21. Engineering magnetic nanosystem for TRPV1 and TRPV4 channel activation.

Abstract

Recently, physical tools for remotely stimulating mechanical force‐sensitive and temperature‐sensitive proteins to regulate intracellular pathways have opened up novel and exciting avenues for basic research and clinical applications. Among the numerous modes of physical stimulation, magnetic stimulation is significantly attractive for biological applications due to the advantages of depth penetration and spatial‐temporally controlled transduction. Herein, the physicochemical parameters (e.g., shape, size, composition) that influence the magnetic properties of magnetic nanosystems as well as the characteristics of transient receptor potential vanilloid‐1 (TRPV1) and transient receptor potential vanilloid‐4 (TRPV4) channels are systematically summarized, which offer opportunities for magnetic manipulation of cell fate in a precise and effective manner. In addition, representative regulatory applications involving magnetic nanosystem‐based TRPV1 and TRPV4 channel activation are highlighted, both at the cellular level and in animal models. Furthermore, perspectives on the further development of this magnetic stimulation mode are commented on, with emphasis on scientific limitations and possible directions for exploitation. 47. Development and Evaluation of the Biological Adjuvant Bivalent Vaccine for Preventing Newcastle Disease and Infectious Bursal Disease

Abstract

Newcastle disease (ND) and infectious bursal disease (IBD) are both one of the most economically important infectious diseases, which cause high infection rate and mortality of chickens and restrict the development of the poultry industry. Currently, vaccination is the most effective method to prevent ND and IBD. In this study, the biological adjuvant bivalent vaccine (rClone30-VP2L-chGM-CSF) was developed for preventing ND and IBD. Furthermore, the immune and protective effects of rClone30-VP2L-chGM-CSF were evaluated in specific-pathogen-free chickens. The experimental results indicated that chickens immunized with rClone30-VP2L-chGM-CSF vaccine significantly improved anti-IBDV and anti-NDV antibody titer; the level of CD4+ T, CD8+ T, B+, MHC-II+, and monocyte/macrophagocyte+ cells; the concentrations of IL-1β, IL-4, IL-17, IFN-α, IFN-β, and IFN-γ; and the splenocyte proliferation rate. Anti-NDV antibody titer reached the theoretical protection value of 4log2 at 7 days after immunization in chickens of rClone30-VP2L-chGM-CSF group; however, which at 14 days after immunization in chickens of rClone30-VP2L and rClone30. These results showed that chickens immunized with rClone30-VP2L-chGM-CSF stimulated stronger immune response than those with the rClone30-VP2L and rClone30. Chickens were challenged with virulent IBDV BC6/85, which were protected in the rClone30-VP2L-chGM-CSF group. Furthermore, IBDV RNA was not measured, and there appeared to be little apoptosis in the bursa of Fabricius in the rClone30-VP2L-chGM-CSF group. Therefore, rClone30-VP2L-chGM-CSF is a promising vaccine candidate against infectious bursal disease virus (IBDV) and newcastle disease virus (NDV), and it provides an idea for developing other poultry vaccines.

48. Efficient Organic Pollutant Removal by Bio/MNs Collaborating with Pseudomonas aeruginosa PAO1.

Abstract

Organic pollution is one of the main sources of environmental pollution, which poses a serious threat to the ecological environment and human health. In this study, we synthesized a composite material consisting of biochar-supported magnetite nanoparticles (Bio/MNs) and collaborated with Pseudomonas aeruginosa PAO1 (P. aeruginosa PAO1) to conduct a bio-chemical composite remediation approach for organic pollution. The results of the scanning electron microscope (SEM) and X-ray diffractometer (XRD) show that Bio/MNs composites have been prepared successfully. Under light conditions, the highest removal rate of organic pollution by Bio/MNs synergistic P. aeruginosa PAO1 reached 81.5%. Gradient experiments revealed a direct correlation between the removal rate of organic pollution and the dosage of P. aeruginosa PAO1, as well as the input of Bio/MNs, within a specific range. Moreover, due to the positively charged nature of organic pollution, its maximum removal rate reaches 98.6% at pH = 11, exhibiting a 1.76-fold increase compared to that at pH = 3. The experimental results show that the collaboration between Bio/MNs and P. aeruginosa PAO1 expedited the electron transfer rate and increased the generation of ·OH and O2−, consequently facilitating the efficient degradation of organic pollutants. These findings inspire Bio/MNs collaborative microorganisms for providing new methods for the green and sustainable removal of organic pollutants. [ABSTRACT FROM AUTHOR]

Abstract

Petal size is a critical factor in plant reproduction and horticulture, and is largely determined by cell expansion. Gerbera hybrida is an important horticultural plant and serves as a model for studying petal organogenesis. We have previously characterized GhWIP2, a Trp-Ile-Pro (WIP)-type zinc protein, that constrains petal size by suppressing cell expansion. However, the underlying molecular mechanism remains largely unclear. Using yeast two-hybrid screening, bimolecular fluorescence complementation, and co-immunoprecipitation, we identified a TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR (TCP) family transcription factor, GhTCP7, that interacts with GhWIP2 both in vitro and in vivo. Using reverse genetic approaches, we elucidated the function of the GhTCP7-GhWIP2 complex in controlling petal expansion. GhTCP7 overexpression severely reduced cell expansion and petal size, whereas GhTCP7 silencing resulted in increased cell expansion and petal size. GhTCP7 showed similar expression patterns to GhWIP2 in various types of G. hybrida petals. We further identified GhIAA26 , which encodes an auxin signalling regulator, that is activated by the GhTCP7-GhWIP2 complex, leading to the suppression of petal expansion. Our findings reveal a previously unknown transcriptional regulatory mechanism that involves protein-protein interactions between two different transcription factor families to activate a negative regulator of petal organogenesis.
