The eukaryotic eIF4F complex, formed mainly by eIF4E and eIF4G subunits, is responsible for the canonical selection of mRNAs required for the initiation of mRNA translation. The best-known complexes implicated in translation in trypanosomatids are based on two related pairs of eIF4E and eIF4G subunits (EIF4E3/EIF4G4 and EIF4E4/EIF4G3), whose functional distinctions remain to be fully described. Here, to define interactomes associated with both complexes in Trypanosoma brucei procyclic forms, we performed parallel immunoprecipitation experiments followed by identification of proteins co-precipitated with the four tagged eIF4E and eIF4G subunits. A number of different protein partners, including RNA binding proteins and helicases, specifically co-precipitate with each complex. Highlights with the EIF4E4/EIF4G3 pair include RBP23, PABP1, EIF4AI and the CRK1 kinase. Co-precipitated partners with the EIF4E3/EIF4G4 pair are more diverse and include DRBD2, PABP2 and different zinc-finger proteins and RNA helicases. EIF4E3/EIF4G4 are essential for viability and to better define their role, we further investigated their phenotypes after knockdown. Depletion of either EIF4E3/EIF4G4 mRNAs lead to aberrant morphology with a more direct impact on events associated with cytokinesis. We also sought to identify those mRNAs differentially associated with each complex through CLIP-seq with the two eIF4E subunits. Predominant among EIF4E4-bound transcripts are those encoding ribosomal proteins, absent from those found with EIF4E3, which are generally more diverse. RNAi mediated depletion of EIF4E4, which does not affect proliferation, does not lead to changes in mRNAs or proteins associated with EIF4E3, confirming a lack of redundancy and distinct roles for the two complexes.

Magnetic bead handling is a common tool in purification of biomolecules and subsequent analysis of molecule concentrations. The knowledge of forces acting on magnetizable beads in magnetic fields enables to use them not only as a purification instrument but also for analytical purposes. We wanted to use this to design and build up a novel bioanalytical instrument, which is able to analyze proteins in unpurified samples. Therefore magnetic beads are forced to describe certain trajectories via oscillating magnetic fields. This allows us to move magnetic beads periodically in a hydrodynamic system. This movement creates a dynamic fluorescence signal, which gives an insight about how many molecules are bound to the particle surface. This MHF technology can be combined with several assays, which are already on the market, maximizing signal to noise ratio.

Die individualisierte Krebstherapie als Teil der personalisierten Medizin zielt auf eine optimale Behandlung eines Patienten mit minimalen Nebenwirkungen ab. Bei der Chemotherapie ist es daher ein Ansatz, die am besten wirksamen Chemotherapeutika bzw. die beste Kombination unterschiedlicher Chemotherapeutika vorab zu bestimmen, um die Lebensqualität des Patienten zu erhöhen und die therapeutische Effizienz zu steigern. Doch die herkömmlichen Verfahren sind aufwändig und teuer. Außerdem konnte bislang der gesundheitsökonomische Nutzen solcher Verfahren noch nicht ausreichend nachgewiesen werden. Mit DiagnoSYS konnte gezeigt werden, dass mittels Automatisierung solcher Verfahren eine höhere Reproduzierbarkeit bei Chemosensitivitätstests möglich ist. Beispielhafte Grundlage hierfür ist ein ATP/TCA Test, der auf der DiagnoSYS Plattform durchgeführt wurde. DiagnoSYS ermöglicht damit erstmalig die durchgehend automatisierte Testung individueller Patientenproben von der Gewebeaufarbeitung bis hin zur Testdurchführung und Ergebnisauswertung. Zusätzlich konnte durch Integration einer magnetischen Zellseparation das Signal zu Rausch Verhältnis des ATP/TCA Test wesentlich verbessert werden. Die DiagnoSYS Plattform arbeitet mit dem international anerkannten SBS-Format. Daher sind alle Prozessschritte von der Gewebeaufarbeitung bis hin zur Lumineszenz-basierten Signalerfassung modular austauschbar und können an andere Biomarker wie beispielsweise die Bestimmung von uPA/PAI-1 angepasst werden.

Das Aufbringen von Flüssigkeiten auf verschiedenste Zielformate stellt für Produktionsprozesse jeglicher Art oft eine technologische Hürde dar. Ein besonders robustes Dosierverfahren, welches mit günstigen Wegwerfdüsen und einer einfachen pneumatischen Druckpulserzeugung auf Basis von Schnellschaltventilen arbeitet, soll hier Abhilfe leisten. Am Beispiel der technologisch besonders komplexen Herstellung von synthetischen Wundauflagen wird dies deutlich. Synthetische Wundauflagen bieten im Gegensatz zu Wundauflagen aus tierischen Biopolymeren, wie z.B. Kollagen, Vorteile bezüglich ihrer Verarbeitbarkeit. Sie lassen sich bei hohen Temperaturen in etablierten Verfahren und in großer Stückzahl am Fließband herstellen. Um synthetische Wundauflagen zusätzlich mit biologischen Materialien zu funktionalisieren, müssen neue Produktionstechnologien entwickelt werden, damit die temperatursensitiven funktionalisierenden Materialien bei der Verarbeitung nicht zerstört werden. Einer dieser neuen Produktionstechnologien ist die Dosierung hochviskoser Materialien mittels Immediate Drop On Demand Technology (I-DOT). Durch die Flexibilität dieses Verfahrens ist es möglich, unterschiedliche Materialien während des Produktionsprozesses auf eine synthetische Wundauflage aufzubringen. In dieser Anwendung wurde eine synthetische Wundauflage aus einem Co-Polymer mit einem Array aus hochkonzentriertem mikroverkapselten Antibiotikum bedruckt. Für die Verkapselung wurde in DMSO gelöstes Polylactid gewählt. Zum Schutz des Arrays wurde eine zweite Schicht aus PLA-PLGA Co-Polymer darüber laminiert. Hierdurch entstand eine synthetische Sandwich-Wundauflage mit integriertem Antibiotikum-Array.

Christian Reis ist Biotechnologe am Fraunhofer IPA. Der dreißigjährige Naturwissenschaftler leitet stellvertretend die Projektgruppe für Automatisierung in der Medizin und Biotechnologie in Mannheim, die neben zahlreichen Ingenieuren auch aus vier Biologen besteht. Hauptverantwortlich leitet er die Gruppe Bioprozesstechnik, die sich mit der technischen Umsetzung biologischer Prozesse beschäftigt. In einem Interview mit Dr. Ariane Pott erklärt er, welche Aufgaben Biologen in der Automatisierungstechnik übernehmen können.

Embolisation - the blocking of vessels - is a key procedure in Interventional Radiology. It plays a steadily growing role in the treatment of various tumour lesions, with hepatic cellular carcinoma and uterine fibroids the main focus. We produced multimodal visible embolization particles, which can be visualized in X-ray based fluoroscopy and computer tomography (CT) as well as radiation free magnetic resonance imaging (MRI).

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Os tripanossomatídeos são caracterizados por processos moleculares atípicos como o controle pós-transcricional da expressão gênica, através da degradação de mRNA e controle da tradução. Na maioria dos eucariotos, a iniciação da tradução começa com a ligação do complexo eIF4F (composto pelas subunidades eIF4A, eIF4E e eIF4G) ao cap monometilado presente na região 5 dos mRNAs, permitindo seu reconhecimento pelo ribossomo e outros eIFs participantes. A atividade do eIF4F é reforçada pela proteína de ligação a cauda poli-A (PABP), que atua através de uma interação direta com o eIF4G. Vários homólogos às subunidades do eIF4F e PABP foram previamente identificados nos tripanossomatídeos, como cinco homólogos do eIF4G (EIF4G1-5) e múltiplos homólogos das demais subunidades (EIF4E1-4, EIF4AI e III e PABP1-2) e suas funções vem sendo investigadas em Leishmania major e Trypanosoma brucei. Como parte deste estudo, nós demonstramos que tanto o LmEIF4G3 como o LmEIF4G4 interagem com o LmEIF4AI, embora mutações nos aminoácidos LNK destas proteínas abolam suas interações com o LmEIF4AI. As proteínas LmEIF4G3 e LmEIF4G4 também se ligam respectivamente aos homólogos LmEIF4E4 e LmEIF4E3 e diferentes aminoácidos foram associados a estas interações. A substituição dos resíduos FSL no LmEIF4G3 abole sua interação com o LmEIF4E4 e a mutagênese dos resíduos IL no LmEIF4G4 abole sua ligação ao LmEIF4E3. Adicionalmente, nós demonstramos que as proteínas LmEIF4G3 e LmEIF4G4 ligam-se à LmPABP1 e nós também sugerimos funções distintas para o TbEIF4AI e TbEIF4AIII em ambas as formas procíclica e sanguínea de T. brucei. Em síntese, nossos resultados indicam uma maior complexidade na iniciação da tradução nos tripanossomatídeos e provavelmente a mesma é regulada por pelo menos dois diferentes complexos eIF4F

One of the basic steps in the field of bioprocess engineering is the handling of liquids. The processing of those media as well as the cleaning of the dosing system is an important time, cost and quality factor for conventional systems. The Fraunhofer IPA developed a method that avoids this extensive process. This method allows a fast, cost-effective and flexible accomplishment due to a compact device, which performs the majority of the pipetting steps fully automated.

Einer der grundlegendsten Schritte in der Bioprozesstechnik ist die Handhabung von Flüssigkeiten. Die Verarbeitung dieser Medien, sowie die Reinigung des Dosiersystems stellt für herkömmliche Dosiersysteme einen enormen Zeit, Kosten und Qualitätsfaktor dar. Ein Verfahren, mit dem sich dieser aufwändige Prozess umgehen lässt, wurde am Fraunhofer IPA entwickelt. Mit dem Verfahren lassen sich ein Großteil der Pipettierschritte in einem kompakten Gerät vollautomatisiert und damit schnell, kostengünstig und flexibel durchführen.
