and is also a very popular ingredient in South Asian cuisine. A study involving 53 turmeric genotypes evaluated for rhizome yield and related traits at Spices Research Center, Bogura, Bangladesh over three years (2019-22). A randomized complete block design was followed with two replications. ANOVA revealed significant trait variations among genotypes. Genotype T0015 emerged as the highest yielder at 28.04 t/ha. High heritability (0.58-0.99) and genetic advance characterized plant height (PH), mother rhizome weight (WMR), primary and secondary finger weights (WPF and WSF), and yield per plant (YPP) across seasons. Genetic gain (GG) was prominent in these traits. Genotypic and phenotypic coefficient variations (GCV and PCV) (6.24-89.46 and 8.18-90.88, respectively) across three years highlighted mother rhizome weight's importance followed by numbers of primary finger (NPF), and WPF. Positive and significant correlations, especially with PH, WMR, WPF, and YPP, emphasized their relevance to fresh yield (FY). Multiple linear regression identified PH, number of mother rhizome (NMR) and WMR as key contributors, explaining 37-79% of FY variability. Cluster analysis grouped genotypes into five clusters with maximum distance observed between clusters II and III. The geometric adaptability index (GAI) assessed adaptability and superiority, revealing nine genotypes outperforming the best existing cultivar. Genotype T0117 as the top performer based on GAI, followed by T0103 and T0094. Mean rank analysis favoured T0121 as the best performer, succeeded by T0117, T0082 and T0106. The top ten genotypes (T0015, T0061, T0082, T0085, T0094, T0103, T0106, T0117, T0121 and T0129) were identified as superior based on yield and overall ranking, warranting further evaluation. These findings may induce a window for improving turmeric research and ultimately play a role in enhancing its cultivation and productivity.

Abstract The silver pride of Bangladesh, migratory shad, Tenualosa ilisha (Hilsa), makes the highest contribution to the total fish production of Bangladesh. Despite its noteworthy contribution, a well-annotated transcriptome data is not available. Here we report a transcriptomic catalog of Hilsa, constructed by assembling RNA-Seq reads from different tissues of the fish including brain, gill, kidney, liver, and muscle. Hilsa fish were collected from different aquatic habitats (fresh, brackish, and sea water) and the sequencing was performed in the next generation sequencing (NGS) platform. De novo assembly of the sequences obtained from 46 cDNA libraries revealed 462,085 transcript isoforms that were subsequently annotated using the Universal Protein Resource Knowledgebase (UniPortKB) as a reference. Starting from the sampling to final annotation, all the steps along with the workflow are reported here. This study will provide a significant resource for ongoing and future research on Hilsa for transcriptome based expression profiling and identification of candidate genes.

Driven by the rapid escalation of its utilization, as well as ramping commercialization, Internet of Things (IoT) devices increasingly face security threats. Apart from denial of service, privacy, and safety concerns, compromised devices can be used as enablers for committing a variety of crime and e-crime. Despite ongoing research and study, there remains a significant gap in the thorough analysis of security challenges, feasible solutions, and open secure problems for IoT. To bridge this gap, we provide a comprehensive overview of the state of the art in IoT security with a critical investigation-based approach. This includes a detailed analysis of vulnerabilities in IoT-based systems and potential attacks. We present a holistic review of the security properties required to be adopted by IoT devices, applications, and services to mitigate IoT vulnerabilities and, thus, successful attacks. Moreover, we identify challenges to the design of security protocols for IoT systems in which constituent devices vary markedly in capability (such as storage, computation speed, hardware architecture, and communication interfaces). Next, we review existing research and feasible solutions for IoT security. We highlight a set of open problems not yet addressed among existing security solutions. We provide a set of new perspectives for future research on such issues including secure service discovery, on-device credential security, and network anomaly detection. We also provide directions for designing a forensic investigation framework for IoT infrastructures to inspect relevant criminal cases, execute a cyber forensic process, and determine the facts about a given incident. This framework offers a means to better capture information on successful attacks as part of a feedback mechanism to thwart future vulnerabilities and threats. This systematic holistic review will both inform on current challenges in IoT security and ideally motivate their future resolution.
