The complex wound healing process involves activating and synchronizing intracellular, intercellular, and extracellular components. Adipose tissue is attracting attention to promote wound healing. Within subcutaneous adipose tissue, stromal vascular cells and their subsets release growth factors and cytokines critical for neovascularization and wound repair.This study evaluated human placental collagen/adipose-derived stem cells (ADSCs) hydrogel for wound healing in rats.In this study, ADSCs were harvested, cultured, and mixed with placental collagen. Twelve rats were used, and their backs were excised three times each. Group one received collagen/ADSCs, group two collagen, and group three non-filled (control) excisions. The healing processes were assessed by histological analysis, taking photographs, and calculating the percentage of wound contraction in mentioned times.Histopathological analysis revealed that the content of fibroblasts, follicles of the hair, and angiogenesis in group one was significantly more than in other groups. Group one had a significant result compared with the collagen and control groups. In group one, significant wound healing and wound contraction were observed with 52% and 80% wound contraction at 7 and 14 days, respectively.Collagen/ADSCs can be considered a suitable candidate hydrogel in wound healing with a high potential for enhancing wound repairing.

Many factors including growth factors (GF), scaffold materials, and chemical and physical cues determine the cell behaviors. For many years, growth factors have been considered as the pivotal cell behavior regulators, whereas recent studies emphasize also the key role of physical factors such as mechanical forces, cell shape, surface topographies, and extracellular matrix (ECM) in regulating the cell proliferation, apoptosis, differentiation, etc. through mechanotransduction pathways. In this process, the cell morphology and mechanical properties of the cell's micro/ nano-environments and ECM can be conveyed to the nucleus by regulating transcriptional factors such as Yes-associated protein and transcriptional coactivator with PDZ-binding motif (TAZ). Generally, YAP/TAZ activity is considered as the key factor for the growth of whole organs, however, recent studies have also repeatedly addressed the role of YAP/TAZ in mechanotransduction. In this review, the biological functions of the YAP/TAZ pathway and its contribution to the mechanotransduction and cell behavior regulation in response to the mechanical cues have been summarized. Also, the role of key mechanical checkpoints in the cell including focal adhesions, cytoskeletal tension, Rho small GTPases, and nuclear membrane protein elements involved in the transfer of environmental mechanical cues from the cell surface to the nucleus and their effect in regulating the YAP/TAZ activity are discussed.

Successful non‐surgical orthograde retreatment of a tooth with external root resorption in apical third of root and overfilling material beyond the apical barrier which was diagnosed with the help of cone beam computed tomography (CBCT), was reported. Biodentine was used as the treatment of choice for obturation of resorption area., Overfilling management can be done by performing nonsurgical endodontic procedure. Gutta‐percha of coronal and middle third of root should be removed by mechanical instruments; however, in apical, removal should be done by hand instrument. Biodentine can be considered as the treatment of choice for filling the perforating external root resorption.

Synthetic scaffolds, as an alternative to allograft and xenograft scaffolds, are suitable for bone regeneration. This study aimed to synthesize a composite biomaterial of zeolite and beta-tricalcium phosphate (βTCP) to obtain a biocompatible material with physical and mechanical properties in bone regeneration. One scaffold without zeolite (βZG 0) and two scaffolds with different amounts of zeolite (βZG 1 and βZG 2) were synthesized. The scaffolds were evaluated by FTIR, XRD, compressive strength test, MTT assay, and radiographic and histological analyses. The XRD results confirmed the presence of βTCP and ZSM-5 phases in the composite scaffolds and also, indicated that the addition of gelatin decrease the crystallinity of composite scaffolds. FTIR revealed the gelatin, β-TCP and ZSM-5 functional groups in the composite structure. βZG 2 group had the maximum porosity among the scaffolds (74%) ranging in size from 61-600 µm. Compressive strength test showed that the Young’s modulus changed from 23 MPa to 59 MPa, and the zeolite nanostructure was the most influential factor responsible for this change. The MTT assay showed the superiority of βZG 2, and the macroscopic and microscopic results at 4, 8, and 12 weeks revealed the maximum bone regeneration and formation of bone trabeculae in the βZG 2 and βZG 1 groups, respectively. The zeolite scaffold showed the superior mechanical, radiographic and histological properties compared with the control and non-zeolite scaffold. βTCP/ Zeolite/ Gelatin scaffold can be an appropriate candidate for medical application in bone regeneration.

In this study, a novel Polycaprolactone suture assembled with Tadalafil was investigated to improve wound healing processes via vascular stimulation. Tadalafil/Polycaprolactone (TP) suture was developed by the electrospinning method. The designed suture was characterized by SEM, mechanical properties assessments, tensile strength measurements and the drug release study. For in vivo tests, rats were classified into two study groups. An incision was made on their back skin and they were sutured with TP suture and Polycaprolactone suture as control. Rats were sacrificed at 7 days following surgery for histopathological examinations with Hematoxylin and Eosin staining. Results of Tensile test demonstrated that the lowest tensile strength belonged to 3 and 4 % wt and the highest tensile strength belonged to 1 and 2 % wt of TP suture. The rate of Tadalafil release showed that the highest drug release was related to 3 and 4% wt which were about 125 to 210 μg during 15 days. The histopathology revealed that the number of blood vessels, collagen fibers, fibroblast, polymorphonuclear leukocytes, and epithelization was remarkable in Tadalafil/Polycaprolactone group during 7-day. A novel Tadalafil/Polycaprolactone suture improved the processes of wound healing by releasing the Tadalafil drug around the sutured wound and can be used in medical applications.

Different composite materials have been investigated in bone regeneration but none of them have a significant regeneration in a short time. In this study, the novel scaffold with the osteoinductive characteristic in order to accelerate bone regeneration for 6 weeks. Tadalafil/β-TCP/Collagen (TβC) and β-TCP/Collagen (βC) composite scaffolds were prepared and analyzed by porosity, biodegradability and MTT tests. And then, three bone defects (8 mm diameter, n = 6 group) were produced and filled with TβC, βC scaffolds and the third defect was unfilled as a control. Samples were taken and evaluated by histological, radiological and histomorphometric evaluation at 4 and 6 weeks. In vitro tests showed that both scaffold approximately had the same results in the percentage of porosity and in vitro cytotoxicity. Biodegradability of the βC scaffold was more than TβC scaffold. In vivo test showed bone regeneration was more in TβC scaffold at 6 weeks based on radiological and histopathologic analysis compared with βC scaffold and control groups. This case report describes the oral rehabilitation of a patient with cleidocranial dysplasia who received a removable partial denture along with silicone-based permanent soft liner to improve esthetic and masticatory function. This patient was the candidate of neither implant nor orthodontic treatment due to medical conditions, history of mandible fracture, age, and risk of fracture after mandibular teeth extractions. Cone-beam computed tomography has made it possible to obtain comprehensive information regarding the morphology and positional relationship of impacted supernumerary teeth. Also, proper collaboration between surgeon and prosthodontist helped to achieve significant improvements in patient’s self-esteem, masticatory function, and esthetic.

Polylactide-co-glycolide acid (PLGA) is known as a biodegradable and biocompatible polymer. This polymer has been highly used in tissue engineering. In this study, the biological behavior of Schwann cells (Rat) was investigated in co-culture with L lysine/gelatine coated PLGA nano-fiber. In this study, PLGA was dissolved in a hexafluoro propanol based solvent and nanofiber prepared by an electronic method. They were coated with gelatin and poly-L-lysine individually. These polymer properties were investigated by Scanning Electron Microscopy (SEM) analysis and contact angle measurement. After extraction of rat Schwann cells, the cells were cultured in three groups of nano-fiber; nano-fiber PLGA, nano-fiber gelatine coated PLGA and nano-fiber poly-L-lysine coated PLGA. Cell death and Cell proliferation were evaluated by Acridine orange staining (living cell with a green nucleus and dead cell with an orange nucleus) and morphology was investigated by SEM in 2, 4 and 6 days. The diameter of electronic nanofiber PLGA was between 270 to 700 nm. Background and Aim: Prevention of dental caries and periodontal disease is an important health priority. Oral health instruction can help to achieve this goal. This study sought to assess the effect of a short-term, targeted, well-structured comprehensive oral health instruction on the level of the knowledge and behavior of patients. Materials and Methods: This study was conducted on 120 patients who were randomly divided into two groups of case and control (n=60). The control group received routine oral health instructions, while the case group received a two-session comprehensive, well-structured oral health instruction provided by trained dental hygienists. The level of the knowledge in the two groups was assessed before and after the intervention using a questionnaire. To assess the effect of the instructions on the oral health behavior of the patients, bleeding on probing (BOP) and periodontal pocket depth (PPD) were measured before and after the instructions. Data were analyzed using generalized estimating equations (GEE). Results: The level of knowledge, PPD, and BOP were not significantly different between the two groups at the baseline (P>0.05). There were significant differences in the level of knowledge (P=0.02), PPD (P=0.03), and BOP (P=0.03) between the two groups after the intervention such that the patients in the case group experienced a reduction in PPD by 2 mm, while BOP decreased by 45%, and knowledge was enhanced by 16% in the case group, compared to the control group, two months after the intervention. Conclusion: Well-structured, targeted, comprehensive short-term oral health instructions can greatly enhance the knowledge and change the behavior of patients.

BACKGROUND Collagen is the main product in pharmaceutics and food industry with a high demand. Collagen can be extracted from several tissues such as skin, bone and tendon, etc. Collagen can be used in tissue engineering researches as a substrate of wound healing and nerve regeneration. Extraction methods of collagen are various with different purities. In this research, we aimed to extract collagen from human placenta with a modified method. METHODS This modified approach was used for extracting of collagen from human placenta with acetic acid and NaCl treatment using different concentrations. RESULTS SDS page showed three different bands that reflected two alpha-chains and one beta-chain with molecular weights of 102, 118 and 220 kDa, respectively. There was no significant difference between extracted collagen from human placenta and standard collagen in western blot analysis. CONCLUSION It was concluded that human placenta can be an alternative source of collagen with high purity for biomedical applications such as tissue engineering, stem cell therapy and research.
