Abstract

We present experimental data illustrating how heat load affects the performance of a cryogenically cooled monochromator under intense X-ray Free-Electron Laser pulses. Abstract

Employing X-ray photon correlation spectroscopy we measure the kinetics and dynamics of a pressure-induced liquid-liquid phase separation (LLPS) in a water-lysozyme solution. Scattering invariants and kinetic information provide evidence that the system reaches the phase boundary upon pressure-induced LLPS with no sign of arrest. The coarsening slows down with increasing quench depths. The $g_2$-functions display a two-step decay with a gradually increasing non-ergodicity parameter typical for gelation. We observe fast superdiffusive ($\gamma \geq 3/2$) and slow subdiffusive ($\gamma < 0.6$) motion associated with fast viscoelastic fluctuations of the network and a slow viscous coarsening process, respectively. The dynamics age linear with time $\tau \propto t_\mathrm{w}$ and we observe the onset of viscoelastic relaxation for deeper quenches. Our results suggest that the protein solution gels upon reaching the phase boundary.

Abstract

We use machine learning methods for an automated classification of experimental XPCS two-time correlation functions from an arrested liquid-liquid phase separation of a protein solution. We couple simulations based on the Cahn-Hilliard equation with a glass transition scenario and classify the measured correlation maps automatically according to quench depth and critical concentration at a glass/gel transition. We introduce routines and methodologies using an autoencoder network and a differential evolution based algorithm for classification of the measured correlation functions. The here presented method is a first step towards handling large amounts of dynamic data measured at high brilliance synchrotron and X-ray free-electron laser sources facilitating fast comparison to phase field models of phase separation.

Abstract

Solutions of wormlike micelles can form cross-linked networks on microscopic length scales. The unique mechanical properties of these complex fluids are driven by the interplay between the network structure and dynamics which are investigated by plate-plate rheometry and X-ray photon correlation spectroscopy~(XPCS) nanorheology. Intensity auto-correlation functions of tracer nanoparticles~(NPs) dispersed in micelle solutions were recorded which captured both the slow structural network relaxation and the short-time dynamics of NPs trapped in the network. The results are indicative of a resonance-like dynamic behavior of the network on the nanoscale that develops as a consequence of the intrinsic short-range rigidity of individual micelle chains., Comment: 5 pages, 5 figures

Abstract

We use time-resolved momentum microscopy at a free-electron laser (FEL) and extend orbital tomography into the time domain to image the electronic wave functions of excited molecular orbitals. This technique provides unprecedented insight into the ultrafast interplay between structural and electronic dynamics. In this work we prove general applicability and establish the experimental conditions at FEL sources to minimize space charge effects and radiation damage. We investigate a bilayer pentacene film on Ag(110) by optical laser pump and FEL probe experiments. From the momentum microscopy signal, we obtain time-dependent momentum maps of the excited-state dynamics of both pentacene layers separately. Combining experimental observations with a theoretical study, we interpret the observed signal for the bottom layer as resulting from the charge redistribution between the molecule and the substrate induced by excitation. We identify that the dynamics of the top pentacene layer resembles excited-state molecular dynamics., Comment: 10 pages, 4 figures

Abstract

A process of energy transfer is demonstrated in non-equilibrium charged particle beams with anisotropy and space charge. Equipartitioning of energy between available degrees of freedom occurs in just a few betatron wavelengths, without halo formation. Collective space charge modes similar to those observed in recent experiments provide the underlying coupling mechanism. Since laboratory beams are commonly far from equilibrium, the traditional K-V stability analysis does not necessarily apply, implying that selection of an operating point based on theory does not necessarily avoid equipartitioning. Furthermore, the rate of equipartitioning is shown to depend on a single free parameter related space charge content of the final (equipartitioned) beam, and does not depend on how the kinetic energy is initially distributed between the two planes., Comment: LINAC 2000 Conference Proceedings Paper ID: MOE19 3 pages, 3 figures

Zusammenfassung: Die radiologische Bildgebung stellt einen unerlässlichen Bestandteil in der Diagnostik von Wirbelsäulenerkrankungen dar. Als Basisdiagnostik steht initial unverändert die konventionelle Röntgenaufnahme (CR) im Vordergrund, mit der bereits in vielen Fällen eine richtungweisende Diagnose ermöglicht wird. Weiterführende Modalitäten wie die Computertomographie (CT) oder die Magnetresonanztomographie (MRT) ermöglichen eine differenziertere Abklärung. Durch die schnelle Innovation und breite Verfügbarkeit hat die Multidetektor-CT (MDCT) in den letzten Jahren eine wichtige Stellung in der Akutdiagnostik traumatischer Wirbelsäulenverletzungen eingenommen und die CR als diagnostisches Mittel der Wahl abgelöst. Nuklearmedizinische Verfahren wie die Knochenszintigraphie oder die Positronenemissionstomographie (PET) dienen als Screeningverfahren und ergänzen die morphologischen bildgebenden Verfahren der Radiologie. Die vorliegende Arbeit liefert eine synoptische Übersicht über die aktuell verfügbaren radiologischen und nuklearmedizinischen Verfahren sowie deren Indikation und Stellenwert bei der Bildgebung von Wirbelsäulenerkrankungen.

Abstract

Zusammenfassung: Die traumatische thorakale Aortenruptur stellt eine lebensbedrohliche Verletzung der Aorta dar, die als Begleitverletzung nach stumpfem Thoraxtrauma oder im Rahmen eines Polytraumas vorkommen kann. In Abhängigkeit vom Ausmaß der Zerreißung der Aortenwand ist diese Gefäßverletzung mit einer hohen primären Letalität und einem relevanten sekundären Rupturrisiko im weiteren Verlauf verbunden. Bei der präklinischen Beurteilung können der linksthorakale Schmerz, ein abgeschwächtes linksseitiges Atemgeräusch mit zunehmender Dyspnoe und Tachykardie sowie ein abgeschwächter Puls an den unteren Extremitäten entsprechend einer Pseudokoarktation Hinweise für eine thorakale Aortenruptur geben. Die präklinische Therapie besteht in der Stabilisierung der Vitalfunktionen und der Behandlung des möglichen Schockzustands. Während die offene operative Therapie der traumatischen Aortenruptur bisher vorwiegend durch Direktnaht oder Gefäßinterponat mit kurzer Abklemmung der thorakalen Aorta oder extrakorporaler Zirkulation durchgeführt wurde, gewinnt die endovaskuläre thorakale Stentgraft-Implantation zunehmend an Bedeutung. Die bisher publizierten Ergebnisse zur endovaskulären Therapie der traumatischen Aortenruptur und auch eigene Erfahrungen weisen darauf hin, dass die Einführung der thorakalen Stentgraft-Implantation zu einem Verfahrenswechsel mit der Möglichkeit der frühzeitigen Primärversorgung der lebensbedrohlichen Verletzung ohne zusätzlichen Thoraxeingriff geführt hat. Regelmäßige Kontrolluntersuchungen sind erforderlich, um mögliche Dislokationen der Stentgrafts oder Leckagen zu erkennen und notwendige endovaskuläre oder offen chirurgische Sekundäreingriffe durchführen zu können.

31. Neue bildgebende Strategien zum Monitoring molekularer Pharmakotherapien bei GIST

Abstract

Zusammenfassung: Prognose und klinisches Management von Patienten mit gastrointestinalem Stromatumor (GIST) haben sich nach Einführung neuer molekularer Pharmakotherapien wie Imatinib dramatisch geändert. Damit einhergegangen ist die Notwendigkeit, bisher etablierte bildgebende Methoden zur Beurteilung des Ansprechens auf die Therapie zu überdenken. Die derzeit am häufigsten eingesetzten Responsekriterien, die RECIST- (Response-evaluation-criteria-in-solid-tumors-)Kriterien, beruhen wesentlich auf der unidirektionalen Messung tumoröser Läsionen. Änderungen in der CT-Dichte oder der Anzahl intratumoraler Blutgefäße werden hierbei nicht berücksichtigt. Die Positronenemissionstomographie (PET) ist ein sensitives Verfahren zur Erfassung des frühen und zur Prädiktion des längerfristigen Therapieansprechens auf Imatinib. Die Anwendung der PET ist jedoch aufgrund begrenzter Verfügbarkeit und der hohen Kosten eingeschränkt. Modifizierte CT-Kriterien, die sowohl Größe- als auch Dichteänderungen berücksichtigen, sind besonders bei der Evaluation des frühen Therapieansprechens viel versprechend. Die weitere Optimierung von Responsekriterien sowie Evaluation möglicher neuer Responsemarker, wie die quantitative Bewertung der Perfusion, können vermutlich zu einem verbesserten Monitoring molekularer Pharmakotherapien bei GIST beitragen.

Die periphere arterielle Verschlusskrankheit (pAVK) lässt sich in die Stadien Claudicatio intermittens, chronisch-kritische Ischämie und akute Ischämie unterteilen. Da diese Krankheitsbilder häufig und auch vital bedrohlich auftreten, sind aktuelle Leitlinien zur Diagnostik und stadiengerechten Therapie für den behandelnden Arzt auch aus forensischen Gründen von großer Bedeutung. Die vorliegende Übersicht stellt aktuelle Prinzipien der Diagnostik und Therapie sowie aktuelle Entwicklungen auf dem Gebiet der peripheren Gefäßintervention unter Berücksichtigung aktueller nationaler und internationaler Leitlinien, insbesondere des Transatlantic Intersociety Consensus (TASC) dar.
