The coevolutionary dynamic of host–parasite associations varies from strictly local adaptations to diffuse guild coevolution. How innate and acquired immune responses modulate host resistance to parasitism and how host specificity and geographic scaling affect the efficacy of host response have consequences for range dynamics and biological invasions.Using reciprocal transplant experiments, we tested whether local or diffuse coevolution shapes host response to parasitism in the host–parasite association between bitterling fishes and larval stages (glochidia) of freshwater mussels from Europe and East Asia.We found that glochidia initially indiscriminately attached to all study host species, but immune responses elicited significant differences in host responses within 24 h of infection, which intensified during glochidial development. European bitterlings were more resistant to European glochidia and Asian bitterlings to Asian glochidia, with the strongest geographic bias in resistance in the Asian bitterling hosts.This finding suggests a strong effect of local adaptation but also indicates the non‐negligible role of coevolutionary hotspots. Low natural glochidia load on bitterling species overall hence arise from competent immune response rather than glochidia avoidance behaviour. Bitterling immune response is primarily innate and not acquired.Overall, our data demonstrate the complex outcome of parasite pressure on the evolution of host resistance, and important role of geographically structured coevolution in shaping host response. Zona radiata is formed in vitellogenic oocytes between the plasmalemma and the follicular layer. Light microscopy, scanning electron microscopy, and transmission electron microscopy were used to study on the formation of oocytes and zona radiata. The morphological architecture of zona radiata is classified into four types: (1) villous structure-covered massive villi seen only in the genus Rhodeus (R. ocellatus, R. pseudosericeus, R. uyekii, and R. notatus); (2) mesh-shaped structures with porous shapes in Acheilognathus rhombeus and A. macropterus; (3) vine-like structure with longer and thicker materials in A. chankaensis; (4) smooth structure with no any structural modifications in A. yamatsutae, A. lanceolatus, A. koreensis, A. majusculus, A. signifier, and A. somjinensis. Given the fact that threatened species are often composed of isolated small populations, spatial continuity or demography of the populations may be major factors that have shaped the species' genetic diversity. Thus, neutral loci have been the most commonly-used markers in conservation genetics. However, the populations under the influence of different environmental factors may have evolved in response to different selective pressures, which cannot be fully reflected in neutral genetic variation. Rhodeus pseudosericeus, a bitterling species (Acheilognathidae; Cypriniformes) endemic to the Korean Peninsula, are only found in some limited areas of three rivers, Daecheon, Han and Muhan, that flow into the west coast. Here, we genotyped 24 microsatellite loci and two loci (DAB1 and DAB3) of MHC class II peptide-binding β1 domain for 222 individuals collected from seven populations. Our microsatellite analysis revealed distinctive differentiation between the populations of Daecheon and Muhan Rivers and the Han River populations, and populations were structured into two subgroups within the Han River. Apparent positive selection signatures were found in the peptide-binding residues (PBRs) of the MHC loci. The allelic distribution of MHC showed a degree of differentiation between the populations of Daecheon and Muhan Rivers and the Han River populations, partially similar to the results obtained for microsatellites, however showed rather complex patterns among populations in the Han River. Considering the apparent differences in the distribution of supertypes obtained based on the physicochemical differences induced by the polymorphisms of these PBRs, the differentiation in DAB1 between the two regional groups may result in the differences in immune function. No differentiation between these two regions was observed in the supertyping of DAB3, probably indicating that only DAB1 was associated with the response to locally specialized antigenic peptides. The modern-day distribution of freshwater fishes throughout multiple rivers is likely the result of past migration during times when currently separate drainages were once connected. Here, we used mitochondrial and microsatellite analyses for 248 individuals of Rhodeus notatus collected from seven different rivers to obtain better understand historical gene flow of freshwater fish on the Korean Peninsula. Based on our phylogenetic analyses, this Korean species originated through the paleo-Yellow River from China and first colonized near the west coast. These genetic data also provided evidence of estuary coalescences among the rivers flowing to the west and southwest coast on well-developed continental shelf. In addition, the pattern of population structure revealed the biogeodispersal route from the west coast to the south coast. It could be inferred that massive migration was not involved in the formation of southern populations, since the signature of historical genetic drift was clearly observed. Our study is the first genetic attempt to confirm hypotheses describing the migration of freshwater species towards the end of East Asia, which have previously been developed using only geological reasoning. The tripartite motif-containing (TRIM) proteins are involved in a wide range of cellular processes, and the role of TRIM1 in immunity has been explored. However, fundamental studies on fish TRIM1 are lacking. In this study, we cloned and characterized TRIM1 cDNA from the Korean rose bitterling, Rhodeus uyekii (RuTRIM1). Two RuTRIM1 isoforms (RuTRIM1-X1 and RuTRIM1-X2) were identified. The coding sequence (CDS) of RuTRIM1-X1 comprised 2157 bp encoding a 718-aa protein, and the CDS of RuTRIM1-X2 comprised 1929 bp encoding a 642-aa protein. Both RuTRIM1 isoforms contained a RING finger domain, B-box 1, B-box 2, coiled-coil domain, COS box, FN3 motif, and PRY/SPRY domain. The deduced RuTRIM1-X1 and RuTRIM1-X2 proteins had high amino acid identity (76.27–98.89%) with orthologs from various other species, and a phylogenetic tree was constructed. RuTRIM1-X1 and RuTRIM1-X2 mRNA were expressed in all tissues examined, with the highest expression levels detected in the hepatopancreas. During early development, RuTRIM1-X1 and RuTRIM1-X2 mRNA levels changed differently from the gastrula period to the first feeding stage. An in vivo ubiquitination assay showed that RuTRIM1 exhibited RING-dependent E3 ubiquitin ligase activity, mainly by comparing RuTRIM1-X2 to RuTRIM1-X1. The subcellular localization of the two RuTRIM1 protein isoforms was characterized, revealing that they formed aggregates in cytoplasmic bodies in Raw264.7 cells. Interferon-γ/lipopolysaccharide-induced nuclear factor-κB signaling was negatively regulated by RuTRIM1-X1 and RuTRIM1-X2, and the negative effect was reversed in RING deletion mutants. In order to investigate the effect of methyl testosterone on the body color, Rhodeus sinensis of juveniles (fifty days postfertilization, 50-DPF) and adults (eighty days post-fertilization, 80-DPF) were divided into six groups, each treatment group cultured in duplicate, randomly and separately, with 80 ind each. All juveniles and adults were raised, respectively, in the recirculation aquarium tanks with 500 liters separately. Five treatment groups (TG1, TG2, TG3, TG4, and TG5) were dieted with 17a-methyl testosterone (17α-MT) at different concentration of 10, 20, 30, 40 and 50 mg/kg, while the control group was dieted without 17α-MT. During the 56 days of test duration, the carotenoid content of the test fishes and the carotenoid deposited in the muscle were researched comparatively. The results showed there was significant pigmentation for 50-DPF Rhodeus sinensis dieted with 17α-MT at concentration of 40 mg/kg and 50 mg/kg (p <0.05). When Rhodeus sinensis dieted with 17α-MT at concentration of more than 30 mg/kg, the body color of 50-DPF Rhodeus sinensis started to change obviously at the 28th day (p <0.05), then sustained coloring to the maximum at the 56th day. The rosy bitterling, Rhodeus ocellatus, is a small freshwater fish belonging to the family Cyprinidae. This species lives in ponds where freshwater mussels are abundant and female lays eggs inside mussels. To understand whether the reproduction mode influenced the phylogeography, our study examined the genetic structure of R. ocellatus using sequences from the mitochondrial DNA (cytochrome b gene and control region) and nuclear DNA (the first intron of ribosomal protein S7 gene). In total, 213 specimens were collected from twelve populations in south of Yangtze River, including Yangtze River, Taiwan and Hainan Islands. The phylogenetic analyses based on mitochondrial and nuclear sequences both showed the lack of a population genetic structure, but all results, including the approximate Bayesian computation approaches, showed that these two markers revealed incongruent historical signals. Our study found that (1) the discordance between these two markers were accounted for admixtures by introduced; (2) the phylogeographic pattern of R. ocellatus and that of other freshwater fish were identical; (3) the reproduction mode was due to that the gene flows among populations were limited; and (4) R. ocellatus colonized in south of the Yangtze River (including) after southeast coastal districts formed and before Taiwan Island reached its present shape by comparing our results with that of the previous studies. The presence of non-native species can affect coevolved relationships. However, rapid reciprocal changes in coevolutionary associations provide the potential to quickly respond to a new situation. We studied a system where bitterling fish ( Rhodeus amarus) parasitize unionid mussels by laying their eggs onto their gills. This association is affected by the infestation of unionid shells by the non-native zebra mussel ( Dreissena polymorpha). In a series of experiments under experimental, semi-natural and natural conditions, we compared the behavioural response to zebra mussel infestation of unionid shells, its effect on oviposition decisions and their population consequences between bitterling populations naïve to zebra mussels and those recently sympatric with zebra mussels. We found no effect of recent sympatry on bitterling preoviposition behaviour and oviposition decisions and only a weak effect on their reproductive success. Bitterling from both populations inspected infested and non-infested mussels at the same rate but preferred to oviposit into non-infested unionid hosts. However, neither bitterling population completely avoided oviposition into infested unionids and three ovipositions into zebra mussels were observed. Overall, there was a clear negative relationship between the number of zebra mussels on unionid host shells and the number of juvenile bitterling emerging from the mussels. Patterns of movement are related to life history and/or species traits. Understanding movement patterns of multiple species sharing the same niche may clarify coexistence mechanisms. Therefore, the movement patterns of three bitterling species, Acheilognathus tabira erythropterus, Acheilognathus melanogaster, and Rhodeus ocellatus ocellatus, were examined between June and November 2009 in an agricultural ditch system associated with Lake Kasumigaura in central Japan. The mark-recapture method was used with a fixed releasing point in an approximately 5.5 km-long ditch. Movement distances of A. t. erythropterus and A. melanogaster were longer than that of R. o. ocellatus, but the direction of movement was similar among the three species. However, R. o. ocellatus moved to locations with a low dissolved oxygen concentration and deeper water. These results suggest that R. o. ocellatus may adapt better to environmental fluctuations, because its tolerance to hypoxia is the highest of the three species. Compared with R. o. ocellatus, A. t. erythropterus and A. melanogaster may move relatively longer distances to search for suitable habitats following an environmental fluctuation. Oviposition decisions can have important fitness consequences for offspring. We investigated the responses of European bitterling, Rhodeus amarus , a freshwater fish that spawns in the gills of living unionid mussels, to oviposition site cues. Using an artificial mussel, we manipulated the flow velocity, dissolved oxygen concentration and odour cues of mussels presented to pairs of R. amarus . Females responded positively to mussel odour and to dissolved oxygen cues. Male response was dependent on mussel odour and the flow velocity of water emerging from the artificial mussel. These responses are potentially adaptive, with females responding to cues that indicate the quality of oviposition sites for incubation of eggs. Thioredoxin is a multifunctional antioxidant enzyme that belongs to the reductase family. In this study, we cloned and characterized thioredoxin 1 cDNA from the Korean rose bitterling Rhodeus uyekii (RuTrx). The full-length RuTrx cDNA consists of 674 bp with a 324 nt open reading frame (ORF) encoding a 107 aa protein. The deduced RuTrx amino acid sequence indicated a characteristic redox active site, 31WCGPC35. Pairwise alignment revealed RuTrx amino acid identity (55.1%-83.2%) with orthologs from various species of mammalia, amphibia, fish and bird. Phylogenetic analysis was conducted to determine the evolutionary position of RuTrx. Expression analysis showed that RuTrx transcripts were present in all of the tissues examined, and was high in the hepatopancreas of R. uyekii. During early development, the expression of RuTrx transcripts was increased. Recombinant RuTrx protein (rRuTrx) was tested for its capacity to serve as an antioxidant enzyme using a metal-catalyzed oxidation (MCO) system. The ability of rRuTrx to protect against supercoiled DNA cleavage due to oxidative nicking increased in a dose-dependent manner. In Raw264.7 cells, Dihydroethidium (DHE) staining for ROS production indicated the antioxidant activity of rRuTrx. Genetic structure and its artificial disturbance in the endangered Japanese bitterling Acheilognathus cyanostigma were examined based on mitochondrial cytochrome b gene sequences. The haplotype network included three major clades: the dominant clade consisted of haplotypes found from the Lake Biwa-Yodo River system and other sites, including a known site of introduction; the other two clades consisted of haplotypes found exclusively or almost exclusively from the Ise Bay area, and the Yura and Kako River systems, respectively. Their divergence times, estimated using teleost molecular clocks, yielded results concordant with geological events such as uplifting of the Suzuka Mountains during the early Pleistocene. The widespread haplotypes were suggested to be introduced from the Lake Biwa-Yodo River system during stocking of Ayu from this system. The Lake Biwa-Yodo River population is critically endangered in its original habitats, yet is invasive in habitats where it has been introduced as it hybridizes with or replaces indigenous populations and competes with native bitterlings. Utilizing introduced populations for reintroduction and genetic rescue of the original population may be possible after careful examination for factors that could negatively impact the local community. The fish Vitellogenin (Vg) gene has been applied as a biomarker for exposure to estrogenic compounds in the aquatic environment. In this study, we cloned and characterized Vg cDNA from the Korean rose bitterling Rhodeus uyekii (Ru-Vg). The Ru-Vg cDNA encodes a 1424-amino-acid polypeptide that belongs to the VgAo1 family and contains a putative signal peptide, lipovitellin I, phosvitin, and lipovitellin II, but does not contain the vWFD domain or the C-terminal peptide. The deduced Ru-Vg protein has high amino acid identity (73.97%-32.17%) with fish Vg proteins. Pairwise alignment and phylogenetic analysis revealed that Ru-Vg is most closely related to Acheilognathusyamatsutae Vg. Ru-Vg transcripts were detected using quantitative polymerase chain reaction in all tissues tested, with the highest level of expression observed in the ovary. Ru-Vg mRNA was upregulated in R. uyekii hepatopancreas cells in response to treatment with 17ß-estradiol (E2) or 17a-ethinylestradiol (EE2). Luciferase reporter expression, driven by the 5'-regulatory region of the Ru-Vg gene spanning from -1020 bp to the start codon was induced by the estrogen receptor and was synergistically activated by treatment with E2 or EE2. While the effect of Operational Sex Ratio (OSR) on reproductive behaviour of males has been studied extensively, little is known of the response of females facing a female-biased OSR. We investigated the effect of different OSRs on female reproductive behaviour using the rosy bitter-ling, Rhodeus ocellatus, a freshwater fish that lays its eggs inside the gills of living freshwater mussels. Three levels of OSR (male/female ratio 1:1, 1:3 and 1:5) were tested. We demonstrated that inspection of the mussel (spawning substrate) by individual females increased with increas-ingly female-biased OSR, but that the rate of following territorial male decreased. Aggression towards other females was not affected by the OSR. Interestingly, when a male bitterling led a non-dominant female towards the mussel, the dominant female would become aggressive to the male and chase the non-dominant female away. Aggression towards male followed a bell-shaped pattern and was highest at an OSR of 1:3. In both the female-biased OSRs examined, almost 50% of dominant females tended to chase away other females and defend the mussel, showing territori-ality in a similar manner to males. These observations suggest that female reproductive behaviour is strongly affected by the OSR, and their reproductive tactics during courtship change from a passive role in courtship (following a male) to an active role in courtship (approaching a male), with pres-ence of female territorial behaviour as the OSR becomes increasingly female-biased. Hybridization between native and non-native fishes is a serious global problem. Thus, there is a need to develop monitoring methods for predicting potential hybridization to evaluate the risk of genetic introgression and to identify important areas for conservation of pure native populations. Here, we developed a prediction model for intersubspecific hybridization, based on distribution and genetic data. We selected Rhodeus ocellatus kurumeus and R. ocellatus ocellatus as the native and non-native subspecies, respectively. First, we developed generalized linear models (GLMs) for the species habitat requirements by using presence/absence data and environmental variables. The best-fit models showed river length gave conflicting effects for the two subspecies. Next, we developed a GLM using the ratio of non-native haplotypes in mitochondrial DNA as an objective variable, with the predicted probabilities of the occurrence of each fish and spatial information as explanatory variables. The best-fit model selected the distance from the center of native distribution and the non-native habitat requirement as key factors. Our findings indicate that hybridization occurs highly and/or initially near the margin of native distribution where non-native habitat requirements are available. Our model could identify sites in native habitats with very low potential risk for genetic invasion as important areas for conservation of pure native populations. [ABSTRACT FROM AUTHOR]

Research on sexual selection has tended to focus on indirect benefits of female mating decisions, and few attempts have been made to quantify the relative effect of direct and indirect selection simultaneously. Here we compared direct and indirect selection on female mating decisions in the rose bitterling (Rhodeus ocellatus), a fish with a resource-based mating system, using experimental treatments with equivalent consequences for female reproductive success. Direct selection was varied by manipulating the quality of sites available to females for oviposition, and indirect selection by presenting females with males of known genetic compatibility. Manipulating the strength of direct and indirect selection had specific, quantified consequences for embryo survival during incubation. There was a significant effect of both direct and indirect selection on female mating decisions, though direct selection accounted for more variance in female oviposition rate compared with indirect. No interaction between direct and indirect selection was detected. Although effects on female reproductive success were additive, selection for direct mate choice benefits appears to be stronger than for indirect benefits. A possible explanation for weaker selection on indirect benefits in the study species is because females are constrained in making mate choice decisions through alternative male mating tactics, which generate a sexual conflict. Abstract: Tripartite motif-containing 25 (TRIM25), also known as estrogen-responsive finger protein (EFP), plays an essential role in cell proliferation and innate immunity. In the present study, we isolated and characterized the TRIM25 cDNA of the Korean rose bitterling Rhodeus uyekii, designated RuTRIM25. It encodes an open reading frame of 669 amino acids containing an N-terminal RBCC motif composed of a RING domain, two B boxes, and a coiled-coil domain and a C-terminal B30.2 (PRY/SPRY) domain. RuTRIM25 shows strong homology (79.7%) to zebrafish TRIM25 and shared 32.4–28.8% homology with TRIM25 from other species, including mammals. RuTRIM25 mRNA was expressed ubiquitously. It was highly expressed in the ovary, spleen, and liver and moderately in the stomach and intestine of normal Korean rose bitterling. The intracellular localization of RuTRIM25 in HEK293T cells was diffusely localized in the cytoplasm and its RING domain deletion mutant (RuTRIM25ΔR) was detected diffusely with some aggregates in the cytoplasm. RuTRIM25, but not RuTRIM25ΔR, is ubiquitinated in vivo. Ectopic expression of RuTRIM25 synergistically activated the estrogen receptor (ER)-mediated luciferase reporter activity in a dose-dependent manner in HEK293T cells. Together, these results suggest that the RuTRIM25 regulates the ER-mediated transcription in fish similarly to its mammalian counterpart. [Copyright &y& Elsevier]

Ecologically specialized species may be more susceptible to anthropogenic impacts than generalist species. Japan's native bitterlings (subfamily Acheilognathinae), which are specialized to spawn on the gills of certain freshwater mussels, have been declining dramatically during the last few decades. To identify factors affecting the local occurrence of the threatened bitterling species Tanakia lanceolata, we measured its presence and absence, along with several environmental factors, at 68 sites within agricultural canal networks in the Lake Mikata basin, Fukui Prefecture, Japan. Based on the theoretical information approach of Akaike's information criterion, generalized linear mixed models were constructed. These revealed that the species' occurrence is strongly affected by five major factors: the presence of freshwater mussels Anodonta sp., water depth, floating plants coverage, the presence of bullfrogs, and submerged plants coverage. The probability of the presence of T. lanceolata was higher at shallower sites with lower floating plants coverage, located within channels containing mussel beds. Territorial animals sometimes use conspicuous natural landmarks as boundaries to their territories. The utilization of territory-demarcating landmarks may have evolved to limit the costs of territorial defence, since the adoption of clearly defined boundaries by opponents in adjacent territories can reduce the overall rate of aggressive encounters, which can be energetically expensive or might result in injury. Here the role of artificial landmarks as boundaries was tested in territorial male rose bitterling (Rhodeus ocellatus), a fish with a resourcebased mating system. Pairs of size-matched territorial males were permitted to interact for short periods in an otherwise featureless aquarium with an obvious landmark at the shared boundary of their territory either present or absent. The presence of the territory-demarcating landmark significantly reduced both the frequency of territorial incursions by males into adjacent territories and the rate of territorial displays. Males showed individual differences in their propensity to enter the territory of a rival, irrespective of the presence of a territorydemarcating landmark. These results suggest that the cost of defence of a territory may be reduced by utilizing territory-demarcating landmarks, in accordance with the predictions of theoretical models. The European bitterling, Rhodeus amarus, is a non-indigenous fish species in British fresh waters. It lays its eggs in unionid mussels which themselves are vulnerable to fouling by the non-indigenous zebra mussel, Dreissena polymorpha. Observations from an unmanipulated natural system showed that only 27% of zebra mussel-fouled Unio pictorum hosted bitterling, while 47% of unfouled U. pictorum hosted bitterling. We conducted a field experiment in the River Great Ouse catchment, Cambridgeshire, England in May–June 2007 and 2008 to quantify the impact of zebra mussels on bitterling load in host mussels. Zebra mussel-fouled unionids were significantly less likely to host bitterling than unfouled unionids. The number of unionids hosting bitterling did not differ significantly whether the zebra mussels fouling the unionid were alive or dead. Bitterling appeared to discriminate against zebra mussel-fouled unionids less as the 2007 breeding season advanced, potentially because preferred unfouled unionids had a higher bitterling load, and were therefore relatively lower quality hosts than at the start of the breeding season. Female density and resource availability are two key variables that shape mating systems. Theory predicts that reproductive skew will amplify with increased male density and decreasing availability of resources, though limited empirical evidence suggests that this may not always be the case. Here we tested mean crowding, defined as the number of males per unit of resource, and density per se, defined as the number of individuals present per unit area, to investigate their effect on the mating system of Rhodeus ocellatus, a fish with a promiscuous, resource-based mating system. Males were exposed to combinations of high and low levels of crowding and density, while the operational sex ratio was held constant. High levels of crowding significantly affected the proportion of mussel spawning sites defended by males and the proportion of mussels into which sperm was released. In contrast to theoretical predictions, neither density nor crowding influenced overall male aggressive behaviours. Density, but not crowding, had a significant effect on male courtship rate, which arose as a possible trade-off between intra-sexual competition and inter-sexual behaviour. Good genes models of mate choice predict additive genetic benefits of choice whereas the compatibility hypothesis predicts nonadditive fitness benefits. Here the Chinese rose bitterling, Rhodeus ocellatus, a freshwater fish with a resource-based mating system, was used to separate additive and nonadditive genetic benefits of female mate choice. A sequential blocked mating design was used to test female mate preferences, and a cross-classified breeding design coupled with in vitro fertilizations for fitness benefits of mate choice. In addition, the offspring produced by the pairing of preferred and nonpreferred males were reared to maturity and their fitness traits were compared. Finally, the MHC DAB1 gene was typed and male MHC genotypes were correlated with female mate choice. Females showed significant mate preferences but preferences were not congruent among females. There was a significant interaction of male and female genotype on offspring survival, rate of development, growth rate, and body size. No significant male additive effects on offspring fitness were observed. Female mate preferences corresponded with male genetic compatibility, which correlated with MHC dissimilarity. It is proposed that in the rose bitterling genetic compatibility is the mechanism by which females obtain a fitness benefit through mate choice and that male MHC dissimilarity, likely mediated by odor cues, indicates genetic compatibility. Rhodeus pseudosericeusis a native bitterling to the Korean Peninsula and found in very limited areas with small census size. Here, its complete mitochondrial genome was analyzed to provide novel data for the reconstruction of phylogenetic relationship among Acheilognathinae species. The genome was a 16,574 bp long consisting of 1 putative control region, 2 rRNA genes, 22 tRNA and 13 protein-coding genes. The gene arrangement was completely identical to those observed in other Acheilognathinae species as well as in other cyprinid species. In our phylogenetic analyses, three major genera of Acheilognathinae indepedently formed monophyletic groups in the tree reconstructed based on the whole genome sequences, whereasRhodeuswas not recovered as a single monophyly when solely considering protein-coding genes, indicating that the taxonomic reevaluation is still required in this subfamily. An intersexual conflict arises when males and females differ in their reproductive interests. Although experimental studies have shown that females often mate with dominant males, it may not always be in the interest of a female to do so. Here we investigated the impact of male dominance on female mate choice and offspring growth and survival in the rose bitterling ( Rhodeus ocellatus), a freshwater fish with a resource-based mating system. Three experimental mating trials were conducted using males of known dominance rank, but with different levels of constraint on male behavior. Thus, females were able to choose among; (1) males that were isolated from each other; (2) males that could see and smell each other, but could not directly interact; (3) males that could interact fully. Using a combination of behavioral observation and parentage analyses it was shown that female preferences did not correspond with male dominance and that male aggression and dominance constrained female mate choice, resulting in a potential intersexual conflict. The survival of offspring to independence was significantly correlated with female mate preferences, but not with male dominance. The spatial distribution of breeding resources can have pronounced demographic and evolutionary consequences. We used 20 experimental groups of the bitterling ( Rhodeus amarus), an annual fish with a promiscuous, resource-based mating system, and extended breeding season to investigate how the spatial distribution (clumped or regular) of bitterling oviposition sites (live freshwater mussels) affected offspring production, variation in reproductive success, and directional selection on phenotypic traits over their entire reproductive lifetime. We did not detect any effect of resource distribution on offspring production or variation in reproductive success among individual fish, although variation between replicates was higher with a clumped distribution. This finding is discussed with regard to the incidence of alternative mating behaviors (sneaking) within the limitations imposed by our experimental design. Breeding resource distribution had a significant effect on selection on male phenotypic traits. Stronger directional selection on traits associated with intrasexual competition for fertilizations, gonad mass (an indicator of sperm competition), and the extent of red, carotenoid-based pigment in the iris (an index of dominance status), was detected with a clumped resource distribution. Environmental and population parameters that influence the strength of sexual selection may vary considerably over the course of the reproductive season. However, the potential for sexual selection frequently fails to translate into variation in reproductive success among individuals. We investigated seasonal changes in variation in reproductive success, measured as the opportunity for sexual selection, using parentage analysis in 20 experimental populations of the European bitterling ( Rhodeus amarus, Cyprinidae), a small freshwater fish with a promiscuous, resource-based mating system. We showed that although the largest males sired most offspring over the entire reproductive season, variation in reproductive success and hence the opportunity for sexual selection was low at the start of the season but increased significantly at its end. This seasonal difference probably arose from the superior competitive endurance of large males and from a higher temporal clustering of reproductively active females at the start of the breeding season than later in the season. The spatial distribution of oviposition sites had a negligible effect on the variation in reproductive success. Sexual selection involves two main mechanisms: intrasexual competition for mates and intersexual mate choice. We experimentally separated intrasexual (male–male interference competition) and intersexual (female choice) components of sexual selection in a freshwater fish, the European bitterling (Rhodeus sericeus). We compared the roles of multiple morphological and behavioural traits in male success in both components of sexual competition, and their relation to male reproductive success, measured as paternity of offspring. Body size was important for both female choice and male–male competition, though females also preferred males that courted more vigorously. However, dominant males often monopolized females regardless of female preference. Subordinate males were not excluded from reproduction and sired some offspring, possibly through sneaked ejaculations. Male dominance and a greater intensity of carotenoid-based red colouration in their iris were the best predictors of male reproductive success. The extent of red iris colouration and parasite load did not have significant effects on female choice, male dominance or male reproductive success. No effect of parasite load on the expression of red eye colouration was detected, though this may have been due to low parasite prevalence in males overall. In conclusion, we showed that even though larger body size was favoured in both intersexual and intrasexual selection, male–male interference competition reduced opportunities for female choice. Females, despite being choosy, had limited control over the paternity of their offspring. I investigated the seasonal change in factors affecting embryonic mortality in the rosy bitterling,Rhodeus ocellatus kurumeus, a freshwater fish that spawns on the gills of living unionid mussels. Research was conducted in a small pond during 1999 and 2001 in which bitterling were provided withAnodontasp. mussels for spawning. Bitterling spawned between April and July, peaking mid-late May. Seasonal survival rate of bitterling embryos in their mussel hosts was unimodal, with a peak between late April and mid May (about 70% of total spawnings). In mid April, survival was about 50%. The lowest survival was from late May to July (0%). Losses of bitterling embryos from mussels were identified by ejections from the mussel host. Ejections were categorized as either ejections of live embryos, or ejections of embryos that died in the mussel and were subsequently expelled from the mussel. Ejection rates of live embryos were higher in the earlier part of the spawning period (early-mid April) and dead embryo ejections in the later period (after June). The ejection rate of live embryos was higher among younger embryos earlier in the season, probably because of the incomplete development of morphological and behavioural traits associated with maintaining the embryo inside the mussel gill chambers, and as a consequence of a more protracted developmental period at low temperatures making them more susceptible to ejection. The ejection rate of dead embryos was higher in older embryos later in the season, and in larger mussels and at high embryo densities. The survival of embryos in mussels was probably related to oxygen availability, with mortalities probably caused by asphyxiation. Increased embryo mortalities may arise through competition among embryos, between embryos and mussel, and ambient dissolved oxygen levels. The optimal period for bitterling to spawn may represent a balance between two opposing factors; with positive and negative effects of a seasonal rise in temperature directly affecting embryonic growth rate and oxygen availability. The European bitterling Rhodeus sericeus Pallas is a freshwater fish belonging to the Acheilognathinae, a subfamily that has an unusual spawning symbiosis with freshwater mussels. Female bitterling possess long ovipositors that they use to place their eggs onto the gills of a mussel through the mussels' exhalant siphon. Males fertilize the eggs by releasing sperm into the inhalant siphon of the mussel. The embryos develop inside the mussel for approximately a month, eventually leaving the mussel as actively swimming larvae. Because they use a discrete spawning site that can be readily manipulated they have recently been used in studies to understand oviposition choice with respect to mate and spawning site quality, host–parasite coevolution, alternative male mating tactics, and linking behavioural decisions with population dynamics.
