Abstract

Summary: Starch is synthesized as insoluble, semicrystalline particles within plant chloroplast and amyloplast, which are referred to as starch grains (SGs). The size and morphology of SGs in the cereal endosperm are diverse and species–specific, representing a key determinant of the suitability of starch for industrial applications. However, the molecular mechanisms modulating SG size in cereal endosperm remain elusive. Here, we functionally characterized the rice (Oryza sativa) mutant substandard starch grain7 (ssg7), which exhibits enlarged SGs and defective endosperm development. SSG7 encodes a plant–specific DUF1001 domain‐containing protein homologous to Arabidopsis (Arabidopsis thaliana) CRUMPLED LEAF (AtCRL). SSG7 localizes to the amyloplast membrane in developing endosperm. Several lines of evidence suggest that SSG7 functions together with SSG4 and SSG6, known as two regulators essential for SG development, to control SG size, by interacting with translocon‐associated components, which unveils a molecular link between SG development and protein import. Genetically, SSG7 acts synergistically with SSG4 and appears to be functional redundancy with SSG6 in modulating SG size and endosperm development. Collectively, our findings uncover a multimeric functional protein complex involved in SG development in rice. Abstract

Soil moisture is the most direct evaluation index for agricultural drought. It is not only directly affected by meteorological conditions such as precipitation and temperature but is also indirectly influenced by environmental factors such as climate zone, surface vegetation type, soil type, elevation, and irrigation conditions. These influencing factors have a complex, nonlinear relationship with soil moisture. It is difficult to accurately describe this non-linear relationship using a single indicator constructed from meteorological data, remote sensing data, and other data. It is also difficult to fully consider environmental factors using a single drought index on a large scale. Machine learning (ML) models provide new technology for nonlinear problems such as soil moisture retrieval. Based on the multi-source drought indexes calculated by meteorological, remote sensing, and land surface model data, and environmental factors, and using the Cubist algorithm based on a classification decision tree (CART), a comprehensive agricultural drought monitoring model at 10 cm, 20 cm, and 50 cm depth in Gansu Province is established. The influence of environmental factors and meteorological factors on the accuracy of the comprehensive model is discussed, and the accuracy of the comprehensive model is evaluated. The results show that the comprehensive model has a significant improvement in accuracy compared to the single variable model, which is a decrease of about 26% and 28% in RMSE and MAPE, respectively, compared to the best MCI model. Environmental factors such as season, DEM, and climate zone, especially the DEM, play a crucial role in improving the accuracy of the integrated model. These three environmental factors can comprehensively reduce the average RMSE of the comprehensive model by about 25%. Compared to environmental factors, meteorological factors have a slightly weaker effect on improving the accuracy of comprehensive models, which is a decrease of about 6.5% in RMSE. The fitting accuracy of the comprehensive model in humid and semi-humid areas, as well as semi-arid and semi-humid areas, is significantly higher than that in arid and semi-arid areas. Abstract

Summary: Endosperm is the main storage organ in cereal grain and determines grain yield and quality. The molecular mechanisms of heat shock proteins in regulating starch biosynthesis and endosperm development remain obscure.Here, we report a rice floury endosperm mutant flo24 that develops abnormal starch grains in the central starchy endosperm cells. Map‐based cloning and complementation test showed that FLO24 encodes a heat shock protein HSP101, which is localized in plastids. The mutated protein FLO24T296I dramatically lost its ability to hydrolyze ATP and to rescue the thermotolerance defects of the yeast hsp104 mutant.The flo24 mutant develops more severe floury endosperm when grown under high‐temperature conditions than normal conditions. And the FLO24 protein was dramatically induced at high temperature. FLO24 physically interacts with several key enzymes required for starch biosynthesis, including AGPL1, AGPL3 and PHO1. Combined biochemical and genetic evidence suggests that FLO24 acts cooperatively with HSP70cp‐2 to regulate starch biosynthesis and endosperm development in rice.Our results reveal that FLO24 acts as an important regulator of endosperm development, which might function in maintaining the activities of enzymes involved in starch biosynthesis in rice. Abstract

Strigolactones (SLs), a newly discovered class of carotenoid-derived phytohormones, are essential for developmental processes that shape plant architecture and interactions with parasitic weeds and symbiotic arbuscular mycorrhizal fungi. Despite the rapid progress in elucidating the SL biosynthetic pathway, the perception and signalling mechanisms of SL remain poorly understood. Here we show that DWARF 53 (D53) acts as a repressor of SL signalling and that SLs induce its degradation. We find that the rice (Oryza sativa) d53 mutant, which produces an exaggerated number of tillers compared to wild-type plants, is caused by a gain-of-function mutation and is insensitive to exogenous SL treatment. The D53 gene product shares predicted features with the class I Clp ATPase proteins and can form a complex with the α/β hydrolase protein DWARF 14 (D14) and the F-box protein DWARF 3 (D3), two previously identified signalling components potentially responsible for SL perception. We demonstrate that, in a D14- and D3-dependent manner, SLs induce D53 degradation by the proteasome and abrogate its activity in promoting axillary bud outgrowth. Our combined genetic and biochemical data reveal that D53 acts as a repressor of the SL signalling pathway, whose hormone-induced degradation represents a key molecular link between SL perception and responses.

Abstract

Corrugated damage to bearings is a common fault in electrical facilities such as new energy vehicles, wind power, and high-speed railways. The aim of this article is to reveal the microscopic characteristics and formation mechanism of such damages. The corrugation with alternating "light" and "dark" shape was produced on GCr15 bearing races in the experimental conditions. Compared to the light area, the dark area (in the images generated by optical microscope) has more severe electrical erosion, lower hardness, more concave morphology, and lower oxidation. As the voltage increases, the width of the corrugation, the height difference between corrugation, and surface roughness all increase. It is believed that the formation of corrugated damage requires a sufficiently high voltage to induce the periodic destruction and reconstruction of the lubrication film. When the bearing is in a metal-lubrication film–metal contact state, the high voltage causes the lubrication film to break down and induce electrical erosion. Then, the contact area is in metal–metal contact, and the surface is mainly damaged by mechanical rolling. After the reconstruction of lubrication film, the next round of electrical erosion begins.
