9. Synthesis of two dopamine D4 receptor ligands: <SUP>11</SUP>C labelled chromeno[3,4-c]pyridin-5-ones

Abstract

The synthesis of two 11C labelled chromeno[3,4-c]pyridin-5-ones for the visualisation of the dopamine D4 receptor subtype has been developed. The production entailed an O-methylation of the O-desmethyl precursor with [11C]iodomethane in the presence of tetrabutylammonium hydroxide. Subsequent purification by RP-HPLC and formulation by tracer enrichment on a C18 Sep Pak provided a solution which was suitable for human iv injection. Specific activity of the tracer averaged 37 GBq/μmol at EOS and the radiochemical yields were 65% (decay-corrected, based on [11C]CH3I). Total activity obtained was 5·6–7·4 GBq. The preparations have been demonstrated to be chemically and radiochemically pure by HPLC. Copyright © 2000 John Wiley & Sons, Ltd.

Abstract

Donepezil (1-benzyl-4-[(5,6-dimethoxy-1-oxoindan-2-yl)methyl-]piperidine), a potent acetylcholine esterase inhibitor, was labelled with [11C]iodomethane by O-methylation of the O-6'-desmethyl precursor. Synthesis, purification and formulation were performed in approximately 30 min with an average specific activity of 39.5 GBq/μmol at EOS. 16. Cobalt57 as a SPET tracer in the visualization of ischaemic brain damage in patients with middle cerebral artery stroke

Abstract

In PET studies we have shown the usefulness of cobalt radionuclides for the visualization and quantification of ischaemic damage in stroke. In the present study, we explored 57Co2+as a SPET tracer. Uptake of radioactivity was estimated by using a cobalt enhancement ratio defined as the ratio of cobalt uptake in the affected region versus a similar volume in the non-affected contralateral side. Clinical status was assessed with the Orgogozo stroke score at the time of scanning and at least 60 days after admission. Nineteen patients (11 men, 8 women) with a middle cerebral artery stroke were examined with 57Co2+SPET 0–30 days after stroke onset. Our investigations show enhanced cobalt uptake in the infarcted brain tissue in patients with a major stroke and little clinical improvement. There was a significant correlation between the cobalt enhancement ratio and the Orgogozo score at the time of scanning and discharge. Our results suggest that 57Co2+SPET is suitable for determining the extent of (possibly calcium-mediated) damage in stroke and in the assessment of potential therapeutic interventions.

33. Hippocampal microbleed on a post-mortem t(2)∗-weighted gradient-echo 7.0-tesla magnetic resonance imaging?

Abstract

The present post-mortem study of a brain from an Alzheimer patient showed on a T(2)∗-weighted gradient-echo 7.0-T MRI of a coronal brain section a hyposignal in the hippocampus, suggesting a microbleed. On the corresponding histological examination, only iron deposits around the granular cellular layer and in blood vessel walls of the hippocampus were observed without evidence of a bleeding. This case report illustrates that the detection of microbleeds on MRI has to be interpreted with caution.
